DNA sequencing of the coding region of the erythroid 5-aminolaevulinate synthase (ALAS2) cDNA from a male with pyridoxine-responsive sideroblastic anaemia revealed a missense mutation, a G561T transversion in exon 5 of the gene. Previously, the mutation G561A has been shown to be responsible for sideroblastic anaemia in females and thought to be lethal in males (1). The mutation G561T results in the loss of an MspA1-I cutting site. Analysis of MspA1-I restriction enzyme digests of amplified exon 5 genomic DNA from other family members revealed that the proband's mother, aunt and youngest siste

Treacher Collins Syndrome (TCS) is the most common of the human mandibulofacial dysostosis disorders. Recently, a partial TCOF1 cDNA was identified and shown to contain mutations in TCS families. Here we present the entire exon/intron genomic structure and the complete coding sequence of TCOF1. TCOF1 encodes a low complexity protein of 1,411 amino acids, whose predicted protein structure reveals repeated motifs that mirror the organization of its exons. These motifs are shared with nucleolar trafficking proteins in other species and are predicted to be highly phosphorylated by casein kinase. C…

Glucocorticoid-suppressible hyperaldosteronism (GSH) is an uncommon form of dominantly inherited hypertension caused by the inheritence of a chimaeric 11 beta-hydroxylase/aldosterone synthase gene. Affected individuals appear to have an increased risk of premature morbidity and mortality from stroke, but treatment with low doses of dexamethasone can completely reverse the biochemical and clinical features. We assessed the clinical and genetic features of 5 British kindreds with GSH, the largest collection outwith the United States, and determined the location of the crossover regions in the ch

Leydig cell hypoplasia (LCH) is a form of male pseudohermaphroditism in which Leydig cell differentiation and testosterone production are impaired. This report describes the first case of a nonsense mutation (A1635C) in exon 11 of the human luteinizing hormone receptor (hLHR) gene in two sisters with LCH. This mutation causes loss of function of the receptor by introducing a stop codon at residue 545 in transmembrane helix 5 of the hLHR. Surface expression of the truncated hLHR (hLHR-t545) in human embryonic kidney cells stably transfected with cDNA encoding hLHR-t545 was diminished compared t

A deficiency of the enzyme iduronate-2-sulfatase (IDS) is the cause of Hunter syndrome (mucopolysaccharidosis type II). Here, we report a study of the human IDS locus at Xq28. An unexpected finding was an IDS-related region (IDS2) which is located on the telomeric side of the IDS gene within 80 kb. We have identified sequences in this locus that are homologous to exons 2 and 3 as well as sequences homologous to introns 2, 3 and 7 of the IDS gene. The exon 3 sequences in the IDS gene and in the IDS2 locus showed 100% identity. The overall identities of the other identified regions were 96%. A l…

Liver factor B1 [LFB1, also called hepatocyte nuclear factor 1 (HNF1)] is a tissue-specific vertebrate transcription factor that is present in the liver, intestine, stomach and kidney. The LFB1 protein contains an unusual homeobox that is characterized by an insertion of 21 amino acids (aa) not found in any other homeodomain protein. We have isolated and characterized the genomic sequences encoding the LFB1 of Xenopus laevis. By comparing the genomic sequences with the cDNA clones, we could identify nine exons. In general, the position of the introns is identical to the one previously found in…

Complete deficiency of hypoxanthine guanine phosphoribosyltransferase (HPRT) causes Lesch-Nyhan syndrome. A single nucleotide substitution of G to T at the 3'-end of intron 3 in the splicing consensus region has been identified in one allele of the HPRT gene from a mother predicted to be a heterozygous Lesch-Nyhan carrier. Utilizing a BfaI restriction site which was lost in the mutation as an indicator, family study showed that the mother and her only daughter were heterozygotes but the mother's sister did not have the mutant allele. The mutation generated splicing error and resulted in two ty

Several heterozygous mutations within the gene encoding the alpha-subunit of Gs (GNAS1), the G protein that stimulates adenylyl cyclase, have been previously identified in patients with Albright hereditary osteodystrophy (AHO). We have now identified a fourth GNAS1 mutation from an AHO patient. Amplification by the polymerase chain reaction (PCR) of a genomic fragment encompassing GNAS1 exons 7 and 8 from one patient resulted in a product with aberrant migration on nondenaturing polyacrylamide and agarose gels. Direct DNA sequencing identified a 4-bp deletion in one allele of exon 7 encoding a…

PURPOSE: Drug resistance is a major cause of treatment failure in breast cancer patients. Numerous genetic and epigenetic markers have been proposed to influence Neoadjuvant Chemotherapy (NACT) response, and genes that regulate the effectiveness of chemotherapeutic agents undergo expression regulation via methylation. In this study, we aimed to investigate the methylation levels of KDM5A, KDM5B, and KDM5C, as well as their associated genes, EZH2, TFAP2C, ZMYND8, and SIN3B (Exon 1-2 and 19-20), in blood samples collected from patients before and after NACT treatment. Besides, we also examined p

Interleukin-37 (IL-37) is an anti-inflammatory member of interleukin-1 family. Alternative splicing generates five isoforms (IL-37a-e) with distinct exon compositions. IL-37a has recently attracted increasing attention because it uniquely retains exon 3. Exon 3 encodes an N-terminal nuclear localization sequence (NLS) that enables autonomous nuclear translocation and harbors an elastase cleavage site at L21/R22. Elastase cleavage produces N-terminal IL-37a (NIL-37a) and mature IL-37a (mIL-37a). Full-length IL-37a and NIL-37a can exert IL-1R8-independent nuclear regulatory and anti-inflammatory

Atypical Epidermal Growth Factor Receptor (EGFR) mutations in non-small-cell lung cancer (NSCLC) account for approximately 12% of all EGFR alterations. In contrast to classical mutations (exon 19 deletions and exon 21 L858R) and exon 20 insertions, for which therapeutic strategies have significantly evolved, relatively little progress has been achieved in the management of patients with atypical EGFR-mutated NSCLC. To date, afatinib remains the only approved agent specifically indicated for tumors harboring selected atypical variants-namely S768I, L861Q, and G719X-based on regulatory approvals

Even in the era of tyrosine kinase inhibitor (TKI) therapy, the prognostic landscape of atypical transcripts in chronic myeloid leukemia (CML) remains poorly defined. In our preliminary work, we identified two atypical transcripts harboring distinct DNA breakpoints that ultimately translate into an identical truncated protein. To investigate the DNA breakpoints of the BCR::ABL fusion gene in two patients with novel e13a3-type CML and to clarify the underlying mechanism of their generation, we conducted a breakpoint analysis. Target DNA fragments were amplified via nested PCR using specific pri

In chronic myeloid leukemia (CML), less than 2% of cases express atypical or rare BCR::ABL1 transcripts. The e8a2 BCR::ABL1 fusion transcript, a rare variant, has been reported in only 20 cases to date, primarily in case reports or case series. The direct junction between BCR exon 8 and ABL1 exon 2 generates a premature stop codon at position 7 after the fusion, while the insertion of certain sequences can result in the formation of an in-frame e8a2 transcript. To date, the insertion of SPECC1L gene sequences into e8a2 BCR::ABL1 fusion transcripts has been reported in two CML cases, and the V3

BACKGROUND: Systemic lupus erythematosus (SLE) is an autoimmune disease characterized by loss of immune tolerance, leading to systemic inflammation and organ damage. The Mediterranean fever (MEFV) gene, primarily linked to familial Mediterranean fever (FMF), has been suggested to have a protective role against SLE. However, comprehensive whole-exon analyses of MEFV and research on MEFV or FMF in non-Mediterranean populations, where MEFV exon 10 mutations are relatively rare, are limited.
METHODS: We conducted a whole-exon analysis of the MEFV gene in 55 Japanese patients with SLE. Patients we
