Physiology and pathophysiology of the calcium-sensing receptor in the kidney.

Angiotensin-(1-7) [ANG-(1-7)], one of the bioactive peptides produced in the renin-angiotensin system, plays a pivotal role in cardiovascular physiology by providing a counterbalance to the function of ANG II. Recently, it has been considered as a potential candidate for therapeutic use in the treatment of various types of cardiovascular diseases. The aim of the present study is to explain the modulatory role of ANG-(1-7) in atrial natriuretic peptide (ANP) secretion and investigate the functional relationship between two peptides to induce cardiovascular effects using isolated perfused beatin

Glutamate regulates Ca2+ signals in smooth muscle cells of newborn piglet brain slice arterioles through astrocyte- and heme oxygenase-dependent mechanisms.

Glutamate is the principal cerebral excitatory neurotransmitter and dilates cerebral arterioles to match blood flow to neural activity. Arterial contractility is regulated by local and global Ca(2+) signals that occur in smooth muscle cells, but modulation of these signals by glutamate is poorly understood. Here, using high-speed confocal imaging, we measured the Ca(2+) signals that occur in arteriole smooth muscle cells of newborn piglet tangential brain slices, studied signal regulation by glutamate, and investigated the physiological function of heme oxygenase (HO) and carbon monoxide (CO)

Translation initiation factor 4E blocks endoplasmic reticulum-mediated apoptosis.

Eukaryotic translation initiation factor 4E (eIF4E) is the mRNA cap-binding protein required for translation of cellular mRNAs utilizing the 5' cap structure. The rate-limiting factor for mRNA recruitment to ribosomes, eIF4E is a major target for regulation of translation by growth factors, hormones, and other extracellular stimuli. When overexpressed, eIF4E exerts profound effects on cell growth and survival, leading to suppression of oncogene-dependent apoptosis, causing malignant transformation and conferring tumors with multiple drug resistance. We found previously that overexpressed eIF4E

Ca(2+)-independent spine dynamics in cultured hippocampal neurons.

Developing spines are highly dynamic processes that undergo rapid morphologic changes. We have investigated whether basal developmentally regulated spine motility is triggered by spontaneous changes in intracellular Ca(2+) ([Ca(2+)](i)). To address the link between Ca(2+) transients and motility, we have used the cell-permeable chelator BAPTA-AM. Consistent with others, we found that young cultured hippocampal neurons [7-13 days in vitro (DIV)] display significantly more spine motility than older neurons (14-21 days in vitro). Control experiments indicate that BAPTA-AM can lower basal [Ca(2+)]

Effects of aldosterone on transient outward K+ current density in rat ventricular myocytes.

1. Aldosterone, a major ionic homeostasis regulator, might also regulate cardiac ion currents. Using the whole-cell patch-clamp technique, we investigated whether aldosterone affects the 4-aminopyridine-sensitive transient outward K+ current (I(to1)). 2. Exposure to 100 nM aldosterone for 48 h at 37 degrees C produced a 1.6-fold decrease in the I(to1) density compared to control myocytes incubated without aldosterone. Neither the time- nor voltage-dependent properties of the current were significantly altered after aldosterone treatment. RU28318 (1 microM), a specific mineralocorticoid recepto

Two-Step Design of Triazole-Stapled Opioid Agonists Exhibiting Potent and Prolonged Peripheral Analgesic Activity.

This study establishes a two-step strategy for developing triazole-stapled multifunctional agonists that integrate opioid and neuropeptide FF (NPFF) pharmacophores. Initially, calcium mobilization assays, enzymatic stability tests, and analgesic activity screens identified triazole-stapled opioid peptide 16, derived from the opioid pharmacophore of the multifunctional opioid/NPFF agonist DN-9. Subsequently, chimeric conjugation of 16 with the NPFF pharmacophore yielded the novel multitarget analgesic agonist, 16a. Functional assays demonstrated that 16 functions as a μ-opioid receptor (MOR) ag

Osteocyte-Derived Extracellular Vesicles: Bridging the Gap in Skeletal Physiology and Pathophysiology.

PURPOSE OF REVIEW: Osteocytes are the most abundant cells in bone and serve as central skeletal regulators. Recent findings from our and other groups demonstrate that beyond soluble mediators, osteocytes communicate by releasing extracellular vesicles (EVs). This review provides a systematic overview of osteocyte-derived EVs and highlights their importance in diagnostic and therapeutic innovation. RECENT FINDINGS: Accumulating evidence has linked osteocyte mechanotransduction to EV biogenesis and secretion. Mechanically induced calcium oscillations and signaling pathways impact load-induced E

Design and Exploration of a Tricyclic Series of CNS-Penetrant CD38 Small Molecule Inhibitors.

Cluster of Differentiation 38 (CD38) plays a key role in NAD+ metabolism and calcium signaling, and recent reports highlight therapeutic opportunities for CD38 modulation in central nervous system (CNS) disorders. In this report, we describe the rational design, synthesis, and exploration of structure-activity relationships (SAR) for a novel and brain-penetrant series of tricyclic compounds that form an adduct with ADPR to inhibit CD38 in an uncompetitive manner. Rigidification of the bioactive conformation via covalent tethering of a 6-membered heteroaromatic core to an imidazole warhead serv…

Astrocytic facilitation of somatic NMDARs underpins hippocampal E-S potentiation.

EPSP-spike (E-S) potentiation is a form of plasticity defined by a persistent increase in neuronal spike output for a given synaptic input, reflecting enhanced E-S coupling independent of synaptic strength. However, its cellular mechanisms remain poorly understood. Here, we show that brief induction of E-S potentiation triggers a sustained calcium activity increase in astrocytes adjacent to the hippocampal CA1 pyramidal layer. Mechanistically, E-S potentiation critically requires GluN2B-containing N-methyl-D-aspartate receptors (NMDARs) localized to the neuronal soma. Sustained astrocytic acti

The Glucagonocentric Basis of Diabetic Cardiomyopathy: Pancreas-Heart Crosstalk Linking α-Cell Dysregulation to Cardiac Metabolic Stress and Fibrosis.

Type 2 diabetes mellitus (T2DM) is increasingly recognized as a disorder of inappropriate glucagon secretion in addition to β-cell dysfunction and insulin resistance. Chronic hyperglucagonemia, driven by α-cell dysregulation, contributes directly to the development of diabetic cardiomyopathy (DCM) through hepatic, systemic, and myocardial mechanisms. Emerging evidence shows that glucagon receptor (GCGR) signaling is active in cardiomyocytes, endothelial cells, and vascular smooth muscle, where persistent activation promotes cAMP/PKA-mediated stress, calcium dysregulation, oxidative injury, and

Tolylfluanid induces mitochondrial dysfunction through excessive ROS generation and inhibits cell growth in bovine mammary epithelial cells.

Tolylfluanid is a phenylsulfamide fungicide commonly used to manage fungal diseases in crops; however, evidence has shown its unintended toxicity in non-target organisms. To elucidate its cellular impact, we investigated how tolylfluanid affects bovine mammary epithelial (MAC-T) cells. The results revealed that exposure to tolylfluanid suppressed MAC-T cell viability, enhanced apoptotic responses, and disrupted 3D spheroid development compared with the control group. The treatment provoked excessive generation of ROS, elevated mitochondrial calcium accumulation, and triggered a reduction in mi

Postmenopausal osteoporosis and vascular calcification: The estrogen regulation network and calcification paradox.

The decline in estrogen levels in postmenopausal women is a major risk factor for the high prevalence of osteoporosis and vascular calcification in this population. Estrogen deficiency leads to an imbalance in the RANKL/OPG ratio, abnormalities in the Wnt/β-catenin pathway, and disruptions in bone morphogenetic protein (BMP) signaling, thereby inducing osteoporosis and vascular calcification. Moreover, estrogen deficiency affects calcium and phosphorus metabolism and induces the release of extracellular vesicles from the aging bone matrix, which mediate interactions between bone and vascular t

Serotonin neurons in the dorsal raphe control food-craving-like behavior during pregnancy in mice.

During pregnancy, physiological changes can alter food intake behaviors. However, the underlying neural mechanisms remain unclear. Here we show that female mice exhibit food-craving-like behavior during pregnancy, mirroring many patterns described in humans. We show that 5-HTDRN neuronal firing activity is reduced during pregnancy through increased small-conductance calcium-activated potassium type 3 (SK3) ion-channel activity. Genetic knockout of SK3 ion channels from 5-HTDRN neurons abolishes the pregnancy-associated suppression in 5-HTDRN neuronal firing activity and reduces food-craving-li

OPA1-dependent mitochondrial remodeling coordinates TCR signaling and metabolic adaptation during iNKT cell differentiation.

Invariant natural killer T (iNKT) cells require mitochondrial metabolism for terminal effector development. We found iNKT cells express elevated levels of proteins regulating mitochondrial membrane dynamics, and identified Opa1, but not Drp1, as selectively required for iNKT cell differentiation. OPA1 deficiency disrupted mitochondrial cristae organization, reduced mitochondrial membrane potential, increased mitochondrial mass, and altered calcium homeostasis in iNKT cells. Bulk and single-cell transcriptomic analyses revealed impaired TCR-responsive gene expression, activation of mitochondria

Ultrastructural membrane dynamics of mouse and human cortical synapses.

Live human brain tissues provide unique opportunities for understanding synaptic transmission. Investigations have been limited to anatomy, electrophysiology, and protein localization-while crucial parameters such as synaptic vesicle dynamics were not visualized. Here we utilize zap-and-freeze time-resolved electron microscopy to overcome this hurdle. First, we validate the approach with acute mouse brain slices to demonstrate that slices can be stimulated to produce calcium signaling. Next, we show that synaptic vesicle endocytosis is induced in both mouse and human brain slices. Crucially, c…

Sensory neurons drive pancreatic cancer progression through glutamatergic neuron-cancer pseudo-synapses.

Cancers thrive on neuronal input. Here, we demonstrate the presence of pseudo-synaptic connections between sensory nerve endings and cancer cells in an extracerebral cancer, i.e., pancreatic ductal adenocarcinoma (PDAC). These synaptic sites exhibit a selective enrichment of the glutamatergic N-methyl-D-aspartate receptor (NMDA) receptor subunit NMDAR2D (GRIN2D) on the cancer cells, which turns PDAC cells responsive to neuron-derived glutamate and promotes tumor growth and spread. Intriguingly, neurons transform a subset of co-cultured PDAC cells into calcium-responsive cells via GRIN2D-type g
