Chronic stress-induced gut dysfunction exacerbates Parkinson's disease phenotype and pathology in a rotenone-induced mouse model of Parkinson's disease.

Recent evidence provides support for involvement of the microbiota-gut-brain axis in Parkinson's disease (PD) pathogenesis. We propose that a pro-inflammatory intestinal milieu, due to intestinal hyper-permeability and/or microbial dysbiosis, initiates or exacerbates PD pathogenesis. One factor that can cause intestinal hyper-permeability and dysbiosis is chronic stress which has been shown to accelerate neuronal degeneration and motor deficits in Parkinsonism rodent models. We hypothesized that stress-induced intestinal barrier dysfunction and microbial dysbiosis lead to a pro-inflammatory mi

Association of Autonomic Dysfunction With Disease Progression and Survival in Parkinson Disease.

IMPORTANCE: Evidence suggests that development of autonomic dysfunction (AutD) may negatively affect disease course and survival in patients with synucleinopathies. However, the few available studies on Parkinson disease (PD) have conflicting results, comprise a small number of patients, have short follow-up periods, and lack pathologic confirmation of the diagnosis. OBJECTIVE: To examine the association of time of onset of AutD with disease progression and survival in PD. DESIGN, SETTING, AND PARTICIPANTS: This retrospective review of clinical data from 100 consecutive patients with an auto

A critical reappraisal of current staging of Lewy-related pathology in human brain.

Sporadic Parkinson disease (sPD) or brainstem-predominant type of Lewy body (LB) disease, and dementia with Lewy bodies (DLB), the two most frequent alpha-synucleinopathies, are progressive multisystem neurodegenerative disorders with widespread occurrence of alpha-synuclein (AS) deposits in the central, peripheral, and autonomic nervous system. For both LB-related disorders, staging/classification systems based on semiquantitative assessment of the distribution and progression pattern of Lewy-related/AS pathology are used that are considered to be linked to clinical dysfunctions. In PD, a six

How cytochrome P450 enzymes in humans are involved in Parkinson's disease: a literature review.

This review synthesizes three decades of evidence regarding the role of cytochrome P450 enzymes (CYPs) in Parkinson's disease (PD), revealing their multifaceted roles beyond traditional pesticide metabolism. While CYP2D6 remains the most studied enzyme due to its association with PD risk in poor metabolizer phenotypes and its dual role in dopamine (DA) synthesis (directly via tyramine hydroxylation and indirectly through precursor demethylation), recent research has highlighted less-studied CYPs with critical pathological implications. Another focal enzyme, CYP2E1, mediates the bioactivation o

Postmortem Associations Between Alzheimer Disease Pathology and Plasma pTau217, GFAP, and NfL in AD and AD-Related Dementias.

BACKGROUND AND OBJECTIVES: Alzheimer disease (AD) and its related disorders (ADRDs) are characterized by a high frequency of copathologies. We aimed to determine the specificity of plasma pTau217, glial fibrillary acidic protein (GFAP), and neurofilament light chain (NfL) for AD neuropathological change (ADNC) in the presence of common ADRD copathologies. METHODS: pTau217, GFAP, and NfL were measured using S-PLEX immunoassays from Meso Scale Discovery in banked plasma samples from 2 groups of participants in the Massachusetts Alzheimer's Disease Research Center (MADRC) Longitudinal Cohort stu

Region-specific variations in the cerebrovasculature underlie disease progression in Parkinson's disease.

Parkinson's disease is a progressive neurodegenerative disorder characterized by motor dysfunction, dopaminergic neuronal loss in the substantia nigra and abnormal accumulation of α-synuclein Lewy bodies. Research suggests that the cerebrovascular system plays a role in fluid dynamics, waste clearance and removal of abnormal proteins. Imaging studies show that this waste clearance system, known as the glymphatic system, is disrupted in Parkinson's disease, highlighting its involvement in the disease. This immunohistochemical human brain tissue study quantified changes in the cerebrovascular sy

Vicarious defeat stress (VDS) is not a simple substitute for chronic social defeat stress (CSDS): a comparative study on behavioral and molecular mechanisms in both stress-induced anxiety- and depression-like behaviors.

Social stress is a fundamental environmental factor driving the pathophysiology of neuropsychiatric disorders. Chronic social defeat stress (CSDS) and vicarious defeat stress (VDS) are two widely used social stress paradigms, with VDS initially proposed as an alternative to CSDS. The present study integrated behavioral phenotypes, hippocampal proteomics, synaptic morphology, energy metabolism, neurotransmitter and tryptophan-kynurenine (TRP-KYN) metabolism, and inflammatory indicators to directly compare responses in adult male mice subjected to CSDS and VDS. Both CSDS and VDS mice showed sign

Autopsy findings in Alzheimer's disease clinical trial participants demonstrate a high frequency of off-target coexisting pathologic features.

BACKGROUND: Having multiple comorbid neuropathologic features may confound the results of interventional trials that were designed to target a specific pathophysiologic mechanism in Alzheimer's disease (AD) and or related dementias (ADRD). However, it is unknown what percentage of individuals undergoing AD/ADRD interventional studies have mixed pathologies. OBJECTIVE: To characterize the spectrum of coexisting neuropathologies in brains of AD/ADRD clinical trial participants to inform trial design and therapeutic strategies METHODS: Autopsied participants from the University of Kentucky Alzhe

Astragenol alleviates neuroinflammation and improves Parkinson's symptoms through amino acid metabolism pathway and inhibition of ferroptosis.

ETHNOPHARMACOLOGICAL RELEVANCE: Astragalus membranaceus (Fisch.) Bunge, as a tonifying medicine, is widely used in the traditional Chinese medicine treatment of Parkinson's disease (PD). We found Astragenol (AST) in the acid hydrolysis product of Astragalus membranaceus, which has anti-inflammatory and neuroprotective effects. However, the pharmacological effects and molecular mechanisms of AST in MPTP induced PD models have not been elucidated. AIM OF THE STUDY: This study aims to verify whether AST can improve the symptoms of MPTP induced PD model and elucidate the molecular mechanisms unde

Relationships among cortical activation, cognition, and blood biomarkers in two types of dementia determined using functional near-infrared spectroscopy.

OBJECTIVE: The most prevalent types of dementia in older adults are Alzheimer's disease (AD) and Lewy body dementia (LBD), and they have overlapping clinical symptoms. We aimed to define amounts of cortical activation and to identify indicators of brain function to better distinguish between these types of dementia and aid diagnosis using functional near-infrared spectroscopy (fNIRS). METHODS: Oxygenated hemoglobin (HbO) concentrations in the brains of patients with AD and LBD were detected using fNIRS. Brain function was assessed using a verbal fluency task (VFT). Chronic sleep deprivation induces plasma exosome-derived miR-150-5p downregulation as a novel mechanism involved in Parkinson's disease progression by targeting DCLK1.

BACKGROUND: Researches have suggested that chronic sleep deprivation (SD) can lead to neurological dysfunction and facilitate the onset and progression of Parkinson's disease (PD). However, the association between SD and PD remains unclear. Exosome (exo) cargo comprises microRNAs (miRNAs), which are potential regulators of PD. This study focused on assessing the role and related mechanisms of SD on PD. METHODS: SD plus 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced PD mice were used to investigate effects of SD on PD. Exos were extracted from plasma by polymer precipitation metho

Decoding Parkinson's Disease: The interplay of cell death pathways, oxidative stress, and therapeutic innovations.

Parkinson's disease (PD), a complex neurodegenerative disorder characterized by selective loss of substantia nigra (SN) dopaminergic neurons, pathological aggregation of α-synuclein (α-syn), and chronic neuroinflammation, is fundamentally driven by redox imbalance and oxidative stress. Recent studies reveal that a dynamic interplay of programmed and non-programmed cell death mechanisms-amplified by oxidative damage-drives PD progression. Programmed cell death pathways include apoptosis (caspase-dependent mitochondrial/extrinsic pathways), necroptosis (eceptor-interacting serine/threonine-prote

Restoration of Lysosomal Hydrolase Activities by LRRK2 Inhibition in GBA1- and LRRK2-Associated Parkinson's Disease Patient-Derived Cells.

Mutations in the GBA1 gene, encoding the lysosomal enzyme glucocerebrosidase (GCase), and the LRRK2 gene, encoding leucine-rich repeat kinase 2 (LRRK2) are the most common genetic risk factors for Parkinson's disease (PD). The potential use of LRRK2 inhibitors for treating not only LRRK2-associated PD (LRRK2-PD) but also GBA1-associated PD (GBA1-PD) is currently under discussion. In the present study, we aimed to evaluate whether LRRK2 inhibition affects lysosomal hydrolase enzymatic activities, autophagy, and alpha-synuclein levels in various cell types derived from LRRK2-PD and GBA1-PD patie

Early proteasome downregulation and dysfunction drive proteostasis failure in Alzheimer's disease.

Alzheimer's disease (AD) is characterized by the accumulation of pathogenic proteins, notably amyloid-beta and hyperphosphorylated tau, which disrupt neuronal function and contribute to cognitive decline. Although proteotoxic stress is well established in AD, the role of the ubiquitin-proteasome system in maintaining neuronal proteostasis and how it becomes compromised during disease progression remain incompletely understood. Here, we integrated multiple approaches to characterize proteasome function, composition and regulation in post-mortem human AD brain tissue compared with age-matched co

Neuroprotective activity of mung bean (Vigna radiata) coat extract via AMPK-dependent autophagy in Alzheimer's and Parkinson's models.

OBJECTIVE: Alzheimer's disease (AD) and Parkinson's disease (PD) are major age-related neurodegenerative disorders that currently lack effective disease-modifying therapies. This study investigated the neuroprotective potential and underlying mechanisms of natural products derived from medicine-food homology (MFH) plants, with a focus on autophagy modulation in AD and PD models. METHODS: Twenty MFH plant extracts were screened using the Caenorhabditis elegans amyloid-β peptide (Aβ) proteotoxicity model CL4176. Mung bean coat extract (MBCE) was identified as a promising candidate and subsequen

Curcumin-enhanced stem cell exosomes: A novel approach to modulating neuroinflammation and improving cognitive function in a rat model of Alzheimer's disease.

The effect of Curcumin-enhanced stem cell exosomes on the learning and memory impairment induced by streptozotocin (STZ) and neuro-inflammation in rats was evaluated. An animal model of Alzheimer's disease (AD) was established by intracerebroventricular (ICV) injection of STZ (3 mg/kg) in male Wistar rats (250 ± 50 g). ICV STZ injections chronically reduce cerebral glucose uptake and produce other effects similar to pathological, molecular and behavioral features of AD. Numerous studies confirmed the anti-inflammatory and antioxidant properties of curcumin (a natural polyphenol) against free r

Neuropathologic correlates of cognitive impairment in Alzheimer's disease with discordant CSF biomarker profiles: co-pathologies in focus.

CSF Aβ reflects Alzheimer's disease neuropathologic change (ADNC), while CSF p-tau offers an indirect indication of tangle pathology. However, interpretation can be challenging when cognitive impairment is present alongside Aβ positivity (Α +) but p-tau negativity (T -). We examined neuropathologic differences between CSF A + T - and A + T + profiles, defined by CSF Aβ42 and p-tau181 levels, hypothesizing that cognitively impaired older adults with a CSF A + T - profile would exhibit a greater co-pathology burden, suggesting alternative contributing disease processes. We identified 77 ADNI par

Acetylcholinesterase: A Versatile Template to Coin Potent Modulators of Multiple Therapeutic Targets.

ConspectusThe enzyme acetylcholinesterase (AChE) hydrolyzes the neurotransmitter acetylcholine (ACh) at cholinergic synapses of the peripheral and central nervous system. Thus, it is a prime therapeutic target for diseases that occur with a cholinergic deficit, prominently Alzheimer's disease (AD). Working at a rate near the diffusion limit, it is considered one of nature's most efficient enzymes. This is particularly meritorious considering that its catalytic site is buried at the bottom of a 20-Å-deep cavity, which is preceded by a bottleneck with a diameter shorter than that of the trimethy
