T-cell receptors and autoimmune thyroid disease--signposts for T-cell-antigen driven diseases.

The human autoimmune thyroid diseases (AITDs) are characterized by profuse infiltrates of both CD4+ and CD8+ T cells. The intrathyroidal T-cell-receptor repertoire in Graves' disease, more than in Hashimoto's disease, has been shown to be biased as evidenced by phenotypic analysis and by the use of a restricted T-cell-receptor variable (V) gene repertoire seen in both TCR alpha and beta chains. Evidence for a bias in the T-cell repertoire has also been observed in animal models of induced and spontaneous autoimmune thyroiditis. We found a similar phenomenon of autoimmune thyroid-related T-cell

Regulated cell death in the temporomandibular joint: From disc internal derangement to osteoarthritis and tissue repair.

OBJECTIVE: This narrative review examines regulated cell death programmes underlying disc internal derangement and condylar osteoarthritis in the temporomandibular joint (TMJ), evaluating the balance between cell loss and endogenous repair capacity across both compartments. DESIGN: A narrative, mechanism-oriented review of PubMed-indexed literature, supplemented by citation tracking from core TMJ cell death studies, with evidence stratified by an explicit hierarchy prioritising human TMJ tissue and synovial fluid findings over animal, in vitro, and non-TMJ osteoarthritis data. RESULTS: In th

Marine-Derived bioactive compounds from Aquaculture: Receptor-Mediated neuroprotection in neurodegenerative disorders.

Neurodegenerative diseases, like Alzheimer's disease (AD), Parkinson's disease (PD), Huntington's disease (HD), amyotrophic lateral sclerosis (ALS), and ischemic stroke (IS), are a major global health challenge because of their complex, multifactorial pathology, and the lack of effective disease-modifying therapies. In recent years, aquaculture-derived marine bioactive molecules like fucoidan, phlorotannins, fucoxanthin, laminarin, alginate oligosaccharides, and C-phycocyanin have developed as promising agents for neuroprotection with their structural diversity and multi-target biological acti

Placental mRNA and miRNA dynamics associated with lipid metabolism pathways in pregnancies affected by obesity.

INTRODUCTION: Maternal obesity (pregravid body mass index >30 kg/m2), which has reached epidemic levels in the US, increases the incidence of cardiovascular disease and all cause premature death in the offspring. The placenta modulates fetal access to lipids and other nutrients and is considered a key player in fetal growth and maturation. However, the complex interplay between dysregulated metabolism in mothers with obesity and placental pathways mediating impacts on fetal development that predispose offspring to morbidities later in life, is poorly understood. METHODS: We used unbiased Whol

Environmental virotoxicology: how pollutant exposure modulates viral infections - a systematic review of wildlife-associated evidence.

Within a One Health framework, pollution is no longer a passive backdrop to host-virus interactions but actively reshapes disease dynamics across humans, animals, and ecosystems. The aim of this review was to synthesize evidence across free-ranging wildlife and wild-derived models to define environmental virotoxicology, which is the study of how environmental contaminants alter susceptibility, replication, shedding, transmission, pathogenesis, persistence, and viral evolution in hosts. A systematic search of PubMed, Scopus, and Web of Science (26 Oct 2025), augmented by citation-chasing, yield

Neonatal testosterone exposure modulates exploration and object avoidance without exacerbating autism-like behavior in Shank3b-deficient mice.

A rapid increase in autism spectrum disorder (ASD) prevalence, high heritability, and higher incidence in boys suggest a role of gene-environment interactions, likely involving sex hormones in its etiology. Prenatal exposure to high testosterone concentration has been linked to risk of ASD, however, experimental proof in genetically predisposed animals is lacking. Since neonatal development in mice mirrors late prenatal neurodevelopment in humans, we investigated the effects of neonatal testosterone administration on the behavior of female and male Shank3b(-/-) mice. On postnatal day 1, neonat

Yolk sac development in dogs: from morphology to functional aspects.

The yolk sac (YS) is a multifunctional and essential structure for early maternal-fetal interactions, playing roles in nutrient supply, organogenesis, immune modulation, and hematopoiesis. Unlike most species, dogs maintain the YS until the end of pregnancy, yet little is known about its functional activity, particularly in later stages. Animal models are crucial for exploring maternal-fetal processes and offering insights into comparative development, including in humans. This study aimed to analyze the YS structure in canine embryos and fetuses at early (up to 20 days, n = 6), middle (21-40

NO modulates human airway smooth muscle function by altering glucose-6-phosphate dehydrogenase effects on sGC function in asthma.

Since NO can modulate mesenchymal cell function, we posit that NO can modulate gene expression associated with excitation-contraction coupling. Our study shows that treating asthma-derived HASMCs with a low dose of NO plus sGC stimulator BAY-41, in most cases sensitized smooth muscle sGC towards activation via an elevated sGC heterodimer and in some cases also improved sGCβ1, catalase, Cyb5r3 or Trx1 expression (n=24 non-asthma and n=25 asthma). Interestingly we found that majority of asthma HASMCs showed a marked downregulation of G6PD expression inducing a low GSH/GSSG ratio in asthma, and t

Gestational and lactational exposure to glyphosate and a glyphosate-based herbicide causes sex-dependent effects on neurodevelopment and monoaminergic markers in rats.

Glyphosate (Glyph) exposure during the gestational and lactational periods have been linked to neurodevelopmental alterations in humans and animal models, and it has been proposed to induce autistic-like traits in rodents. This study evaluated the effects of gestational and lactational exposure to Glyph and a glyphosate-based herbicide (GBH) on neurodevelopmental and monoaminergic outcomes in male and female rat offspring, using valproic acid (VPA) as a pharmacological model of autism. Pregnant dams were orally treated with 50 mg Glyph/L in distilled water or with GBH throughout gestation and

Reverse vaccinology and immunoinformatics approaches driven designing of a novel multi-epitope mRNA vaccine against Toxoplasma gondii.

Toxoplasmosis is a zoonotic infectious disease caused by the intracellular apicomplexan parasite Toxoplasma gondii and primarily affects warm-blooded animals. Despite its global spread, the disease has limited therapeutic interventions. Chemotherapy can be used as a treatment, but there are potential side effects and contraindications. Despite several epitope-based in-silico vaccine studies, no mRNA-based vaccine has yet been explored. This study aims to design an mRNA vaccine using reverse vaccinology and immunoinformatics based on MIC1, MIC3, ROP29, and SAG1 proteins, which contribute to the

Integrated cortical and plasma proteomic analysis of Cntnap2 knockout mice and human models of autism spectrum disorder: Potential involvement of galectin-3-binding protein.

Despite decades of research, the etiology of autism spectrum disorder (ASD) remains largely uncomprehended, probably due to its clinical and phenotypic heterogeneity. Animal models, particularly contactin-associated protein-like 2 (Cntnap2) knockout (KO) mice, have been instrumental in elucidating ASD-related neurobiological mechanisms, as they exhibit ASD-like phenotypes, such as impaired social interactions in sociability paradigms. This provides the possibility for identifying detectable protein-based biomarkers that may assist in ASD diagnosis. Herein, we implemented an integrated approach

Gut Microbiota and Autism: Unlocking Connections.

Background: Autism Spectrum Disorder (ASD) is a multifactorial neurodevelopmental condition in which genetic predisposition interacts with environmental factors. Among these, the gut microbiota has emerged as a crucial modulator of the microbiota-gut-brain axis (MGBA), influencing neuroinflammation, neurotransmission, and behavior. This review aims to provide an updated and integrative overview of the relationship between gut microbiota, diet, and neurodevelopment in ASD. Methods: A comprehensive search was conducted in PubMed, Scopus, and Web of Science for articles published between 2010 and

A systematic review on distribution, sources and impacts of heavy metals in mangrove ecosystems.

Mangrove ecosystems play a vital role in providing habitat for numerous plant and animal species, protecting coastlines from erosion and storm damage, and supporting coastal communities with natural resources. Several studies have been conducted on heavy metals due to advancements in technology, which have improved the accuracy in detecting, identifying, and quantifying the metals. However, our understanding on the distribution, sources, and impacts of heavy metals on mangrove ecosystem health and productivity is currently scattered and unorganized. This review aims to bridge knowledge gaps, i

Replicon family of Vibrionaceae plasmids as a reservoir of antimicrobial and phage resistance genes in marine ecosystems.

Plasmids are mobile genetic elements that drive horizontal gene transfer among bacteria, influencing microbial community composition and functional traits in marine ecosystems. However, many marine plasmids remain unclassified due to unknown replication mechanisms. Here, we describe VBR1, a novel plasmid replicon family, widespread among species of the family Vibrionaceae. The minimal VBR1 replicon comprises a 570-bp AT-rich origin of replication (oriV) and two genes, vrp1AB, sufficient for autonomous replication in Escherichia coli and Photobacterium damselae. A comprehensive GenBank search r

Trimethylamine-N-oxide (TMAO) and risk of incident cardiovascular events in the multi ethnic study of Atherosclerosis.

Trimethylamine-N-oxide (TMAO) is a gut microbiome-derived metabolite of choline, L-carnitine and lecithin, abundant in animal source foods. In experimental models, higher blood TMAO levels enhance atherosclerotic cardiovascular disease (ASCVD). However in humans, most prior studies have evaluated high risk or secondary prevention populations, and no studies have investigated relationships in a diverse, multi-ethnic population. We evaluated 6,767 US adults free of ASCVD at baseline in the community-based Multi-Ethnic Study of Atherosclerosis (MESA), including 38% identifying as White; 28%, as B

α-Synuclein Pathology in Synucleinopathies: Mechanisms, Biomarkers, and Therapeutic Challenges.

Parkinson's disease and related synucleinopathies, including dementia with Lewy bodies and multiple system atrophy, are characterised by the pathological aggregation of the α-synuclein (aSyn) protein in neuronal and glial cells, leading to cellular dysfunction and neurodegeneration. This review synthesizes knowledge of aSyn biology, including its structure, aggregation mechanisms, cellular interactions, and systemic influences. We highlight the structural diversity of aSyn aggregates, ranging from oligomers to fibrils, their strain-like properties, and their prion-like propagation. While the r
