The TRIB1-PPARγ Axis Regulates Cholesterol Metabolism in Pancreatic Ductal Adenocarcinoma.

Metabolic reprogramming, particularly the dysregulation of cholesterol metabolism, is a hallmark of pancreatic ductal adenocarcinoma (PDAC). However, the underlying molecular drivers remain largely elusive. In this study, we demonstrate that Tribbles homolog 1 (TRIB1) is elevated in PDAC tissues and is significantly associated with poor prognosis. Functionally, TRIB1 mRNA knockdown suppresses PDAC cell growth and tumor formation, while its overexpression promotes both. Mechanistically, TRIB1 protein binds to the DNA-binding domain of PPARγ and inhibits its transcriptional activity. This intera

SOX4: A pivotal transcriptional hub and potential therapeutic target in hepatocellular carcinoma progression.

Hepatocellular carcinoma (HCC) carries high global morbidity and mortality, with dismal prognosis due to hidden onset, limited advanced therapies and prevalent drug resistance. SOX4, a conserved transcription factor, maintains hepatic homeostasis physiologically but undergoes abnormal overexpression in HCC via transcriptional, epigenetic and ceRNA modulation. It acts as a pivotal oncogenic driver, facilitating tumor metastasis, metabolic reprogramming, stem cell retention, immune evasion and treatment resistance. Clinically, SOX4 upregulation is tightly associated with advanced tumor stage and

Synovial fibroblasts modulate endothelial activation in an acute injury-on-a-chip model.

Most patients who sustain an acute joint injury develop degenerative joint disease or osteoarthritis (OA). Animal models have informed the design of OA therapeutics; however, no disease-modifying therapy has successfully translated to human patients. Thus, there is a strong motivation to develop humanized in vitro platforms to fill a critical gap in knowledge of disease progression postinjury. Here, we develop an acute injury-on-a-chip model of the synovium, a vascularized, joint-lining tissue that has been implicated in OA progression and as a key driver of joint disease. We apply this chip-b

Targeting an atypical G protein-coupled receptor signaling pathway for cardiac fibrosis therapy.

Cardiac fibrosis independently predicts adverse outcomes in heart failure (HF), yet no Food and Drug Administration-approved therapy directly targets fibrotic remodeling in the heart. To address this unmet clinical need, we used a multidimensional drug discovery pipeline centered on a human induced pluripotent stem cell (iPSC)-based platform. Through high-throughput screening, we identified CGS15943 (CGS) as the lead antifibrotic compound and validated its activity in human cardiac fibroblasts, three-dimensional engineered heart tissues, and animal models of HF. Mechanistic studies revealed an

Circular RNA circDCUN1D4 suppresses hepatocellular carcinoma development via targeting the miR-590-5p/ TIMP3 axis.

Источник пометил публикацию как отозванную

Hepatocellular carcinoma (HCC) is a major global health concern, necessitating innovative therapeutic strategies. In this study, we investigated the functional role of circular RNA circDCUN1D4 in HCC progression and its potential therapeutic implications. It was found that HCC patients exhibiting higher levels of circDCUN1D4 demonstrated a more favorable survival rate. Furthermore, we revealed that circDCUN1D4 suppressed HCC cell proliferation, migration, and invasion. Mechanistically, circDCUN1D4 was identified as a sponge for miR-590-5p, leading to the downregulation of its downstream target

Regenerative treatment of zinc-based nanoenzyme incorporated injectable hydrogel with mesenchymal stem cells alleviates spinal cord injury.

Mesenchymal stem cells (MSCs)-derived therapy has surfaced as a viable approach for addressing spinal cord injury (SCI). The unfavorable milieu of SCI significantly impairs the paracrine and survival function of implanted MSCs, hence greatly restricting the therapeutic efficacy of this method. We present a zinc-based nanoenzyme (ZnNZs)-combined in situ forming thermoresponsive hydrogels (ZnNZ-HGL) that facilitate dual improvement of MSCs paracrine and viability effects, resulting in highly effective spinal cord reconstruction. The sol-gel transitions characteristic of ZnNZ-HGL at the physiolog

Paraneoplastic Neurological Syndromes Associated With Parotid Cancer Expressing the Zic4 Antibody: A Case Report.

Paraneoplastic neurological syndromes (PNS) are rare, immune-mediated disorders associated with cancer, often presenting with various neurological symptoms, such as encephalomyelitis, limbic encephalitis, and rapidly progressive cerebellar syndrome. A 66-year-old woman presented to our department with rapidly increasing swelling in her right parotid gland. Fine needle aspiration cytology confirmed parotid cancer, and a right parotidectomy and dermectomy were planned. Preoperatively, she experienced symptoms of nausea and vertigo and was later diagnosed with PNS based on the presence of Zic4 an

IgG4-Related Disease in a 90-Year-Old Man with an 18-Year Disease Course: A Case Report.

IgG4-related disease (IgG4-RD) is a multi-organ inflammatory immune-mediated illness caused by IgG4-secreting plasma cells infiltrating the tissue. This condition usually affects elderly men. A 90-year-old Chinese male was diagnosed with IgG4-RD based on the new 2019 ACR/EULAR classification criteria, as he had multiple organ involvement. After receiving treatment with glucocorticoids, leflunomide, and gamma-globulin, the patient's clinical symptoms significantly improved, confirming the accuracy of the diagnosis. The patient had an 18-year medical history during which the disease progressivel

Current approaches for the regeneration and reconstruction of ocular surface in dry eye.

Significant research revealed the preocular tear film composition and regulations that remain vital for maintaining Ocular surface functional integrity. Inflammation triggered by many factors is the hallmark of Ocular surface disorders or dry eyes syndrome (DES). The tear deficiencies may lead to ocular surface desiccation, corneal ulceration and/or perforation, higher rates of infectious disease, and the risk of severe visual impairment and blindness. Clinical management remains largely supportive, palliative, and frequent, lifelong use of different lubricating agents. However, few advancemen

Immunotherapy against programmed death-1/programmed death ligand 1 in hepatocellular carcinoma: Importance of molecular variations, cellular heterogeneity, and cancer stem cells.

Hepatocellular carcinoma (HCC) is a heterogeneous malignancy related to diverse etiological factors. Different oncogenic mechanisms and genetic variations lead to multiple HCC molecular classifications. Recently, an immune-based strategy using immune checkpoint inhibitors (ICIs) was presented in HCC therapy, especially with ICIs against the programmed death-1 (PD-1) and its ligand PD-L1. However, despite the success of anti-PD-1/PD-L1 in other cancers, a substantial proportion of HCC patients fail to respond. In this review, we gather current information on biomarkers of anti-PD-1/PD-L1 treatm

Small Interfering RNAs and their Delivery Systems: A Novel Powerful Tool for the Potential Treatment of HIV Infections.

Small interfering RNAs (siRNAs) have rapidly developed into biomedical research as a novel tool for the potential treatment of various human diseases. They are based on altered gene expression. In spite of the availability of highly active antiretroviral therapy (HAART), there is a specific interest in developing siRNAs as a therapeutic agent for human immunodeficiency virus (HIV) due to several problems including toxicity and drug resistance along with long term treatment. The successful use of siRNAs for therapeutic goals needs safe and effective delivery to specific cells and tissues. Indee

Bacteria-induced aggregation of bioorthogonal gold nanoparticles for SERS imaging and enhanced photothermal ablation of Gram-positive bacteria.

The challenge in antimicrobial photothermal therapy (PTT) is to develop strategies for decreasing the damage to cells and increasing the antibacterial efficiency. Herein, we report a novel theranostic strategy based on bacteria-induced gold nanoparticle (GNP) aggregation, in which GNPs in situ aggregated on the bacterial surface via specific targeting of vancomycin and bioorthogonal cycloaddition. Plasmonic coupling between adjacent GNPs exhibited a strong "hot spot" effect, enabling effective surface enhanced Raman scattering (SERS) imaging of bacterial pathogens. More importantly, in situ ag

A Tumor-pH-Responsive Supramolecular Photosensitizer for Activatable Photodynamic Therapy with Minimal In Vivo Skin Phototoxicity.

A major challenge in photodynamic therapy (PDT) is the development of new tumor-targeting photosensitizers. The tumor-specific activation is considered to be an effective strategy for designing these photosensitizers. Herein, we describe a novel tumor-pH-responsive supramolecular photosensitizer, LDH-ZnPcS8, which is not photoactive under neutral conditions but is precisely and efficiently activated in a slightly acidic environment (pH 6.5). LDH-ZnPcS8 is prepared by using a simple coprecipitation method based on the electrostatic interaction between negatively charged octasulfonate-modified z

Occult primary head and neck carcinoma.

Unknown primary carcinoma presenting as cervical lymph node metastasis accounts for approximately 5% of all head and neck malignancies. The typical presentation involves a middle-aged man with a painless neck mass that has been present for several months. Over 90% of these malignancies represent squamous cell carcinoma originating within Waldeyer's ring (lymphoid tissue of the nasopharynx, tonsil, and base of tongue). The remainder are comprised of adenocarcinoma, melanoma, and other rare histologic variants. The ability to identify the occult primary tumor is imperative because identification

Macrophages in tertiary lymphoid structures promote apoptosis of ATF4-positive gastric cancer cells via IL18.

BACKGROUND: Gastric cancer (GC) is a globally prevalent and highly fatal malignancy of the digestive system. Despite its transformative role in gastric cancer, immunotherapy’s efficacy is limited by the complexity and incomplete understanding of the heterogeneous tumor microenvironment, necessitating a deeper analysis to uncover mechanisms of resistance. METHOD: We analyzed the immune response status of GC patients and generated a single-cell transcriptome landscape of GC patients under various immunotherapy regimens. Dynamic changes in prognosis, biological functions, developmental trajectori

Early dynamics of a neutrophil interferon program drive immunotherapy resistance and limit durable anti-tumor immunity.

Despite the remarkable success of immune checkpoint blockade (ICB), durable clinical benefit remains limited to a subset of patients, highlighting the need for predictive biomarkers and new therapeutic targets. Here, we investigated the early immune dynamics associated with anti-PD1 responsiveness using longitudinal single-cell transcriptomics, spatial transcriptomics, and neutrophil-specific genetic perturbations in preclinical models of triple-negative breast cancer. We identified a conserved neutrophil interferon-stimulated gene (ISG) program that emerges before overt tumor divergence and p

Triple-targeted artificial extracellular vesicles for enhanced glioma therapy via synergistic chemomagnetic penetration of the blood-brain barrier.

Malignant glioma remains one of the most aggressive and therapy-resistant brain tumors, with limited treatment options and a poor prognosis. Overcoming the blood-brain barrier (BBB) is a major challenge for effective drug delivery. Here, we developed a triple-targeted nanodelivery system, denoted BCNU-FeSeH@EVLF, which integrates magnetic guidance, chemotactic homing, and receptor-mediated transport for enhanced brain penetration and glioma treatment. The system consists of carmustine (BCNU)-loaded, hyaluronic acid-modified superparamagnetic Fe3O4 nanoparticles with a mesoporous selenium shell

Living materials for gas therapy.

The clinical translation of gas therapy, which employs medical gases such as nitric oxide (NO), carbon monoxide (CO), hydrogen sulfide (H2S), hydrogen (H2), and sulfur dioxide (SO2), is mainly limited by the absence of delivery systems that can provide precise spatiotemporal control in complex pathological environments. While conventional nanocarriers have improved in gas delivery, they often suffer from limited biocompatibility, poor targeting, and insufficient responsiveness. Recently, living materials emerged as a promising and innovative paradigm. Engineered from biological entities such a

Targeted DNA Nanomachine Enables Specific miRNA-Responsive Singlet Oxygen Amplification for Precise Cutaneous Squamous Cancer Therapy.

Photodynamic therapy (PDT) is a promising strategy for the treatment of skin-related tumors including cutaneous squamous cells carcinoma (cSCC). However, it is hard to balance the dosage off-target phototoxicity while maintaining satisfactory therapeutic effect. In addition, oxygen-dependent photosensitizers (PSs) are affected by tumor hypoxic environment, which further causes inefficient photocatalysis and reduces therapeutic effect. Herein, an intelligent DNA nanomachine based on tetrahedral DNA framework is proposed, incorporated with tumor-targeted aptamer and specific miRNA-responsive hai
