Mixed infections and In Vivo evolution in the human fungal pathogen Cryptococcus neoformans.

Koch's postulates are criteria establishing a causal relationship between a microbe and a disease that lead to the assumption that diseases are caused by a single strain or its evolved forms. Cryptococcus neoformans is a life-threatening human fungal pathogen responsible for an estimated 1 million cases of cryptococcosis/year, predominantly meningoencephalitis. To assess the molecular diversity of clinical isolates and gain knowledge of C. neoformans biology in the host, we analyzed clinical cultures collected during the prospective CryptoA/D study. Using molecular analysis of unpurified isola

Venom from the centipede Scolopendra viridis Say: purification, gene cloning and phylogenetic analysis of a phospholipase A2.

Venom components from the centipede Scolopendra viridis Say were studied, using both the soluble venom and a cDNA library prepared from mRNA of the venomous glands. Separation of the soluble venom by high performance liquid chromatography (HPLC) permitted to obtain at least 54 different fractions. The fraction eluting at 46.24 min showed phospholipase activity. The enzyme was purified to homogeneity and the first 25 amino acid residues were identified by Edman degradation. From the cDNA library several genes were cloned, one of which codes for a protein with identical amino acid sequence as th

Angelman syndrome phenotype associated with mutations in MECP2, a gene encoding a methyl CpG binding protein.

Angelman syndrome (AS) is a neurodevelopmental disorder characterised by severe mental retardation, absent speech, ataxia, sociable affect, and dysmorphic facial features. Eighty five percent of patients with AS have an identifiable genetic abnormality of chromosome 15q11-13. Mutations within the X linked MECP2 gene have been identified in patients with Rett syndrome (RTT), a neurodevelopmental disorder which affects females almost exclusively and which shares phenotypic overlap with AS. RTT is usually associated with normal development in infancy followed by loss of acquired skills and evolut

The puzzle of Hox genes.

During the last few years, the regionalization of the rostral-caudal axis has been extensively studied through treatments with RA and genetic manipulations of Hox-C genes. RA shifts several Hox expression boundaries rostrally, deletes anterior rhombomeres but expands the caudal ones, and induces homeotic transformations in the vertebral column. These phenotypes indicate that retinoids may act in a graded fashion in the A-P axis, with maximum activity caudally. This excludes forebrain and midbrain, which apparently depend on neither Hox-C genes nor RA modulations, at least during early developm

Mercuric chloride induces autoantibodies against U3 small nuclear ribonucleoprotein in susceptible mice.

Autoantibodies to nucleolar components are a common serological feature of patients suffering from scleroderma, a collagen vascular autoimmune disease. While animal models, which spontaneously develop abundant anti-nucleolar antibodies, have not yet been described, high titers of such antibodies may be induced by treating susceptible strains of mice with mercuric chloride. We have identified the nucleolar autoantigen against which the HgCl2-induced IgG autoantibodies from mice of strain B10.S are directed. It is a protein with an apparent molecular mass of 36 kDa and a pI value of approximatel

Single-sequence based gFET-aptasensors for the discrimination of apo- and holo-RBP4 in human serum.

Retinol-binding protein 4 (RBP4) is a key transporter of all-trans-retinol (vitamin A), circulating in blood as either holo-RBP4 (retinol-bound) or apo-RBP4 (retinol-free). Dysregulated RBP4 levels, particularly an imbalance between apo- and holo-RBP4, have been implicated in a range of metabolic and cardiovascular diseases. Current detection methods such as ELISA and Western blot lack the specificity to distinguish these two forms. Here, we present an aptamer-based biosensing strategy that overcomes this limitation by using selected single aptamer sequences for the precise and selective quant

The relative congruence of cranial and genetic estimates of hominoid taxon relationships: implications for the reconstruction of hominin phylogeny.

Previous analyses of extant catarrhine craniodental morphology have often failed to recover their molecular relationships, casting doubt on the accuracy of hominin phylogenies based on anatomical data. However, on the basis of genetic, morphometric and environmental affinity patterns, a growing body of literature has demonstrated that particular aspects of cranial morphology are remarkably reliable proxies for neutral modern human population history. Hence, it is important to test whether these intra-specific patterns can be extrapolated to a broader primate taxon level such that inference rul

Large-cell transformation following detection of minimal residual disease in cutaneous T-cell lymphoma: molecular and in situ analysis of a single neoplastic T-cell clone expressing the identical T-cell receptor.

PURPOSE: One of the unique characteristics of cutaneous T-cell lymphoma (CTCL) is its ability to undergo cytologic transformation in which the malignant T cells develop the morphologic appearance of a large-cell lymphoma. Reported to occur in up to 20% of advanced cases, large-cell transformation (LCT) is associated with an aggressive clinical course. Little is known about the risk factors or the molecular mechanisms of LCT. Before current immunohistochemical and molecular techniques, it was not possible to determine if LCT represented changes of the initial neoplastic T-cell clone or, in fact

Dynamic Interactions and Deformation Mechanisms in Nanocutting of Carbon Nanotube Reinforced Ni-Based Composites.

Molecular dynamics simulations were employed to investigate the effects of cutting speed and cutting depth on the nanocutting behavior of carbon nanotube (CNT)-reinforced Ni composites. The results show that increasing cutting speed from 50 to 200 m/s reduces the average tangential cutting force by approximately 16.5%, while the cutting temperature increases by 26.7% at a cutting distance of 200 Å. Cutting depth produces a stronger influence on subsurface deformation, with deeper cutting accompanied by increased stress localization, structural disorder, and dislocation activity. In contrast, d

Molecular and cellular correlates of human cortical lateralization.

Hemispheric specialization is a fundamental feature of human brain organization, with most individuals showing left hemisphere dominance for language and right hemisphere dominance for visuospatial attention. Although lateralized functions exist in other species, humans exhibit a pronounced, species-wide bias. Despite its evolutionary and functional significance, the molecular and cellular basis of this asymmetry remains unclear. Here, we identify neurochemical and cellular features associated with cortical lateralization. We show lateralized gradients in neurotransmitter receptor densities al

Advances in the study of the role of epigenetic mechanisms in plant growth and response to stress.

Plants face the challenge of balancing dynamic environmental stresses with internal developmental demands during growth, necessitating the precise regulation of gene expression networks to optimize resource allocation. Recent studies have shown that epigenetic regulatory systems, which serve as bridges between genotype and phenotype, enable plants to respond rapidly to environmental changes and facilitate adaptive evolution by establishing heritable epigenetic memory. This article systematically elucidates the molecular mechanisms by which core epigenetic processes coordinate plant growth and

Invisible Threats, Relentless Hunters: Biosurveillance of Airborne Plant Pathogens.

Airborne dispersal enables plant pathogens to travel across fields, regions, and continents, fueling rapid epidemics and emerging disease threats. Biosurveillance, the systematic monitoring of airborne inoculum, offers the opportunity to detect pathogens before symptoms appear and informs timely, risk-based management. Recent advances in air sampling, molecular diagnostics, metagenomics, and imaging technologies have expanded the scale and resolution of pathogen monitoring, from single-species qPCR assays to community-level aerobiome surveys. Integration of biosurveillance data with decision-s

An "Oxidative Stress Clock" Nanoprobe Reveals the Therapeutic Time Window for Antioxidant Intervention in Atherosclerosis.

The antioxidant treatment for atherosclerosis has presented a paradox: theoretically feasible but clinically ineffective. In this study, we developed an "oxidative stress clock" nanoprobe to dynamically monitor oxidative stress at different stages of atherosclerosis, thereby dissecting the optimal time window for antioxidant intervention. This probe could simultaneously and specifically recognize hydrogen peroxide, protein sulfenylation, and protein phosphorylation in vitro and in vivo, enabling accurate mapping of the evolution of oxidative stress during disease progression. Using this probe,

Thermal Decomposition Mechanism of β‑HMX under Irradiation Damage Based on Molecular Dynamics Simulations.

Energetic materials undergo chemical bond cleavage and structural reorganization under external stimuli such as heat and irradiation, leading to performance degradation or change. This study employs molecular dynamics simulations to investigate the atomic-scale mechanisms of irradiation-induced damage in β-HMX and its subsequent thermal decomposition mechanism. Results revealed that irradiation defects are primarily localized within collision cascade regions. Irradiation-generated nitrogen oxides and HNO x radicals provide new reaction sites during HMX thermal decomposition, significantly alte

A single-point mutation in TRPA1 drives heat resilience in oviparous embryos.

The molecular basis for species-specific thermal adaptation in vertebrate sensory systems is not well understood. We show that a single-amino-acid change in the pore domain of ion channel transient receptor potential ankyrin 1 (TRPA1) during the synapsid-diapsid split rewired its heat sensitivity. Many oviparous vertebrates retaining the ancestral residue display TRPA1 heat activation, protecting embryonic development under high temperatures, whereas selection for an aspartate in mammals reduced thermal responsiveness. Blocking heat-activated TRPA1 in oviparous embryos impaired dorsal-root-gan

How I treat advanced phases of CML.

Advanced phases of Philadelphia chromosome-positive chronic myeloid leukemia (CML), classically encompassing de novo presentation of accelerated-phase disease and blast-phase disease (myeloid and lymphoid "blast crisis") and progression to these from antecedent chronic-phase disease, have diminished in incidence but remain a challenge. Despite continued development of additional and novel kinase inhibitors of BCR::ABL1, global limitations on access to diagnostics and therapy persist and account for continued incidence of advanced disease at initial presentation and progressive disease. Mechanisms and biomarkers of immune checkpoint inhibitor-associated myocarditis: from T cell imbalance to multicellular crosstalk.

Immune checkpoint inhibitors (ICIs) have transformed cancer therapy but can induce immune-related myocarditis (ICI-MC), a rare yet life-threatening toxicity. This review elucidates the cellular and molecular mechanisms underlying ICI-MC, integrating insights from T-cell dysregulation, innate immunity, and stromal interactions. Pathogenesis involves α-myosin-driven clonal expansion of cytotoxic CD8+ T cells, loss of regulatory subsets, and macrophage polarization toward M1 phenotypes via cGAS-STING and STAT1/NF-κB pathways. Crosstalk among macrophages, fibroblasts, endothelial cells, neutrophil

Comparative single-cell transcriptomics highlights species-biased developmental trajectories and candidate regulatory modules in primate spermatogenesis.

Understanding primate testicular development is essential for elucidating the evolution of male fertility. Here, we integrated publicly available single-cell RNA sequencing datasets from 23 human and 4 macaque testes spanning infant to adult stages. Integrated analysis identified 22 conserved germ cell subtypes, including spermatogonial stem cells, spermatocytes, and spermatids. Notably, a human-enriched late spermatid-like transcriptional state, Sperm.C2, was enriched for genes involved in chromatin condensation, acrosome formation, and fertilization readiness, but was not detected in the mac

From Physicochemical Classification to Multidimensional Insights: A Comprehensive Review of Uremic Toxin Research.

Chronic kidney disease (CKD) is a global health burden, with uremic toxins (UTs) playing a central role in its pathophysiology. In this review, we systematically examined the evolution of UT classification from the 2003 European Uremic Toxin Work Group (EUTox) system based on molecular weight and protein-binding properties to the 2023 multidimensional framework integrating clinical outcomes, clearance technologies, and artificial intelligence. We highlighted the toxicity mechanisms of UTs across the cardiovascular, immune, and nervous systems and evaluated traditional (e.g., low-/high-flux hem
