Longitudinal associations between PM2.5 with gestational diabetes mellitus mediated by gut microbiome and potential mechanism: based on a prospective pregnant women cohort in China.

BACKGROUND: Exposure to particulate matter pollution with aerodynamic diameters < 2.5 μm (PM2.5) has been linked to gestational diabetes mellitus (GDM) and gut microbiota dysbiosis. However, few studies have illustrated the associations among PM2.5 exposure, gut microbiota, blood metabolites, circular RNAs (circRNAs) and GDM risk. This study aimed to explore the moderating effects of the gut microbiota on the association between PM2.5 exposure and GDM, and to analyze the interaction network of PM2.5 exposure, gut microbiota, blood metabolites and circRNAs. METHODS: Participants (n = 1,248) we…

Matched case-control study of air pollution and infant mortality in North Carolina.

BACKGROUND: Infant mortality is an indicator of overall population health but has been evaluated in relatively few studies of air pollution. We estimated associations between postnatal exposure to air pollution and postneonatal infant mortality in a North Carolina birth cohort. METHODS: We identified cases of infant mortality with dates of birth between 2003 and 2015. For each case, we randomly selected 10 controls from at-risk infants without replacement, matched on year of birth and geographic region. Air pollutant concentrations - fine particulate matter (PM2.5); 5 PM2.5 components; nitrog…

Water-insoluble components of PM2.5 are major triggers of pulmonary ferroptosis by inducing mitochondrial damage via Nrf2/Prdx6 pathway.

Fine particulate matter (PM2.5) is a major environmental pollutant that poses a significant risk to respiratory health. Ferroptosis represents a key pathogenic mechanism underlying this process. However, the relative contributions of specific PM2.5 components and the potential mechanisms through which they induce pulmonary ferroptosis remain unclear. This study aimed to compare pulmonary ferroptosis triggered by PM2.5 components and to elucidate the underlying mechanisms via in vivo and in vitro experiments. The cytotoxicity of PM2.5 exposure on BEAS-2B cells was measured through the CCK-8 ass…

Subchronic Residual Oil Fly Ash (ROFA) exposure induces oxidative stress in brain, lung, and cardiac tissues and promotes neuroinflammation, with partial attenuation by taurine.

BACKGROUND: Exposure to particulate matter (PM₂.₅), including Residual Oil Fly Ash (ROFA), triggers oxidative stress and inflammation. Taurine, an antioxidant amino acid, may protect against these effects, but its efficacy against subchronic ROFA exposure is unclear. This study evaluated taurine's protective role in ROFA-induced multi-organ damage and neurobehavioral changes. METHODS: Male Wistar rats received ROFA (250 µg/day, intranasally) for six weeks, with or without taurine (400 mg/kg/day). Oxidative markers (CAT, SOD, TBARS) were measured in the brain, lungs, heart, and liver. Cytokine…

Progressive lung fibrosis in aged mice induced by repetitive bleomycin: Exacerbation by long-term PM₂.₅ inhalation.

Idiopathic pulmonary fibrosis (IPF) is a progressive and fatal interstitial lung disease marked by spatially and temporally heterogeneous fibrotic remodeling. Although epidemiological studies suggest that prolonged exposure to fine particulate matter (PM2.5) accelerates IPF progression, the molecular mechanisms underlying this effect remain poorly defined. The objective of this study was to establish an experimental model that better reflects the chronicity and pathological complexity of IPF. To this end, we developed a repetitive low-dose bleomycin (BLM) model in aged mice and subjected them …

PM2.5 augments cigarette smoke-induced lung inflammation in mice by driving a stronger immune response: Potential beneficial effects of oleanolic acid.

Though cigarette smoke (CS) is primary risk factor for Chronic obstructive pulmonary disease (COPD), rising air pollution and higher concentrations of particulate matter (PM2.5) in ambient air contribute substantially to COPD cases, particularly in smokers. However, the pathogenesis of COPD upon dual exposure to CS and PM2.5 is not entirely known. Therefore, the impact of combined exposure to CS (9 cigarettes/day for 4 days) and PM2.5 (single dose of 50 μg) on COPD pathogenesis was examined using mouse model in order to understand the key players behind the process. The data suggest that singl…

Traffic-emitted ultrafine particles disrupt macrophage efferocytosis and resolution of allergic lung inflammation.

BACKGROUND: Particulate matter (PM) in air pollution is a major health concern. PM includes ultrafine particles (UFPs; PM0.1 and particles of ≤0.1 μm), which can evoke lung inflammation. However, the impact of UFPs on the resolution of lung inflammation, a potentially important link to chronic inflammatory diseases, remains to be determined. OBJECTIVE: We sought to investigate the impact of UFPs on the resolution of allergic lung inflammation and to identify potential therapeutic interventions to mitigate these effects. METHODS: UFPs were collected from urban Boston. Using a mouse model, tra…

Genetic sensitivity moderates the effects of joint exposure to air pollution on depression risk: a genome-wide by environment interaction analysis.

How ambient air pollutants and genetic factors interact to influence depression risk remains unclear. This study aims to investigate how genetic factors regulate the relationship between air pollution and depression. Using UK Biobank data, an air pollution index was constructed from NO2, NOx, PM2.5, PM2.5-10 and PM10 to assess joint exposure. A genome-wide interaction analysis identified 189 air pollution-sensitive SNPs (APS-SNPs) enriched in neurodevelopment. Polygenic scores (PGS) constructed by APS-SNPs showed significant interaction with the air pollution index, NO2, NOx, PM2.5 and PM10 on
