Predicting Antileishmanial Activity of Plant-Derived Compounds Using Random Forest Modeling.

Leishmaniasis is a parasitic disease for which existing treatments can be toxic and costly, and resistance to current therapies is becoming increasingly common. This study implemented a systematic review of data regarding antileishmanial activity in plant-derived compounds and used machine learning techniques to train and test a Random Forest algorithm to predict the antileishmanial activity of plant-derived compounds. Asteraceae, Euphorbiaceae, Lamiaceae, and Myrtaceae plant families were identified for their high or moderate antileishmanial activity and nativity to areas of high leishmaniasi

Systematic analysis of genome-wide fitness data in yeast reveals novel gene function and drug action.

We systematically analyzed the relationships between gene fitness profiles (co-fitness) and drug inhibition profiles (co-inhibition) from several hundred chemogenomic screens in yeast. Co-fitness predicted gene functions distinct from those derived from other assays and identified conditionally dependent protein complexes. Co-inhibitory compounds were weakly correlated by structure and therapeutic class. We developed an algorithm predicting protein targets of chemical compounds and verified its accuracy with experimental testing. Fitness data provide a novel, systems-level perspective on the c

Analysis of Virologic and Clinical Predictors for Hospitalization, Supported by Machine Learning, in a Phase 3 Molnupiravir COVID-19 Treatment Trial.

Machine learning (ML) is increasingly being used in clinical medicine. The objective of this study was to identify factors predictive of hospitalization, with the support of ML, in outpatients with mild-to-moderate COVID-19. Unvaccinated adults with mild-to-moderate COVID-19 were randomized 1:1 to molnupiravir or placebo in MOVe-OUT (NCT04575597). In this post hoc analysis, various factors were evaluated among participants who progressed or did not progress to hospitalization. ML was used as an unbiased method to evaluate multiple factors for their association with hospitalization in placebo-t

Development of a lipid-glycaemic prediction model for gestational diabetes mellitus: a single-Centre retrospective study.

BACKGROUND: Gestational diabetes mellitus (GDM) is a common pregnancy complication. Although lipid parameters, the triglyceride-glucose (TyG) index, and TyG-body mass index (TyG-BMI) have been associated with GDM, their relative value within an integrated lipid-glycaemic prediction framework remains unclear. This study developed and internally validated an interpretable model for GDM risk estimation. METHODS: This single-centre retrospective study included 287 pregnant women who underwent 75 g oral glucose tolerance testing and fasting lipid assessment at 24-28 gestational weeks, including 72

HELIX: a scalable model for predicting context-dependent regulation of RNA splicing and isoform usage.

Context-dependent alternative splicing plays a critical role in disease pathogenesis and organ development, but its complex regulation remains challenging to predict. Here, to address this, we developed HELIX, a hierarchical deep learning framework that integrates pre-mRNA sequence and RNA-binding protein expression profiles to predict tissue- and condition-specific splicing patterns and transcript isoform usage simultaneously. By leveraging both short-read and long-read RNA sequencing data during training, HELIX achieves greater accuracy than existing splicing prediction models and convention

AI-enabled forecasting of prehospital transfusion needs in patients with trauma: a multinational, registry-based, retrospective, machine learning development and validation study.

BACKGROUND: Trauma is a major global cause of morbidity and mortality, with haemorrhage representing a leading preventable cause of early death. Timely blood transfusion is a crucial intervention, but current prehospital decision-making tools are scarce. Conventional triggers, such as haemoglobin concentrations, are often unreliable in the acute setting. There is a clear need for more robust, data-driven methods to guide transfusion decisions before hospital arrival. METHODS: We conducted a retrospective, machine learning development and validation study to predict the need for prehospital tr

Continuous predictive mortality risk monitoring after allogeneic hematopoietic stem cell transplantation.

Allogeneic hematopoietic stem cell transplantation remains critical for treating high-risk hematological malignancies like leukemia. Despite advances in treatment, early mortality remains clinically significant, with approximately 6-11% of patients dying within the first 100 days after transplantation. This highlights the need for dynamic monitoring strategies beyond static pre-transplant risk assessments. This study introduces a novel, interpretable, real-time proof-of-concept monitoring framework that continuously assesses individualized mortality risk during treatment using routinely collec

Protocol for the external validation of machine learning models for risk prediction in early-onset preeclampsia: a multicenter prospective cohort study.

BACKGROUND: Early-onset preeclampsia (eoPE), defined as preeclampsia occurring before 34 weeks of gestation, remains one of the leading causes of severe maternal morbidity, iatrogenic prematurity, and maternal mortality worldwide. Its heterogeneous and rapidly evolving clinical course makes the prediction of adverse outcomes particularly challenging. Although machine learning (ML)-based models have shown promise in stratifying eoPE risk and supporting clinical decision-making, robust prospective external validation is still lacking. The present study aims to address this gap through a multicen

Machine learning prediction of tuberculosis mortality: a comparative analysis of random survival forest and cox regression models.

BACKGROUND: Survival analysis is widely used to predict time-to-event outcomes, with the Cox regression model being a standard approach. However, machine learning methods such as Random Survival Forests (RSF) can capture complex, non-linear relationships that traditional models may miss. OBJECTIVE: This study compared the predictive performance of RSF and Cox regression in modelling tuberculosis (TB) mortality. METHODS: We conducted a retrospective study of TB patients treated at the East London Central Clinic in South Africa. Patient data included demographic, clinical, and treatment-related

Light Bladder Net: Non-invasive Bladder Cancer Prediction by Weighted Deep Learning Approaches and Graphical Data Transformation.

BACKGROUND/AIM: Bladder cancer (BCa) is associated with high recurrence rates, emphasizing the importance of early and accurate detection. This study aimed to develop a lightweight and fast deep learning model, Light-Bladder-Net (LBN), for non-invasive BCa detection using conventional urine data. MATERIALS AND METHODS: We improved LBN's generalization by applying data transformations, adding uniform noise, and employing feature selection methods (mRMR, PCA, SVD, t-SNE) to extract key vectors from its fully connected layer. These vectors were integrated into the original dataset, and multiple

Exploring prognostic factors on vascular outcomes among maintenance dialysis patients and establishing a prognosis prediction model using machine learning methods.

BACKGROUND: Cardiovascular diseases (CVDs) remain the leading cause of morbidity and mortality in end-stage kidney disease patients, with persistently high rates of major adverse cardiovascular events (MACEs). Traditional risk factors such as diabetes and hypertension have limited predictive value in this population, while chronic kidney disease (CKD)-specific factors including inflammation, disordered mineral metabolism, and vascular calcification play significant roles. Therefore, we developed a machine learning-based model incorporating traditional and CKD-specific variables to improve MACE

Performance of a Semi-Automated Hierarchical Rest Interval Detection Pipeline (actiSleep) for Wrist Actigraphy in Adolescents.

Actigraphy is a popular behavioral sleep assessment tool in research and clinical practice. Hierarchical hand-scoring approaches remain the standard for actigraphy rest interval estimation, but can be impractical for large cohort studies and suffer from reproducibility problems. We developed a semi-automated pipeline (actiSleep) to set rest intervals consistent with best-practice hand-scoring algorithms incorporating event marker, diary, light, and activity data. To evaluate actiSleep performance, we used data from an observational study of 51 adolescents (14-19yr), with and without family his

Evaluation of an algorithm ascertaining cases of osteonecrosis of the jaw in the Swedish National Patient Register.

BACKGROUND: Osteonecrosis of the jaw (ONJ) is a medical condition associated with antiresorptive drugs, among others, used to treat osteoporosis and bone metastasis. Currently, there is no consensus regarding the definition of ONJ, and no ONJ-specific International Classification of Diseases-10 code exists. Therefore, register-based studies of this condition may be troublesome. PURPOSE: To evaluate an algorithm ascertaining ONJ cases in an attempt to facilitate future assessments of ONJ in clinical and epidemiological studies. METHODS: By means of the Patient Register and the Prescribed Drug
