Experimental Dirofilaria immitis infection in dogs: effects of doxycycline and Advantage Multi® administration on immature adult parasites.

To better understand the efficacy of doxycycline and 10% imidacloprid+2.5% moxidectin (Advantage Multi(®); Bayer Animal Health, Shawnee Mission, Kansas) on immature adult Dirofilaria immitis parasites and the results of antigen tests, 12 healthy, randomly selected dogs were experimentally infected with D. immitis and monitored for 407 days. Two dogs in each of three subgroups of four dogs were each infected with six (total of 6 dogs) or 12 (total of 6 dogs) D. immitis infective third-stage larvae (L3) obtained from infected mosquitoes. Doxycycline (10mg/kg per os twice daily×30 days) and 10% i

Spatial distribution of G6PD deficiency variants across malaria-endemic regions.

BACKGROUND: Primaquine is essential for malaria control and elimination since it is the only available drug preventing multiple clinical attacks by relapses of Plasmodium vivax. It is also the only therapy against the sexual stages of Plasmodium falciparum infectious to mosquitoes, and is thus useful in preventing malaria transmission. However, the difficulties of diagnosing glucose-6-phosphate dehydrogenase deficiency (G6PDd) greatly hinder primaquine's widespread use, as this common genetic disorder makes patients susceptible to potentially severe and fatal primaquine-induced haemolysis. The

The effect of improved rural sanitation on diarrhoea and helminth infection: design of a cluster-randomized trial in Orissa, India.

BACKGROUND: Infectious diseases associated with poor sanitation such as diarrhoea, intestinal worms, trachoma and lymphatic filariasis continue to cause a large disease burden in low income settings and contribute substantially to child mortality and morbidity. Obtaining health impact data for rural sanitation campaigns poses a number of methodological challenges. Here we describe the design of a village-level cluster-randomised trial in the state of Orissa, India to evaluate the impact of an ongoing rural sanitation campaign conducted under the umbrella of India's Total Sanitation Campaign (T

Understanding the factors contributing to dengue virus and chikungunya virus seropositivity and seroconversion among children in Kenya.

Dengue virus (DENV) and chikungunya virus (CHIKV) are causes of endemic febrile disease among Kenyan children. The exposure risk to these infections is highly multifactorial and linked to environmental factors and human behavior. We investigated relationships between household, socio-economic, demographic, and behavioral risk factors for DENV and CHIKV seropositivity and seroconversion in four settlements in Kenya. We prospectively followed a pediatric cohort of 3,445 children between 2014-2018. We utilized the Kaplan-Meier curves to describe the temporal patterns of seroconversion among teste

VAMP8 Contributes to the TRIM6-Mediated Type I Interferon Antiviral Response during West Nile Virus Infection.

Several members of the tripartite motif (TRIM) family of E3 ubiquitin ligases regulate immune pathways, including the antiviral type I interferon (IFN-I) system. Previously, we demonstrated that TRIM6 is involved in IFN-I induction and signaling. In the absence of TRIM6, optimal IFN-I signaling is reduced, allowing increased replication of interferon-sensitive viruses. Despite having evolved numerous mechanisms to restrict the vertebrate host's IFN-I response, West Nile virus (WNV) replication is sensitive to pretreatment with IFN-I. However, the regulators and products of the IFN-I pathway th

Seasonal and spatial distribution of Aedes aegypti and Aedes albopictus in a municipal urban park in São Paulo, SP, Brazil.

Dengue, yellow fever, chikungunya and Zika are important arboviruses present in various countries of the world, the etiological agents of which are transmitted to human-beings by the bite of infected females of Aedes aegypti and Aedes albopictus. Biological aspects of these vectors, such as their distribution and abundance, are influenced by climatic variables such as rainfall and temperature. We assess the spatial and seasonal distribution of Ae. aegypti and Ae. albopictus, during spring 2014 and spring 2015 and autumn 2015 and autumn 2016, in an urban Municipal Park, São Paulo (SP, Brazil),

Reduced Genetic Diversity of Key Fertility and Vector Competency Related Genes in Anopheles gambiae s.l. Across Sub-Saharan Africa.

BACKGROUND: Insecticide resistance challenges the vector control efforts towards malaria elimination and proving the development of complementary tools. Targeting the genes that are involved in mosquito fertility and susceptibility to Plasmodium with small molecule inhibitors has been a promising alternative to curb the vector population and drive the transmission down. However, such an approach would require a comprehensive knowledge of the genetic diversity of the targeted genes to ensure the broad efficacy of new tools across the natural vector populations. METHODS: Four fertility and para

BACKGROUND: Aedes aegypti is an important vector that transmits dengue, Zika, chikungunya, and yellow fever viruses. Although research on Aedes aegypti has been conducted for decades, scientometric studies on Aedes aegypti are scarce, are limited to regions, and cover short periods. Thus, there is still a knowledge gap in the current trend, research focuses and directions, leading authors and collaboration, journal and citation impacts, countries, and worldwide collaborations. OBJECTIVE: The objectives of the study are to investigate the research trend, focus and directions, citation impact,

Irrigated agriculture is an important risk factor for West Nile virus disease in the hyperendemic Larimer-Boulder-Weld area of north central Colorado.

This study focused on two West Nile virus (WNV) disease outbreak years, 2003 and 2007, and included a three-county area (Larimer, Boulder, and Weld) in North Central Colorado that is hyperendemic for WNV disease. We used epidemiological data for reported WNV disease cases at the census tract scale to: (1) elucidate whether WNV disease incidence differs between census tracts classified as having high versus lower human population density (based on a threshold value of 580 persons/km2) and (2) determine associations between WNV disease incidence and habitat types suitable as development sites fo

Unravelling the distribution of vectors of major vector-borne diseases in Koshi Province of Nepal: A concern of expansion in diverse geo-ecological and climatic regions.

BACKGROUND: Vector-borne diseases (VBDs), including malaria, visceral leishmaniasis, lymphatic filariasis, Japanese encephalitis, and dengue, are major public health concerns and are either slated for elimination or projected for control in Nepal. One of the major challenges in controlling these VBDs is halting their emergence and expansion in diverse geo-ecological and climatic regions. In this study, we collected vectors of major VBDs to assess their distribution, diversity, and associations with ecological variations and to provide an updated understanding of the current situation. Phase I/IIa study to assess the safety, immunogenicity and efficacy of ChAdOx1-MVA vectored vaccines expressing a novel liver-stage malaria dual antigen LS2 by sporozoite challenge in malaria-naïve adults.

BACKGROUND: Induction of CD8 + T-cells using viral vectors is a promising strategy in developing effective vaccines against pre-erythrocytic malaria. A recent comparative assessment of candidate antigens using this approach in a mouse model had identified Liver Stage Antigen 1 (LSA1) and Liver Stage Associated Protein 2 (LSAP2) as more protective than TRAP and CSP antigens, which have been the dominant focus of clinical testing. We proposed that combining these within a novel dual antigenic insert (LS2), encoded alongside an orthologous immunogenic domain from invariant chain in ChAdOx1, and t

Single-cell transcriptomics reveals a multiphasic Wolbachia host infection trajectory.

Intracellular bacterial symbionts must navigate host cellular environments, co-opt host biology, and evade immune clearance to establish persistent infections, yet the molecular mechanisms of infection establishment remain poorly characterized. The endosymbiont Wolbachia pipientis, prevalent across arthropods and nematodes and widely used for biological control, exemplifies this challenge: transinfected into mosquitoes, it blocks viral transmission to humans and suppresses reproduction. Yet how w Mel establishes infection in its native host, Drosophila melanogaster , remains unclear, obscured

Efficacy of a fixed combination of permethrin 54.5% and fipronil 6.1% (Effitix) in dogs experimentally infested with Ixodes ricinus.

BACKGROUND: Ticks are the most important vectors of disease-causing pathogens in domestic animals and are considered to be second worldwide to mosquitoes as vectors of human diseases. In Europe, Ixodes ricinus, the sheep tick, plays an important role as companion animal parasite but is also the primary vector of medically important diseases such as tick-borne encephalitis and Lyme borreliosis. The present study was designed to evaluate the efficacy under laboratory conditions of a new fixed spot-on combination of fipronil and permethrin (Effitix, Virbac) in treating and preventing tick infesta

Enhancing Entomological Surveillance to Combat Aedes-Borne Diseases in Southern Iran.

BACKGROUND: Vector-borne diseases, particularly those transmitted by Aedes mosquitoes, including dengue, chikungunya, Zika, and yellow fever, represent significant public health challenges globally. The rapid spread of these diseases in urban environments, driven by climate change and human activities, necessitates effective surveillance and control strategies. In Iran, the emergence of Aedes aegypti has raised concerns about potential outbreaks of the disease, particularly in southern regions such as Hormozgan Province. METHODS: A retrospective descriptive study was conducted using routinely

Imported malaria: a silent and forgotten threat in malaria free zones?

BACKGROUND: Countries that declared free of malaria remain vulnerable to the reintroduction and re-establishment of malaria transmission, primarily triggered by the influx of imported malaria. This risk is particularly acute in regions with population movement from endemic zones, limited tracking capacity, or continued presence of competent vectors. Experiences from countries navigating these vulnerabilities can provide valuable lessons for regions currently progressing or sustaining malaria elimination. This study aims to present various strategies and lessons learned in controlling imported

Genome and Transcriptome Analyses Facilitate Genetic Control of Wohlfahrtia magnifica, a Myiasis-Causing Flesh Fly.

Myiasis caused by Wohlfahrtia magnifica is a widespread parasitic infestation in mammals. The infested host suffers from damage as the developing larvae feed on its tissues. For the control of myiasis infestation, genetic methods have been shown to be effective and promising as an alternative to insecticides. Combining genome, isoform sequencing (Iso-Seq), and RNA sequencing (RNA-seq) data, we isolated and characterized two sex-determination genes, W. magnifica transformer (Wmtra) and W. magnifica transformer2 (Wmtra2), whose orthologs in a number of insect pests have been utilized to develop

Isolation and identification of Xenorhabdus and Photorhabdus bacteria associated with entomopathogenic nematodes and their larvicidal activity against Aedes aegypti.

BACKGROUND: Aedes aegypti is a potential vector of West Nile, Japanese encephalitis, chikungunya, dengue and Zika viruses. Alternative control measurements of the vector are needed to overcome the problems of environmental contamination and chemical resistance. Xenorhabdus and Photorhabdus are symbionts in the intestine of entomopathogenic nematodes (EPNs) Steinernema spp. and Heterorhabditis spp. These bacteria are able to produce a broad range of bioactive compounds including antimicrobial, antiparasitic, cytotoxic and insecticidal compounds. The objectives of this study were to identify Xen
