BACKGROUND: Long-standing pulmonary hypertension (PH) leads to structural alterations of the pulmonary vasculature and its endothelium, and occlusion of small vessels by microthrombi. In patients with PH, the search for factors inducing or worsening endothelium damage and in situ thrombi is still ongoing. Thrombomodulin (TM), an endothelial cell membrane protein, is a receptor for thrombin and a major anticoagulant proteoglycan.
PURPOSE: To analyze plasma TM levels in patients with different forms of severe PH.
PATIENTS: We prospectively studied 32 consecutive patients with PH referred for h

BACKGROUND AND OBJECTIVES: Neuronal ceroid lipofuscinosis type 1 (CLN1) is a rare autosomal recessive lysosomal storage disorder caused by pathogenic variants in the PPT1 gene, leading to lipofuscin accumulation, neurodegeneration, psychomotor regression, seizures, and vision loss, with early death in infancy. Currently, no curative treatment exists. Neuroinflammation plays a major role in CLN1 pathophysiology, making anti-inflammatory treatments a potential option. Fingolimod, an immune modulator approved for multiple sclerosis (MS), has shown efficacy in reducing neurodegeneration in CLN1 mo

BACKGROUND: Among the salivary gland carcinomas, carcinoma in pleomorphic adenoma has been regarded as a representative carcinoma type which arises secondarily in the background of a pre-existent benign pleomorphic adenoma. It is still poorly understood how and which benign pleomorphic adenoma cells transform into its malignant form, carcinoma ex pleomorphic adenoma.
METHODS: We have established five cell systems from a benign pleomorphic adenoma of the parotid gland of a 61-year-old woman. They were characterized by immunofluorescence, classical cytogenetics, p53 gene mutational analysis, fl

The skull bone marrow contributes to brain immune homeostasis via recently discovered skull-meningeal channels, enabling the bidirectional trafficking of immune cells between skull bone and underlying dura mater. In multiple sclerosis, autoreactive T cells migrate to the bone marrow and shift its haematopoietic output towards myeloid differentiation, contributing to disease progression. However, the role of the skull bone marrow in multiple sclerosis pathophysiology, and its relationship to brain damage and clinical disability remain largely unexplored. We utilized simultaneous MR-PET with the

BACKGROUND: Maternal nutrition is an important factor in the health of lactating women and their children. Due to increased energy and nutrient needs during lactation, women in middle- and low-income nations are more nutritionally vulnerable than other women of reproductive age. Inadequate energy and macronutrient intake can also contribute to stunting, muscle wasting, edema, loss of bone density, anemia, and increased susceptibility to infections and nutritional deficiencies in women and babies. Some previous studies reported only the descriptive results of energy and macronutrient intake. Bu

Androgens are important steroid hormones for expression of the male phenotype. They have characteristic roles during male sexual differentiation, during development and maintenance of secondary male characteristics, and during the initiation and maintenance of spermatogenesis. The two most important androgens in this respect are testosterone and 5 alpha-dihydrotestosterone. Each androgen has its own specific role during male sexual differentiation, testosterone is involved in the development and differentiation of Wolffian duct derived structures, whereas 5 alpha-dihydrotestosterone, a metabol

INTRODUCTION: Suboptimal prenatal nutrition is associated with adverse pregnancy outcomes, yet the contribution of the lived food environment on prenatal diet quality remains incompletely understood. This study evaluated differences in prenatal macro- and micronutrient intake, glycemic and lipid intake patterns, and dietary quality indices among pregnant individuals living in food desert (FD) versus non-FD areas.
METHODS: This secondary analysis utilized data from an urban maternal-child cohort enrolled between 2014 and 2024. Eligible participants were aged 18-40 years, had a singleton pregna

The prominent role of the CD40 receptor in B cell responses led us to investigate the role of the gp39-CD40 interaction in a group of primary immunodeficient patients with defective antibody production. Here we report that patients with hyper-IgM syndrome (HIM) have a defective gp39-CD40 interaction. B cells from HIM patients express functional CD40, but their T cells do not bind CD40-Ig. These patients expressed normal levels of gp39 mRNA, but these mRNAs encode defective gp39 proteins owing to mutations in the extracellular domain of gp39. Soluble recombinant forms of gp39 containing these m

BACKGROUND: Abnormal glucose tolerance (AGT) is surging rapidly in sub-Saharan Africa, yet evidence on effective interventions remain limited. Physical activity improves glycemic control, but self-reported measures are prone to bias. Device-based monitoring enables objective assessment and may enhance adherence. This pilot randomized controlled trial assessed the preliminary efficacy of a structured, device-measured physical activity intervention on cardiometabolic outcomes among Ghanaian adults with AGT.
METHODS: An open-label, parallel-group pilot RCT with blinded outcome assessment randoml

Stochastic optical reconstruction microscopy (STORM) is a single-molecule localization microscopy technique that enables visualization of cellular structures beyond the diffraction limit. This approach has revealed previously inaccessible ultrastructural details in a wide range of cellular components, including the actin cytoskeleton, clathrin-coated pits, mitochondria, and bacterial nucleoid-associated proteins. STORM relies on the sequential emission of single photons from photosensitive fluorophores, which are precisely localized before entering a dark state or undergoing photobleaching. By

ETHNOPHARMACOLOGICAL RELEVANCE: The incidence of cholestatic liver disease (CLD), which is primarily marked by abnormal bile acids (BAs) metabolism and can result in significant hepatic injury, is rising. Nevertheless, there remains a lack of effective treatments and drugs in clinical practice. Silphium perfoliatum L. (SP) is rich in various structural types of caffeoylquinic acid (CQA) compounds, and it is a traditional herb of North American Indians with hepatobiliary therapy effects. However, its therapeutic effect and mechanism of action on CLD have never been studied.

Ag-specific CD4(+) Th cells play a key role in the development, maturation, and maintenance of pathogen-specific humoral and cellular immune responses. To define the fine specificity of broadly reactive Th responses associated with mature immunity in a lentiviral system, we analyzed peptide-specific Th responses in eight macaques chronically infected with a reference live attenuated SIV at 12-14 mo postinoculation. All macaques had stable immunocompetent Th cells at the time of analysis, and a unique array of Th responses to 20-mer overlapping peptides from envelope (Env) and Gag was identifie

The use of nucleic acids as potential therapeutic tools, sensors, or biomaterials, among other applications, has dramatically increased. Among these, RNA aptamers are of interest due to an innate high specificity toward their cognate targets, which include small molecules, proteins, ions, or cells. In this work, we took advantage of the ability that 8-oxo-7,8-dihydroguanine (8-oxoG) has to participate in unique H-bonding interactions, and probed its use to increase/control the selectivity/affinity of aptamers of RNA and DNA. The chosen model is a 23-nt long RNA (Neo61/Neo1-5'-GGA CUG GGC GAG A

OBJECTIVES: Intracerebral hemorrhage (ICH) is a type of stroke associated with high rates of disability and mortality. Mitochondrial dysfunction plays an important role in secondary brain injury following ICH, and preservation of mitochondrial function is considered a potential therapeutic strategy. Ubiquitin-specific protease 30 (USP30) is a deubiquitinase located on the outer mitochondrial membrane and plays an important role in regulating mitochondrial homeostasis. This study aims to investigate the role of USP30 after ICH and its potential molecular mechanisms.
METHODS: In vitro and in vi

The looming depletion of global phosphate rock reserves necessitates an urgent transition to phosphorus (P) recovery from secondary resources. Wet waste represents a massive yet largely untapped reservoir of P in China. However, efficient P extraction is constrained by the complex and heterogeneous speciation of P within these waste matrices. This review comprehensively examined P speciation in wet waste, emphasizing the predominance of inorganic fractions and the structural challenges posed by organic forms, particularly phytate and phosphate esters. Advanced analytical techniques were summar

Ischemic stroke creates a hypoxic and nutrient-deprived microenvironment that rapidly activates microglia, a central driver of post-ischemic inflammation and a key determinant of secondary tissue damage and neurological recovery. G-quadruplex (G4) structures are non-canonical nucleic acid conformations that can reshape stress responses in diverse settings, yet whether microglial DNA G4 relates to ischemic pathology remains unknown. Here, using a transient middle cerebral artery occlusion (tMCAO) mouse model and an oxygen-glucose deprivation/reoxygenation (OGD/R) primary microglia model, we exa

Lymphoplasmacytic gastritis (LPG) is one of the most prevalent chronic diseases affecting cheetahs (Acinonyx jubatus) under human care, yet its underlying cause remains unresolved. Gastric inflammation occurs in the majority of adult captive cheetahs but is uncommon in free-ranging populations, suggesting that management-related factors contribute to disease pathogenesis. This review proposes that LPG represents a bile acid-driven gastroenteropathy arising from disruption of the natural feeding ecology of the cheetah. In free-ranging systems, cheetahs consume large episodic meals separated by

INTRODUCTION: Essential tremor (ET) is one of the most prevalent yet mechanistically unresolved movement disorders. Although traditionally attributed to cerebellar dysfunction, accumulating evidence indicates the involvement of broader systemic processes, including neuroinflammation, immune dysregulation, and neurodegeneration. However, the role of inflammation in ET remains insufficiently defined, particularly regarding whether it constitutes a primary pathogenic mechanism or a secondary response.
MATERIAL AND METHODS: This systematic review aimed to comprehensively evaluate current evidence

This study explored the effectiveness of secondary metabolites of referred traditional Ayurvedic plants in treating fungal infections, particularly targeting Candida auris. Recognized as a global health threat, this fungus is notorious for its resistance to several antifungal treatments. The inhibition of lanosterol 14α-demethylase causes the depletion of ergosterol, ultimately resulting in the inhibition of fungal cell growth. A total of 469 metabolites, including alkaloids, flavonoids, and tannins from Ayurvedic plants, were screened against CYP51 (PDB ID: 4UYL) using molecular docking. Key

The traumatic penumbra (TP) is a secondary injury zone surrounding the core area of traumatic brain injury (TBI) and plays a critical role in determining TBI outcomes. The primary pathological change in the TP is brain edema, which includes both vasogenic and intracellular edema. Brain edema is closely associated with the expression of aquaporin-4 (AQP4). Bevacizumab, a vascular endothelial growth factor (VEGF) inhibitor, reduces vascular permeability. However, the effect of bevacizumab on traumatic brain edema remains unclear, as does its potential molecular mechanism in relation to AQP4. To
