Enzymatic production of low-molecular-weight chitosan and chitooligosaccharides (COS) with consistent properties is challenging since chitosanases are unstable under acidic, high-temperature conditions. To address this, a recombinant chitosanase was covalently attached to glyoxyl-activated agarose via multipoint attachment to improve stability and influence its hydrolysis pattern. The immobilized enzyme showed increased activity and stability at pH 4.5 and 55 °C, where the free enzyme quickly lost activity. Immobilization also reduced temperature sensitivity and enabled sustained catalysis up

Interfacial regulation of molecular oxygen activation is critical for controlling reactive oxygen species generation in photocatalytic systems. Here, a copper 4-cyanophenylacetylide/copper phenylacetylide heterointerface (CN-PhC2Cu/PhC2Cu) was constructed through a one-step in situ strategy to regulate interfacial charge transfer and excited-state processes. Cyano functionalization created an electronic difference between the two copper acetylide components, while their related CCCu coordination frameworks favored intimate interfacial coupling. The optimized 75CNPC removed 95.2 ± 3.9% of diclo

Against the backdrop of an accelerating global aging population, the epidemiological correlation between cognitive impairment and osteoporosis has become increasingly prominent. These two conditions exhibit a profound pathological coupling mediated by the bidirectional regulatory network of the "bone-brain axis." The operation of this axis is rooted in an intricate neuro-skeletal signaling network involving hormonal dysregulation, systemic inflammatory cascades, and the aberrant regulation of core molecular pathways, such as Wnt/β-catenin and RANKL/OPG. Together, these factors synergistically

Impaired wound healing in diabetes is closely associated with cellular ferroptosis. Microneedle-assisted gas therapy exerts molecular regulation of ferroptosis, representing a promising approach for accelerating diabetic wound healing. To date, O2 and H2 gas therapies have consistently been conducted independently and have never been integrated into a single system. Whether and how their synergy might be realized remains a defining challenge. Herein, we propose a concept of integrated O2 and H2 gas therapy, demonstrated by fabricating a gas-producing microneedle material. Platinum@MIL-101(Fe)-

Although modern therapies have significantly improved survival, multiple myeloma (MM) remains incurable and biologically heterogeneous, resulting in substantial variability in treatment response and outcomes. Effective risk stratification is therefore critical to guide therapy intensity, predict relapses, and inform prognosis. This review critically examines the current biological determinants of high-risk MM and their implications for treatment intensification, selection of novel therapeutic agents, and stratified clinical trial enrollment. High-risk MM is driven by three major biological det

Hybrid catalysts combining semiconductors with molecular metal complexes have emerged as a promising strategy for efficient solar-to-chemical conversion under mild conditions; however, the mechanistic origin of their catalytic selectivity remains poorly understood. Here, we report a TiO2/[Co(bpy)3]2+ hybrid photocatalyst that exhibits significantly enhanced activity and selectivity for CO2-to-CO conversion compared with pristine TiO2. By combining electrochemical analysis, DFT, and TAS, we address consecutive elementary steps of the photocatalytic cycle: electrochemical analysis establishes th

NPM1-mutated acute myeloid leukemia (AML) represents one of the most frequent and biologically distinct AML entities, characterized by the aberrant cytoplasmic localization of the NPM1 mutant protein. The recent advances in molecular biology and translational research have progressively redefined the clinical management of this disease. This narrative review summarizes the evolution of NPM1-mutated AML over the last two decades, focusing on the transition from immunohistochemical observations to modern genetics-based and measurable residual disease (MRD)-driven approaches. We discuss the biolo

COVID-19, caused by SARS-CoV-2, remains a global health challenge because of viral evolution and immune escape. Although current therapies primarily target viral entry and replication, agents that directly interfere with the Spike receptor-binding domain (S-RBD) remain limited. Here, we combined virtual screening with biological validation to identify compounds capable of interfering with S-RBD function. A total of 3014 compounds from a customized drug library were screened by molecular docking. Candidate binding and functional activity were subsequently evaluated using cellular thermal shift

AIMS: Sulfadoxine-pyrimethamine (SP) is widely used as a partner drug in artemisinin combination therapy for Plasmodium falciparum malaria in India, except in the North-Eastern states. Drug resistance arises through mutations in Pfdhfr and Pfdhps, compromising treatment efficacy. The study investigated geographic distribution, temporal trends and molecular evolution of SP resistance markers in P. falciparum field isolates across India over three decades.
MATERIALS & METHODS: A total of 307 isolates were collected across 14 states between 1993 and 2023, with successful sequencing of 243 Pfdhfr

Pancreatic ductal adenocarcinoma (PDAC) remains a highly lethal malignancy, largely because of its profound and evolving therapeutic resistance. Resistance is not determined by a single molecular alteration but arises from interconnected mechanisms, including intrinsic resistance, treatment-induced adaptive resistance, acquired resistance, genomic evolution, clonal selection, cancer stemness, phenotypic plasticity, metabolic adaptation, and tumor microenvironment-mediated effects. Emerging therapeutic approaches targeting KRAS/RAS signaling, stromal and immune components, metabolic dependencie

Developing high-performance n-type organic mixed ionic-electronic conducting (OMIEC) polymers with simple structural motifs is still challenging. We show that high-performance, low-threshold-voltage n-type OMIEC polymers can be achieved using a simple diketopyrrolopyrrole unit flanked by thiazole groups, which is functionalized with glycolated side chains. Interestingly, the regiospecific sp2-N position in the repeating unit's thiazole governs the polymer chains' solvation and molecular packing. This specific backbone chemistry enhances conjugation efficiency, reduces trap density, and improve

Post-transplant lymphoproliferative disorders (PTLDs) typically arise months to years after solid-organ transplantation and are frequently driven by Epstein-Barr virus (EBV). However, EBV-negative monomorphic PTLDs are increasingly recognized as an alternative pathogenesis. We report a case of a 64-year-old man who underwent liver transplantation and was found to have a minute focus of diffuse large B-cell lymphoma (DLBCL) in the resected gallbladder. Given the absence of residual disease and the patient's postoperative frailty, close observation was chosen. Three months later, the patient dev

Japanese encephalitis virus (JEV) is a mosquito-borne zoonotic flavivirus causing severe neurological disease across Asia, and genotype V (GV) is now predominant in Korea. Despite frequent detection of GV in mosquitoes, human-derived complete genome data remain scarce. To elucidate the molecular and antigenic characteristics of human GV infections, cerebrospinal fluid samples from unvaccinated patients positive for JEV RNA during 2018-2023 were subjected to virus isolation in LLC-MK2 cells (rhesus monkey kidney-derived epithelial cell line). Three human GV isolates (K18P80, K23P84, K23P88) wer

Dietary specializations in animals lead to adaptations in morphology, anatomy and physiology. Neotropical bats, with their high taxonomic and trophic diversity, offer a unique perspective on diet-driven evolutionary adaptations. Here we assess the metabolic response to different dietary sugars among wild-caught bats. We found that insectivorous bats had a pronounced metabolic response to trehalose, whereas bats with nectar and fruit-based diets showed significantly higher blood glucose levels in response to glucose and sucrose, reaching levels over 750 mg dl-1. The genomic analysis of 22 focal

Despite emerging molecular information on chronic myelomonocytic leukemia (CMML), patient outcome remains unsatisfactory and little is known about the transformation to acute myeloid leukemia (AML). In a single-center cohort of 219 CMML patients, we explored the potential correlation between clinical features, gene mutations, and treatment regimens with overall survival (OS) and clonal evolution into AML. The most commonly detected mutations were TET2, SRSF2, ASXL1, and RUNX1. Median OS was 34 months and varied according to age, cytogenetic risk, FAB, CPSS and CPSS-Mol categories, and number o

Specific proteins and processes have been identified in post-myocardial infarction (MI) pathological remodeling, but a comprehensive understanding of the complete molecular evolution is lacking. We generated microarray data from swine heart biopsies at baseline and 6, 30, and 45 days after infarction to feed machine-learning algorithms. We cross-validated the results using available clinical and experimental information. MI progression was accompanied by the regulation of adipogenesis, fatty acid metabolism, and epithelial-mesenchymal transition. The infarct core region was enriched in process

Trichuriasis is among the most prevalent worldwide parasitism caused by helminths. For many years, Trichuris spp. have been described with a relatively narrow range of both morphological and biometrical features. The use of the complete mitochondrial genome (mitogenome) is an alternative and powerful molecular method for inferring phylogenies. Here, we present an overview of the contributions of mitogenome for Trichuris spp. from human and non-human primates. In addition, we carry out structural and phylogenetic comparative analyses with genomes of Trichuris species available in public dataset

BACKGROUND/PURPOSE: With the evolution of assisted fertility and prenatal diagnostic technology, the prevalence of multi-fetal pregnancy increased, followed by the demand for prenatal intervention if genomic aberration was detected. How to distinguish the affected foetus from the normal co-twin before selective fetal reduction is therefore challenging.
OBJECTIVES: We retrospectively reviewed the cases of dichorionic twins at our centre during 2004-2018, where selective fetal reduction was requested because one foetus carried a pathogenic genomic aberration. Five cases were enrolled, including

Parasites with complex life cycles have developed numerous phenotypic strategies, closely associated with developmental events, to enable the exploitation of different ecological niches and facilitate transmission between hosts. How these environmental shifts are regulated from a metabolic and physiological standpoint, however, still remain to be fully elucidated. We examined the transcriptomic response of Schistocephalus solidus, a trophically transmitted parasite with a complex life cycle, over the course of its development in an intermediate host, the threespine stickleback, and the final a

Background: Molecular typing is essential for inferring genetic relatedness between bacterial pathogens. In this study, we applied whole genome sequencing (WGS) for rapid prediction of sequence type and antibiotic resistance for invasive pneumococcal isolates. Methods: 240 isolates from adults (≥50 years old) in Ontario, Canada during 2009 to 2013 were subjected to WGS. Sequence type, antibiotic susceptibility and resistance were predicted directly from short reads. Emerging non-vaccine serotype 22F was further characterized by WGS. Results: Sequence type was successfully determined for 98.3%
