Mechanical performance of hybrid polymer-lipid vesicles with leaflet asymmetry engineered using microfluidics.

Lipid vesicles consist of aqueous cores surrounded by a bilayer of phospholipids. Hybrid polymer-lipid vesicles incorporate both polymers and lipids, offering promising properties for developing pharmaceuticals, biosensors, and artificial cells. The hybrid vesicles can be symmetric, with two leaflets of identical compositions, or asymmetric, in with leaflets of dissimilar compositions, which can lead to dramatically altered properties. However, existing methods for producing symmetric and asymmetric hybrid vesicles often result in heterogenous compositions and sizes, making it challenging to q

Evaluation of MWCNTs as Nanoadditives in a Propanol-Biodiesel-Diesel Mixture on Compression Ignition Engine Characteristics.

Cleaner alternatives to internal combustion engines are essential for reducing pollution emissions while maintaining performance. In this study, a ternary blend of diesel, biodiesel, and propanol (80:10:10 by volume) was prepared and enriched with multiwalled carbon nanotubes (MWCNTs) at concentrations of 40, 60, and 80 ppm to evaluate its combustion, performance, and emission in a monocylinder diesel engine. Compared to diesel fuel, the ternary blends with MWCNTs demonstrated an increase of up to 6.73% in the net heat release rate and an average decrease of up to 14.74% in cylinder pressure.

Engineered Bacteria-Driven Biohybrid Microrobots Potentiate IL-2 Immunotherapy by Evoking Pyroptosis.

Interleukin-2 (IL-2) immunotherapy offers considerable potential for metastatic cancers; however, its efficacy is limited by low response rates, dose-dependent toxicity, short half-life, poor tumor accumulation, and off-target immune activation. In this work, we present a biohybrid microrobot (IL-2@Z/C/A) engineered through biomimetic mineralization of IL-2 variant-secreting Escherichia coli Nissle 1917 (EcN) with a zeolitic imidazolate framework-8 (ZIF-8) shell coloaded with an aggregation-induced emission photosensitizer and catalase. The ZIF-8 coating preserves bacterial viability tumor-hom

Engineered polymer micelles with tunable gas release for effective reduction of local fat deposits.

Demand is rising for non-surgical, site-specific fat reduction. This study presents fully organic biodegradable polymer micelles capable of intrinsic carbon dioxide (CO2) gas generation for adipocytolysis. Amphiphilic methoxy-poly(ethylene glycol) (mPEG) derivatives containing tri-carbonate groups (mPEG-T-OC) are synthesized to enhance CO2 release. These amphiphilic polymers form micellar structures and exhibit sustained CO2 release profiles under physiological conditions. To improve adipose tissue uptake, micelles are functionalized with nona‑arginine (r9), a cell-penetrating peptide that pro

DNA sensor-engineered nanovesicles for multi-logic-cascaded signal-amplified miRNA imaging.

The low abundance of tumor-related miRNA and significant background signal interference highlight the necessity for sensitive and accurate miRNA imaging. Herein, we report the DNA sensor-engineered nanovesicles that achieve sensitive and accurate imaging of target miRNA-21 by combining multi-logic-cascaded response strategy with catalytic hairpin assembly (CHA)-based signal amplification strategy. The DNA sensor was constructed by the glutathione (GSH)-responsive sequence, target recognition sequence, CHA hairpins and a DNA bridge sequence. Co-existence of GSH and target miRNA-21 could trigger

Single-Molecule Monitoring of Formaldehyde-Lysine Residue Chemistry using an Engineered MspA Nanopore.

Formaldehyde (HCHO) is a reactive electrophile that covalently modifies diverse amino acid residues in proteins. Although ensemble studies have characterized reactions between HCHO and amino acids, the reaction dynamics at individual residue sites have yet to be resolved at the single-molecule level. Here, we engineer a Mycobacterium smegmatis porin A (MspA) nanopore by positioning a single lysine residue at its constriction, enabling real-time single-molecule monitoring of HCHO-lysine chemistry. Distinct current blockade states and reversible transitions between them are observed, consistent

AI-engineered AAV capsid enables intravitreal delivery for the treatment of diverse retinal degenerations.

Retinal degenerations, including Leber's congenital amaurosis type 1 (LCA1) and wet age-related macular degeneration (wAMD), represent leading causes of vision impairment and blindness, driven by a range of factors, including genetic mutations and pathological neovascularization. While adeno-associated viruses (AAVs) have emerged as a promising platform for sustained gene therapy, most AAVs typically require subretinal injections to reach target cells, which carry risks to retinal integrity. To overcome this limitation, we employed an AI-guided approach to AAV capsid engineering and developed …

Microenvironment-engineered hydrogels drive osteo-angiogenic coupling to accelerate osteoporotic bone regeneration.

Osteoporotic bone defects repair remians significant clinical challenges, characterized by impaired osteogenic differentiation and dysregulated angiogenesis within a microenvironment of oxidative stress and chronic inflammation. Modulating this pathological microenvironment to create favorable conditions is a pivotal strategy for treating osteoporotic bone defects. In this study, we developed a multifunctional composite hydrogel (SMm) through photo-crosslinking, incorporating Metformin (Met) and magnesium oxide nanoparticles (MgO), to promote osteoporotic bone regeneration. The SMm hydrogel de

Interfacial Friction-Controlled Fiber Failure Modes for Toughness Enhancement of Engineered Cementitious Composites.

Despite extensive advancements in Engineered Cementitious Composites (ECCs), mixture design remains predominantly empirical, due to the absence of a quantitative parameter directly linking fiber-matrix interfacial mechanics to strain-hardening performance. This study identifies fiber-matrix interfacial friction as a quantifiable parameter and establishes a micromechanics-guided interfacial regulation framework to enhance the toughness of ECC by regulating fiber failure modes. First, a critical fiber-matrix interfacial frictional stress, (τ0)crit, corresponding to the transition between fiber p

Bioprinting of Microtissues Within Mechanically Tunable Support Baths to Engineer Anisotropic Musculoskeletal Tissues.

Bioprinting is a powerful tool for engineering living grafts, however replicating the composition, structure and function of native tissues remains a major challenge. During morphogenesis, cellular self-organization and matrix development are strongly influenced by the mechanical constraints provided by surrounding tissues, suggesting that such biophysical cues should be integrated into bioprinting strategies to engineer more biomimetic grafts. Here, we introduce a novel bioprinting platform that spatially patterns mesenchymal stem/stromal cell (MSC)-derived microtissues into mechanically tuna…

Spatial Confinement-Engineered Heteronuclear Diatomic Nanozymes for Modulation of Immune Homeostasis in Rheumatoid Arthritis.

The progression of rheumatoid arthritis (RA) is critically aggravated by synovial M1 macrophage-mediated inflammatory infiltration and excessive reactive oxygen species (ROS) accumulation. While emerging heteronuclear diatomic catalysts show potential in inflammatory modulation, their therapeutic efficacy remains constrained by insufficient control over interatomic synergy and multi-active sites regulatory precision. This work pioneers a spatial confinement strategy to engineer heteronuclear diatomic nanozymes (HDNs) with dual-pathway therapeutic capabilities for RA. We investigate Fe and Co s

Engineered CAR-monocytes coordinate fibrosis clearance and cardiac regeneration following myocardial infarction.

Overwhelming cardiomyocyte death and excessive cardiac fibrosis post myocardial infarction (MI) collectively lead to heart failure and mortality. For treating this devastating disease, it is essential to eliminate fibrosis and reconstitute the damaged myocardium, yet effective strategies remain elusive. Here, we created pleiotropic chimeric antigen receptor-monocytes (pCAR-Mos), revitalizing the injured heart via synergistic fibrosis clearance and myocardial reconstitution. Specifically, we engineered monocytes to express fibroblast activation protein (FAP)-chimeric antigen receptor (CAR) and

A synergistic strategy involving reverse-adaptation and engineered MSC exosomes against ferroptosis in osteoarthritis.

Osteoarthritis (OA) progression is driven by persistent oxidative stress and ferroptosis, which erode chondrocyte viability and extracellular matrix integrity. Although mesenchymal stem cell-derived exosomes (MSC-EXO) hold regenerative promise, their native cargo lacks the adaptability to withstand such a hostile microenvironment, limiting therapeutic efficacy. Here, we demonstrated a synergistic strategy involving reverse-adaptation and engineered MSC exosomes against ferroptosis in osteoarthritis. Firstly, the reverse-adaptation strategy in which OA-like oxidative stress was harnessed to pre

Enhancing exosomes efficacy with engineered nanozymes for a multi-targeted combination strategy in osteoarthritis treatment.

Osteoarthritis (OA) is a prevalent degenerative joint disease characterized by multifactorial pathological mechanisms, and remains a significant clinical challenge. Exosome therapy represents a future direction for delaying OA progression, yet its efficacy is often compromised by inflammatory microenvironment within the joints. To overcome these limitations, we present a novel combinatorial therapeutic platform that alleviates OA through a multi-targeted strategy, including the scavenging of reactive oxygen species (ROS), suppression of macrophage-driven inflammation, and inhibition of chondro

Quantifying the Viscoelastic Properties of Pancreatic Tissue: A Comparative Study of Human, Porcine and Engineered Hydrogel.

Pancreatoduodenectomy (PD) is among the most complex abdominal procedures, with a leak from the pancreatic anastomosis (postoperative pancreatic fistula, POPF) continuing to be a major source of morbidity and mortality. The mechanical characteristics of the pancreas, particularly tissue texture, are closely linked to POPF risk, yet remain poorly defined, limiting the accuracy of currently available training models. This study sought to quantify the viscoelastic behaviour of human pancreatic tissue and to compare it with porcine pancreas and a commercially available synthetic substitute. Fresh

Engineered Nestin+ TSPC-Derived Exosomes Promote Tendon Repair Via Metabolic Reprogramming.

Tendon repair remains a substantial clinical problem with limited treatment options. While Nestin+ tendon stem/progenitor cells (TSPCs) are known to regulate tendon healing, their clinical translation faces challenges. Here, we show that exosomes derived from engineered Nestin-overexpressing TSPCs (Nes-EVs) significantly ameliorate tendinopathy in multiple mouse models. We isolated Nes-EVs following lentiviral Nestin overexpression and demonstrated their capacity to reduce cellular senescence and promote tenogenic differentiation in vitro.
