Heterozygous RAB3A variants cause cerebellar ataxia by a partial loss-of-function mechanism.

RAB3A encodes a small GTP-binding protein that is abundant in brain synaptic vesicles and crucial for the release of neurotransmitters and synaptic plasticity. Here, we identified RAB3A as a candidate gene for autosomal dominant cerebellar ataxia by two independent approaches: linkage in a large dominant ataxia family and, in parallel, an untargeted computational genetic association approach, analysing the 100 000 Genomes Project datasets. To validate the role of RAB3A in ataxia, we next screened large rare disease databases for rare heterozygous RAB3A variants in probands with ataxia features

PR/SET domain 1 targeting glutathione peroxidase 4 regulates chronic hepatitis B liver fibrosis through ferroptosis.

OBJECTIVE: Addressing the inhibition and reversal of chronic hepatitis B fibrosis is an urgent global challenge, which highlights the critical need to understand its underlying mechanisms. Inhibiting the activation of hepatic stellate cells (HSCs) is an important strategy for fibrosis reversal. In particular, the induction of ferroptosis in HSCs presents a promising avenue for curtailing liver fibrosis. Therefore, this study explores the influence of PR/SET domain 1 (PRDM1), which is a transcriptional regulator, on the progression of liver fibrosis by regulating HSC ferroptosis through glutath

A missense variant in human perilipin 2 (PLIN2 Ser251Pro) reduces hepatic steatosis in mice.

BACKGROUND & AIMS: Non-alcoholic fatty liver disease (NAFLD) is characterised by the accumulation of lipid droplets (LDs) within hepatocytes. Perilipin 2 (PLIN2) is the most abundant protein in hepatic LDs and its expression correlates with intracellular lipid accumulation. A recently discovered PLIN2 coding variant, Ser251Pro (rs35568725), was found to promote the accumulation of small LDs in embryonic kidney cells. In this study, we investigate the role of PLIN2-Ser251Pro (PLIN2-Pro251) on hepatic LD metabolism in vivo and research the metabolic phenotypes associated with this variant in hum

Outcomes reported in randomized controlled trials for mixed and non-IgE-mediated food allergy: Systematic review.

BACKGROUND: Mixed and non-IgE-mediated food allergy is a subset of immune-mediated adverse food reactions that can impose a major burden on the quality of life of affected patients and their families. Clinical trials to study these diseases are reliant upon consistent and valid outcome measures that are relevant to both patients and clinicians, but the degree to which such stringent outcome reporting takes place is poorly studied. OBJECTIVE: As part of the Core Outcome Measures for Food Allergy (COMFA) project, we identified outcomes reported in randomized clinical trials (RCT) of treatments

The genetics of occupational asthma development among workers exposed to diisocyanates: A systematic literature review with meta-analysis.

Diisocyanates are widely used compounds that pose a safety concern for workers in occupations within the spray-paint, spray-foam insulation, and furniture varnish industries. Epidemiological studies show that only a subset of workers exposed to diisocyanates develop diisocyanate-induced occupational asthma (diisocyanate asthma, DA), indicating that genetic susceptibility may play a role. The purpose of this systematic literature review was to compile and meta-analyze the reported data on genetic susceptibility markers for DA. Three databases (Embase, Pubmed, and Scopus) were searched and 169 n

Prevention and Management of Cow's Milk Allergy in Non-Exclusively Breastfed Infants.

Introduction: The prevention and management of cow milk allergy (CMA) is still debated. Since CMA is much less frequent in breastfed infants, breastfeeding should be stimulated. Method: Literature was searched using databases to find original papers and reviews on this topic. Results: Hydrolysates with a clinical proof of efficacy are recommended in the prevention and treatment of CMA. However, not all meta-analyses conclude that hydrolysates do prevent CMA or other atopic manifestations such as atopic dermatitis. There are pros and cons to consider partially hydrolysed protein as an option fo

Identification and characterization of 40S ribosomal protein S27a as an antimicrobial protein from starfish, Patiria pectinifera.

Antimicrobial proteins/peptides (AMPs) are promising bioactive molecules with potent and broad-spectrum activity, representing a key alternative to conventional antibiotics. Among these host defense molecules, ribosomal proteins represent a largely unexplored reservoir with potential antimicrobial capacity, as recent studies suggest that certain members possess moonlighting antimicrobial activity. Thus, studies on the antimicrobial activity of ribosomal protein subunits broaden our knowledge of these antimicrobial moonlighting proteins and contribute to better strategies to combat antimicrobia

Pathway-level proteomics identifies subcellular trafficking dysregulation as a molecular signature in severe traumatic brain injury.

BACKGROUND: Traumatic brain injury (TBI) remains a leading cause of death and disability worldwide in intensive care units, yet no targeted neuroprotective therapies exist despite over 300 clinical trials. This therapeutic failure stems partly from biological heterogeneity and the limitations of single-biomarker approaches that cannot capture the multifaceted pathophysiology of secondary brain injury. Systems biology approaches examining dysregulated pathways rather than isolated proteins may reveal candidate endotypes suitable for patient stratification and identify potential therapeutic targ

Follistatin-like protein 1 (FSTL1) modulates bone remodeling and attenuates bone loss in a mouse model of postmenopausal osteoporosis.

This study systematically delineates the expression dynamics of follistatin-like protein 1 (FSTL1) in postmenopausal osteoporosis (PMOP) and elucidates its role in skeletal homeostasis. Integrated analyses of clinical serum samples and the GTEx database revealed a pronounced reduction in circulating FSTL1 levels in PMOP patients. In vivo, Fstl1+/- ovariectomized mice displayed exacerbated bone loss-evidenced by diminished BMD and trabecular volume-together with impaired osteoblastic activity and heightened osteoclast activation. Complementary in vitro experiments showed that FSTL1 over-express

Identification of potential SARS-CoV-2 Mpro inhibitors from a Thai natural product database via interaction fingerprint-based virtual screening.

The SARS-CoV-2 main protease (Mpro) is a promising target for coronavirus disease 2019 (COVID-19) therapy due to its high conservation and essential role in viral replication. In this study, 148 crystal structures of SARS-CoV-2 Mpro complexed with non-covalent inhibitors from the Protein Data Bank (PDB) were used to map the inhibitor binding site using protein-ligand interaction fingerprints (IFPs). The analysis revealed that the S2 binding site serves as a key region to anchor most non-covalent inhibitors. This mapping was used as a guide to virtually screen an in-house Thai natural product d

The hsa_circ_0084050/miR-373-3p Axis Controls Bone Sialoprotein-Induced ADAM9 Upregulation to Drive Lung Cancer Progression and Metastasis.

Lung cancer carries a high mortality burden, with metastatic dissemination accounting for much of its poor prognosis. Bone sialoprotein (BSP), a matricellular protein belonging to the SIBLING family, has been associated with tumor invasion; nonetheless, its role in lung cancer remains inadequately characterized. Clinical database analysis indicates that BSP is the most critical SIBLING protein associated with lung cancer proliferation and metastasis. Our clinical data also confirmed that BSP levels are higher in metastatic lung cancer compared to non-metastatic cases. We further demonstrate th

Functional Analyses of SATB2 Variants Reveal Pathogenicity Mechanisms Linked With SATB2-Associated Syndrome.

SATB2-associated syndrome (SAS) is characterized by intellectual disability, neurodevelopmental disorders, cleft palate, and dental abnormalities. SAS is caused by variants in the special AT-rich sequence-binding protein 2 (SATB2), which encodes a transcription factor containing two CUT domains and a homeobox (HOX) domain. Here, we report the case of a 16-year-old male diagnosed with SAS using exome sequencing and investigate the functional consequences of previously reported SATB2 variants, including those in this case. The patient carried a heterozygous missense variant (c.1147G>C, p.A383P)
