Target-oriented anti-implantation approaches for pregnancy interception: experiences in the rhesus monkey model.

Blastocyst implantation is a critical process in the establishment of pregnancy in eutherian mammals and requires a harmonious symbiosis bet … ry cytokine, together with interleukins 1 and 6, during the process of implantation-placentation in primates, and its immunoneutralization with MAb resulted in inhibition (p<.04) of pregnancy establishment in the rhesus monkey. However, timed administration of l-NAME or aminoguanidine failed to inhibit blastocyst implantation in a significant manner. Also, …

Distinct Functional Circuits in the Superior Colliculus and Their Connection to Behavior.

The superior colliculus is a conserved midbrain structure that converts sensory input into motor output to guide orienting, defensive, and a … ts, contexts, and brain states. Drawing primarily on work in mice, and using selected findings from primates, birds, and other vertebrates for comparison, we argue that the superior colliculus is organized by three principles: laminar organization, topographic alignment of sensory and motor maps, and modular specialization of cell types and pathways. Toget …

Cutting a nerve of the hand alters the organization of digit maps in primary somatosensory cortex.

In non-human primates, forelimb nerve transection and repair alters the otherwise orderly and highly conserved organization of the digit maps in primary somatosensory cortex (S1). These same changes are presumed to occur in humans and to have meaningful implications for patient recovery. Here, using functional magnetic resonance imaging, we map digit responses in 21 patients with surgically repaired hand nerves and 30 controls. In S1 contralateral to the repaired hand, digit map arrangement is systematically altered and response amplitudes are elevated. At the individual level, map organizatio…

Preclinical assessment of broadly neutralizing HIV-1 antibody BNT351 with optimized pharmacokinetics and potent antiviral activity.

Broadly neutralizing antibody (bNAb) 1-18 is a promising tool for future clinical strategies against HIV-1 infection. To enhance 1-18's clinical potential, we introduced half-life-extending LS mutations and evaluated the resulting investigational bNAb candidate, BNT351. LS mutations increased the affinity of BNT351 to human neonatal Fc receptor by 20-fold, resulting in a long half-life of 10-14 days in Tg32 mice and 18 days in non-human primates, with a predicted human half-life of ∼50 days. BNT351 retained 1-18's exceptional neutralization potency and breadth against a 119 multiclade panel, a…

Pragmatic representations of self- and others' action in the monkey putamen.

Social coordination in primates relies on parieto-frontal networks encoding self- and others' actions. These areas send convergent projections to the putamen, but its role in representing self- and others' actions remains unknown. We recorded neuronal activity from anatomically characterized putamen regions of two male macaques during a Mutual Action Task (MAT), where a monkey and a human took turns grasping a multi-affordance object based on sensory cues. Cortico-striatal synaptic input, indexed by local field potentials, mirrored known cortical dynamics during sensory instructions and moveme…

Maternal obesity decreases offspring lifespan.

Data in mice, nonhuman primates, and in humans demonstrate that exposure to maternal obesity increases the risk of multiple diseases in offspring. However, little is known about the aging effects of maternal obesity on the offspring. This study shows that maternal obesity significantly reduced the lifespan of both male and female mice born to obese dams despite being weaned onto a healthy diet at three weeks of age. This reduction in longevity was linked to an increase in age-related fibrotic pathology across multiple organs, e.g., liver, heart, and kidney. Gompertz analysis of the lifespan da…

Overcoming the blood-brain barrier using central nervous system-accessing lipid nanoparticles for enhanced mRNA therapeutics.

Messenger RNA (mRNA) therapeutics hold potential for central nervous system (CNS) disease treatment. However, the blood-brain barrier (BBB) presents a major obstacle, preventing efficient delivery of mRNA into the brain. To overcome this challenge, we designed, synthesized, and tested a series of ionizable lipids and formulated them into CNS-accessing lipid nanoparticles (CA LNPs) to deliver mRNA. The lead candidate among them, CA2d LNP, demonstrated efficient mRNA delivery across the BBB following intravenous injection. In wild-type mice, Ai14 mice, and nonhuman primates, CA2d LNPs effectivel…

A single-cell atlas of porcine hematopoietic development.

Domestic pigs are both a globally important agricultural species and an increasingly valuable biomedical model that bridges the gap between rodents and non-human primates. Thus, a thorough understanding of the developing porcine immune system is important to improve pig health and advance preclinical research. Here, we coupled single-cell transcriptomic analyses with T and B cell receptor profiling to create a comprehensive cell atlas of pig hematopoietic development across fetal liver, bone marrow, and thymus tissues from fetal to adult life. We highlight tissue-specific and age-related featu…

Skeletal-muscle-targeted non-viral delivery of full-length DMD mRNA for Duchenne muscular dystrophy.

Duchenne muscular dystrophy (DMD) is a severe, progressive muscle-wasting disorder caused by mutations in the DMD gene, which encodes dystro … s muscle function. We further demonstrate the safety and biocompatibility of DMD t-EVs in non-human primates, supporting their translational potential. These findings highlight the promise of mRNA-loaded extracellular vesicles as a therapeutic platform for treating genetic disorders involving large, difficult-to-package genes.

Fatty-acid-based antimiR-23b delivery in the DMSXL model: A potential therapeutic strategy for brain dysfunction in myotonic dystrophy type 1.

Myotonic dystrophy type 1 (DM1) is a severe neuromuscular disorder caused by CTG repeat expansions in the DMPK gene, leading to the formation of toxic RNA foci that sequester essential splicing regulators MBNL1/2. Beyond muscle impairment, DM1 affects also the brain, leading to significant cognitive deficits, behavioral abnormalities, and intellectual disabilities. This study evaluates the therapeutic potential of the lipid-conjugated antimiR-23b, X82108, designed to promote MBNL1/2 upregulation through inhibition of miR-23b. Systemic administration of X82108 in mice and non-human primates eff…

Species-dependent activities of the PINK1-parkin axis.

In vitro studies have established that PTEN-induced putative kinase 1 (PINK1) and parkin are central regulators of mitophagy, and loss-of-function mutations in either gene can cause early-onset Parkinson's disease (PD). Although various animal models, including mice and pigs with PINK1 or PRKN knockout, have largely failed to recapitulate the neurodegeneration seen in PD patients, effective knockdown of PINK1 or PRKN in non-human primates, when achieving substantial protein depletion, does induce dopaminergic neuron loss in the substantia nigra and α‑synuclein pathology, suggesting that both t…

Yellow fever beyond the Amazon: surveillance challenges and the role of Callithrix spp. in sylvatic cycles in Brazil.

Yellow fever virus (YFV) remains a major public health concern in tropical and subtropical regions of the Americas and Africa. Since late 2016, sylvatic yellow fever (YF) outbreaks and sporadic cases affecting both humans and non-human primates (NHP) have been recorded in Southern Brazil. These events raised concerns about viral persistence beyond the Amazon Basin, where YFV is endemic, and the potential reemergence of urban transmission. Neotropical NHPs are central to the ecology of sylvatic YF, but many aspects of their infection dynamics and species-specific roles remain poorly understood.…

Molecular and serological evidence of systemic Treponema pallidum infection in wild western chimpanzees.

Human treponematoses are chronic diseases caused by closely related Treponema species. Increasing evidence of yaws-like Treponema pallidum subsp. pertenue infections in nonhuman primates raises concerns about pathogen persistence in natural ecosystems. Here, we investigated treponemal infection in western chimpanzees (Pan troglodytes verus) using molecular and serological analyses of postmortem skeletal remains from three deceased individuals. Anti-T. pallidum antibodies were detected in all chimpanzees using validated serological assays, while molecular assays identified T. pallidum DNA in a …

Natural Hepacivirus Infection in Tree Shrews: A Call for Routine Screening in Hepatitis Virus Research.

Hepatitis viruses continue to pose major global health challenges, necessitating the development of reliable and well-characterized experimental models. Tree shrews are increasingly recognized as a valuable small animal model because of their natural susceptibility to hepatitis viruses and close phylogenetic relationship with primates. Recent identification of a high prevalence of natural hepacivirus infections in tree shrews underscores the urgent need for routine viral screening of the animals used in hepatitis studies. Undetected infections may confound experimental results, undermine data …
