IL-13 is necessary, not simply sufficient, for epicutaneously induced Th2 responses to soluble protein antigen.

Th2 responses are clearly involved in the pathogenesis of atopic disease. Thus, understanding the factors responsible for Th2 sensitization at sites of allergen exposure, such as airway and skin, is crucial for directing therapeutic or preventive strategies. Contrary to other models of Th2 sensitization to proteins, we have reported that Th2 responses induced by epicutaneous exposure to OVA are IL-4 independent. Combined deficiency of both IL-4 and IL-13 signaling did prevent Th2 generation, suggesting that IL-13 was mediating these IL-4-independent responses. It was not clear, however, whethe

Cathepsin S: a double-edged protease in barrier damage, neuroimmune inflammation, and tissue repair, with implications for dermatology.

Cathepsin S (CTSS) is a lysosomal cysteine protease traditionally recognized for its role in invariant-chain degradation and major histocompatibility complex class II antigen presentation. However, unlike many lysosomal cathepsins, CTSS remains active at neutral or mildly acidic pH, enabling proteolytic functions at the cell surface and within extracellular inflammatory microenvironments. This property makes CTSS particularly relevant to skin biology, where barrier disruption, immune activation, neuroimmune signaling, extracellular matrix remodeling, angiogenesis, fibrosis, tumor progression,

Response of advanced cutaneous squamous cell carcinoma to immunotherapy: case report.

The most common cancer in the United States is non-melanoma skin cancer. Cutaneous squamous cell carcinoma (cSCC) is the second most common non-melanoma skin cancer after basal cell carcinoma. It develops in the middle and outer layers of the skin. Its precursor is actinic keratosis, which can progress to squamous cell carcinoma in situ, invasive cSCC, and finally metastatic cSCC. About 20% of non-melanoma skin cancers are squamous cell and the remaining 80% are basal cell. Unlike basal cell, squamous cell carcinoma has the propensity to metastasize. This commonly occurs with squamous cell car

Immune-mediated side-effects of cytokines in humans.

A large body of clinical experience on the adverse consequences of cytokine administration has accumulated since the last decade. Side-effects reported after the therapeutic use of cytokines has provided evidence that activation of the immune response may sometimes have deleterious consequences. Several effects appeared as a direct consequence of the immune activation induced by cytokines, e.g. flu-like reactions, vascular leak syndrome. Cytokine-induced exacerbation of underlying diseases or immune dysregulation were other complications of growing concern. Interferon-alpha (IFN-alpha) treatme

Topical ionic liquid-mediated GLUT1 gene editing ameliorates psoriasis and prevents recurrence.

Psoriasis is a chronic inflammatory skin disorder characterized by immune dysregulation and a high relapse rate. Current therapies seldom achieve lasting remission. Aberrant overexpression of glucose transporter 1 (GLUT1) in keratinocytes enhances glycolysis, fueling inflammation and immune imbalance, thus positioning GLUT1 as a promising therapeutic target. In this work, a composite ionic liquid-mediated transdermal platform was established for the delivery of CRISPR-Cas9 ribonucleoprotein (CIL-RNP), aiming to achieve efficient GLUT1 gene editing in keratinocytes. The CIL-RNP achieved 76.6% e

Alterations in Apoptotic Cell Populations, Protein Markers, and Gene Expression Patterns in Rats with Sulfur Mustard-induced Pulmonary Injuries.

Sulfur mustard (SM) is a potent chemical warfare agent that causes severe cutaneous, ocular, and pulmonary injuries, with respiratory tract damage being the most life-threatening. Despite its well-documented toxicity, the cellular mechanisms driving SM-induced apoptosis remain poorly understood. This study seeks to elucidate the apoptotic pathways involved in SM-induced pulmonary injury using a rat model. We induced acute lung injury through two delivery methods: intraperitoneal injection (8 mg/kg) and intratracheal instillation (2 mg/kg) of SM, with both doses representing 1 LD50. We assessed

Neferine ameliorates psoriasis through inducing oxidative stress-mediated keratinocyte apoptosis and suppressing inflammatory response via the Keap1-Nrf2 pathway.

Psoriasis is a chronic inflammatory skin disease characterized by keratinocytes (KCs) hyperproliferation and persistent inflammation. Current biologics are limited by high relapse rates and long-term safety concerns, prompting the search for multi-target natural small molecules. A murine psoriasis model was induced by 6-day continuous topical imiquimod (IMQ) application; cellular models were established by stimulating keratinocyte lines with tumor necrosis factor-alpha (TNF-α)/interleukin-17A (IL-17A). Network pharmacology and molecular docking were employed to predict targets, and small inter

Recurrent severe hypersensitivity reaction following repeated paclitaxel-eluting balloon angioplasty in the coronary arteries: a case report.

BACKGROUND: Paclitaxel drug-coated balloons (P-DCBs) are widely used for selected coronary lesions, but severe hypersensitivity reactions during coronary P-DCB angioplasty are rarely recognized. We report a rare case of recurrent severe hypersensitivity reaction temporally associated with repeated coronary P-DCB (Note: For ease of expression, the "Paclitaxel-eluting balloon" used in this case will be uniformly referred to as "P-DCB".) angioplasty. Disseminated herpes zoster with pulmonary lesions mimicking miliary tuberculosis in an HIV-infected patient previously treated with adalimumab: A case report.

Disseminated herpes zoster (DHZ) in immunocompromised patients is uncommon but potentially life-threatening. Pulmonary involvement is particularly rare and can mimic miliary tuberculosis, complicating the diagnosis. We report herein a 51-year-old, male, Japanese patient with a human immunodeficiency virus (HIV) infection, uncontrolled diabetes mellitus, and hidradenitis suppurativa in whom DHZ developed five months after he discontinued adalimumab therapy. He presented with a five-day history of fever, abdominal pain, and generalized vesicular eruptions. Chest computed tomography revealed smal

Gene Augmentation Techniques to Stimulate Wound Healing Process: Progress and Prospects.

Gene therapy has traditionally been used to treat individuals with late-stage cancers or congenital abnormalities. Numerous prospects for therapeutic genetic modifications have emerged with the discovery that gene therapy applications are far more extensive, particularly in skin and exterior wounds. Cutaneous wound healing is a complex, multistep process involving multiple steps and mediators that operate in a network of activation and inhibition processes. This setting presents a unique obstacle for gene delivery. Many gene delivery strategies have been developed, including liposomal administ

Nanofibrous patches for targeted therapy of cutaneous leishmaniasis caused by Leishmania major: a preclinical amphotericin B platform.

Cutaneous leishmaniasis (CL) remains a significant public health challenge in endemic regions, particularly where access to safe and patient-friendly treatments is limited. Amphotericin B (AmB), although highly active against Leishmania, is restricted by systemic toxicity and the need for parenteral administration. In this study, AmB-loaded bioactive nanofibrous patches were fabricated using a dual-nozzle electrospinning method incorporating chitosan, gelatin, and polyvinyl alcohol. The resulting nanofibers were structurally characterized by scanning electron microscopy, Fourier-transform infr

Measuring Psoriasis Severity at Home.

Psoriasis plaque severity metrics, such as induration (thickness), erythema (redness), and desquamation (scaliness), are associated with the subsequent development of psoriatic arthritis (PsA) among cutaneous-only psoriasis patients (patients with skin or nail psoriasis but no psoriatic arthritis). These metrics can be used for PsA screening. However, a key challenge in PsA screening is to optimize accessibility and minimize costs for patients, while also reducing the burden on healthcare systems. Therefore, an ideal screening tool consists of questions that patients can answer without a physi
