Functional Connectivity in Patients With Parkinson's Disease and Freezing of Gait Using Resting-State EEG and Graph Theory.

BACKGROUND: Although many studies have shown abnormalities in brain structure and function in people with Parkinson's disease (PD), we still have a poor understanding of how brain structure and function relates to freezing of gait (FOG). Graph theory analysis of electroencephalography (EEG) can explore the relationship between brain network structure and gait function in PD. METHODS: Scalp EEG signals of 83 PD (42 PDFOG+ and 41 PDFOG-) and 42 healthy controls were recorded in an eyes-opened resting-state. The phase lag index was calculated for each electrode pair in different frequency bands,

Sequence analysis of nucleoprotein gene reveals the co-circulation of lineages and sublineages of rabies virus in herbivorous in Rio Grande do Sul state, Brazil.

An unprecedented outbreak of rabies occurred in Rio Grande do Sul state (RS) from 2012 onward, resulting in thousands of bovine deaths, important economic losses, and posing risk to human health. This article describes a genetic analysis of 145 rabies viruses (RABV) recovered from herbivorous from RS between 2012 and 2017, based on partial sequence analysis of the nucleoprotein (N) gene. High nucleotide (nt) identity (95.5 to 100%) and amino acid (aa) similarity (96.7 to 100%) were observed among the analyzed sequences. These sequences displayed a high sequence nt identity/aa similarity with b

The application of DM-MSTP method on Tunisian financial market: 2024 case study.

The Novel Dhouib Matrix Minimum Spanning Tree Problem (DM-MSTP) method is used to illustrate a topological relationship between all Tunisian financial market for 2024. The results reveal a clear hierarchical organization with the financial sector playing a central bridging role, particularly through banks and financial services, which connect consumer-oriented and real-economy sectors. A two-cluster configuration highlights a tightly integrated group of cyclical sectors basic materials, construction and building materials, and industry characterized by strong co-movements and limited diversifi

Site-directed chromosome rearrangements in skin fibroblasts from persons carrying genes for hereditary neoplasms.

Chromosomal variability was studied in cultured skin fibroblasts in members of two unrelated families associated with hereditary neoplasms, one with familial childhood leukemia and the other with medullary thyroid cancer syndrome. Nonconstitutional chromosome rearrangements occurred with consistent frequency in the patients and obligate carriers. The G-banding analysis showed that th chromosome rearrangements were not random, and site of rearrangements tended to cluster to band p22 of chromosome 1 in the carriers of childhood leukemia gene and to band q23 of chromosome 17 in the patient with m

Transmission of tuberculosis in rural Henan, China: a prospective population-based genomic spatial epidemiological study.

The incidence of tuberculosis (TB) has declined more slowly in rural than urban areas in China, and data on the patterns of transmission and the high-risk populations in rural areas remains scarce. We conducted a population-based study of culture-positive pulmonary TB patients diagnosed in rural Linzhou City, Henan Province from July 2018 to February 2023. Genomic clusters were defined based on whole-genome sequencing and risk factors for clustering were identified by logistic regression. Transmission events were inferred with phybreak and transmission links were sought through epidemiological

Adherence predictor variables in AIDS patients: an empirical study using the data mining-based RFM model.

BACKGROUND: Highly active antiretroviral therapy (ART) is still the only effective method to stop the disease progression in acquired immunodeficiency syndrome (AIDS) patients. However, poor adherence to the therapy makes it ineffective. In this work, we construct an adherence prediction model of AIDS patients using the classical recency, frequency and monetary value (RFM) model in the data mining-based customer relationship management model to obtain adherence predictor variables. METHODS: We cleaned 257,305 diagnostic data elements of AIDS outpatients in Shanghai from August 2009 to Decembe

Analytical Prediction of Critical Transitions in Oscillator Networks With Non-Local Links.

A wide range of dynamical systems can be modeled as oscillators coupled through an interaction network. Such frameworks have been widely used to explore the emergence of collective phenomena across diverse applications. Here, we develop an analytical framework for oscillator-based Ising systems with Kuramoto-type pairwise coupling and a second harmonic term. Spectral dimension reduction identifies a stability boundary governed by the balance between global coupling and second-harmonic strength, while a mean-field Fokker-Planck analysis predicts the transition between single-cluster and two-clu

Synergistic action of specialized metabolites from divergent biosynthesis in the human oral microbiome.

Despite extensive efforts, our understanding of the virulence factors contributing to oral biofilm formation-a hallmark of dental caries-remains incomplete. We present evidence that the specialized metabolism of the oral microbiome is a critical yet underexplored factor in oral biofilm formation. Through microbiome analysis, we identified a hybrid nonribosomal peptide synthetase (NRPS) and polyketide synthase (PKS) encoding biosynthetic gene cluster that correlates with dental caries and is widely represented in oral pathogens, including Streptococcus mutans. This gene cluster produces two maj

Adolescent BCG revaccination induces a phenotypic shift in CD4+ T cell responses to Mycobacterium tuberculosis.

A recent clinical trial demonstrated that Bacille Calmette-Guérin (BCG) revaccination of adolescents reduced the risk of sustained infection with Mycobacterium tuberculosis (M.tb). In a companion phase 1b trial, HVTN 602/Aeras A-042, we characterize in-depth the cellular responses to BCG revaccination or to a H4:IC31 vaccine boost to identify T cell subsets that could be responsible for the protection observed. High-dimensional clustering analysis of cells profiled using a 26-color flow cytometric panel show marked increases in five effector memory CD4+ T cell subpopulations (TEM) after BCG re

Advanced Tool for Chiral Separations of Anionic and Zwitterionic (Metalla)carboranes: Supercritical Fluid Chromatography.

The continuous expansion of research in the field of stable carboranes and their wide potential in the drug design require carrying out fundamental studies regarding their chiral separations. Although supercritical fluid chromatography (SFC) is a viable technique for fast enantioseparations, no investigation concerning boron cluster compounds has been done yet. We aimed at the development of a straightforward method enabling chiral separations of racemic mixtures of anionic cluster carboranes and metallacarboranes that represent an analytical challenge. The fast gradient screening testing nine

Genomic fingerprinting and genotyping of Helicobacter pylori strains from patients with duodenal ulcer or gastric cancer from different geographic regions.

Continuing attempts have been made to classify pathogenic strains within bacterial populations based on DNA fingerprints and to identify virulence factors in H. pylori. We studied 287 H. pylori isolates from patients with duodenal ulcer or gastric cancer from three different geographic regions. DNA fingerprints were generated using REP-PCR and analyzed by cluster analysis. The status of three candidate virulence factors-vacA polymorphism, cagA and iceA,-were examined by PCR amplification. Cluster analysis of the REP-PCR fingerprints showed clustering by geographic region but not by disease pre…

A Multi-Task Deep Learning Framework for Characterizing Beating Behavior and Synchrony in Cardiomyocyte Clusters.

Beat-level synchrony among cardiomyocyte clusters is a critical indicator of cardiac electromechanical function. Traditional invasive approaches have substantial limitations, and conventional computer vision methods are poorly suited for resolving densely packed, adherent clusters. To address these challenges, we developed an analysis framework to characterize the beating characteristics of cardiomyocyte clusters from microscopic imaging data. Specifically, we propose CardioSegNet, a multi-task deep learning model that combines attention mechanisms with three prediction heads (semantic segment

Spatial clustering of multiple hyperplastic, adenomatous, and malignant colonic polyps in individual patients.

Analysis of relative polyp locations in 426 consecutive patients with multiple colonic polyps found on colonoscopy showed novel findings. First, synchronous and metachronous neoplastic polyps showed spatial clustering in individual patients. For example, patients with their largest neoplasm in the cecum or proximal ascending colon, had 34.3 percent +/- 4.6 percent (standard error) of their other colonic neoplasms in the same location. Second, hyperplastic polyps showed spatial clustering in individuals that was statistically significantly greater than expected from the increased hyperplastic p

Plasmacytoid dendritic cell clusters are associated with poor prognosis in chronic myelomonocytic leukemia.

Chronic myelomonocytic leukemia (CMML) is associated with high rates of transformation to acute myeloid leukemia (AML) and poor overall survival. Plasmacytoid dendritic cell (pDC) proliferations in bone marrow biopsies have been proposed to be prognostically significant in CMML. In this multi-institutional study, we quantified pDC clusters in bone marrow biopsies of patients with CMML by interferon regulatory factor 8 (IRF8) immunohistochemical staining performed in separate laboratories and demonstrated the association of pDC clusters with overall survival. Two hundred five newly diagnosed CM

Combined 5' UTR RFLP analysis and VP1 sequencing for epidemic investigation of enteroviruses.

Enteroviruses, the main cause of aseptic meningitis, consist of 100 serotypes, and many of them have been associated with large outbreaks. In the present study, a comparison of RFLP analysis of the 5' untranslated region (5'UTR) and sequencing of both the 5'UTR and VP1 regions was conducted for epidemiological linkage of 27 clinical enterovirus strains. The clinical enterovirus strains were clustered into five restriction profile groups. Even though the restriction profile clusters of clinical isolates were not related to those of the respective prototype strains, epidemiological relationships

Dumulmycin, an Antitubercular Bicyclic Macrolide from a Riverine Sediment-Derived Streptomyces sp.

Dumulmycin (1) was isolated from Streptomyces sp. DM28, a bacterial strain from a riverine sediment sample. The structure of 1 was elucidated as a bicyclic macrolide possessing 19-membered and 5-membered rings by spectroscopic analysis. The stereochemistry of 1 was determined by J-based configuration analysis, ROESY NMR data, DP4 calculations, and the modified Mosher's method. Genetic analysis identified a trans-acyltransferase polyketide biosynthetic gene cluster for 1. Dumulmycin exhibited in vitro antitubercular activity (MIC50 = 27.1 μM).
