[Novel approach for management of age-related macular degeneration--antiangiogenic therapy and retinal regenerative therapy].

Age-related macular degeneration (AMD) is a leading cause of legal blindness in developed countries. Even with the recent advent of several treatment options, treatment of exudative AMD, characterized by choroidal neovascularization (CNV), remains difficult. Thus, in this review article, we report on the investigation of novel approaches for the management of AMD, antiangiogenic therapy for CNV, and retinal regenerative therapy. Polyion complex(PIC) micelles have a range in size of several tens of nanometers formed through an electrostatic interaction, and accumulate in solid tumors through an

Promotion of PDT efficacy by a Bcl-2 antagonist.

Photodynamic therapy (PDT) directed against the endoplasmic reticulum (ER) is also known to target antiapoptotic Bcl-2 family proteins. This effect is associated with the initiation of both apoptosis, a cell death pathway, and autophagy, an organelle recycling system that can lead to survival or cell death. In this study, we examined the ability of the Bcl-2 antagonist HA14-1 to promote the photodynamic efficacy of PDT directed at the ER. At concentrations that independently caused only a small loss of viability, HA14-1 markedly enhanced the proapoptotic and phototoxic effects of ER photodamag

A non-laser light source for photodynamic therapy: in vitro effects on normal and malignant cells.

AIM: Photodynamic therapy (PDT) involves the use of photosensitizing drugs combined with light to treat tumors. Laser systems, the current source of light for PDT, have several inherent drawbacks: the spectrum is essentially monochromatic which may be problematic for second generation photosensitizers, the systems are bulky and nearly impossible to move between hospital locations and require complicated electrical and cooling installations, the cost of a typical system is enormous, and its maintenance and operation require highly trained personnel. We now introduce a new non-laser light system

Photodynamic therapy with violet light and topical 6-aminolaevulinic acid in the treatment of actinic keratosis, Bowen's disease and basal cell carcinoma.

BACKGROUND: Most clinical studies using photodynamic therapy (PDT) with topical application of delta-aminolaevulinic acid (delta-ALA) use red light because it allows greater depth of penetration. However, given the porphyrin-like spectrum of delta-ALA-induced photosensitivity, violet light provides a maximal overlap with the excitation spectrum of protoporphyrin IX, meaning that PDT with violet light uses less light energy to induce the phototoxic reaction. AIM: To study the efficacy of violet light in combination with topical delta-ALA PDT in the treatment of pre-malignant and malignant skin

Potential efficacy of a delta 5-aminolevulinic acid bioadhesive gel formulation for the photodynamic treatment of lesions of the gastrointestinal tract in mice.

A delta 5-aminolevulinic acid (ALA) bioadhesive gel has been developed and evaluated in an in-vivo mouse model for photodynamic treatment of gastric cancer or Barrett's oesophagus. Four gels were tested: noveon AA-1, keltrol T, lutrol and blanose. An initial in-vitro study of gel adhesion showed that noveon and keltrol had longer polyethylene transit times than lutrol and blanose. In-vivo assays indicated that protoporphyrin IX was synthesized by gastric mucosa when ALA-noveon and ALA-lutrol were used (preferable results for noveon). Keltrol was eliminated from the study after these investigat

Repetitive photodynamic therapy with topical delta-aminolaevulinic acid as an appropriate approach to the routine treatment of superficial non-melanoma skin tumours.

Photodynamic therapy (PDT) with topical delta-aminolaevulinic acid (ALA) is considered as a valuable tool for treating non-melanoma skin tumours, but there is no consensus about the methods of treatment, where they should be used and the rates of complete responses. This study reports the treatment of 50 actinic keratoses (AKs), 23 superficial basal cell carcinomas (BCCs), 30 nodular BCCs, 4 pigmented BCCs, 12 superficial squamous cell carcinomas (SCCs), 6 nodular SCCs, 6 Bowen's diseases and 4 keratoacanthomas (KAs). A 20% ALA emulsion was applied under an occlusive dressing for 6-8 h and the

The effect of photodynamic therapy on a retinoblastoma-like tumour. An experimental in vitro and in vivo study on the potential use of photodynamic therapy in the treatment of retinoblastoma.

A retinoblastoma-like tumour has been established and characterized in terms of growth rates in vitro and in vivo, and by histopathology and chromosome analysis. Injected tumour cells grew regularly in the vitreous body with a blood supply from the retinal vessels. The tumour tissue was histopathologically similar to that of anaplastic human retinoblastomas. Almost all the cells had a triploid chromosome number and the DNA amount in tumours was stable, suggesting a stable tumour system without drift against more anaplastic degrees. Tumour cells plated in culture flasks were grown in colonies.

CAIX-targeted red-light-activated osmium(II) polypyridyl photosensitizer inducing pyroptosis and ferroptosis to combat hypoxia tumors.

To overcome the limitations imposed by hypoxia in photodynamic therapy (PDT), we rationally designed and synthesized three carbonic anhydrase IX (CAIX)-targeted osmium(II) polypyridyl complexes (Os1-Os3) by conjugating a benzenesulfonamide CAIX inhibitor to the photosensitizer core. Among them, Os3 exhibited excellent photophysical properties and efficient singlet oxygen generation. This complex showed remarkable selectivity for CAIX-overexpressing MDA-MB-231 cancer cells, with very low dark toxicity (IC50 > 100 μM) and outstanding phototoxicity (IC50 = 2.2 μM, phototoxicity index = 45) upon r

Endogenous-Activatable Nanotheranostic Platform for Multimodal Bioimaging-Guided Photodynamic and Gas Synergistic Cancer Therapy.

Based on gold nanoclusters (AuNCs), l-Arginine (l-Arg), dendritic macroporous silica nanoparticles (DSiO2), MnO2 nanosheets, and Sgc8 aptamers, a multifunctional nanotheranostic platform (DALMS) was developed for endogenous-activatable multimodal bioimaging-guided synergistic targeted cancer therapy. The DALMS precisely targets cancer cells via Sgc8 and responds to the high concentration GSH in the tumor microenvironment (TME). The subsequent release of AuNCs and l-Arg enables endogenous-activatable, controlled drug delivery. In tumor tissues and cells, DALMS exhibits efficient two-photon near

Dual-masked cathepsin B-activated 5-aminolevulinic acid prodrugs with enhanced photodynamic cytotoxicity.

5-Aminolevulinic acid-mediated photodynamic therapy (ALA-PDT) exploits tumor-selective accumulation of protoporphyrin IX (PpIX). However, its clinical utility is constrained by poor physicochemical properties and suboptimal tumor selectivity. To address these limitations, we rationally designed and synthesized 17 N-terminal dipeptide-modified 5-ALA prodrugs using a dual-masked strategy. This approach integrates lipophilic shielding at the carboxyl terminus with a cathepsin B (Cath B)-cleavable dipeptide at the amino terminus, enabling tumor-specific release of active 5-ALA. Structure-activity

DNA Nanoflowers Efficiently Encapsulate Photodynamic Agents and CRISPR/Cas9 for Synergistic Pancreatic Cancer Therapy.

Photodynamic therapy (PDT) holds significant promise for treating pancreatic cancer by utilizing photosensitizers to generate reactive oxygen species (ROS) that induce tumor cell death. However, the therapeutic efficacy of PDT is hindered by inadequate ROS accumulation. Herein, we develop a DNA nanoflower that enables the controlled codelivery of Cas9 ribonucleoprotein (RNP), hemin, and chlorin e6 for synergistic PDT. The Cas9 RNP selectively knocks out the antioxidant regulator nuclear factor E2-related factor 2 (Nrf2), thereby increasing cancer cells' sensitivity to ROS. Simultaneously, the
