Safety Concerns, Mechanistic Pathways, and Knowledge Gaps in the Clinical Use of Selective Serotonin Reuptake Inhibitors.

Selective serotonin reuptake inhibitors (SSRIs) are widely prescribed for the management of depressive and anxiety disorders and represent one of the most significant pharmacological advances in psychiatry. SSRIs are effective and better tolerated than older antidepressants; however, long-term safety issues, interactions, and withdrawal effects remain a concern. This review combines data from clinical trials, safety databases, and biological studies to provide a clear overview of the safety of SSRIs. The key risks include gastrointestinal bleeding, sexual dysfunction, hyponatremia, serotonin s

Parental knowledge and access barriers in the management of cleft lip and/or palate: A cross-sectional study from Saudi Arabia.

Cleft lip and/or cleft palate (CLP) are congenital anomalies caused by improper fusion of facial structures, influenced by genetic, environmental, and nutritional factors. Understanding parental knowledge and attitudes is essential, as misconceptions and limited awareness can affect the care and development of affected children. This study aimed to comprehensively evaluate Saudi parents' understanding, perspectives, and behaviors concerning CLP in their children. Additionally, it sought to identify prevalent misconceptions and obstacles encountered by these parents in accessing necessary care.

Exploring the Involvement of RSPO1 Gene Variations in 46,XX DSD Patients With Mullerian Agenesis and/or Gonadal Dysgenesis-An Indian Study.

INTRODUCTION: There is a dearth of knowledge on the genetic background of Indian 46,XX DSD patients with Mullerian agenesis (MA) and/or gonadal dysgenesis (GD). Being one of the key controlling genes in female sex determination/differentiation, there is a paucity of study regarding the association of RSPO1 variations with MA/GD. Hence in this study, we screened proximal as well as exonic regions of the RSPO1 gene in 25 Indian 46,XX DSD patients with MA and/or GD. METHODOLOGY: Bidirectional PCR-Sanger sequencing was performed with RSPO1 proximal promoter and coding sequence-specific primers. T

Complex in vitro fertilization laboratory factors affecting sperm DNA integrity: A narrative review.

Sperm DNA fragmentation (SDF) is increasingly recognized as a key biomarker of male fertility, influencing natural conception and assisted reproduction clinical outcomes. Unlike conventional semen parameters, SDF directly reflects chromatin integrity and the sperm's genetic potential. Mechanistically, oxidative stress and abortive apoptosis represent the main pathways leading to DNA damage. Laboratory practice, such as centrifugation, prolonged incubation, cryopreservation, and inappropriate temperature or light exposure can elevate reactive oxygen species and compromise DNA stability. Similar

Outcomes of GnRH Agonist Therapy in Boys with Central Precocious Puberty: A Systematic Review of Final Height and Long-Term Health Effects.

BACKGROUND: Central precocious puberty (CPP) in boys can result in accelerated growth, early epiphyseal fusion, and reduced adult height. Gonadotropin-releasing hormone agonists (GnRHa) are commonly used to delay pubertal progression and preserve growth potential. However, data on their long-term effects on final height, bone health, and body mass index (BMI) remain limited. This systematic review aimed to evaluate the outcomes of GnRH agonist therapy in boys with CPP, focusing on final height and reported side effects. METHODS: A systematic literature search was conducted across five databas

Redescription of the Larval Trematode Taxon Monilicaecum ventricosum (Hemiuroidea: Didymozoidae) from Type Host Cololabis saira, with Inference of the Definitive Host Based on DNA Analysis.

The larval trematode taxon Monilicaecum ventricosum Yamaguti, 1942 (Didymozoidae) was redescribed from its type host, the Pacific saury Cololabis saira (Brevoort, 1856), collected from the North Pacific off Eastern Hokkaido, Japan. Because didymozoid larvae and adults differ markedly in morphology, linking these two stages using morphological data alone has traditionally been difficult. Consequently, the monotypic genus Monilicaecum Yamaguti, 1942 is currently regarded as a collective group, similar to genera established solely on larval forms of other trematodes. However, recent advances in m

An Overview of the Genetic Mechanisms of Colistin-Resistance in Bacterial Pathogens: An Indian Perspective.

Colistin resistance in bacteria is a growing global issue, given its role as a critical last-resort antibiotic, particularly for treating Gram-negative bacterial infections. Pathogens adopt multiple resistance mechanisms, mediated either by plasmids or chromosomal changes. Some of the most frequently observed strategies include the occurrence of plasmid-borne mobile colistin resistance (mcr) genes, enhanced efflux pump activity, mutations in the regulatory systems, and alterations in the lipid A structure. This article provides an overview of the studies investigating the genetic mechanisms un

Strategies to Mitigate Cisplatin-Induced Ototoxicity: A Literature Review of Protective Agents, Mechanisms, and Clinical Gaps.

BackgroundCisplatin, a widely used chemotherapeutic agent, is associated with significant ototoxicity, leading to progressive and irreversible sensorineural hearing loss in up to 93% of patients. Cisplatin generates reactive oxygen species (ROS) in the cochlea, activating apoptotic and necroptotic pathways that result in hair cell death. Inflammatory processes and nitrative stress also contribute to cochlear damage.MethodsThis literature review was conducted to explore the mechanisms underlying cisplatin-induced ototoxicity and evaluate protective strategies, including both current and emergin

Phenotypic and genetically predicted leukocyte telomere length and prostate cancer risk: results from a large-scale longitudinal cohort study.

BACKGROUND: Previous studies on the correlation between leukocyte telomere length (LTL) and prostate cancer (PCa) have shown inconsistent results. We aimed to clarify this association by leveraging a large-scale prospective design and Mendelian randomisation. METHODS: We enrolled a total of 229 022 male individuals from the UK Biobank (UKB) to investigate the association between LTL and PCa risk. We employed both unadjusted and covariates-adjusted Cox proportional hazards regression models to assess this relationship. We defined the primary outcome as the diagnosis of incident PCa using in-pa
