Mesenchymal stem cell-derived small extracellular vesicles in spinal cord injury: From molecular repair mechanisms to standardized translational development.

Spinal cord injury (SCI) causes permanent neurological disability through a complex sequence of primary mechanical damage and secondary injury cascades, including neuroinflammation, blood-spinal cord barrier disruption, oxidative stress, apoptotic and ferroptotic cell death, demyelination, glial scar formation, and limited axonal regeneration. Mesenchymal stem/stromal cell-derived extracellular vesicles (MSC-EVs), particularly small EV preparations frequently reported in the SCI literature as exosomes, have emerged as cell-free therapeutic candidates because they can transfer regulatory protei

Comparison of standard care versus a biomarker algorithm to guide antibiotic stewardship in pediatric intensive care unit: a prospective observational cohort study with historical control (the BASIP study).

UNLABELLED: The purpose of this study is to evaluate the effectiveness and safety of an integrated antimicrobial stewardship approach in critically ill children with suspected infection admitted to a pediatric intensive care unit (PICU). The approach combined procalcitonin (PCT)-guided decision-making with serial clinical and microbiological assessment to reduce antibiotic duration. Secondary aims included evaluating additional biomarkers, such as pancreatic stone protein (PSP) and C-reactive protein (CRP) in the context of antibiotic de-escalation. This prospective observational cohort study

Metabolic markers of myocardium insulin resistance in dogs with heart failure.

BACKGROUND: Heart failure syndrome is an aspect of primary or secondary heart disease and is associated with decompensation, formation, and activation of pathological interactions between regulation systems. This results in myocardial energy metabolism alteration. This study was carried out to defy some metabolic aspects of myocardial tissue insulin resistance (IRM) development in canine heart failure. AIM: To investigate the myocardial tissue concentration of adenosine triphosphate (ATP), glucose transporters 1 and 4, pyruvate dehydrogenase (PDH), hexokinase 2, insulin receptor (InsR), and a

A CsrA-Binding, trans-Acting sRNA of Coxiella burnetii Is Necessary for Optimal Intracellular Growth and Vacuole Formation during Early Infection of Host Cells.

Coxiella burnetii is an obligate intracellular gammaproteobacterium and zoonotic agent of Q fever. We previously identified 15 small noncoding RNAs (sRNAs) of C. burnetii One of them, CbsR12 (Coxiella burnetiismall RNA 12), is highly transcribed during axenic growth and becomes more prominent during infection of cultured mammalian cells. Secondary structure predictions of CbsR12 revealed four putative CsrA-binding sites in stem loops with consensus AGGA/ANGGA motifs. We subsequently determined that CbsR12 binds to recombinant C. burnetii CsrA-2, but not CsrA-1, proteins in vitro Moreover, thro

Genome sequence and comparative analysis of a putative entomopathogenic Serratia isolated from Caenorhabditis briggsae.

BACKGROUND: Entomopathogenic associations between nematodes in the genera Steinernema and Heterorhabdus with their cognate bacteria from the bacterial genera Xenorhabdus and Photorhabdus, respectively, are extensively studied for their potential as biological control agents against invasive insect species. These two highly coevolved associations were results of convergent evolution. Given the natural abundance of bacteria, nematodes and insects, it is surprising that only these two associations with no intermediate forms are widely studied in the entomopathogenic context. Discovering analogous

Amyloidosis: review of 236 cases.

From 1960 through 1972, 236 cases of amyloidosis with histologic proof were found. The amyloidosis was primary (without evidence of preceding or coexisting disease) in 132 cases (group 1) and associated with multiple myeloma in 61 (group 2). Secondary amyloidosis appeared in 19 cases (associated with rheumatoid arthritis or osteomyelitis in two-thirds of them). There were 22 patients with amyloid localized to a single organ (bladder, lung, skin, or larynx in more than half of them). Two patients had familial amyloidosis. In group 1 and group 2, the most common presenting symptoms were fatigue,

In silico analyses of the involvement of GPR55, CB1R and TRPV1: response to THC, contribution to temporal lobe epilepsy, structural modeling and updated evolution.

INTRODUCTION: The endocannabinoid (eCB) system is named after the discovery that endogenous cannabinoids bind to the same receptors as the phytochemical compounds found in Cannabis. While endogenous cannabinoids include anandamide (AEA) and 2-arachidonoylglycerol (2-AG), exogenous phytocannabinoids include Δ-9 tetrahydrocannabinol (THC) and cannabidiol (CBD). These compounds finely tune neurotransmission following synapse activation, via retrograde signaling that activates cannabinoid receptor 1 (CB1R) and/or transient receptor potential cation channel subfamily V member 1 (TRPV1). Molecular water pumps.

There is good evidence that cotransporters of the symport type behave as molecular water pumps, in which a water flux is coupled to the substrate fluxes. The free energy stored in the substrate gradients is utilized, by a mechanism within the protein, for the transport of water. Accordingly, the water flux is secondary active and can proceed uphill against the water chemical potential difference. The effect has been recognized in all symports studied so far (Table 1). It has been studied in details for the K+/Cl- cotransporter in the choroid plexus epithelium, the H+/lactate cotransporter in t

Localized Pulmonary Amyloidosis Associated With Sjögren's Syndrome, Coexisting Lymphoid Interstitial Pneumonia, and a Severe Double Aortic Lesion: A Case Report and Literature Review.

Amyloidosis is a protein-folding disease that results in the deposition of misfolded proteins into various tissues of the body, resulting in dysfunction of the affected organ. It is caused by a heterogeneous group of disorders caused by extracellular deposition of misfolded proteins as insoluble fibrils. Involvement of the lungs can present as nodular, tracheobronchial, or diffuse interstitial, and can be mistaken for cancer or infection on imaging. The combination of Sjögren's syndrome, chronic activation of B cells, and the risk of developing a lymphoproliferative disorder can lead to the ra

Enhanced Neurophysiological Benefits of Magnesium-Acetyl-Taurate Over Magnesium-L-Threonate: A Comparative pre-clinical Study on Bioavailability, Synaptic Plasticity and Cognitive Functions.

The study examines how magnesium compounds, especially Magnesium-L-Threonate (MLT) and Magnesium-Acetyl-Taurate (MAT), affect neurophysiological functions in adult Wistar rats. Magnesium is essential for many cellular processes in the brain & peripheral system, such as neurotransmitter regulation, muscle function, and energy metabolism. Tissue Mg²⁺ levels were defined as the primary endpoint of the study, as they directly reflect the central objective of evaluating magnesium-based interventions. All other outcomes, including behavioral, biochemical, and molecular parameters, were considered se

Sintered indium-tin-oxide (ITO) particles: a new pneumotoxic entity.

Indium-Tin-Oxide (ITO) is a sintered mixture of indium- (In(2)O(3)) and tin-oxide (SnO(2)) in a ratio of 90:10 (wt:wt) that is used for the manufacture of LCD screens and related high technology applications. Interstitial pulmonary diseases have recently been reported in workers from ITO producing plants. The present study was conducted to identify experimentally the exact chemical component responsible for this toxicity and to address possible mechanisms of action. The reactivity of respirable ITO particles was compared with that of its single components alone or their unsintered 90:10 mixtur

Neutralization of influenza virus by low concentrations of hemagglutinin-specific polymeric immunoglobulin A inhibits viral fusion activity, but activation of the ribonucleoprotein is also inhibited.

High concentrations of hemagglutinin-specific neutralizing polymeric monoclonal immunoglobulin A (IgA) inhibit attachment of the majority of type A influenza virus virions to cell monolayers and tracheal epithelium (H. P. Taylor and N. J. Dimmock, J. Exp. Med. 161:198-209, 1985; M. C. Outlaw and N. J. Dimmock, J. Gen. Virol. 71:69-76, 1990). A minority of virions attaches but is not infectious. Here, we report that a different mechanism operates when influenza virus A/Puerto Rico/8/34 (H1N1) is neutralized by low concentrations of monoclonal polymeric IgA or when A/fowl plague virus/Rostock/34

Lipocalin-2 is an essential component of the innate immune response to Acinetobacter baumannii infection.

Acinetobacter baumannii is an opportunistic pathogen and an emerging global health threat. Within healthcare settings, major presentations of A. baumannii include bloodstream infections and ventilator-associated pneumonia. The increased prevalence of ventilated patients during the COVID-19 pandemic has led to a rise in secondary bacterial pneumonia caused by multidrug resistant (MDR) A. baumannii. Additionally, due to its MDR status and the lack of antimicrobial drugs in the development pipeline, the World Health Organization has designated carbapenem-resistant A. baumannii to be its priority

Development of an aptamer-based rapid diagnostic assay for Salmonella detection using whole-cell SELEX technique.

UNLABELLED: Rapid and accurate detection of Salmonella is essential for improving food safety and reducing the burden of enteric infections, especially in low-resource settings. Aptamer-based diagnostic platforms offer a promising alternative to antibody-based methods, given their high specificity, stability, and low production cost. A DNA aptamer targeting Salmonella enterica was developed through Systematic Evolution of Ligands by Exponential Enrichment (SELEX) iterative rounds of selection. Target-binding efficiency was monitored by colorimetric and spectrophotometric binding tests. Silver

Influence of yoga on the epigenetics of obesity: a study on its short- and medium-term benefits in the treatment of patients with severe/morbid obesity eligible for bariatric surgery.

BACKGROUND: Obesity is a growing global health problem characterised by chronic low-grade inflammation and epigenetic dysregulation. Epigenetic mechanisms-potentially reversible changes in gene expression occurring without alteration of the DNA sequence, most notably DNA methylation-are increasingly implicated in obesity-related inflammation. Yoga may act as complementary therapy. OBJECTIVE: To explore whether participation in a structured yoga program is associated with changes in DNA methylation of TNF-α, and with clinical, hormonal, metabolic and psychological parameters, in patients with

Pragmatic randomised trial of a 12-week exercise and nutrition program for Aboriginal and Torres Strait Islander women: clinical results immediate post and 3 months follow-up.

BACKGROUND: Aboriginal and Torres Strait Islander women experience higher rates of heart disease and type 2 diabetes than non-Indigenous Australian women. Increasing physical activity, improving diets and losing weight have been shown to reduce cardio metabolic risk. The primary aim was to evaluate the effectiveness of a 12-week structured exercise and nutrition program in a cohort of urban Indigenous Australian women on waist circumference, weight and biomedical markers of metabolic functioning from baseline (T1) to program completion (T2). The secondary aim assessed whether these outcomes we

New targets for antithrombotic drugs.

The normal hemostatic process is initiated by disruption in the vascular continuity and exposure of the subendothelial components. Platelets adhere to subendothelium-bound von Willebrand factor via glycoprotein (GP) Ib complex. This initial interaction per se and the release of platelet agonists transduce signals that lead to the rise in intracellular Ca(2+). The rise in Ca(2+) induces shape change, prostaglandin synthesis, release of granular contents and conformational changes in platelet Gp IIb-IIIa. Gp IIb-IIIa in activated platelets becomes competent to bind fibrinogen and other adhesive
