Detection of equine antibodies to babesia caballi by recombinant B. caballi rhoptry-associated protein 1 in a competitive-inhibition enzyme-linked immunosorbent assay.

A competitive-inhibition enzyme-linked immunosorbent assay (cELISA) was developed for detection of equine antibodies specific for Babesia caballi. The assay used recombinant B. caballi rhoptry-associated protein 1 (RAP-1) and monoclonal antibody (MAb) 79/17.18.5, which is reactive with a peptide epitope of a native 60-kDa B. caballi antigen. The gene encoding the recombinant antigen was sequenced, and database analysis revealed that the gene product is a rhoptry-associated protein. Cloning and expression of a truncated copy of the gene demonstrated that MAb 79/17.18.5 reacts with the C-termina

Isolation and characterization of a clone encoding a major allergen (Bla g Bd90K) involved in IgE-mediated cockroach hypersensitivity.

Previous studies have established that atopic individuals living in cockroach-infested housing become sensitized to cockroach aeroallergens and produce IgE antibodies to a variety of proteins. We describe the isolation of a complementary DNA clone from an expression library, constructed with messenger RNA from German (Blattella germanica) cockroaches, which encodes a major allergen involved in mediating cockroach hypersensitivity. Approximately 0.2% of the clones from a lambda ZAP XR cDNA library bound IgE from a patient with cockroach sensitivity. A randomly selected subset of these clones re

Polar zippers: their role in human disease.

Ascaris hemoglobin consists of 8 subunits, each of which contains a C-terminal peptide with the sequence Glu-Glu-Lys-His repeated 4 times. When plotted on a beta-strand, this sequence leads to alternate lysines and glutamates on one side of the strand, and alternate glutamates and histidines on the other side, suggestive of a polar zipper that links the subunits together. A computer search of the protein database showed that the same or similar sequences also occur in other proteins. Some contain long repeats of Asp-Arg or Glu-Arg, among them the small nuclear ribonucleo-U1 70K protein, which

Isolation from opossum serum of a metalloproteinase inhibitor homologous to human alpha 1B-glycoprotein.

Fractionation of opossum (Didelphis virginiana) serum with (NH4)2SO4, followed by chromatography on DEAE-Sepharose, phenyl-Sepharose, and Mono Q HR 5/5, has resulted in the isolation in homogeneous condition of a metalloproteinase inhibitor designated oprin (opossum proteinase inhibitor). Oprin is a single-chain glycoprotein (26% carbohydrate) with an estimated Mr = 52,000, pI = 3.5, and E(1%/1 cm) = 11. Oprin inhibited snake venom metalloproteinases, but showed no activity on venom serine proteinases or on bacterial metalloproteinases. Incubation of Crotalus atrox alpha-proteinase (EC 3.4.24.

Proteomics-Based Approach for Detailing the Allergenic Profile of Cannabis Chemotypes.

Allergic sensitization to cannabis is an emerging public health concern and is difficult to clinically establish owing to lack of standardized diagnostic approaches. Attempts to develop diagnostic tools were largely hampered by the Schedule I restrictions on cannabis, which limited accessibility for research. Recently, however, hemp was removed from the classified list, and increased accessibility to hemp allows for the evaluation of its practical clinical value for allergy diagnosis. We hypothesized that the proteomic profile is preserved across different cannabis chemotypes and that hemp wou

HNRNPC as a Novel Therapeutic Target for Ischemic Heart Disease: Evidence From Mendelian Randomization and Experimental Validation.

BACKGROUND: Several studies have suggested that N6-methyladenosine (m6A) plays an essential role in cardiovascular disease, but the causality of m6A on ischemic heart disease (IHD) remains unknown. Therefore, this study investigated the potential relationship between m6A and IHD using a 2-sample Mendelian randomization method. METHODS: The publicly available genome-wide association study data for m6A-related proteins were obtained from the INTERVAL study, a large population-based cohort of healthy blood donors in the United Kingdom, whereas the genome-wide association study database (includin

Autoantibody reactome profiling reveals distinct humoral immune signatures induced by simulated microgravity, radiation and combined exposure in mice.

Microgravity (μG) and space radiation are major environmental stressors leading to immune dysfunction in astronauts, but the mechanisms governing humoral immunity and effective interventions remain unclear. In this study, we used a high-throughput autoantigen microarray derived from the AAgAtlas 1.0 database to profile humoral immune responses in mice exposed to μG and proton irradiation. We generated 70,490 autoantibody reactivity measurements and observed combined effects of radiation and μG on humoral immunity. Specifically, 38 IgM and 57 IgG autoantibody candidates showed dose-associated c

Non-Random Association of Ultraconserved Genomic Elements (UCE) with Human Genes.

Ultraconserved elements (UCEs) are among the most evolutionarily conserved DNA sequences in vertebrate genomes, yet the biological mechanisms underlying their extraordinary conservation remain poorly understood. Using the recently developed dedUCE database comprising 12,813 human UCEs, we performed a comprehensive genome-wide analysis of their distribution relative to protein-coding genes, transcription factor (TF) genes, and long noncoding RNA (lncRNA) genes. UCEs showed a highly non-random genomic organization, with approximately 40% occurring in clusters within 20 kb genomic intervals. Non-

Dynamic protein expression in golden hamster oocytes and embryos reveals extensive post-translational regulation of PIWIL1 during the oocyte-to-embryo transition.

The golden hamster, renowned for its representative PIWI-piRNA pathway profile among mammals, has emerged as a promising animal model for investigating female mammalian fertility. Prior research uncovered that the disruption of Piwil1 leads to complete infertility of female golden hamsters, with embryos derived from homozygous knockout (Piwil1-/-) females mated with wild-type males arresting at the two-cell stage. However, the underlying deep mechanisms remain unexplored. In the current study, we utilized highly sensitive single-cell mass spectrometry to generate proteome data of five developm

Transcriptomic Signatures of Microglial Phagocytosis Reveal Diagnostic Markers and Immune Landscape in Alzheimer's Disease.

The core pathological features of Alzheimer's disease (AD) include Aβ plaques and neurofibrillary tangles, which collectively drive the neurodegenerative process. Meanwhile, the phagocytic function of microglia plays a dual role in AD: it attempts to clear pathological proteins such as Aβ, but its chronic activation may also exacerbate neuroinflammation and synaptic damage. This study integrated microarray data from AD cohorts in the Gene Expression Omnibus (GEO) database. Through differential expression analysis, protein-protein interaction network construction, and three machine learning alg

Targeting D-alanine ligase from methicillin-resistant Staphylococcus aureus using natural compounds.

Methicillin-resistant Staphylococcus aureus (MRSA) infections pose a critical threat in healthcare and community settings. Meanwhile, the effectiveness of treatment is limited by vancomycin, which remains the only frontline drug against MRSA. The emergence of new strains of MRSA, along with the development of vancomycin-resistant S. aureus (VRSA), emphasises the need for alternative therapies. This study explores the use of natural compounds as a sustainable alternative to treat MRSA. The inhibition potential of natural compounds against D-alanine-d-alanine ligase, a key enzyme in the peptidog

ADAMDEC1 promotes the malignant progression of cholangiocarcinoma by regulating NF-κB signaling pathway.

Cholangiocarcinoma (CCA), a highly aggressive form of cancer, is known for its high mortality rate. A Disintegrin and Metalloprotease Domain-like Protein Decysin-1 (ADAMDEC1) can promote the development and metastasis in various tumors by degrading the extracellular matrix. However, its regulatory mechanism in CCA remains unclear. Public databases and clinical tissue samples were used to evaluate whether ADAMDEC1 expression was correlated with the prognosis of CCA. We investigated the expression of ADAMDEC1-related regulatory genes and proteins in CCA and assessed the biological behaviors of C

Transcriptional Regulatory Systems in Pseudomonas: A Comparative Analysis of Helix-Turn-Helix Domains and Two-Component Signal Transduction Networks.

Bacterial communities in diverse environmental niches respond to various external stimuli for survival. A primary means of communication between bacterial cells involves one-component (OC) and two-component signal transduction systems (TCSs). These systems are key for sensing environmental changes and regulating bacterial physiology. TCSs, which are the more complex of the two, consist of a sensor histidine kinase for receiving an external input and a response regulator to convey changes in bacterial cell physiology. For numerous reasons, TCSs have emerged as significant targets for antibacter
