UNLABELLED: Aging and pathologic conditions cause intracellular aggregation of macromolecules and the dysfunction and degeneration of neurons, but the mechanisms are largely unknown. Prime examples are lysosomal storage disorders such as Niemann-Pick type C (NPC) disease, where defects in the endosomal-lysosomal protein NPC1 or NPC2 cause intracellular accumulation of unesterified cholesterol and other lipids leading to neurodegeneration and fatal neurovisceral symptoms. Here, we investigated the impact of NPC1 deficiency on rodent neurons using pharmacologic and genetic models of the disease.

Extracellular vesicles (EVs) are gaining increasing attention among studies focusing on biomarker research, elucidating disease mechanisms, and exploring therapeutic approaches. Here, we present a method for size-exclusion chromatography (SEC)-based enrichment of human plasma EVs, and describe steps for assessing the quality of obtained EV samples. For characterizing the EVs, we provide detailed procedures for western blot, transmission electron microscopy, and nanoparticle tracking analysis techniques.

The primary treatment for ameloblastoma is curettage or marsupialization, with a relatively high recurrence. Presently, the authors demonstrate the potential efficacy of topical 5-fluorouracil (5-FU) as a promising treatment modality for ameloblastoma after curettage in 4 cases. Transmission electron microscopy of 5-FU-treated ameloblastoma tissue showed apoptosis within cells. Topical application of 5-FU after curettage efficiently treats ameloblastoma without recurrence within 2 years.

Legionella utilizes specialized protein secretion machinery called the type IV secretion system encoded by dot/icm genes to modulate host cellular systems. We describe here the procedure to isolate the core complex of the Dot/Icm type IV secretion system of L. pneumophila based on detergent lysis of bacteria and ultracentrifugation. The isolated protein complex can be applied for biochemical and transmission electron microscopy analysis.

The 58 cervical biopsies were studied by cytological, histological, immunomorphological methods, electron microscopy and PCR. Expression of L1 was observed only in the differentiated cells of squamous and metaplastic cervical epithelium. At increase of grade of cervical epithelial lesion decrease expression L1 from 75% of cases of CIN1 up to 28.5% of cases of SCC. Not all capsid structures connect with DNA HPV in case of CIN3.

Two outbreaks of mortality in juvenile barramundi (Lates calcarifer) with a high prevalence of gastrointestinal cryptosporidiosis are described. Juvenile barramundi at a separate hatchery and grow-out facility were demonstrating markedly increased mortality rates. Histological examination in all cases indicated a heavy infection of Cryptosporidium-like organisms within the distal stomach and proximal small intestine. Electron microscopy confirmed a diagnosis of cryptosporidiosis; however, speciation was not possible.

We present a case of juvenile dermatomyositis with unusual histopathologic findings. The child presented with a course consistent with dermatomyositis, a diagnosis confirmed by finding reticulotubular aggregates in endothelial cells on electron microscopy. However, histopathology of his muscle biopsy revealed a striking pattern of glycogen accumulation, to an extent similar to that seen in glycogen storage diseases; this degree of accumulation could potentially confound histopathologic diagnosis.

Burkholderia pseudomallei causes melioidosis and is believed to be an intracellular pathogen in human and animal disease. The uptake of B. pseudomallei by mouse peritoneal macrophages and cells in tissue culture was examined by electron microscopy. In all the systems studied B. pseudomallei were phagocytosed and apparently inhibited the normal processes of intracellular killing. Destruction of the phagosome membrane occurred and the bacteria escaped into the cytoplasm.

An 82-year-old male presented with a small parasagittal meningioma associated with disproportionately severe perifocal edema. Histological examination including immunohistochemical staining and electron microscopy resulted in a diagnosis of secretory meningioma. In addition to tumor size, the edema could not be explained by location, growth rate, vascular involvement, or other factors. We conclude that secretory meningiomas may possess an innate ability to cause brain edema.

A 43-year-old woman with progressive external ophthalmoplegia developed a bifascicular block and dilatation of the right ventricle during 4 years of follow-up. Histochemical and electron microscopy studies detected mitochondrial abnormalities in ocular, skeletal muscle and cardiac biopsies. This case registers disease progression from the external ocular to the skeletal and cardiac muscles. Mitochondrial DNA was deleted in relation to the morphological abnormality.

A female infant, younger than any other case in the literature, with ACTH-producing microadenoma of the pituitary is reported. She had full-blown symptoms and signs as well as laboratory evidence of Cushing's disease. The tumor was investigated by histology, immunocytology (avidin-biotin-peroxidase complex technique), and electron microscopy. The possibility that the tumor was derived from the fetal intermediate lobe is discussed.

In light of the recent finding of the S-100 antigen in satellite cells of the rat adrenal medulla, we looked for S-100-labelled cells in both the normal human adrenal medulla and in pheochromocytomas. Immunostaining enabled us to detect S-100-labelled satellite cells by both light and electron microscopy in a significant number of pheochromocytomas and, as expected, in the normal human adrenal medulla.

Special interaction between Salmonella typhimurium (STM) and Schistosoma mansoni is considered under two complementary aspects, in vivo and in vitro, using scanning and transmission electron microscopy. The resulting observations have obviously permitted us to discover a large specificity in these adhesion phenomena that seems to lead to the fusion of membranes between the two organisms. Microanalysis trials executed in these areas of strong affinity were attempted.

A 12-year-old, castrated male cat with a history of respiratory disease developed bilateral endophthalmitis, retinal detachment, and granulomatous choroiditis, and unilateral glaucoma, and was killed. Other lesions included granulomatous optic neuritis, myositis involving extraocular muscles, and focal retinitis. Light and electron microscopy showed many intracellular organisms, interpreted to be Histoplasma. Granulomatous inflammatory lesions and organisms were present in lung, liver, lymphoid tissues, and adrenals.

Skin biopsies from a collodion baby, spontaneously healing at the end of the third month, were taken on the 1st and 15th day after delivery and examined by light and electron microscopy. The microscopical features observed were different from those known to occur in collodion babies evolving into lamellar ichthyosis and may contribute to a more precise, early diagnosis and prognosis of this heterogeneous neonatal syndrome.
