Directed differentiation of embryonic P19 cells and neural stem cells into neural lineage on conducting PEDOT-PEG and ITO glass substrates.

Differentiation of pluripotent and lineage restricted stem cells such as neural stem cells (NSCs) was studied on conducting substrates of various nature without perturbation of the genome with exogenous genetic material or chemical stimuli. Primary mouse adult neural stem cells (NSCs) and P19 pluripotent embryonal (P19 EC) carcinoma cells were used. Expression levels of neuronal markers β-III-tubulin and neurofilament were evaluated by immunochemistry and flow cytometry. It was shown that the ability of the substrate to induce differentiation directly correlated with its conductivity. Conducti

Functional Shift of NLRP3 Inflammasome Activity in Aged Macrophages.

The present study investigates the functional remodeling of the NLRP3 inflammasome in bone marrow-derived macrophages during aging. Upon LPS stimulation, no increase in proinflammatory cytokine secretion was detected; however, NLRP3 activation was accompanied by an increase in DNA fragmentation. Pharmacological inhibition of the inflammasome with MCC950 attenuated apoptosis while simultaneously increasing the proportion of senescent cells, indicating a shift from apoptosis toward senescence. These findings suggest a functional shift in NLRP3 activity during aging: rather than playing a predomi

Efficacy of venetoclax combined with azacitidine for advanced blastic plasmacytoid dendritic cell neoplasm: experience from two Japanese cases.

Blastic plasmacytoid dendritic cell neoplasm (BPDCN) is a rare and aggressive hematologic malignancy with no established standard treatment. Recently, the combination of venetoclax (VEN) and azacitidine (Aza) has shown promising efficacy in elderly or unfit patients; however, clinical data in Japanese patients remain limited. We report two Japanese cases with BPDCN that responded favorably to VEN plus Aza therapy. The first case involved a 50-year-old man who relapsed shortly after Hyper-CVAD/MA therapy with leukemic transformation. Despite requiring dose reductions due to prolonged cytopenia

Venetoclax or Pirtobrutinib in Relapsed/Refractory Waldenström Macroglobulinemia: Clinical and Molecular Predictors and Sequencing Implications.

Venetoclax and pirtobrutinib have emerged as two chemotherapy-free options for relapsed or refractory Waldenström macroglobulinemia (WM). However, evidence to guide treatment sequencing or identify molecular subsets most likely to benefit from each agent remains limited. We retrospectively evaluated consecutive WM patients treated with venetoclax or pirtobrutinib. Major response rate (MRR) and progression-free survival (PFS) were assessed for each agent, with predictors of response and PFS analyzed using logistic and Cox regression. Comparative efficacy was examined in unmatched analyses and i

The ATF4/PSAT1/JNK signaling axis suppresses ferroptosis to drive venetoclax resistance in AML.

Metabolic reprogramming has emerged as a key driver of therapy resistance in acute myeloid leukemia (AML). Here, we identify phosphoserine aminotransferase 1 (PSAT1) as a critical metabolic determinant of venetoclax (VEN) resistance through the suppression of ferroptosis. PSAT1 was consistently upregulated in VEN-resistant cell lines and relapsed patient samples. Mechanistically, the transcription factor ATF4 directly bound the PSAT1 promoter, enhancing its expression and subsequently promoting glutathione synthesis, depleting the labile iron pool, and attenuating lipid peroxidation. Oxaliplatin/Norcantharidin Prodrug Co-Loaded Nanoparticles: Synergistic DNA-Repair Blockade Plus PD-L1 Downregulation for Inhibit Tumor Growth and Metastasis.

The combination of phototherapy with oxaliplatin (OXA) represents a potent strategy for triggering immunogenic cell death (ICD) and elicit robust antitumor immunity. In this study, we construct IROTD nanoparticles (IROTD NPs) through self-assembly of a norcantharidin (NCTD)-OXA-conjugated polymer (P2) and the near-infrared (NIR) photosensitizer DTIR780. Under 808 nm laser irradiation, IROTD NPs generate heat and reactive oxygen species (ROS) while release OXA and NCTD, result in severe DNA damage and potent induction of ICD within tumor cells. Concurrently, NCTD inhibits DNA repair mechanisms

Extended Versus Standard Hypomethylating Agent Dosing in Combination With Venetoclax in Patients With Newly Diagnosed Acute Myeloid Leukemia.

The optimal duration of hypomethylating agent (HMA) therapy combined with venetoclax (Ven) in newly diagnosed acute myeloid leukemia (ND-AML) remains uncertain. Standard of care (SOC) uses 5-day decitabine (Dec-5) or 7-day azacitidine (Aza-7), while extended HMA regimens (e.g., 10-day decitabine [Dec-10] during cycle 1) have also been explored, although direct comparative data between these approaches are limited. We conducted a large retrospective analysis of 335 patients with ND-AML treated at five US centers with Dec-10/Ven (n = 102) or SOC HMA/Ven (n = 233). Baseline characteristics were l
