The effect of trypanocidal drug treatment on the haematological changes in Trypanosoma brucei brucei infected splenectomised dogs.

Twenty adult mongrel dogs of both sexes were used. Ten of the dogs were splenectomised and the remaining dogs were left intact. Five dogs each from the splenectomised and non-splenectomised (intact) animals were infected intravenously with Trypanosoma brucei brucei while the rest served as uninfected controls. All the infected dogs developed trypanosomosis between Days 4 and 8 postinfection. The packed cell volume, haemoglobin concentration, total red blood cell count and white blood cell count decreased progressively indicating anaemia and leucopenia. The absolute reticulocyte counts were inc

Pharmacomodulations on new organometallic complexes of Ir, Pt, Rh, Pd, Os: in vitro and in vivo trypanocidal study against Trypanosoma brucei brucei.

New organometallic complexes have been synthesized by association of an active organic molecule with a metallic element such as Pt, Rh, Ir, Pd, Os. Their trypanocidal activity was studied in vitro and in vivo against T. b. brucei. The more active compounds were pentamidine derivatives. The Ir- COD-pentamidine complex, and Iridium (I) cationic and organometallic complex showed and in vitro activity at 60 micrograms/l. Moreover, all infected mice were cured by this compound subcutaneously administered in a single dose at 0.5 mg/kg (0.317 mumol/kg). In the same conditions, pentamidine cured all t

Uganda faces health crisis.

This article reports on the health crisis confronting Uganda, which results from the breakdown of disease control. The breakdown of disease control here has led to the reappearance of diseases that were once eliminated. Malaria has come back with full force and now accounts for nearly 20% of all deaths in hospitals. Hospital sources say that 30% of the cases they treat are resistant to chloroquine drugs. Diarrheal diseases have also made a comeback, and UNICEF reports that they are now the second biggest killer and the second cause of hospital consultations. Sleeping sickness has now also reac

Suppressive macrophages occurring in murine Trypanosoma brucei infection inhibit T-cell responses in vivo and in vitro.

Intraperitoneal injection of Trypanosoma brucei AnTat 1.1 into mice of the C3H.He, BALB/c or C57BL/6 strains resulted in impaired immune responses from day 3 onwards, as measured by the reduction in DNA synthesis in spleen cell populations stimulated with concanavalin A (Con-A) in vitro. Adherent cells from the peritoneum (PC) or from the spleen of infected mice, consisting predominantly of macrophages, caused a 60-80% reduction of the Con-A response in spleen cells from syngeneic recipients 3-4 days after transfer in vivo. Adherent PC from irradiated or athymic mice were equally suppressive.

Detection of circulating trypanosomal antigens in Trypanosoma evansi-infected animals using a T. brucei group-specific monoclonal antibody.

An antigen-detection enzyme immunoassay based on a T. brucei group-specific monoclonal antibody was used for the detection of circulating antigens in several animal species experimentally infected with T. evansi stocks from Sudan, Indonesia, Thailand and South America. Circulating antigens were detected as early as 6 days after infection, and they persisted throughout the observation period of up to 60 days postinfection. In an analysis of sera from naturally infected water buffaloes from Thailand, the test identified all the animals with positive parasitological findings, and 3 additional cas

Polyclonal B-cell-activating factors produced by spleen cells of mice stimulated with a cell homogenate of Trypanosoma gambiense.

The effect of injection of a cell homogenate of Trypanosoma gambiense into mice on the production of soluble factors responsible for the induction of polyclonal B-cell activation (PBA) by their spleen cells was examined. PBA was induced by injection of the cell homogenate and was also detected in mice treated with either the serum or the spleen cells of mice treated with the cell homogenate. PBA-inducing activity became detectable in the serum and spleen cells as early as 12 h after injection of the cell homogenate, reached a peak on day 2, and then decreased. This activity was also detected i

Pathobiochemical alterations in experimental chronic and acute trypanosomal infection in mice.

During an experimental chronic infection of inbred mice with Trypanosoma congolense several physiological parameters become altered. Splenomegaly followed later by hepatomegaly are predominant. Lactate dehydrogenase and aminotransferase activities of the plasma are elevated, the number of erythrocytes and thrombocytes decreases, whereas monocytic cells are detected in higher concentrations. Gamma-Globulins and transferrin become elevated. Some of the pathobiochemical alterations depend directly on the parasitaemia and are reversed to normal values after chemotherapy with diminazene aceturate (

Complete two-dimensional gel electrophoresis pattern of de novo synthesized acute phase reactants.

The early stages of inflammatory responses are characterized by the rapid synthesis of a heterogenous group of plasma proteins known as acute phase reactants. We show that the complex changes in the serum protein composition of mice in response to infections can easily be analysed by two-dimensional (2D) gel electrophoresis, which allows simultaneous analysis of numerous proteins from small volume samples. While changes in the protein composition can be visualized by silver staining, radiofluorography of gels loaded with serum samples from mice that had been labelled in vivo with 35S-methionin

Identification of an antigen specific to Trypanosoma congolense by using monoclonal antibodies.

Using indirect immunofluorescence assays on acetone-fixed smears of a series of different parasites, we have shown that two monoclonal antibodies bind specifically to Trypanosoma congolense organisms. The antibodies bind to both bloodstream trypomastigotes and procyclic culture forms of the parasite and are thus not stage specific. Immunoprecipitation and immunoblot analysis showed that both monoclonal reagents bound a protein of approximately 31,000 m.w. This antigen appeared to be located on the plasma membrane of T. congolense, but the epitope was not exposed on the surface of living bloods

Comparative Pathogenicity and Semi-quantitative Multi-organ Histopathology of Trypanosoma congolense and Trypanosoma brucei Infection in Mice.

BACKGROUND: African animal trypanosomosis remains a persistent constraint on livestock production in sub-Saharan Africa, yet the combined influence of parasite species, inoculation route and host immune response on disease severity has rarely been examined within a single controlled experiment. We compared Trypanosoma congolense and Trypanosoma brucei infections in BALB/c mice following intraperitoneal or intramuscular inoculation. Four infected groups of fifteen mice and five uninfected controls were followed for parasitaemia, body weight, rectal temperature and survival. Packed cell volume,

Global distribution, host range and prevalence of Trypanosoma vivax: a systematic review and meta-analysis.

BACKGROUND: Trypanosomosis caused by Trypanosoma vivax is one of the diseases threatening the health and productivity of livestock in Africa and Latin America. Trypanosoma vivax is mainly transmitted by tsetse flies; however, the parasite has also acquired the ability to be transmitted mechanically by hematophagous dipterans. Understanding its distribution, host range and prevalence is a key step in local and global efforts to control the disease. METHODS: The study was conducted according to the methodological recommendations of the Preferred Reporting Items for Systematic Reviews and Meta-A

Effect of vegetable oils on the experimental infection of mice with Trypanosoma congolense.

Vegetable oils are frequently used as solvents for lipophilic materials; accordingly, the effects of their components should be considered in animal experiments. In this study, the effects of various vegetable oils on the course of Trypanosoma congolense infection were examined in mice. C57BL/6J mice were orally administered four kinds of oils (i.e., coconut oil, olive oil, high oleic safflower oil, and high linoleic safflower oil) with different fatty acid compositions and infected with T. congolense IL-3000. Oil-treated mice infected with T. congolense showed significantly higher survival ra

Hepatocyte-derived IL-10 plays a crucial role in attenuating pathogenicity during the chronic phase of T. congolense infection.

Bovine African Trypanosomosis is an infectious parasitic disease affecting livestock productivity and thereby impairing the economic development of Sub-Saharan Africa. The most important trypanosome species implicated is T. congolense, causing anemia as most important pathological feature. Using murine models, it was shown that due to the parasite's efficient immune evasion mechanisms, including (i) antigenic variation of the variable surface glycoprotein (VSG) coat, (ii) induction of polyclonal B cell activation, (iii) loss of B cell memory and (iv) T cell mediated immunosuppression, disease

Genetic diversity of trypanosome species in tsetse flies (Glossina spp.) in Nigeria.

BACKGROUND: Trypanosomes cause disease in humans and livestock in sub-Saharan Africa and rely on tsetse flies as their main insect vector. Nigeria is the most populous country in Africa; however, only limited information about the occurrence and diversity of trypanosomes circulating in the country is available. METHODS: Tsetse flies were collected from five different locations in or adjacent to protected areas, i.e. national parks and game reserves, in Nigeria. Proboscis and gut samples were analysed for trypanosome DNA by molecular amplification of the internal transcribed spacer 1 (ITS1) re

TLR-2 and MyD88-Dependent Activation of MAPK and STAT Proteins Regulates Proinflammatory Cytokine Response and Immunity to Experimental Trypanosoma congolense Infection.

It is known that Trypanosoma congolense infection in mice is associated with increased production of proinflammatory cytokines by macrophages and monocytes. However, the intracellular signaling pathways leading to the production of these cytokines still remain unknown. In this paper, we have investigated the innate receptors and intracellular signaling pathways that are associated with T. congolense-induced proinflammatory cytokine production in macrophages. We show that the production of IL-6, IL-12, and TNF-α by macrophages in vitro and in vivo following interaction with T. congolense is dep

Recombinant and native TviCATL from Trypanosoma vivax: Enzymatic characterisation and evaluation as a diagnostic target for animal African trypanosomosis.

African animal trypanosomosis (nagana) is caused by tsetse-transmitted protozoan parasites. Their cysteine proteases are potential chemotherapeutic and diagnostic targets. The N-glycosylated catalytic domain of Trypanosoma vivax cathepsin L-like cysteine protease, rTviCATLcat, was recombinantly expressed and purified from culture supernatants while native TviCATL was purified from T. vivax Y486 parasite lysates. Typical of Clan CA, family C1 proteases, TviCATL activity is sensitive to E-64 and cystatin and substrate specificity is defined by the S2 pocket. Leucine was preferred in P2 and basic
