Shared Neural Correlates Underlying Addictive Disorders and Negative Urgency.

Negative urgency is a personality trait reflecting the tendency to act rashly in response to extreme negative emotions and is considered a transdiagnostic endophenotype for problematic levels of addictive behaviors. Recent research has begun to identify the neural correlates of negative urgency, many of which appear to overlap with neural circuitry underlying addictive disorders associated with negative urgency. The goal of this qualitative review is to summarize the extant literature concerning the neural correlates of negative urgency, to compare these correlates with those implicated with a

Acquired mutation of the tyrosine kinase JAK2 in human myeloproliferative disorders.

BACKGROUND: Human myeloproliferative disorders form a range of clonal haematological malignant diseases, the main members of which are polycythaemia vera, essential thrombocythaemia, and idiopathic myelofibrosis. The molecular pathogenesis of these disorders is unknown, but tyrosine kinases have been implicated in several related disorders. We investigated the role of the cytoplasmic tyrosine kinase JAK2 in patients with a myeloproliferative disorder. METHODS: We obtained DNA samples from patients with polycythaemia vera, essential thrombocythaemia, or idiopathic myelofibrosis. The coding exo

Cellular and molecular neurosurgery: pathways from concept to reality--part I: target disorders and concept approaches to gene therapy of the central nervous system.

Recent advances in cellular and molecular biology and better understanding of genetic and biochemical bases of different central nervous system (CNS) disorders have made gene therapy of the CNS a realistic goal. Concept approaches for gene therapy of CNS disorders are reviewed and include the following: 1) gene replacement with a single normal allele to correct the inherited global neurodegenerative disorders, such as enzyme deficiencies; 2) brain repair to restore the function of a particular subset of cells that were lost because of a neurodegenerative process; 3) gene therapy of brain tumor

No Significant Changes in Addictive and Problematic Behaviors During the COVID-19 Pandemic and Related Lockdowns: A Three-Wave Longitudinal Study.

INTRODUCTION: The COVID-19 outbreak and related lockdowns brought substantial changes in people's lives and led to concerns about possible increases of addictive behaviors at the initial stages of the pandemic. To examine these concerns, the aim of the present study was to assess longitudinal changes in addictive and problematic behaviors (i.e., problematic social media use, Internet gaming disorder, gambling disorder, problematic pornography use, and compulsive sexual behavior disorder) over time during the COVID-19 pandemic. METHODS: Three waves of data collection took place in different st

Food-Borne Environmental Contaminants, Intestinal Injury, and Early Childhood Stunting: A Developmental Toxicology Perspective.

BACKGROUND: Childhood stunting (height-for-age z-score < -2 SD) remains a global developmental health burden. Traditional interventions centered on nutrition and infection have reached a bottleneck. Emerging evidence identifies chronic low-dose dietary environmental toxins as an independent, modifiable risk factor for early-life growth restriction. METHODS: This review synthesizes cohort studies, meta-analyses, and in vivo toxicological models published over the past decade to characterize exposures to mycotoxins, toxic heavy metals, and food-borne endocrine disruptors that are closely associ

Understanding Posttraumatic Growth in Conflict-Affected Children and Adolescents: A Systematic Scoping Review.

PURPOSE: The United Nations High Commissioner for Refugees reports that armed conflicts burden children, comprising nearly half of the population in most war and conflict regions. Despite experiencing trauma from exposure to armed conflict, some children experience positive psychological change as well. The study aims to systematically review existing literature to explore the factors that promote posttraumatic growth in children and adolescents exposed to armed conflict. MATERIAL AND METHOD: This systematic scoping review followed five consecutive steps specified in the Arksey and O'Malley f

A review of the TGF-β1 pathway in Alzheimer's disease and depression: Possible restoration potential of antidepressants.

Alzheimer's Disease (AD), the most common type of dementia, is increasing rapidly in prevalence, while Major Depressive Disorder (MDD), the most frequent psychiatric condition, continues to pose significant global healthcare challenges. Recent hypotheses suggest that AD and MDD might be interrelated, potentially sharing common pathophysiological mechanisms. One possible central link is the Transforming Growth Factor-beta 1 (TGF-β1) pathway. Reduced TGF-β1 levels, a cytokine involved in inflammatory responses, have been observed in both conditions. In AD, diminished TGF-β1 might contribute to n

Use of Aromatase Inhibitors in Short Children and Adolescents to Increase Height Gain: A Current Practice Survey and Review of the Literature.

INTRODUCTION: Management of pediatric patients with short stature or short predicted adult height (PAH) is challenging. The use of aromatase inhibitors (AIs) has become a therapeutic option. Up to date, there is still scarce data about the indications of AI. Our aim was to conduct a survey on the real-world use of AI in pediatric patients with short stature or short PAH. METHODS: We conducted a descriptive, cross-sectional, questionnaire-based study. Participants were collected throughout convenient sampling and by diffusion through different pediatric endocrine societies. RESULTS: Two hundr

Post-traumatic growth in young and middle-aged patients with stroke: a qualitative systematic review and meta-synthesis.

BACKGROUND: With the increasing incidence of stroke among young and middle-aged individuals, addressing their psychological well-being has become a critical concern. Negative emotional states such as depression, anxiety, and conditions like post-traumatic stress disorder (PTSD) can hinder recovery and increase the risk of recurrence. Therefore, this study aims to conduct a meta-synthesis of qualitative research on post-traumatic growth (PTG) in this population, examining the factors that contribute to PTG, its developmental trajectory, and the coping mechanisms involved. METHODS: A systematic

Evaluation of copeptin in children after stimulation with clonidine or L-Dopa.

OBJECTIVES: Arginine-stimulated serum copeptin has been proposed as a new method to diagnose arginine vasopressin (AVP) deficiency in children and adolescents. Herein we investigated the secretagogic potential of clonidine or L-Dopa on the copeptin serum levels in children. METHODS: Eight stimulation tests (4 with clonidine and 4 with L-Dopa) were performed in eight children (5 boys and 3 girls) with a median age of 6.5 years-old, evaluated for short stature due to possible growth hormone deficiency. Serum copeptin levels were measured at 30, 60, 90, and 120 min after administration of clonid

Estrogen Receptor 1 Gene Polymorphism and its Association with Idiopathic Short Stature in a North Indian Population.

OBJECTIVE: In the hypothalamic-pituitary-gonadotrophin axis, estrogen plays a key role in the regulation of bone maturation and growth plate closure. This study was designed to explore the link between single nucleotide polymorphisms (SNPs) in the estrogen receptor 1 (ESR1) gene with idiopathic short stature (ISS) susceptibility in a North Indian population. METHODS: Four SNPs of ESR1 (rs543650, rs6557177, rs2234693 and rs9340799) were genotyped by Sanger sequencing in ISS patients and controls. Linkage disequilibrium (LD) and haplotyping were done by SNPStat and SHEsisPlus software. The exte

Food inflation and child undernutrition in low and middle income countries.

The 21st Century has been marked by increased volatility in food prices, with global price spikes in 2007-08, 2010-11, and again in 2021-22. The impact of food inflation on the risk of child undernutrition is not well understood, however. This study explores the potential impacts of food inflation on wasting and stunting among 1.27 million pre-school children from 44 developing countries. On average, a 5 percent increase in the real price of food increases the risk of wasting by 9 percent and severe wasting by 14 percent. These risks apply to young infants, suggesting a prenatal pathway, as we

Clinical-genetic analysis of selected genes involved in the development of the human skeleton in 128 Czech patients with suspected congenital skeletal abnormalities.

BACKGROUND: Congenital skeletal abnormalities are a heterogeneous group of diseases most commonly associated with small or disproportionate growth, cranial and facial dysmorphisms, delayed bone maturation, etc. Nonetheless, no detailed genotype-phenotype correlation in patients with specific genetic variants is readily available. Ergo, this study focuses on the analysis of patient phenotypes with candidate variants in genes involved in bone growth as detected by molecular genetic analysis. METHODS: In this study we used molecular genetic methods to analyse the ACAN, COL2A1, FGFR3, IGFALS, IGF

Impact of Presence, Level, and Closure of a Stoma on Growth in Young Children: A Retrospective Cohort Study.

INTRODUCTION: A stoma will cause nutrients loss which could result in impaired growth. Impaired growth can negatively impact long-term development. This study aims to evaluate: (1) the effect of stomas on growth comparing small bowel stoma versus colostomy and (2) if early closure (within 6 weeks), proximal small bowel stoma (within 50 cm of Treitz), major small bowel resection (≥ 30 cm), or adequate sodium supplementation (urinary level ≤ 30 mmol/L) influences growth. METHODS: Young children (≤ 3 years) treated with stomas between 1998 and 2018 were retrospectively identified. Growth was mea
