Paternal Zearalenone Exposure Is Associated with Hepatic Dysfunction in F1 Offspring: Insights from Proteomic Analysis.

Zearalenone (ZEN) is a non-steroidal estrogenic mycotoxin that adversely affects directly exposed individuals, yet the intergenerational consequences of paternal ZEN exposure remain poorly understood. In this study, we investigated the impact of paternal ZEN exposure on hepatic outcomes in F1 offspring, with a focus on the underlying molecular mechanisms. Kunming male mice (F0) were fed a ZEN-supplemented diet (10 mg/kg bw/day) for 5 weeks. Their F1 offspring developed hepatic steatosis, elevated oxidative stress, and a chronic inflammatory state. Proteomic analysis of F1 livers revealed signi

Lysine acetyltransferase 8-mediated histone acetylation, regulated by GBA1, is associated with lysosomal function related to α-Synuclein pathology.

Lysosomal defects are closely linked to Parkinson's disease (PD). Mutations in the GBA1 gene, encoding the lysosomal enzyme glucocerebrosidase (GCase), are major genetic risk factors for PD. GBA1 deficiency causes lysosomal dysfunction, leading to α-synuclein (α-syn) accumulation and PD progression. However, the underlying mechanisms remain unclear. In this study, we identified a novel GBA1-KAT8 regulatory pathway that controls lysosomal activity. GBA1 overexpression enhances lysosomal enzyme expression, regulates histone H4 acetylation at K16 via KAT8, and promotes lysosome-associated gene ex

Clusterin ameliorates LPS-induced ARDS by inhibiting the Wnt/β-catenin pathway.

Acute respiratory distress syndrome (ARDS) is a critical illness characterized by endothelial barrier damage, and the present study investigates the specific role of clusterin (CLU). The study found that CLU concentrations were significantly lower in ARDS patients, particularly non-survivors, compared with non-ARDS patients and survivors; similarly, serum CLU levels were decreased in mice with lipopolysaccharide-induced ARDS. Both in vivo and in vitro experiments demonstrated that treatment with recombinant CLU protein significantly alleviated organ injury and suppressed the inflammatory respo

The role of metabolic disorders in major depressive disorder.

Major depressive disorder (MDD) is a highly disabling mental disorder, and its complex etiology remains elusive. Recent studies have shown that there is a close bidirectional relationship between metabolic disorders and MDD. Metabolic pathway disorders are not simply comorbid phenomena but may be deeply involved in the pathophysiological process of MDD. This review provides a systematic evaluation of metabolic dysregulations in MDD, encompassing amino acid, glucose, and lipid pathways. Distinct from existing literature that often treats these systems in isolation, our work uniquely highlights

Neuroprotective effect of sinomenine on parkinson's disease through NLRP3/Caspase-1/GSDMD pathway-mediated pyroptosis inhibition.

Mounting evidence indicates that NLRP3 inflammasome-mediated pyroptosis is critically involved in Parkinson’s disease (PD) pathogenesis. Sinomenine (SN) possesses neuroprotective properties and is known to modulate the NLRP3 inflammasome, but its potential to protect against PD via pyroptosis inhibition remains unexplored. This study demonstrates that SN confers significant neuroprotection in both cellular and animal PD models. Key findings show that SN alleviates motor deficits and nigral pathology in vivo, enhances neuronal viability, and suppresses pyroptosis in vitro. Mechanistically, SN p

A Defined 3D Hydrogel Model Recapitulates Key Transcriptomic Features of Lung Adenocarcinoma.

Three-dimensional (3D) culture systems have emerged as powerful tools to model tumor biology and bridge the gap between conventional two-dimensional (2D) assays and in vivo studies. Here, we evaluate a poly(ethylene glycol)-based hydrogel as a simplified yet functionally relevant platform for modeling premalignant lung adenocarcinoma using the A549 cell line. Cells cultured in hydrogels exhibited transcriptional profiles that more closely resembled xenograft tumors than conventional 2D monolayers. Pathway-level analysis and regression-based benchmarking revealed restoration of critical hallmar

Mechanisms and clinical implications of radiation-induced endothelial dysfunction.

Radiation-induced endothelial dysfunction is a critical mechanism underlying long-term cardiovascular and multiorgan toxicity in cancer survivors after radiotherapy. This review integrates current evidence on the molecular pathogenesis, from initial endothelial apoptosis and senescence to the establishment of chronic pro-thrombotic and pro-inflammatory phenotypes involving various signalling pathways. Clinical data reveal that even low to moderate radiation doses can cause sustained endothelial injury, contributing not only to vascular diseases but also to common cerebrovascular and gastrointe

The role of macrophages in peritoneal dialysis-associated peritoneal fibrosis.

The peritoneum is the largest and most complex serous membrane in the body belonging to the category of semi-permeable membrane. Peritoneal dialysis, which is based on this principle, has become a primary treatment for patients with end-stage kidney disease. Compared to hemodialysis, peritoneal dialysis is not only simple to operate and less expensive, but also better at preserving residual kidney function. However, peritoneal dialysis-associated fibrosis has become the leading cause of peritoneal function loss and treatment withdrawal. Macrophages play a critical role in the pathogenesis of p

ATF4 activates a transcriptional program that chronically suppresses mTOR activity and promotes neurodegeneration in Parkinson disease models.

The integrated stress response (ISR) enables cells to adapt to diverse cellular stresses, but during chronic or unresolved stress it becomes maladaptive and is implicated in neurodegenerative diseases, including Parkinson disease (PD). The mechanisms underlying maladaptive ISR-driven neurodegeneration, however, remain poorly defined. Here, we find a critical pathway by which chronic ISR activation promotes neurodegeneration in neurotoxin and α synucleinopathy models of PD in vitro and in vivo. We show that sustained activation of ATF4, the central ISR transcription factor, induces the coordina

Differential resilience against complete optic nerve transection across visual areas.

Understanding how the central visual system responds to complete optic nerve transection (ONT) is critical for evaluating the feasibility of whole-eye transplantation (WET), as envisioned by the 2024 ARPA-H THETA program. This study evaluates the dynamitic neuronal and synaptic changes within the posterior visual pathway-the superior colliculus (SC), lateral geniculate nucleus (LGN), and primary visual cortex (V1)-after optic nerve crush (ONC) and ONT in mice models. Complementary observations were obtained in non-human primates. Both injury models induced time-dependent alterations, but degen

Genetic mechanisms underlying host susceptibility to central nervous system viral infections.

PURPOSE OF REVIEW: To provide a critical review of the literature on the influence of host genetic variations on the susceptibility to viral encephalitis, and the proposed underlying key immunogenetic mechanisms. RECENT FINDINGS: Easier access to genomic sequencing has led to the identification of an ever-increasing number of monogenic defects and polymorphisms in genes that influence innate viral sensing, interferon signaling, cytotoxic lymphocyte function, antibody responses, cytokine regulation, and blood-brain barrier stability. While many of these genetic abnormalities have been implicat

Pterostilbene restores mitochondrial retrograde signaling to activate protective stress responses in Parkinson's disease.

Parkinson's disease (PD) is the selective demise of dopaminergic neurons in the substantia nigra. Conventional neuroprotective strategies based on exogenous antioxidants have shown minimal clinical efficacy. Emerging evidence suggests that neuronal loss in PD may stem not only from direct mitochondrial damage but, more critically, from the failure of an intrinsic "early-warning system"-the mitochondrial retrograde signaling (MRS) pathway-impairing the nucleus's ability to launch timely protective responses. This review repositions pterostilbene, a bioavailable dietary polyphenol, from a simple

Functional Characterization of Pathway Inhibitors for the Ubiquitin-Proteasome System (UPS) as Tool Compounds for CRBN and VHL-Mediated Targeted Protein Degradation.

Small molecule degraders such as PROteolysis TArgeting Chimeras (PROTACs) and molecular glues are new modalities for drug development and important tools for target validation. When appropriately optimized, both modalities lead to proteasomal degradation of the protein of interest (POI). Due to the complexity of the induced multistep degradation process, controls for degrader evaluation are critical and commonly used in the literature. However, comparative studies and evaluations of cellular potencies of these control compounds have not been published so far. Here, we investigated a diverse se

Expanding the Horizon of Natural Products: The Role of Starter Units in Nonribosomal Lipopeptide Biosynthesis.

Nonribosomal lipopeptides (NRLPs) are structurally complex natural products that play crucial ecological and biological roles. They are also valuable sources and lead structures for developing new pharmaceuticals. These compounds are typically synthesized using a molecular assembly machinery known as nonribosomal peptide synthetases (NRPSs) or hybrid polyketide synthases-NRPSs. Unlike conventional NRPS, NRLPs are characterized by a starter module that loads lipid chains and a substrate synthesis pathway that supplies the necessary substrates during the initiation stages. Unique lipid chains ar

Facet Selectivity of Carbon Dioxide Electroreduction to HCOOH on a Zn Catalyst.

Metallic zinc, regarded as the typical CO selectivity catalyst in CO2 electroreduction, can also exhibit HCOOH selectivity, but the real reasons for HCOOH selectivity remain somewhat unclear. Here, density functional theory calculations were initially employed to elucidate that the Zn(002) facet exhibited the most favorable adsorption energy for the critical HCOO* intermediate in HCOOH formation. Then, the Zn catalyst with a predominant (002) facet was prepared and exhibited a maximum HCOOH Faradaic efficiency of 90%, a partial current density of 40 mA cm-2 and excellent stability without deac

Investigation of the mechanism of methanol electrooxidation: a potential-dependent DFT study.

The methanol electrooxidation reaction (MER), a critical process in direct methanol fuel cells, is systematically investigated through potential-dependent density functional theory (DFT) simulations to unravel its mechanism and potential effects on Pt-based catalysts. For pure Pt, the rate-determining steps (RDSs) are identified as methanol adsorption and CO oxidation, leading to a high overpotential of 0.9 V. Alloying Pt with Cu (PtCu) significantly reduces the overpotential to 0.7 V, with CO oxidation remaining the sole RDS. Potential-dependent analysis reveals that PtCu exhibits enhanced me

Translational approaches manipulating mechanobiology to promote scarless healing in humans.

Wound healing is a complex biological process involving hemostasis, inflammation, proliferation, and remodeling. While this orchestrated response is essential for tissue repair, dysregulated healing often culminates in scar formation, characterized by fibrosis and loss of physiological tissue architecture. Scar formation impairs both aesthetic and functional outcomes, posing significant clinical challenges. We previously identified mechanical signaling, specifically through the Focal Adhesion Kinase (FAK) pathway, as a critical regulator of wound healing and fibrosis across various disease sta

Ferroptosis in Neonatal Hypoxic-Ischemic Encephalopathy: Mechanisms and the Therapeutic Potential of Vitamin D/VDR Signaling.

As a major neonatal brain disorder, hypoxic-ischemic encephalopathy(HIE) presents with elevated risks of long-term disability and neonatal death. Ferroptosis is a distinct mode of regulated cell death marked by excess intracellular iron, oxidative lipid injury, and suppressed GPX4 activity, and has gained attention as a pivotal mechanism in the development of HIE. Signaling pathways such as Nrf2, TLR4/NF-κB, and endoplasmic reticulum stress(ERS) play critical roles.Vitamin D (VD) and its receptor (VDR), beyond their classical roles in calcium-phosphate homeostasis, as neuroprotective modulator

Litomosoides sigmodontis microfilariae-induced eosinophil ETosis is dependent on the canonical inflammasome pathway.

Granulocytes exert several effector mechanisms, including the release of DNA traps during ETosis. While bacteria-induced ETosis has been linked to the non-canonical inflammasome pathway, the role of the inflammasome activation during ETosis in response to extracellular pathogens has not been investigated. The current study demonstrates that microfilariae (MF) of the rodent filarial nematode Litomosoides sigmodontis induce eosinophil ETosis via the canonical inflammasome pathway. The absence of key components of the canonical inflammasome, including gasdermin D, caspase-1, the adaptor molecule
