Role of Transcription Factors in Steatohepatitis and Hypertension after Ethanol: The Epicenter of Metabolism.

Chronic alcohol consumption induces multi-organ damage, including alcoholic liver disease (ALD), pancreatitis and hypertension. Ethanol and ethanol metabolic products play a significant role in the manifestation of its toxicity. Ethanol metabolizes to acetaldehyde and produces reduced nicotinamide adenine dinucleotide (NADH) by cytosolic alcohol dehydrogenase. Ethanol metabolism mediated by cytochrome-P450 2E1 causes oxidative stress due to increased production of reactive oxygen species (ROS). Acetaldehyde, increased redox cellular state and ROS activate transcription factors, which in turn a

Detailed esophageal function and morphological analysis shows high prevalence of gastroesophageal reflux disease and Barrett's esophagus in patients with cervical inlet patch.

Although the pathogenesis of cervical inlet patch (CIP) is not fully understood, most authors consider it as a congenital abnormality, whereas others surmise it to be related to gastroesophageal reflux disease (GERD). We aimed to evaluate esophageal function and the prevalence of GERD and Barrett's esophagus in patients with CIP. GERD is defined by the presence of erosive esophagitis or an abnormal pH monitoring. Seventy-one consecutive patients with endoscopic and histological evidence of CIP were prospectively evaluated. Esophageal symptom analysis, 24-hour simultaneous biliary reflux and do

Etiopathogenesis of atopic dermatitis--an overview.

Atopic eczema/dermatitis syndrome is a term that covers different subtypes of atopic dermatitis. The "intrinsic" type of atopic dermatitis is non-IgE-associated, and the "extrinsic" type is IgE-associated atopic eczema/dermatitis syndrome. In the etiopathogenesis of atopic dermatitis there are well known interactions among genetic, environmental, skin barrier, immune factors, and stress. Genetic factors determine the expression of atopic dermatitis as pure or mixed with concomitant respiratory or intestinal allergy, depending on genetic susceptibility. Immunologic abnormalities of type I and t

New advances in human embryology: morphofunctional relationship between the embryo and the yolk sac.

Organogenesis occurs during the first 8 weeks of human embryonic development; in consequence, early human growth and development take place before and in the absence of fully developed internal organs. During this period, normal development depends on several factors, but two are imperative: nutrition and a functional transport system for the distribution of nutrients and for waste disposal. The yolk sac (YS), a highly differentiated adnexal organ, is known to accomplish this fundamental task during early pregnancy. In this review, we summarize our contribution to the understanding of early hu

Effects of adrenomedullin on cyclic AMP formation and on relaxation in iris sphincter smooth muscle.

PURPOSE: To determine whether iris sphincter and other tissues of the iris-ciliary body secrete adrenomedullin (ADM), a novel hypotensive peptide that is classified into the calcitonin gene-related peptide (CGRP) family and to determine the binding sites for ADM and compare the effects of ADM and CGRP in the absence and presence of their receptor antagonists on cAMP formation and relaxation in the iris sphincter. METHODS: Sphincter muscle was incubated in Krebs-Ringer bicarbonate buffer in the absence and presence of ADM for 10 minutes. Accumulation of cAMP in the tissue extract was determine

Corticotropin-releasing factor receptors: physiology, pharmacology, biochemistry and role in central nervous system and immune disorders.

Corticotropin-releasing factor (CRF) plays a major role in coordinating the endocrine, autonomic, behavioral and immune responses to stress through actions in the brain and the periphery. CRF receptors identified in brain, pituitary and spleen have comparable kinetic and pharmacological characteristics, guanine nucleotide sensitivity and adenylate cyclase-stimulating activity. Differences were observed in the molecular mass of the CRF receptor complex between the brain (58,000 Da) and the pituitary and spleen (75,000 Da), which appeared to be due to differential glycosylation of the receptor p

Escherichia coli cytotoxins and enterotoxins.

Vero cell cytotoxins and cytotonic enterotoxins produced by E. coli are toxic proteins, which have been implicated in a number of specific diseases in humans and animals. Nomenclature for these toxins is complicated by the existence of different names for the same toxin. The Vero cell cytotoxins are called verotoxins because they are lethal for Vero cells in culture; they are also known as Shiga-like toxins (SLTs) because they are clearly related to Shiga toxin in structure, amino acid sequence, mechanism of action, and biological activity. SLTs belong to two classes. SLT-I is identical with S

Dyslipoproteinaemia of liver disease.

Hepatic diseases differ from most other causes of secondary dyslipidaemia in that the circulating lipoproteins are not only present in abnormal amounts but they frequently also have abnormal composition, electrophoretic mobility and appearance. Pre-beta and alpha bands can be absent on electrophoresis in all types of liver disease although material in the VLDL and HDL ranges can be isolated in the ultracentrifuge. Cholestatic liver disease has been the most extensively studied and the hyperlipidaemia can be extreme with marked elevations of free cholesterol and phospholipids. This results larg

DNA methylation inhibitor decitabine ameliorates spondyloarthritis by suppressing Th17 responses and epigenetically upregulating VIPR1 in SKG mice.

BACKGROUND: Spondyloarthritis (SpA) is a chronic inflammatory disorder characterized by enthesitis of axial and peripheral joints. Although genetic factors, bacterial infection, dysbiosis, and mechanical stress contribute to its development, the role of epigenetic regulation remains poorly understood. DNA methylation has been implicated in immune dysregulation, and the DNA methyltransferase inhibitor, decitabine (DAC), has demonstrated therapeutic effects in autoimmune disease models. This study aimed to evaluate the effect of DAC in the SpA-resembling SKG/Jcl (SKG) mouse model and elucidate t

Multi-kingdom gut microbiota analyses identify biomarkers of different types of pediatric short bowel syndrome.

Despite recent advances in bacterial profiling across the three anatomical types of pediatric short bowel syndrome (SBS), the gut multi-kingdom remains unexplored. We characterized the four-kingdom gut microbiota using modified DNA extraction and deep shotgun metagenomic sequencing of fecal samples from 26 healthy controls and 34 pediatric SBS patients comprising three types (8 SBS I, 15 SBS II, and 11 SBS III). Overall, children with SBS exhibited a significant reduction in α-diversity compared with controls, with no difference observed among SBS types. Compared with controls, the proportion

Extracellular vesicle-mediated immune-metabolic crosstalk: research advances in inflammatory regulation and the pathogenesis of metabolic diseases.

Extracellular vesicles (EVs)-lipid bilayer-enclosed nanoparticles secreted by virtually all cell types-have shifted from being viewed as passive cellular byproducts to active signaling nodes orchestrating inter-cellular and inter-organ communication. Yet the field has accumulated faster than it has integrated: hundreds of EV-cargo-phenotype associations exist as isolated edges of a network whose system-level architecture remains poorly defined. Here we propose the vesiculome as an operational framework for that network, resting on three falsifiable axioms: (i) the EV complement of an organism

Serum physiology, fecal microbiota, and metabolome signatures associated with perinatal stage transitions in Mongolian mares.

INTRODUCTION: Lactation is a critical reproductive trait that influences offspring survival and livestock productivity in equine production. Characterizing physiological transitions during the perinatal period can improve our understanding of how mares adapt to late gestation, parturition, and early lactation. However, in horses-a typical monogastric herbivore with unique digestive characteristics-the associations between gut microbiota and perinatal physiological adaptation, as well as related stage-associated microbial and metabolic features, remain largely uncharacterized. METHODS: In this

Non-absorbable antibiotics worsen alcohol-associated liver disease in gastric acid-suppressed mice.

Gastric acid-suppressive medications, particularly proton pump inhibitors (PPIs), are commonly used in patients with alcohol-associated liver disease (ALD) to prevent and manage upper gastrointestinal bleeding, gastroesophageal reflux disease, and non-steroidal anti-inflammatory/aspirin-induced gastroesophageal damage. By inhibiting the gastric H⁺/K⁺-ATPase, PPIs suppress acid secretion and impair bacterial killing, thereby promoting gut dysbiosis that disrupts barrier integrity and enhances bacterial translocation, ultimately exacerbating liver injury. PPIs are frequently co-administered with

Mycobiome dysbiosis and genetic predisposition to elevated IL-17A contribute to fibrosis in MASLD.

BACKGROUND & AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD) is the leading cause of chronic liver disease in Western countries. Progression to metabolic dysfunction-associated steatohepatitis (MASH) occurs when fat accumulation in the liver triggers inflammatory processes including T helper 17 cell (Th17) activation. We aimed to investigate the role of intestinal fungi in MASH-mediating Th17-associated signaling. METHODS: Blood samples from patients with MASLD (n = 451), including 141 with histology-proven MASH, were genotyped for IL17A rs2275913. Microbiome compositio

A novel Vibrio parahaemolyticus strain JM20210818 causing glass post-larvae disease in Litopenaeus vannamei: Histopathology, antimicrobial resistance, genomic and transcriptomic insights.

A new disease commonly referred to as "glass post-larvae disease" (GPD) has emerged in many shrimp hatcheries in China since 2020. This highly infectious and lethal disease primarily affects 6 to 12-day-old post-larvae of Litopenaeus vannamei (L. vannamei). In hatchery environments, GPD spreads rapidly, often leading to a large-scale outbreak within 2-3 days from infection and can cause mortality rates as high as 90-100 %. The clinical signs of diseased post-larvae shrimp include low vitality, feeding cessation, empty digestive tract, white or colorless hepatopancreas and a translucent appeara

Engineered Bacillus subtilis WB600/ZD reduces post-weaning diarrhea in piglets by modulating gut microbiota and aryl hydrocarbon receptor (AHR) signaling.

Post-weaning diarrhea (PWD) causes significant economic losses to the pig industry. A previous study demonstrated that engineered Bacillus subtilis WB600 expressing Zophobas atratus defensin (ZD), termed WB600/ZD, alleviates intestinal inflammation, modulates gut microbiota, and maintains redox homeostasis in Salmonella-challenged mice; however, the precise mechanisms remain unclear. In this study, a total of 50 weaned Landrace × Large White piglets at 21 d of age were assigned to four groups: healthy piglets fed standard diet (H group; 6.60 ± 0.48 kg, n = 15) or 2 × 109 CFU/mL WB600/ZD (H + W
