GWAS-Based Discoveries in IgA Nephropathy, Membranous Nephropathy, and Steroid-Sensitive Nephrotic Syndrome.

Over the past decade, genome-wide association studies (GWAS) have emerged as a powerful tool to understand the genetic basis of complex traits in humans. The GWAS approach has been successfully applied to primary glomerular disorders, providing numerous novel insights into the genetic architecture of IgA nephropathy, membranous nephropathy, and steroid-sensitive nephrotic syndrome. IgA nephropathy appears to have a highly complex polygenic architecture, with nearly 20 genome-wide significant loci of small-to-moderate effects discovered to date. In contrast, the genetic susceptibility to membra

Genetic and environmental contributions to diagnostic fluctuation in anorexia nervosa and bulimia nervosa.

BACKGROUND: Anorexia nervosa and bulimia nervosa are two severe eating disorders associated with high premature mortality, suicidal risk and serious medical complications. Transition between anorexia nervosa and bulimia nervosa over the illness course and familial co-aggregation of the two eating disorders imply aetiological overlap. However, genetic and environmental liabilities to the overlap are poorly understood. Quantitative genetic research using clinical diagnosis is needed. METHODS: We acquired a clinical diagnosis of anorexia nervosa (prevalence = 0.90%) and bulimia nervosa (prevalen

Seasonality shows evidence for polygenic architecture and genetic correlation with schizophrenia and bipolar disorder.

OBJECTIVE: To test common genetic variants for association with seasonality (seasonal changes in mood and behavior) and to investigate whether there are shared genetic risk factors between psychiatric disorders and seasonality. METHOD: Genome-wide association studies (GWASs) were conducted in Australian (between 1988 and 1990 and between 2010 and 2013) and Amish (between May 2010 and December 2011) samples in whom the Seasonal Pattern Assessment Questionnaire (SPAQ) had been administered, and the results were meta-analyzed in a total sample of 4,156 individuals. Genetic risk scores based on r

Molecular Epidemiology of Novel Pathogen "Brachyspira hampsonii" Reveals Relationships between Diverse Genetic Groups, Regions, Host Species, and Other Pathogenic and Commensal Brachyspira Species.

Outbreaks of bloody diarrhea in swine herds in the late 2000s signaled the reemergence of an economically significant disease, swine dysentery, in the United States. Investigations confirmed the emergence of a novel spirochete in swine, provisionally designated "Brachyspira hampsonii," with two genetically distinct clades. Although it has since been detected in swine and migratory birds in Europe and North America, little is known about its genetic diversity or its relationships with other Brachyspira species. This study characterizes B. hampsonii using a newly developed multilocus sequence ty

Alzheimer's disease genes and cognition in the nondemented general population.

BACKGROUND: Genome-wide association studies have established 11 genes for late-onset Alzheimer's disease (AD). We investigated whether these genes jointly affect cognition in a nondemented population and improve prediction of AD. METHODS: In 5171 nondemented people (age 45-99 years) from the population-based Rotterdam Study, we genotyped APOE-ε4 and single nucleotide polymorphisms from the genes CLU, PICALM, BIN1, CR1, ABCA7, MS4A6A, MS4A4E, CD2AP, EPHA1, and CD33. We constructed a genetic risk score by adding the risk alleles per individual weighted by the reported effect sizes. All people u

Development of polymorphic microsatellites for the invasive Asian fish tapeworm Schyzocotyle acheilognathi.

We describe the development of ready-to-use set of fifteen polymorphic microsatellite markers to benefit future population biology and phylogeographic studies on the invasive Asian fish tapeworm Schyzocotyle acheilognathi. The microsatellite loci were selected from partial Illumina shotgun genome sequences of three parasite specimens and their universality tested on a set of 12 geographically distant populations of the parasite. Particularly low levels of heterozygosity have been detected in the Chinese population pointing towards possible hidden population structure that deserves further atte

The interface between rare plant management and genetics: conserving Pitcher's thistle (Cirsium pitcheri) in the Great Lakes region, USA.

BACKGROUND AND AIMS: Tools showing best conservation practices are becoming critical for rare plant conservation as populations become isolated through habitat fragmentation and changing ecosystem processes. We illustrate the importance of reintroduction and assisted gene flow, using reintroduction of the USA federally threatened Pitcher's thistle (Cirsium pitcheri), a monocarpic perennial endemic to the western Great Lakes sand dunes. We evaluate the success of experimental reintroductions of this species into its dynamic coastal environment and address fundamental issues for rare plant reint

Population data of 27 Y-STR loci in indigenous populations from Sabah and Sarawak, Malaysia.

Sabah and Sarawak form East Malaysia on the island of Borneo. They are separated from West Malaysia (i.e., Peninsular Malaysia) by the South China Sea and home to diverse indigenous Austronesian speaking populations. In this study, we analysed 257 male blood samples belonging to Austronesian speaking Murut, Bajau, Kadazan-Dusun and Melanau population groups for 27 Y-chromosome short tandem repeat (Y-STR) loci using the Yfiler™ Plus Polymerase Chain Reaction (PCR) Amplification kit. Forensic parameters such as discrimination capacity and random match probability calculated based on the newly ge

Colonisation history of a vector species of veterinary interest: phylogeography of Culicoides imicola (Diptera: Ceratopogonidae) in the Indian Ocean revealing low genetic structure.

Culicoides imicola (Diptera: Ceratopogonidae) is the main historical vector of livestock viruses such as bluetongue virus (BTV), Epizootic Haemorrhagic Disease virus (EHDV) and African Horse Sickness virus (AHSV), in the Afrotropical region. Despite its wide distribution across Africa, the Mediterranean basin and parts of Asia, the genetic structure and colonisation history of this species in the South-West Indian Ocean (SWIO) territories remain poorly understood. The aim of this study was to investigate how passive wind-driven dispersal and recent animal movements associated with human coloni

The genome of the black-footed cat: Revealing a rich natural history and urgent conservation priorities for small felids.

Habitat degradation and loss of genetic diversity are common threats faced by almost all of today's wild cats. Big cats, such as tigers and lions, are of great concern and have received considerable conservation attention through policies and international actions. However, knowledge of and conservation actions for small wild cats are lagging considerably behind. The black-footed cat, Felis nigripes, one of the smallest felid species, is experiencing increasing threats with a rapid reduction in population size. However, there is a lack of genetic information to assist in developing effective c

Genetic Diversity of Circumsporozoite Surface Protein of Plasmodium vivax from the Central Highlands, Vietnam.

The circumsporozoite surface protein of Plasmodium vivax (PvCSP) plays a critical role in parasite biology. It has been extensively studied as a leading vivax-malaria-vaccine candidate. In this study, the genetic polymorphism and natural selection of pvcsp in P. vivax isolates collected from the Central Highlands, Vietnam were analyzed to understand the genetic structure of the parasite circulating in the endemic area and to provide baseline information for effective vaccine development based on the protein. Only two major alleles, VK210 and VK247, were detected in Vietnamese pvcsp, with VK247

Genetic diversity in two leading Plasmodium vivax malaria vaccine candidates AMA1 and MSP119 at three sites in India.

Plasmodium vivax, a major contributor to the malaria burden in India, has the broadest geographic distribution and shows higher genetic diversity than P. falciparum. Here, we investigated the genetic diversity of two leading P. vivax vaccine candidate antigens, at three geographically diverse malaria-endemic regions in India. Pvama1 and Pvmsp119 partial coding sequences were generated from one hundred P. vivax isolates in India (Chennai n = 28, Nadiad n = 50 and Rourkela n = 22) and ~1100 published sequences from Asia, South America, North America, and Oceania regions included. These data were

Systems biology analysis of human genomes points to key pathways conferring spina bifida risk.

Spina bifida (SB) is a debilitating birth defect caused by multiple gene and environment interactions. Though SB shows non-Mendelian inheritance, genetic factors contribute to an estimated 70% of cases. Nevertheless, identifying human mutations conferring SB risk is challenging due to its relative rarity, genetic heterogeneity, incomplete penetrance, and environmental influences that hamper genome-wide association studies approaches to untargeted discovery. Thus, SB genetic studies may suffer from population substructure and/or selection bias introduced by typical candidate gene searches. We r
