Pro-Apoptotic and Cytotoxic Effects of Melittin on HL-60 Acute Promyelocytic Leukemia Cells: Implications for Retinoid-Independent Therapy.

BACKGROUND: Acute promyelocytic leukemia (APL) is a subtype of acute myeloid leukemia driven by the PML/RARα fusion protein. Standard treatment with all-trans retinoic acid (ATRA) combined with chemotherapy is effective, but resistance and adverse effects remain significant challenges. Melittin, the primary peptide component of bee venom, has demonstrated potent anticancer activity across multiple leukemia subtypes through mitochondrial-dependent mechanisms. Building upon this established evidence, we investigated melittin's therapeutic potential in APL to address the specific clinical challen

Characterization of tau prion seeding activity and strains from formaldehyde-fixed tissue.

Tauopathies such as Alzheimer's disease (AD) feature progressive intraneuronal deposition of aggregated tau protein. The cause is unknown, but in experimental systems trans-cellular propagation of tau pathology resembles prion pathogenesis. Tau aggregate inoculation into mice produces transmissible pathology, and tau forms distinct strains, i.e. conformers that faithfully replicate and create predictable patterns of pathology in vivo. The prion model predicts that tau seed formation will anticipate neurofibrillary tau pathology. To test this idea requires simultaneous assessment of seed titer

Levels of hemoglobin and lipid peroxidation metabolites in blood, catalase activity in erythrocytes and peak expiratory flow rate in subjects with passive exposure to tobacco smoke.

INTRODUCTION: Exposure to tobacco smoke is an extremely important risk factor determining the development of respiratory diseases, cardiovascular diseases and cancer. Passive exposure is common and often not realized by the exposed subjects. Markers of tobacco smoke exposure are nicotine metabolites, i.e. cotinine and trans-3'-hydroxycotinine. OBJECTIVES: The objective of the study was to assess the level of passive exposure to tobacco smoke among students and the exposure impact on the blood hemoglobin level, peak expiratory flow (PEF), lipid peroxidation level and antioxidant enzyme activit

Comparative disposition, receptor affinity, and teratogenic activity of sulfon arotinoids.

To investigate the relationship between sulfon arotinoid biotransformation and teratogenic activity, the potency of the ethyl (Ro 15-1570) and methyl (Ro 14-9706) arotinoid sulfones and their in vivo disposition in pregnant hamsters were studied. Administration of Ro 15-1570 was teratogenic, but Ro 14-9706 showed no such activity. Total absorbed doses of the ethyl and methyl sulfones (measured as maternal plasma AUC) were very similar. Total delivered dose of Ro 14-9706 to liver and lung was 120-160% that of Ro 15-157, and Ro 14-9706 was transferred in greater amounts to the embryo as well. Pl

Epstein-Barr virus (EBV) nuclear antigen 6 induces expression of the EBV latent membrane protein and an activated phenotype in Raji cells.

Epstein-Barr virus (EBV) nuclear antigen (EBNA) 6 (also known as 3c) is a latent nuclear protein with an M(r) of about 160K which is invariably expressed in EBV-immortalized B cells. It includes a putative basic leucine zipper domain; as such it is a good candidate for a regulator of viral gene expression. More than 75% of the EBNA 6 coding sequence is deleted from viral genomes carried in the Burkitt's lymphoma (BL) tumour-derived cell line, Raji. Thus although Raji cells express normal levels of the remaining five EBNAs and low levels of latent membrane protein (LMP), EBNA 6 protein is compl

Telmisartan ameliorates insulin sensitivity by activating the AMPK/SIRT1 pathway in skeletal muscle of obese db/db mice.

BACKGROUND: Telmisartan is a well-established angiotensin II type 1 receptor blocker that improves insulin sensitivity in animal models of obesity and insulin resistance, as well as in humans. Telmisartan has been reported to function as a partial agonist of the peroxisome proliferator-activated receptor (PPAR) γ, which is also targeted by the nicotinamide adenine dinucleotide (NAD)-dependent deacetylase (SIRT1). Here, we investigated the pathways through which telmisartan acts on skeletal muscle, in vitro as well as in vivo. METHODS: Nine-week-old male db/db mice were fed a 60% high-fat diet

Leukemia: A model for drug development.

Early attempts at preclinical model development for cancer drug development relied heavily on mouse leukemias and lymphomas to detect agents with antitumor activity. These models were applied clinically, and the concepts of combination chemotherapy, remission induction, and maintenance treatment all developed in leukemia. Subsequently, the predominant impact of cytogenetics on probability of response to treatment and survival was first illustrated in leukemia. The power of a single drug to change the natural history of a disease was noted in acute myelogenous leukemia, in which a previously in

Interleukin 6 activates androgen receptor-mediated gene expression through a signal transducer and activator of transcription 3-dependent pathway in LNCaP prostate cancer cells.

Interleukin 6 (IL-6) is a cytokine that regulates not only immune and inflammatory responses but also the growth of some tumors, including prostate carcinomas. IL-6 signals through Janus kinase, signal transducer and activator of transcription 3 (STAT3), and mitogen-activated protein kinase and is also able to induce androgen receptor (AR)-mediated gene activation in prostate cancer, which is an important process in prostate cancer androgen-independent progression. We now show that IL-6-induced AR-mediated gene activation requires the activation of STAT3 by IL-6 in LNCaP prostate cancer cells.

Inhibition of WNT signaling sensitizes CRC cells to anti-tumor effects of retinoids: basis for new drug combinations in oncology.

Our Goal is to sensitize human colorectal cancer (CRC) to the differentiation-inducing effects of retinoic acid (RA) agents by suppressing WNT signaling. Previously, we reported that the WNT and RA pathways are functionally linked, and in CRC, APC mutation generates a WNT:RA imbalance leading to incomplete differentiation and stem cell (SC) overpopulation. Hypothesis: Therapeutically inhibiting WNT signaling enhances the ability of retinoid agents to induce growth suppression and differentiation. We tested drug combinations of: (1) anti-WNT agents (Sulindac & Piroxicam) that produce anti-tumor

Modular CRISPR-Cas12a-Activated Gold Nanoparticle Assay for Rapid Visual Detection of Hepatocellular Carcinoma-Related miRNAs.

MicroRNAs (miRNAs) are emerging biomarkers for early hepatocellular carcinoma (HCC) detection, but most CRISPR-Cas12a assays rely on reverse transcription and preamplification and often lack simple visual readouts. Here, we develop a reverse transcription-free, cleavage-guided strategy that converts miRNA recognition into visual outputs. In the presence of the target miRNA, a scaffold RNA forms an active crRNA that triggers Cas12a trans-cleavage using a preformed dsDNA activator, enabling two readout formats. In a homogeneous non-crosslinking colorimetric assay (mC-NCA), Cas12a cleavage regula

Rigidity-Responsive Fluorescence Polarization Detection of Aflatoxin B1 via Programmable RCA-Coupled CRISPR/Cas12a and a Conformation-Restricted Depolarization Reporter.

Aflatoxin B1 (AFB1) is one of the most toxic and carcinogenic mycotoxins, and its trace-level determination in complex food matrices remains a major analytical challenge. Conventional chromatographic methods, while highly accurate, rely on expensive instrumentation and labor-intensive sample pretreatment, whereas most CRISPR/Cas-based biosensors depend on fluorescence intensity turn-on readouts that are vulnerable to matrix autofluorescence, photobleaching, and signal instability, limiting their reliability in real samples. Herein, we propose a rigidity-responsive fluorescence polarization (FP

Bioorthogonally Active Silk Fibroin Patch Facilitate Immunomodulatory Prodrug Activation for Tissue Repair.

Current silk fibroin (SF)-based tissue repair patches often exhibit limited immunomodulatory efficacy, exacerbating inflammatory responses and compromising functional tissue integration. To address this limitation, we developed a bioorthogonal prodrug activation strategy within an SF patch via tetrazine functionalization (SF-Tz). This platform enables the spatiotemporally controlled release of trans-cyclooctene (TCO)-caged dexamethasone (TCO-Dex) via inverse electron-demand Diels-Alder reactions. The SF-Tz platform was engineered through optimized carboxylation and tetrazine conjugation protoc

An electrochemiluminescence biosensor based on the hairpin-mediated exponential amplification and CRISPR/Cas12a amplification for ultrasensitive detection of MMP-2.

In this study, we report a novel electrochemiluminescence (ECL) biosensor for the ultrasensitive detection of matrix metalloproteinase-2 (MMP-2), an important biomarker associated with tumor invasion and metastasis. The biosensor integrates hairpin-mediated exponential amplification with CRISPR/Cas12a-based trans-cleavage for dual-stage signal amplification. In this design, MMP-2 specifically cleaves a peptide sequence (GPLG↓VRGK) on the DNA hairpin probe (HP1), releasing an initiator peptide nucleic acid (PNA) that triggers hairpin-mediated exponential amplification reaction. The amplified DN

Pulsed synchrony regulation of intraocular and cerebrospinal fluid pressure: a novel paradigm for glaucoma pathogenesis and treatment.

Glaucoma, the leading cause of irreversible blindness worldwide, is characterized by the progressive loss of retinal ganglion cells (RGCs) and their axons. While elevated intraocular pressure (IOP) is a core risk factor, the pathogenesis of normal-tension glaucoma (NTG) remains unclear, as static IOP is within the normal range. Based on circadian fluctuations of IOP and cerebrospinal fluid pressure (CSFP), and the pressure-dependent function of the ocular glymphatic system, we propose the "dynamic trans-lamina cribrosa pressure difference (TLCPD) imbalance" hypothesis. This hypothesis posits t

Effect of Interleukin-6 Inhibitors on Rheumatoid Arthritis Pain: Overview of Evidence and Mechanistic Pathways.

Rheumatoid arthritis (RA) is an autoimmune systemic disease in which pain remains a major and often refractory symptom even after clinical remission of the disease. Although historically attributed to joint inflammation, recent evidence reveals a multifactorial pathogenesis of RA pain, involving peripheral sensitization, central sensitization, and neuroimmune crosstalk. In these mechanisms, interleukin-6 (IL-6) plays a central role not only as one of the inflammatory mediators but also as a classic and trans-signaling modulator for pain. This review synthesizes current mechanistic and clinical

Platinum(IV) oxaliplatin analog containing axial benzoate ligands induces mitochondrial hyperpolarization and caspase-dependent apoptosis in pancreatic cancer cell models.

This study investigates the molecular mechanisms underlying the antiproliferative effects of the platinum(IV) complex trans-[Pt(OBz)2(O,C-10-BzODA)(1R,2R-DACH)] (complex 1; OBz = benzoate, 10-BzODA = 10-benzoyloxy-2-decenoate, DACH = diaminocyclohexane) in pancreatic cancer cell models. Initial findings revealed that complex 1 exhibits substantial antiproliferative activity, positioning it as a promising therapeutic agent for pancreatic cancer, where effective treatment options remain limited. Notably, complex 1 demonstrates significantly greater efficacy compared to platinum(II) drugs, such a

Increase in Ia Afferent Synaptic Excitation of SOD1 G93A Mouse Motoneurons by 2-Week Anodal Trans-Spinal Direct Current Stimulation Does Not Ameliorate the Cellular Burden of the Disease.

An imbalance between cells' intrinsic excitability and synaptic excitation levels underlies the spinal motoneuron (MN) pathophysiology in Amyotrophic Lateral Sclerosis. Recently, a transient restoration of the deficient Ia synaptic excitation of spinal MNs in the presymptomatic SOD1 G93A mice was achieved by applying a single trans-spinal direct current stimulation (tsDCS) session. Here we investigate whether two-week repeated tsDCS applied to presymptomatic SOD1 G93A animals can permanently alter spinal MN synaptic excitation levels and in this way affect intracellular metabolic pathways and

Vascular endothelial growth factor A: friend or foe in the pathogenesis of HIV and SARS-CoV-2 infections?

This review article discusses the role of vascular endothelial growth factor A (VEGF-A) in the pathogenesis of SARS-CoV-2 and HIV infection, both conditions being renowned for their impact on the vascular endothelium. The processes involved in vascular homeostasis and angiogenesis are reviewed briefly before exploring the interplay between hypoxia, VEGF-A, neuropilin-1 (NRP-1), and inflammatory pathways. We then focus on SARS-CoV-2 infection and show how the binding of the viral pathogen to the angiotensin-converting enzyme 2 receptor, as well as to NRP-1, leads to elevated levels of VEGF-A an

An integrated multi-mode detection platform based on CRISPR/Cas 12a and aptamers for ultra-sensitive identification of sulfamethazine and genes associated with sulfonamide resistance.

The production and buildup of sulfamethazine (SMZ) and resistance genes for sulfonamide antibiotics (sul1) pose a serious risk to environmental and public health safety. Creating advanced sensing systems that are both highly sensitive and selective for the prolonged observation of SMZ concentrations in the environment, along with the quantification of sul1 gene prevalence, aims to identify trends in resistance, posing a considerable challenge. Here, we devised a platform (SMZ-sul1 multi-mode detection platform) that allows for the fluorescence detection of SMZ in environmental samples. This is

Nanoformulation of a Pin1 inhibitor potentiates the efficacy of liposomal doxorubicin in second-line therapy for ovarian cancer.

The second-line therapy for high-grade serous ovarian cancer (HGSOC) patients is generally ineffective. Drug selection is not aimed at improving overall survival, but rather based on residual toxicities, clinical judgment, and patient adherence. Therefore, the identification of more effective and targeted therapeutic strategies is critically needed. The Peptidyl-prolyl cis-trans isomerase NIMA-interacting 1 (PIN1) has emerged as a key hallmark of cancer progression and represents a promising therapeutic target in ovarian cancer (OC). VS10, a novel PIN1 inhibitor developed by our group, has sho
