BACKGROUND: There is a need to build public support for the conduct of HIV cure research. It is important to enhance the public understanding of HIV cure strategies in resource-constrained settings with high HIV prevalence. Adopting deliberative approaches allows information to be shared interactively, building knowledge on the topic and unpacking the complexity of the science.
METHODS: We conducted deliberative community dialogues in rural KwaZulu-Natal, South Africa, with the Africa Health Research Institute community engagement team and Community Advisory Board members. We discussed the pu

Early detection of tuberculosis (TB) is central to global efforts for TB care and prevention. Conventional symptom-based screening and sputum microbiology fail to identify a substantial proportion of cases, particularly those that are asymptomatic or have low bacillary burden. Non-invasive medical imaging offers a critical solution for visualizing pulmonary pathology before individuals with TB develop characteristic symptoms. Chest X-ray has long been central to community screening, and recent advances in computer-aided detection systems endorsed by the World Health Organization have improved

Chimeric antigen receptor (CAR) T-cell therapy has transformed the treatment of several hematological malignancies, but its broader application remains constrained by the complexity, cost, and time required for conventional ex vivo manufacturing. In vivo CAR T-cell therapy has emerged as a promising next-generation strategy that aims to generate CAR T cells directly within the patient through targeted delivery of CAR-encoding genetic information to endogenous T cells. This approach has the potential to simplify treatment workflows, shorten manufacturing timelines, reduce production costs, and

Interleukin-10 (IL-10) is classically defined as a potent anti-inflammatory cytokine that protects tissues by constraining myeloid activation and limiting inflammatory cytokine production. Yet, accumulating evidence indicates that, in defined contexts, IL-10 can also promote antitumor immunity by enhancing the fitness and cytotoxic programs of CD8+ T cells and natural killer (NK) cells. Conversely, in other tumor contexts, IL-10 may reinforce immunosuppressive myeloid and regulatory programs, and the factors that determine these divergent outcomes remain incompletely understood. Although these

Migraine is a complex neurological disorder involving multiple neuropeptides that modulate nociceptive and sensory pathways. The most studied peptide is calcitonin gene-related peptide (CGRP), which is a well-established migraine trigger and therapeutic target. Recently, another peptide, pituitary adenylate cyclase-activating polypeptide (PACAP), has emerged as an alternative target for migraine therapeutics. This review compares the roles of PACAP and CGRP in preclinical mouse models of migraine. PACAP shares similarities with CGRP, and both are expressed in peripheral and central migraine-re

PURPOSE: Experimental studies of Dry Eye Disease (DED) using animal models are hampered by the lack of reliable, easy-to-use assays that can adequately diagnose disease or monitor effects of novel treatments. The Oculus Keratograph 5 M, an advanced keratography unit (AKU), has shown promise, enjoying recent clinical use. We assessed whether this AKU could be used in DED studies in the rabbit, perhaps the ideal experimental animal for this disease.
METHODS: All measures were made in strictly controlled temperature and humidity spaces. A panel of AKU parameters was measured in 15 New Zealand Wh

The COVID-19 pandemic has revealed significant disparities in health research across regions, particularly in Latin America. This viewpoint explores the challenges encountered during a randomized clinical trial in Colombia designed to evaluate the effectiveness of intravenous Alprostadil for treating moderate-to-severe COVID-19. The trial, aimed to address the urgent need for effective treatments, ultimately became undermined by bureaucratic barriers. This experience of regulatory delays and prolonged setbacks highlighted broader systemic issues in health research across Latin America, such as

Temporal lobe epilepsy (TLE) is increasingly recognized as a network-level disorder, with contemporary strategies shifting focus from localized epileptic lesions to targeting dysfunctional epileptogenic networks. Leveraging recent advancements in neuroimaging genetics and the growing understanding of brain network remodeling in epilepsy, partial least squares regression is employed to integrate the altered synaptic connectome in TLE patients with a human transcriptomics dataset. The findings reveal a strong association between disruptions in synaptic density similarity networks and the spatial

This paper provides an overview of the ethical considerations surrounding the inclusion of underserved populations in later-phase clinical trials. Underserved populations, defined here as those with restricted access to or limited benefits from healthcare, often face systemic, logistical, and social barriers that limit their participation in research. This results in a lack of representation that undermines fairness in research and also hampers the development of effective inclusive healthcare practices. This paper argues that including underserved populations in research is crucial for promot

Immune synapses, which were initially discovered at the interface between antigen-presenting cells (APCs) and T cells, are special structures formed at the contact site between antigen-presenting cells and immune cells and constitute the structural basis for immune cells to kill tumours and synthesise antibodies. Their structures are very similar to those of neural synapses in the nervous system, and they contain different functional structural regions. With the development of cell visualization research, scientists have increasingly conducted in-depth research on immune synapses. At present,

To guide researchers and funders, we sought to identify top priorities for research on increasing HPV vaccine coverage among US children ages 9-12 years. In Survey 1, 60 participants (researchers, health care professionals, and other stakeholders engaged in HPV vaccination research) viewed 57 research topics about increasing HPV vaccination coverage and provided additional topics. We integrated their suggestions into the list and condensed similar items to reach a list of 38 topics. In Survey 2, 230 participants identified their top ten research priorities. The top HPV vaccination research pri

Mycobacterium abscessus complex (MABC) infections are among the most intractable challenges in clinical mycobacteriology because of their extensive intrinsic and acquired antibiotic resistance. Recent studies on compassionate use and a systematic 20-patient cohort study demonstrated that bacteriophage therapy is safe and generally well-tolerated, and it has been proven capable of inducing clinically meaningful improvements. Nevertheless, patient outcomes remain heterogeneous, largely because of antibody-mediated neutralization during intravenous administration and morphotype-dependent suscepti

This article explores female healthcare at the crossroads of bacteriology and obstetric research. Puerperal fever or childbed fever manifested as an epidemic since the nineteenth century, and in both Europe and America, it charted a distinct course for bacteriological research. With the identification of bacteriological causes, new sets of public health regimes were instituted in both regions. The experience of the colonies, however, differed. This paper focusses on how colonial discourse on obstetric nursing, midwifery, clinical hygiene, and maternal healthcare can be positioned in this globa

Animal models, particularly transgenic mice, are extensively used in Alzheimer's disease (AD) research to emulate key disease hallmarks, such as amyloid plaques and neurofibrillary tangles formation. Although these models have contributed to our understanding of AD pathogenesis and can be helpful in testing potential therapeutic interventions, their reliability is dubious. While preclinical studies have shown promise, clinical trials often yield disappointing results, highlighting a notable gap and disparity between animal models and human AD pathology. Existing models frequently overlook earl

An anonymous online survey in French was used to assess if endometriosis patients would be as ready as unaffected women to donate their menstrual blood for biological research on endometriosis and evaluate potential barriers to such donation. It was distributed in September 2022 by social media and two mailing lists, including a French patient organization. The questionnaire assessed participant age and brief medical history (hormonal contraception, endometriosis diagnosis, type of endometriosis), menstrual experience (menstrual blood abundance, dysmenorrhea), and whether participants would do
