Type I interferon induces multiple protein effectors to inhibit viral replication. Among these, the interferon-induced transmembrane protein (IFITM3) and the cholesterol-25-hydroxylase (CH25h) converge on the inhibition of viral entry by altering the behavior of membranes. Here, we dissect the functional and mechanistic relationship between these two membrane-acting effectors using different viruses. When using HIV-1 and vesicular stomatitis virus (VSV), IFITM3 and CH25h restrict viral entry with similar efficiency but act in a largely redundant manner, a finding that is consistent across infe

Genetic diversity of sarbecoviruses phylogenetically related to SARS-CoV-2 has been identified in Malayan pangolins. However, their potential pathogenicity and risk of cross-species transmission to humans remain largely unknown. In the present study, we performed comparative characterization of SARS-CoV-2 and two pangolin SARS-CoV-2-related viruses, named MpCoV-GD and MpCoV-GX, respectively, in human cell lines and organoids and human ACE2 (hACE2) transgenic hamsters. The cell-to-cell fusion assay showed that the Spike (S) protein of MpCoV-GD induces a stronger membrane fusion than the MpCoV-G…

Action potential-evoked neurotransmitter release is impaired in knock-out neurons lacking synaptic cell-adhesion molecules α-neurexins (αNrxns), the extracellularly longer variants of the three vertebrate Nrxn genes. Ca2+ influx through presynaptic high-voltage gated calcium channels like the ubiquitous P/Q-type (CaV2.1) triggers release of fusion-ready vesicles at many boutons. α2δ Auxiliary subunits regulate trafficking and kinetic properties of CaV2.1 pore-forming subunits but it has remained unclear if this involves αNrxns. Using live cell imaging with Ca2+ indicators, we report here that

Previously, we reported that MyoD, a master gene for myogenic cells, could efficiently convert primary skin fibroblasts into myoblasts and m … lasts means that gene transfer from MyoD-transduced cells to host muscle cells could be obtained by cell fusion. In comparison with the iPS method (indirect reprogramming), our transduction method has a low risk for tumorigenesis and carcinogenesis because the starting cells are fibroblasts and the transduced cells are myoblasts, both normal and mortal cells. …

The expression of epithelial features has been investigated from the 9th to the 30th passage in cells derived from the fusion of mouse 3T3.4E cells with normal or hand wart human keratinocytes. These cells stratified and grew either on plastic or collagen substrate without modifications. Such growth characteristics are similar to those observed in the epidermis. The doubling time of hybrid cells is nearer to that of 3T3.4E than to that of keratinocytes. Some epithelial markers were detected by immunofluorescence staining. Bullous pemphigoid antigen characteristic of the basal keratinocyte was

Human papillomavirus infection, the primary cause of cervical cancer (the fourth most common cancer in women), is preventable through prophylactic HPV vaccines. As an alternative to current HPV vaccines, all based on a cocktail of L1 virus-like particles (VLPs) from multiple HPV types, the L2 capsid protein offers cross-protection via a conserved epitope residing at the N-terminus between residues 20-38. The Trx-L2(20-38)-8mer (Trx-8mer) is an L2-based antigen composed of a polytope of L2(20-38) epitopes from eight HPV types inserted into thioredoxin from the hyperthermophilic archaeon Pyrococ

BACKGROUND: Secretory carcinoma (SC) of the salivary gland is a recently described malignant tumor harboring characteristic ETV6-NTRK3 gene fusion. SC generally has a favorable clinical course, and is currently regarded as a low-grade carcinoma. However, a small subset of SCs demonstrates aggressive clinical features with histologically high-grade transformed morphology, the molecular pathogenesis of which has not yet been elucidated. In this study, we performed a clinicopathological and molecular genetic study of patients with SC of the head and neck displaying various clinical characteristic

Radiologic-pathologic correlation is essential for diagnostic accuracy, particularly when dealing with primary bone tumors. This investigation explores the unique radiographic and pathologic features of NFATC2- rearranged bone sarcomas. Inclusion criteria focused on primary bone sarcomas with NFATC2 fusions while excluding soft tissue sarcomas, benign bone cysts, and vascular neoplasms with similar fusions. Our cohort comprised 16 patients (12 males, 4 females) with a mean age of 45.6 years (range: 15 to 77 y). Tumors were located in the femur (n=9), tibia (n=3), humerus (n=2), ulna (n=1), and

A major breakthrough in understanding the steps and signalling that drives the HCV to reach a full life-cycle has been achieved by in vitro models that have facilitated elevated virus production, resulting in the discovery of pathways and factors involved in virus entry, translation and replication. The HCV enters host cells through binding of its envelope glycoproteins to cell receptors, followed by clathrin-mediated endocytosis and fusion with cell membranes, leading to virus uncoating and cell entry. This chain of events is mediated by sequential involvement of different co-receptors, for e

OBJECTIVE: SHP-1 protein tyrosine phosphatase has been implicated in suppressing B-lymphocyte and myeloid cell malignancies; however, there are little data on this role of SHP-1 in T-lymphocyte malignancies. We examined malignant human T cells to identify any abnormalities of SHP-1 that would support a role for this molecule in suppressing T lymphomagenesis.
MATERIALS AND METHODS: Human T-lymphocyte cell lines and primary blood cells were used to examine the expression of SHP-1 mRNA and protein. Reverse transcriptase polymerase chain reaction was used to amplify particular portions of the SHP

Male infertility is rising worldwide. Spermatogenesis is energetically demanding and tightly coupled to mitochondrial quality control (MQC)-including biogenesis, fusion-fission dynamics, and mitophagy. Here we synthesize current evidence across developmental stages of germ cells, highlighting stage-specific shifts from glycolysis-supported ATP production to oxidative phosphorylation (OXPHOS)-dominated metabolism and remodeling of mitochondrial architecture into the midpiece sheath. We summarize genetic and functional data implicating OPA1/MFN1-2 in inner/outer membrane fusion, DRP1/FIS1/MFF in

UNLABELLED: Genes encoding ETS family transcription factors are altered by chromosomal rearrangement in 60% to 70% of prostate cancers and nearly all Ewing sarcomas. Ewing sarcoma rearrangements result in chimeric fusion of ETS proteins to the RNA-binding protein EWSR1. Prostate cancer rearrangements result in aberrant expression of ETS proteins such as ETV1, ETV4, ETV5, or ERG that can interact with wild-type EWSR1, suggesting common mechanisms between these diseases. In this study, we find that ETV1, ETV4, and ETV5 can phenocopy EWSR1::FLI1 in Ewing sarcoma cell lines. However, rescue of EWS

Antibody-drug conjugates (ADCs) are constrained by limited tumor penetration of IgG formats and heterogeneity from stochastic conjugation. We addressed these challenges by engineering a compact trastuzumab Fab-based ADC with site-specific installation of DM1 using the peptide ligase butelase-1 and by extending serum persistence via genetic fusion of an albumin-binding domain (ABD). Butelase-1 ligation proceeded rapidly under mild conditions to afford a homogeneous product with a defined drug-to-antibody ratio. The conjugate selectively killed HER2-positive cells in vitro while sparing HER2-neg

Palatogenesis is a complex developmental process requiring temporospatially coordinated cellular and molecular events. The following review focuses on genetic, epigenetic, and environmental aspects directing palatal formation and their implication in orofacial clefting genesis. Essential for palatal shelf development and elevation (TGF-β, BMP, FGF, and WNT), the subsequent processes of fusion (SHH) and proliferation, migration, differentiation, and apoptosis of neural crest-derived cells are controlled through signaling pathways. Interruptions to these processes may result in the birth defect

Histopathological alterations caused by Thaparocleidus vistulensis (Siwak, 1932) on the European catfish Silurus glanis Linnaeus, 1758 were investigated by histopathology and scanning electron microscopy (SEM). The pathological effects of T. vistulensis on the gills of the European catfish were mainly related to the attachment sites of the parasite to its host, but damage also affected adjacent structures. The primary attachment relies on the anchors at the opisthaptor which commonly adheres both superficially and deeply, into the basal region between adjacent secondary lamellae. At the attach

Many cutaneous adnexal tumors share molecular alterations with other homologous neoplasms occurring in salivary glands. Polymorphous adenocarcinoma (PAC) is a rare salivary gland tumor, usually associated with alterations in the PRKD gene family. In this report, we describe a primary cutaneous sweat gland adenocarcinoma of the scalp occurring in a 65-year-old female. Morphological features of the carcinoma were reminiscent of salivary gland PAC. Tumor cells were CK7+ S100+ SOX10+ p63 focally positive and p40-. Whole-transcriptome sequencing of the lesion showed the presence of an in-frame ATL2

Myoepithelial carcinoma (MECA) is a rare malignant tumor showing myoepithelial differentiation, most commonly found in the salivary glands and soft tissue, with only isolated reports in the female genital tract. We report the first documented case of uterine MECA harboring an EWSR1::ZNF444 fusion. A 55-year-old woman with a history of breast cancer presented with abnormal uterine bleeding. Imaging suggested a benign lesion, but hysteroscopic resection raised suspicion for uterine sarcoma. Hysterectomy revealed an infiltrative neoplasm composed of epithelioid and spindle cells with focal osseou

Allorecognition-the ability of an organism to discriminate between self and nonself-is crucial to colonial marine animals to avoid invasion by other individuals in the same habitat. The cnidarian hydroid Hydractinia has long been a major research model in studying invertebrate allorecognition, establishing a rich knowledge foundation. In this study, we introduce a new cnidarian model Cladonema radiatum (C. radiatum). C. radiatum is a hydroid jellyfish which also forms polyp colonies interconnected with stolons. Allorecognition responses-fusion or regression of stolons-are observed when stolons

Ninein is a centrosome protein that has been implicated in microtubule anchorage and centrosome cohesion. Mutations in the human NINEIN gene have been linked to Seckel syndrome and to a rare form of skeletal dysplasia. However, the role of ninein in skeletal development remains unknown. Here, we describe a ninein knockout mouse with advanced endochondral ossification during embryonic development. Although the long bones maintain a regular size, the absence of ninein delays the formation of the bone marrow cavity in the prenatal tibia. Likewise, intramembranous ossification in the skull is more
