BACKGROUND: Although familial Mediterranean fever (FMF) was originally defined as an autosomal recessive disorder, approximately 10-20% of FMF patients do not carry any FMF gene (MEFV) mutations. Fine phenotype characterization may facilitate the elucidation of the genetic background of the so called "FMF without MEFV mutations". In this study we clinically and demographically characterize this subset.
METHODS: MEFV mutation-negative FMF and control patients were recruited randomly from a cohort followed in a dedicated FMF clinic. The control subjects comprised 2 groups: 1. typical population

BACKGROUND: Visceral leishmaniasis (VL), caused by the members of the Leishmania donovani complex, has been responsible for devastating VL epidemics in the Sudan. Multilocus microsatellite and sequence typing studies can provide valuable insights into the molecular epidemiology of leishmaniasis, when applied at local scales. Here we present population genetic data for a large panel of strains and clones collected in endemic Sudan between 1993 and 2001.
METHODS: Genetic diversity was evaluated at fourteen microsatellite markers and eleven nuclear sequence loci across 124 strains and clones.

INTRODUCTION: Autosomal dominant polycystic kidney disease (ADPKD) is one of the most common genetic kidney disorders with the incidence of 1 in 1,000 births. ADPKD is genetically heterogeneous with two genes identified: PKD1 (16p13.3, 46 exons) and PKD2 (4q21, 15 exons). Eighty five percent of the patients with ADPKD have at least one mutation in the PKD1 gene. Genetic studies have demonstrated an important allelic variability among patients, but very few data are known about the genetic variation among Iranian populations.
METHODS: In this study, exon direct sequencing of PKD1 was performed

The authors aimed to get insights into the population structure of non-(sero)typable pneumococci (NTPn), a specific group of natural atypical pneumococci whose identification is often difficult, and which has remained insufficiently studied. A total of 265 presumptive NTPn, isolated between 1997 and 2003 from the nasopharynx of children, were characterized. Strains were confirmed to be pneumococci on the basis of bile solubility, and PCR detection or Southern blotting hybridization of lytA and psaA, genes ubiquitous in this species. Multilocus sequence typing (MLST) was used to exclude two iso

The genetic qualities of laboratory colonies of phlebotomine sand flies have not been compared with field specimens despite 1) probable genetic shifts due to the colonization process and 2) the problems associated with the extrapolation of experimental data derived from colonized organisms to field populations. The present study compared the genetic profiles of five laboratory colonies of geographic strains of the New World sand fly Lutzomyia longipalpis, and contrasted them with field populations. The profiles were based on the variability exhibited with polyacrylamide gels at 14 enzyme loci.

Stanley Garn's book, Human Races (1965), captured the essence of the new genetic concept of ethnic groups as populations which share the same gene pool, shaped by natural selection. Since then new genetic traits, especially DNA polymorphisms, have been employed to build upon his vision. They have proven useful in the forensic sciences, medicine, and the human biology of adaptation, migration, evolution, and phylogeny. Some diseases like sickle cell anemia are controlled by single genes concentrated in a large ethnic group; their alleles are valuable for understanding human biology. Other condi

Parasitic diseases are widely observed in different geographical and climatic regions and constitute significant problems in terms of animal health, public health, and livestock-based production systems. This study aimed to determine the molecular characterization and population genetic characteristics of Coenurus cerebralis and hydatid cyst cases observed in sheep in Siirt province, located in the Southeastern Anatolia Region of Türkiye, based on the mitochondrial cytochrome b (cytb) gene region, which was selected due to its ability to provide a balance between conserved structure and suffic

BACKGROUND: Replicate population pairs that diverge in response to similar selective regimes allow for an investigation of (a) whether phenotypic traits diverge in a similar and predictable fashion, (b) whether there is gradual variation in phenotypic divergence reflecting variation in the strength of natural selection among populations, (c) whether the extent of this divergence is correlated between multiple character suites (i.e., concerted evolution), and (d) whether gradual variation in phenotypic divergence predicts the degree of reproductive isolation, pointing towards a role for adaptat

This case series explores the genetic and clinical consequences of consanguineous marriage in Bahrain. Two pediatric patients born to first-cousin parents were identified and evaluated at tertiary hospitals. Each underwent detailed clinical assessment, imaging, and genetic testing. The first case involved Müllerian duct agenesis, adrenal insufficiency, and primary ovarian failure without a pathogenic variant. The second case featured growth failure and hypopituitarism linked to a heterozygous nonsense mutation in the Sonic Hedgehog (SHH) gene (7q36.3). These findings highlight the broad phenot

Advances in genetic testing have led to the identification of multiple genes associated with a hereditary risk for developing breast and other cancers. One such gene is the ataxia telangiectasia mutated (ATM) gene, which is available on many genetic panels offered to individuals with suspected hereditary risk. Genetic testing can often lead to improved understanding and clarification of risk for developing cancer, as well as allow affected individuals to make informed choices about management, including the adoption of primary prevention strategies and more aggressive screening than typically

BACKGROUND AND OBJECTIVES: The genetic mechanisms underlying hydrocephalus (HC) risk and pathogenesis are diverse and complex. While many human genetics studies of HC have been performed in largely European ancestry patients, the extent to which these genetic mechanisms are conserved in African ancestry populations is not known. Here we apply complementary and convergent human genetics and functional genomics approaches to identify genes and pathways implicated with HC risk in patients of African ancestry.
METHODS: We perform a transcriptome-wide association study (TWAS) in pediatric patients

The mechanisms by which plants tolerate soil contamination have been studied in details in controlled laboratory conditions, but they still remain largely unexplored in natural conditions where mixtures of contaminants are present in soils and their effects might interact with other environmental variables. This is especially true in high-altitude alpine environments, where abiotic stress is naturally heightened, but which so far have received little attention in environmental pollution studies. As we were interested in the tolerance mechanisms at play on very fine spatiotemporal scales for al

Galactosemias (GALAC) are rare inherited metabolic disorders with highly variable prevalence worldwide. Differences in newborn screening (NBS) implementation, diagnostic methodologies, and population-specific genetic factors contribute to heterogeneity in epidemiological estimates. We aimed to map the estimated global GALAC birth prevalence and explore the role of NBS in shaping epidemiological patterns. A scoping review was conducted following PRISMA-ScR guidelines. Studies reporting prevalence or incidence of GALAC were included regardless of country or screening context. Data were extracted

Severe acute malnutrition (SAM) contributes to the death of millions of children under age five annually. SAM is clinically classified as non-edematous SAM (NESAM) or the more severe edematous SAM (ESAM), which is more common in east-central Africa and the Caribbean. The reason some children develop ESAM while others develop NESAM remains unclear; however, recent studies have identified aberrant one-carbon metabolism (OCM) in ESAM relative to NESAM. Here, we assess genetic variants at 103 loci known to influence OCM, and determine their association with ESAM in 711 samples from Jamaica and Mal

BACKGROUND: The associations of rheumatoid arthritis (RA) with risk of site-specific cancers beyond lymphohematopoietic cancer have been scarcely explored. We conducted a Mendelian randomization investigation of the associations of RA with site-specific cancers in European and East Asian populations.
METHODS: Independent genetic variants strongly associated with RA in European and East Asian populations were selected as instrumental variables from genome-wide association studies of 58,284 European individuals (14,361 cases and 43,923 controls) and 22,515 East Asian individuals (4873 cases and

Numerous specific genetic polymorphisms (PM) in the multi-gene families of cytochromes P450 (CYPs) and glutathione S-transferases (GSTs) have been described in the human population in the past decade. For example, one or more PM have been identified in human CYP1A1, CYP1B1, CYP2C9, CYP2C18, CYP2D6, and CYP2E1. Recent studies using cDNA expressed human CYPs have suggested that CYP3A4 is the principal human CYP involved in the oxidation of parathion and probably other organo(thio)phosphate (OP) insecticides and thus PM in this CYP might influence susceptibility to OP. However, although large (>

Globodera pallida poses a major threat to potato production, with management strategies primarily relying on genetic resistance. However, increasing virulence in field populations across Western Europe raises major concerns for G. pallida control. To investigate the evolutionary mechanisms driving this rise in virulence, we propagated 13 field populations on 30 commercial potato varieties. Our findings indicate that the genetic basis of resistance in potatoes is small, with the major resistance conferred by GpaV from Solanum vernei. The wide application of GpaVvrn has led to continuous selecti

Recent advances in genomics technologies have facilitated the introduction of new tools that broaden the scope of applications for genetic data. Preimplantation genetic testing using polygenic risk scores (PGT-P), a technique based on the fact that complex traits are influenced by multiple genes, is one such technology. Polygenic risk scoring involves the analysis of many genes in a single embryo to assess the odds of expressing diseaseand non-disease-related factors. This technique could alleviate subsequent burdens on healthcare systems, as it could lead to a population with reduced polygeni

The Jordanian Alnawar (Banu Murra) community represents a socially marginalized group within the Middle East. The Alnawar community has long faced systematic discrimination based on occupational stereotyping and hereditary stigma. While the Alnawar people affirm their Arab ancestry, alternative theories have proposed a Roma/(Gypsy) or Dom origin. In this study, we investigated their genetic identity and paternal ancestry by genotyping 27 Y-STR markers and assessing their phylogenetic relationships with selected regional populations. The results revealed a predominant frequency of the predicted

Primary Sjögren's syndrome (pSS) is an autoimmune disease characterized by inflammation and damage to salivary and lacrimal glands. Its etiology involves both genetic and environmental factors. Among susceptibility genes, IRF5 has been highlighted in European populations, but evidence in non-European groups remains limited. This study evaluated whether IRF5 variants rs2004640G/T, rs2070197T/C, rs10954213G/A, and rs59110799G/T are associated with pSS susceptibility, clinical manifestations, or the presence of autoantibodies in a Mexican population. The diagnosis was confirmed by rheumatologists
