Nematode-derived proteins suppress proliferation and cytokine production of antigen-specific T cells via induction of cell death.

In order to establish long-lasting infections in their mammalian host, filarial nematodes have developed sophisticated strategies to dampen their host's immune response. Proteins that are actively secreted by the parasites have been shown to induce the expansion of regulatory T cells and to directly interfere with effector T cell function. Here, we analyze the suppressive capacity of Onchocercavolvulus-derived excreted/secreted proteins. Addition of two recombinant O. volvulus proteins, abundant larval transcript-2 (OvALT-2) and novel larval transcript-1 (OvNLT-1) to cell cultures of T cell re

Dynamic cellular remodeling and symbiont genetic heterogeneity during early bacterial colonization in the deep-sea mussel Bathymodiolus septemdierum.

Symbiotic relationships between animals and microorganisms are widespread and have driven the adaptation and evolution of animals across diverse environments. The establishment of highly integrated endosymbioses often involves dynamic cellular remodeling and management of symbiont populations. Deep-sea mussels of the subfamily Bathymodiolinae horizontally acquire chemosynthetic bacteria in specialized gill cells called bacteriocytes, primarily at the posterior ends of their gills, where new filaments are continuously formed throughout their lifetime. This group exhibits a wide diversity of sym

The role of lncRNAs/miRNAs/Sirt1 axis in myocardial and cerebral injury.

In recent years, researchers have begun to realize the importance of the role of non-coding RNAs in the treatment of cancer and cardiovascular and neurological diseases. LncRNAs and miRNAs are important non-coding RNAs, which regulate gene expression and activate mRNA translation through binding to diverse target sites. Their involvement in the regulation of protein function and the modulation of physiological and pathological conditions continues to be investigated. Sirtuins, especially Sirt1, have a critical function in regulating a variety of physiological processes such as oxidative stress

Genetic adaptations to SIV across chimpanzee populations.

Central and eastern chimpanzees are infected with Simian Immunodeficiency Virus (SIV) in the wild, typically without developing acute immunodeficiency. Yet the recent zoonotic transmission of chimpanzee SIV to humans, which were naïve to the virus, gave rise to the Human Immunodeficiency Virus (HIV), which causes AIDS and is responsible for one of the deadliest pandemics in human history. Chimpanzees have likely been infected with SIV for tens of thousands of years and have likely evolved to reduce its pathogenicity, becoming semi-natural hosts that largely tolerate the virus. In support of th

Conserved Central Intraviral Protein Interactome of the Herpesviridae Family.

Protein interactions are major driving forces behind the functional phenotypes of biological processes. As such, evolutionary footprints are … hroughout their long evolutionary history, individual herpesvirus species have developed remarkable host specificity, while collectively the Herpesviridae family has evolved to infect a large variety of eukaryotic hosts. The development of approaches to fight herpesvirus infections has been hampered by the complexity of herpesviruses' genomes, proteomes, and struc …

Whole transcriptome analysis reveals differential gene expression profile reflecting macrophage polarization in response to influenza A H5N1 virus infection.

BACKGROUND: Avian influenza A H5N1 virus can cause lethal disease in humans. The virus can trigger severe pneumonia and lead to acute respiratory distress syndrome. Data from clinical, in vitro and in vivo suggest that virus-induced cytokine dysregulation could be a contributory factor to the pathogenesis of human H5N1 disease. However, the precise mechanism of H5N1 infection eliciting the unique host response are still not well understood. METHODS: To obtain a better understanding of the molecular events at the earliest time points, we used RNA-Seq to quantify and compare the host mRNA and m

Deconstructing the Bat Skin Microbiome: Influences of the Host and the Environment.

Bats are geographically widespread and play an important role in many ecosystems, but relatively little is known about the ecology of their associated microbial communities and the role microbial taxa play in bat health, development, and evolution. Moreover, few vertebrate animal skin microbiomes have been comprehensively assessed, and thus characterizing the bat skin microbiome will yield valuable insight into the variability of vertebrate skin microbiomes as a whole. The recent emergence of the skin fungal disease white-nose syndrome highlights the potentially important role bat skin microbi

Neuroinvasive flavivirus infections.

Flaviviruses, including Dengue, West Nile, Japanese encephalitis, and Tick-borne encephalitis virus, are major emerging human pathogens, affecting millions of individuals worldwide. Many clinically important flaviviruses elicit CNS diseases in infected hosts, including traditional "hemorrhagic" viruses, such as Dengue. This review focuses on the epidemiology, symptomatology, neuropathology, and, specifically, neuropathogenesis of flavivirus-induced human CNS disease. A detailed insight into specific factors priming towards neuroinvasive disease is of clear clinical significance, as well as imp

Association of a sex-related difference of Strongyloides stercoralis-specific IgG4 antibody titer with the efficacy of treatment of strongyloidiasis.

It is difficult to completely eradicate strongyloidiasis, a human intestinal nematode infection with Strongyloides stercoralis with drugs, especially in males. To find host factors involved in the response to treatment, patients infected with S. stercoralis were examined for S. stercoralis-specific antibody titers and the effect of treatment with albendazole on these titers were determined. The cure rate was slightly but not significantly lower in males than in females (P = 0.108). However, a significantly higher titer of S. stercoralis-specific IgG4 antibody was observed in males than in fema

T cell immunodominance and maintenance of memory regulated by unexpectedly cross-reactive pathogens.

We show here that T cell cross-reactivity between heterologous viruses influences the immunodominance of virus-specific CD8(+) T cells by two mechanisms. First, T cells specific for cross-reactive epitopes dominate acute responses to viral infections; second, within the memory pool, T cells specific for cross-reactive epitopes are maintained while those specific for non-cross-reactive epitopes are selectively lost. These findings suggest an immunological paradigm in which viral infections shape the available T cell repertoire, causing alterations in the hierarchies of both the primary and memo

Spatiotemporal coordination of virulence, metabolism, and stress responses shapes infection dynamics of Xanthomonas perforans.

Plants host diverse microbial communities across spatially distinct foliar niches. For hemibiotrophic pathogens, the epiphytic leaf surface and the apoplast impose contrasting physiological constraints that shape infection strategies. We investigated how these environments program the transcriptional responses of a tomato pathogen, Xanthomonas perforans (Xp), and how virulence, metabolism, and regulation are coordinated across space and time during disease progression. By selectively enriching and profiling the transcriptome of a pathogen from the leaf surface and apoplast, we uncovered pronou

Signatures and likely sources of the male pregnancy microbiome in wild bay pipefish (Syngnathus leptorhynchus).

BACKGROUND: Understanding the origin and structure of microbiomes and their associations with ecologically significant host traits is essential for understanding the evolution of host-microbe interactions. These interactions support a wide range of physiological processes important for development, survival, and reproduction. Syngnathid fishes (seahorses, pipefish, and seadragons) represent a compelling system for investigating host-microbiome interactions due to their unique evolution of male pregnancy. Males harbor a fitness-critical brood pouch that provides embryos with protection, osmoreg

Fluorescent labeling reliably identifies Chlamydia trachomatis in living human endometrial cells and rapidly and accurately quantifies chlamydial inclusion forming units.

Chlamydia replication requires host lipid acquisition, allowing flow cytometry to identify Chlamydia-infected cells that accumulated fluorescent Golgi-specific lipid. Herein, we describe modifications to currently available methods that allow precise differentiation between uninfected and Chlamydia trachomatis-infected human endometrial cells and rapidly and accurately quantify chlamydial inclusion forming units.

Factors of occurrence of ocular toxoplasmosis. A review.

Acquired and congenital toxoplasmosis are frequently complicated by ocular toxoplasmosis. The diagnosis relies on clinical aspects, response to specific treatment and results of biological assays. The incidence and the prevalence of this complication are difficult to establish precisely and depend on the prevalence of the parasite infection in the general population, and are affected by factors such as type of exposure to the parasite, genetic backgrounds of the parasite and the host, and type of immune response elicited by the parasite.

[A cerebral abscess due to Listeria monocytogenes in a 15-month-old infant].

We report a case of brain abscess due to Listeria monocytogenes in an infant. We recall that listeriosis is infrequent in pediatric patients beyond the neonatal period, that most cases occur in immunocompromised hosts, and that clinical features are non-specific in neuromeningeal forms. Management of brain abscesses is discussed. The role of the patient's general health condition seems to have a determinant influence on prognosis.

Targeting TLR2 agonists as immunomodulators with broad antiviral activity.

Toll-like receptor-2 (TLR2) serves as an innate immune sensor that recognizes specific viral proteins, thereby initiating signaling pathways that can either enhance antiviral host defenses or induce pathological inflammatory responses. In this forum, we discuss the role of TLR2 and its agonists in enhancing antiviral and vaccine-induced immune responses.

The spin method for recovering tissue larvae and its use in evaluating C57BL/6 mice as a model for the study of resistance to infection with Ascaris suum.

A centrifugation method for recovering tissue larvae is described and is proven to be superior to both Baerman and tissue digest methods for recovering larvae of Ascaris suum from the liver and lungs of infected C57BL/6 mice. Using the spin method for harvesting larvae, C57BL/6 mice were evaluated for their suitability in studies on specific host immunity to infection with A. suum.
