PURPOSE OF REVIEW: Follicular lymphoma is a diverse disease, with a diverse variable biology, clinical presentation, and prognosis. The identification of factors that can predict the specific outcome of follicular lymphoma patients remains an area of need. Here, we review the significant advances made in follicular lymphoma prognosis in the last two decades, particularly with the advent of new genetic models.
RECENT FINDINGS: Tumor burden remains an important predictor of prognosis and is still a standard upon which treatment initiation decisions are made. Clinical prognostic indices, includi

Immune checkpoint inhibitors (ICIs) have revolutionized anticancer therapy due to their long-term clinical benefits and immune boosting mechanisms. However, despite their consistent therapeutic effects, the use of ICIs is associated with a spectrum of adverse events due to their autoimmune and auto-inflammatory actions. These adverse events can affect any organ system, including the endocrine, neurologic, gastrointestinal, cardiac, skin, pulmonary, and musculoskeletal systems. Of the immune-related adverse events (irAEs), rheumatic complications are common and appear to be distinct from irAEs

Cancer-associated fibroblasts (CAFs) are a key component of the tumour microenvironment and have been shown to play an important role in the progression of cancer. To probe these tumour-stroma interactions, we incorporated CAFs derived from head and neck cancer patients and squamous carcinoma cells of the hypopharynx (FaDu) into the Tissue Roll for the Analysis of Cellular Environment and Response (TRACER) platform to establish a co-culture platform that simulates the CAF-tumour microenvironmental interactions in head and neck tumours. TRACER culture involves infiltrating cells into a thin fib

Cancer cells do create hostile microenvironment (deprivation of nutrients, accumulation of acidity, anoxic habitat). Those cells are not only adapted to this sanctuary environment, blunting of immunity but also, grow, migrate to the distal area (metastasis) and communicate with each other in a unique population structure and organization too (clonal expansion). The adaptation requirements push those types of adaptable cells (cancer cells) to be primitive cells. The prevailing pharmacological approach in treating cancer is developing a chemotherapeutic agent that acts on rapidly proliferating c

Host tissue microenvironment plays key roles in cancer progression and colonization of secondary organs. One example is ovarian cancer, which colonizes the peritoneal cavity and especially the omentum. Our research indicates that the interaction of ovarian cancer cells with the omental microenvironment can activate a stress-kinase pathway involving the mitogen-activated protein kinase kinase 4 (MKK4). A combination of clinical correlative and functional data suggests that MKK4 activation suppresses growth of ovarian cancer cells lodged in omentum. These findings prompted us to turn our focus t

Metabolic crosstalk between tumor cells, cancer-associated fibroblasts, and immune populations promotes immunosuppression through nutrient competition and accumulation of metabolites such as lactate and lipid-derived mediators. Current cancer vaccines mainly focusing on inducing cancer cells-specific immune responses showed very limited efficacy because of the extremely complicated tumor microenvironments. Herein, we facilely construct Se nanoadjuvant-based bivalent DNA nanovaccines targeting both melanoma and cancer-associated fibroblast (CAF), based on the crucial roles of CAF in desmoplastic tumors and Se element in immune regulations. Intriguingly, the bivalent Se nanovaccines significantly induce robust bi-specific immune responses such as more activated macrophages, higher population of OVA+FAP+CD8+ T lymphocyt…

Metastasis is a major cause of cancer-related deaths. A dearth of preclinical models that recapitulate the metastatic microenvironment has impeded the development of therapeutic agents that are effective against metastatic disease. Because the majority of solid tumors metastasize to the lung, we developed a multicellular lung organoid that mimics the lung microenvironment with air sac-like structures and production of lung surfactant protein. We used these cultures, called primitive lung-in-a-dish (PLiD), to recreate metastatic disease using primary and established cancer cells. The metastatic

It has been long recognized that skeleton represents one of the most favored metastatic sites for common cancers like breast and prostate. During the last decade the molecular mechanisms that are responsible for the development of bone metastasis have been gradually illuminated. It appears that the bone microenvironment has a pivotal role in this process. Metastatic tumor cells interact with bone triggering a cascade of molecular events that produce osteolytic and/or osteoblastic phenomena. In this review, we summarize and discuss the most significant factors and signaling pathways implicated

Progressive weight loss combined with skeletal muscle atrophy, termed cachexia, is a common comorbidity associated with cancer that results in adverse consequences for the patient related to decreased chemotherapy responsiveness and increased mortality. Cachexia's complexity has provided a barrier for developing successful therapies to prevent or treat the condition, since a large number of systemic disruptions that can regulate muscle mass are often present. Furthermore, considerable effort has focused on investigating how tumor derived factors and inflammatory mediators directly signal skele

BACKGROUND: Colorectal cancer (CRC) is a highly aggressive and heterogeneous malignancy with a poor prognosis. Mitochondria and programmed cell death (PCD) play crucial roles in CRC tumorigenesis and cancer progression. However, the prognostic significance of mitochondrial programmed cell death (MPCD) remains unclear. This study aims to investigate prognostic value of MPCD-related genes.
METHODS: CRC patient data from The Cancer Genome Atlas (TCGA) and MPCD gene sets from previous studies were collected. A prognostic model based on MPCD-related genes was developed using regression analysis an

The pursuit of effective immunotherapy for lung adenocarcinoma (LUAD) is ongoing. Lactylation is a promising target for cancer management; yet role of lactylation-related genes (LRGs) in LUAD is still largely unexplored. This study was designed to identify genes related to lactylation in LUAD to assist in prognostic treatment. The Cancer Genome Atlas (TCGA), Gene Expression Omnibus (GEO) database, and related literature were accessed to download gene expression and clinical information. To screen LRGs and create a prognostic model, univariate Cox, multivariate Cox, and Lasso regression analyse

Early-onset colorectal cancer (EOCRC), defined as diagnosis before the age of 50 years, is increasing globally and is frequently characterized by aggressive biology and a disproportionate burden of liver metastases. This review synthesizes emerging evidence on the distinct molecular, immunologic and clinical features that differentiate EOCRC liver metastases from those arising in older adults. Genomic studies revealed increased chromosomal instability, increased copy number variation burden and unique amplification patterns involving MYC, RAD21, GNAS and MAPK1, alongside altered frequencies of

Oral squamous cell carcinoma (OSCC) is a highly invasive malignancy marked by poor prognosis and therapeutic resistance. Within its tumor microenvironment (TME), cancer-associated fibroblasts (CAFs) and immune cells form a dynamic network that drives tumor progression. CAFs reshape the extracellular matrix, rewire tumor metabolism, and modulate immune surveillance by recruiting regulatory T cells and promoting macrophage polarization. Recent studies have highlighted CAF heterogeneity, identifying functionally distinct subtypes with differential impacts on prognosis and treatment responsiveness…

PURPOSE: The prognostic significance of tumor-infiltrating lymphocytes (TILs) in colorectal cancer (CRC) is well established; however, existing approaches inadequately capture their spatial distribution. We investigated the prognostic implications of TIL spatial distribution in CRC using an artificial intelligence (AI)-based method.
METHODS: A total of 202 patients with stage II-III CRC were included. TIL densities in intratumoral (iTIL) and stromal (sTIL) regions were quantified using AI-based analysis of hematoxylin and eosin (H&E)-stained images. Based on proximity to the tumor-stromal bor

BACKGROUND: Colorectal cancer (CRC) is a leading cause of cancer-related morbidity and mortality worldwide, driven by complex genetic and ep … CRC risk. The immune infiltration analyses suggested that GLYCTK is associated with an immune-cold tumor microenvironment, whereas functional assays indicated that GLYCTK promotes proliferation, migration and invasion in CRC cells. Drug screening identified phenylbiguanide and hydroxyfasudil as candidate compounds targeting GLYCTK-associated networks.
CONCLUSIONS: WST …

Precision-guided therapy is imperative in the battle against pancreatic ductal adenocarcinoma (PDAC), one of the most lethal solid malignancies with limited improvements in survival despite advances in molecular profiling and systemic therapy. While oncogenic drivers such as KRAS, TP53, CDKN2A, and SMAD4 are nearly ubiquitous, their translation into effective targeted therapies has been constrained by profound tumor heterogeneity, a therapy-resistant tumor microenvironment (TME), and a paucity of predictive biomarkers. In parallel, clinical outcomes are increasingly shaped by extrinsic modifie…
