Nanosensitizer for Cancer Radioimmunotherapy via Anti-IL-35 Blockade Boosted Innate Immunity Activation.

The cGAS-STING signaling pathway has emerged as a promising target for cancer immunotherapy. However, STING agonists have a dual-edged nature. Although they enhance antitumor T cell activity, STING agonists also elicit protumorigenic effects by promoting IL-35-producing B regulatory (Breg) cells, suppressing natural killer (NK) cell density, and fostering immune suppression. To address these challenges, this work develops a tumor microenvironment-responsive hollow mesoporous nanosystem that degrades under high glutathione conditions, thereby releasing the STING agonist MSA-2 and Mn ions. This …

Comprehensive proteomic analysis and multidimensional model construction of peritoneal metastasis in gastric cancer.

Peritoneal metastasis following gastric cancer surgery is often associated with a poor prognosis. This study aimed to investigate the mechanisms underlying peritoneal metastasis and to develop a predictive model for the risk of postoperative peritoneal metastases in gastric cancer. We performed a comprehensive analysis of the protein mass spectra and tumor microenvironment in paraffin-embedded primary tumor sections from gastric cancer patients, both with and without postoperative peritoneal metastases. Using proteomic profiling, we identified 9595 proteins and stratified patients into three d…

Flow cytometry immune profiling of recurrent and newly diagnosed growth hormone secreting pituitary neuroendocrine tumors: comparison of two clinical cases.

Early detection of aggressive pituitary neuroendocrine tumors (PitNETs) remains challenging due to the absence of reliable markers that can predict disease progression. Aggressive tumors are typically identified through long-term observation. Tumor immune microenvironment (TIME) is crucial for understanding PitNETs' heterogeneity and identifying potential predictors of tumor aggressiveness. In this study, we analyzed immune profile of micro- and macroenvironment in two somatotropinomas (aggressive and non-aggressive) using flow cytometry. We observed lymphopenia and elevated neutrophil levels

CD73: a new immune checkpoint for leukemia treatment.

Recent studies on the pathogenesis of leukemia have led to remarkable advances in disease treatment. Numerous studies have shown the potential and viability of immune responses against leukemia. In the classical pathway, this process is often initiated by the upstream activity of CD39, which hydrolyzes extracellular adenosine triphosphate (ATP) and adenosine diphosphate (ADP) to AMP. Subsequently, CD73 acts on AMP to generate adenosine, contributing to an immunosuppressive microenvironment. However, CD73 can also utilize substrates derived from other molecules through the non-canonical NAD+ pa

Grapefruit Extracellular-Vesicle-Derived Nanodrug Loading Three-Functional Platinum(IV) Conjugates with Enhanced Targeting and TME Modulating Properties.

Grapefruit extracellular vesicle (GEVs) derived nanodrug Tf-GEVs@Pt(IV) loading novel three-functional platinum(IV) conjugates was developed, which was effective in modulating the inflammatory, fibrotic, and immunosuppressive tumor microenvironment (TME). The GEVs enhanced the penetration of Tf-GEVs@Pt(IV) into tumor tissues, while the transferrin (Tf) moiety further improved the tumor-targeting capabilities. The active ingredient, platinum(IV) conjugate, was released in a sustained manner within the TME, which was easily reduced to platinum(II), aspirin (ASP), and captopril (CTP) fragments. T…

Tumor Cell-Derived CXCL2 Potentiates Neutrophil-Mediated Antitumor Immunity by Inhibiting Cholesterol Biosynthesis in Hepatocellular Carcinoma.

Hepatocellular carcinoma (HCC) is a major cause of cancer-related death worldwide. Despite the proven efficacy of immunotherapy against malignancies, a large proportion of patients with HCC fail to benefit from these efficacious agents because of their overwhelmingly immunosuppressive microenvironment. Therefore, there is an urgent need to identify key genes and develop effective strategies for reshaping the HCC microenvironment. Here, a significant downregulation of C-X-C motif chemokine ligand 2 (CXCL2) in HCC is identified due to gene copy number loss, which correlates with poor prognosis a

Deletion of FAM76B histidine-rich region enhances macrophage-mediated osteosarcoma inhibition via liquid-liquid phase separation.

BACKGROUND: Osteosarcoma (OS), a prevalent malignancy primarily in young individuals, often evades T cell based immunotherapy due to an immunosuppressive myeloid cell-mediated microenvironment. This study investigated the role of FAM76B in OS and its impact on macrophage polarization and tumor immune response. METHODS: We predicted intrinsically disordered regions (IDRs) of FAM76B and explored its phase separation properties in vitro using fluorescence confocal microscopy and fluorescence recovery after photobleaching (FRAP) experiments. Immunofluorescence staining was also performed. We engi

Downregulation of Decorin in ovarian cancer cells and colonization microenvironment drives progression.

Epithelial ovarian cancer is a gynecological disease in which transformed cells, upon dissemination into the peritoneum colonize locales such as omenta and form metastatic foci. Colonization is an emergent outcome of the interactions between the invading cancer cells and extracellular matrix (ECM) of the peritoneal serosa. Although ECM is known to be remodeled in cancer, the dynamics in ovarian cancer of a major class of ECM-remodeling factors: the proteoglycans remain understudied. Here, we focus on Decorin, a proteoglycan with binding activity to the principal stromal ECM protein Collagen I

Comprehensive Analysis Reveals That ISCA1 Is Correlated with Ferroptosis-Related Genes Across Cancers and Is a Biomarker in Thyroid Carcinoma.

BACKGROUND: ISCA1 (Iron-Sulfur Cluster Assembly 1) is involved in the assembly of iron-sulfur (Fe-S) clusters, which are vital for electron transport and enzyme activity. Some studies suggest the potential involvement of ISCA1 in tumor progression through interactions with ferroptosis-related genes (FRGs) and the tumor immune microenvironment (TME). However, there has been no systematic analysis of its role in FRGs and the TME or its predictive value for prognosis and immunotherapy response across different cancer types. METHODS: In this study, we analyzed the expression and prognosis of ISCA

FEATURES OF DISTRIBUTION OF INTRATUMORAL LYMPHOCYTES IN OVARIAN EPITHELIAL TUMOURS OF DIFFERENT HISTOLOGICAL TYPES AND DEGREE OF MALIGNANCY.

Ovarian cancer is the leading cause of death from gynaecological cancers worldwide. In Georgia, ovarian cancer ranks fourth in terms of prevalence, and third in terms of mortality rate. According to the latest epidemiological data, there will be 21,421 new cases of ovarian cancer and 13,770 deaths from ovarian cancer in the United States in 2021. Due to the lack of early screening methods and asymptomatic clinical courses, most ovarian cancer is diagnosed at an advanced stage, when the prognosis of the disease is already poor. Despite the advances in modern medical treatment and surgery, ovari

Modified C-type natriuretic peptide normalizes tumor vasculature, reinvigorates antitumor immunity, and improves solid tumor therapies.

Deficit of oxygen and nutrients in the tumor microenvironment (TME) triggers abnormal angiogenesis that produces dysfunctional and leaky blood vessels, which fail to adequately perfuse tumor tissues. Resulting hypoxia, exacerbation of metabolic disturbances, and generation of an immunosuppressive TME undermine the efficacy of anticancer therapies. Use of carefully scheduled angiogenesis inhibitors has been suggested to overcome these problems and normalize the TME. Here, we propose an alternative agonist-based normalization approach using a derivative of the C-type natriuretic peptide (dCNP). …

Assessing the Efficacy of Immune Checkpoint Inhibitors in Preclinical Tumor Models.

The identification of novel immune-related targets that can reactivate or enhance antitumor immunity is a very active field of cancer research. In this context, syngeneic tumor models are often used during the preclinical development of immunotherapies to assess their efficacy and analyze the immune system and tumor cell interaction. Here, we present the practical procedures to generate subcutaneous tumors and experimental lung metastases used to evaluate the antitumor activity of your immunotherapy of interest. We also describe a method to quantify contrasted lung metastasis burden by imaging

Persistent mutation burden drives sustained anti-tumor immune responses.

Tumor mutation burden is an imperfect proxy of tumor foreignness and has therefore failed to consistently demonstrate clinical utility in pr … pressure of immunotherapy and tumors with a high pTMB content were characterized by a more inflamed tumor microenvironment. pTMB imposes an evolutionary bottleneck that cancer cells cannot overcome and may thus drive sustained immunologic tumor control in the context of immunotherapy.

In situ hypoxia modulating nano-catalase for amplifying DNA damage in radiation resistive colon tumors.

Radiation therapy (RT) is a mainstream clinical approach in cancer treatment. However, the therapeutic efficacy of RT is greatly hindered by the presence of excessive hydrogen peroxide (H2O2) in the hypoxic region of the solid tumor, thus leading to tumor recurrence and metastasis. Herein, a thioketal-linked amphiphilic nano-assembly (MTS) loaded with hydrophobic manganese oxide (HMO) nanoparticles (MTS@HMO) is examined as a promising multi-purpose reactive oxygen species (ROS)-catalytic nanozyme for transforming an RT-resistant hypoxic tumor microenvironment (TME) into an RT-susceptible one b…

Tumor-Generated Reactive Oxygen Species Storm for High-Performance Ferroptosis Therapy.

Ferroptosis therapy (FT) efficacy of tumors suffers from a relatively low concentration of Fenton agents, limited hydrogen peroxide (H2O2) content, and insufficient acidity in the tumor environment (TME), which are unfavorable for reactive oxygen species (ROS) generation based on Fenton or Fenton-like reactions. The glutathione (GSH) overexpression in TME can scavenge ROS and abate the FT performance. In this study, a strategy of ROS storm generation specifically initiated by the TME and our developed nanoplatforms (TAF-HMON-CuP@PPDG) is proposed for high-performance FT of tumors. The GSH in t
