Induction of endogenous beta-galactosidase by ionizing radiation complicates the analysis of p53-LacZ transgenic mice.

Studies of the response of p53-lacZ transgenic mice have uncovered an unexpected induction of endogenous acid-beta-galactosidase activity following whole body irradiation. Strong induction of endogenous enzyme activity is seen in a variety of mouse strains commonly used in the production of transgenes. The induction of endogenous enzyme activity therefore complicates the analysis of p53-lacZ transgenes and may also influence the analysis of radiation responses in other lacZ-reporter mice.

Reduction of N-acetyl-beta-glucosaminidase activity in the submaxillary glands of streptozotocin diabetic mice.

Streptozotocin diabetes strongly reduced the N-acetyl-beta-glucosaminidase activity in the submaxillary glands of mice. A reciprocal change was observed between the enzyme activity and blood sugar in diabetes. Insulin treatment of th diabetic mice returned the reduced activity to the normal level. Isoelectric focusing analysis showed that the submaxillary gland enzyme was composed of two isozymes having isoelectric points (pI) of 4.8 and 8.8. Only the pI 8.8 isozyme was affected by the diabetes.

[Mechanism of reduction of the phagocytic activity of blood neutrophils in influenza patients].

The authors have anlyzed the results obtained in the study of indices characterizing phagocytosis and the activity of intracellular leukocytic enzymes in patients with uncomplicated influenza and in those with pneumonia complications. For control, the corresponding parameters were studied in patients with an infectious bacterial disease (erysipelas) and in healthy persons. A decrease in the activity of intracellular leukocytic enzymes, acidic and alkaline phosphatases, was found to be one of the causes of deficient phagocytosis (decreased phagocytic activity and the decreased index of phagocyt

AMP-activated protein kinase (AMPK) activator A-769662 increases intracellular calcium and ATP release from astrocytes in an AMPK-independent manner.

AIM: To test the hypothesis that, given the role of AMP-activated protein kinase (AMPK) in regulating intracellular ATP levels, AMPK may alter ATP release from astrocytes, the main sources of extracellular ATP (eATP) within the brain. MATERIALS AND METHODS: Measurements of ATP release were made from human U373 astrocytoma cells, primary mouse hypothalamic (HTAS) and cortical astrocytes (CRTAS) and wild-type and AMPK α1/α2 null mouse embryonic fibroblasts (MEFs). Cells were treated with drugs known to modulate AMPK activity: A-769662, AICAR and metformin, for up to 3 hours. Intracellular calci

AMP-activated protein kinase phosphorylates and inactivates liver glycogen synthase.

Recombinant muscle GYS1 (glycogen synthase 1) and recombinant liver GYS2 were phosphorylated by recombinant AMPK (AMP-activated protein kinase) in a time-dependent manner and to a similar stoichiometry. The phosphorylation site in GYS2 was identified as Ser7, which lies in a favourable consensus for phosphorylation by AMPK. Phosphorylation of GYS1 or GYS2 by AMPK led to enzyme inactivation by decreasing the affinity for both UDP-Glc (UDP-glucose) [assayed in the absence of Glc-6-P (glucose-6-phosphate)] and Glc-6-P (assayed at low UDP-Glc concentrations). Incubation of freshly isolated rat hep

Serotonin-induced MMP-13 production is mediated via phospholipase C, protein kinase C, and ERK1/2 in rat uterine smooth muscle cells.

Serotonin (5-hydroxytryptamine; 5-HT), acting via the 5-HT(2A) receptor, up-regulates the transcription and production of interstitial collagenase (matrix metalloproteinase-13; MMP-13), a critical enzyme responsible for maintaining the integrity of the uterus, after parturition. Serotonin treatment of rat uterine myometrial smooth muscle cells induced inositol phosphate (IP) turnover, which was abolished by the 5-HT(2A) receptor-specific antagonists ketanserin and spiperone. The phospholipase C (PLC) inhibitors and D609 attenuated serotonin-mediated-IP turnover with a corresponding inhibition

Nitric oxide modulates mechanical strain-induced activation of p38 MAPK in mesangial cells.

Mesangial cells (MC), grown on extracellular matrix (ECM) protein-coated plates and stretched, proliferate and produce ECM, recapitulating in vivo responses to increased glomerular capillary pressure (Pgc). Transduction of strain involves mitogen-activated protein kinases (MAPK), and we have shown that p38 MAPK is activated by strain in MC. Because in vivo studies show that nitric oxide (NO) in the remnant kidney limits glomerular injury without reducing Pgc, we studied whether NO attenuated stretch-induced p38 activation in MC. Increasing p38 activation occurred with increasing stretch, maxim

High-throughput fluorescence-based screening assays for tryptophan-catabolizing enzymes.

Indoleamine 2,3-dioxygenase (IDO1) and tryptophan 2,3-dioxygenase (TDO) are two structurally different enzymes that have a different tissue distribution and physiological roles, but both catalyze the conversion of tryptophan to N-formylkynurenine (NFK). IDO1 has been clinically validated as a small-molecule drug target for cancer, while preclinical studies indicate that TDO may be a target for cancer immunotherapy and neurodegenerative disease. We have developed a high-throughput screening assay for IDO1 and TDO based on a novel chemical probe, NFK Green, that reacts specifically with NFK to f

Catalytic divergencies in the mechanism of L-arginine hydroxylating nonheme iron enzymes.

Many enzymes in nature utilize a free arginine (L-Arg) amino acid to initiate the biosynthesis of natural products. Examples include nitric oxide synthases, which generate NO from L-Arg for blood pressure control, and various arginine hydroxylases involved in antibiotic biosynthesis. Among the groups of arginine hydroxylases, several enzymes utilize a nonheme iron(II) active site and let L-Arg react with dioxygen and α-ketoglutarate to perform either C3-hydroxylation, C4-hydroxylation, C5-hydroxylation, or C4-C5-desaturation. How these seemingly similar enzymes can react with high specificity

Heterologous expression of leader-less pga gene in Pichia pastoris: intracellular production of prokaryotic enzyme.

BACKGROUND: Penicillin G acylase of Escherichia coli (PGAEc) is a commercially valuable enzyme for which efficient bacterial expression systems have been developed. The enzyme is used as a catalyst for the hydrolytic production of beta-lactam nuclei or for the synthesis of semi-synthetic penicillins such as ampicillin, amoxicillin and cephalexin. To become a mature, periplasmic enzyme, the inactive prepropeptide of PGA has to undergo complex processing that begins in the cytoplasm (autocatalytic cleavage), continues at crossing the cytoplasmic membrane (signal sequence removing), and it is com

Patterns of inflammation and immune activation by coreceptor use in people living with HIV-1.

INTRODUCTION: Human immunodeficiency virus type 1 (HIV-1) utilizes either the CCR5 (R5) or CXCR4 (X4) coreceptor for host cell entry. Coreceptor switching from R5 to X4 and elevated immune activation have been associated with disease progression. X4-tropic HIV-1 is predominantly observed in the late stage of infection, when the immune environment characterized by chronic activation is optimal for their replication. The aim of this study was to determine viral tropism in late HIV presenters and who have not previously received treatment in Mexico City and its relationship with markers of chroni

Detection of macroenzymes: establishing upper reference limits for eight enzymes after polyethylene glycol precipitation.

INTRODUCTION: The presence of macroenzymes in blood can cause diagnostic confusion. Therefore, confirming the presence of macroenzymes is important to reduce unnecessary (non-)invasive investigations. Polyethylene glycol (PEG) precipitation is a simple and fast first-line method for the detection of macroenzymes. However, there is no consensus on the upper reference limit for the PEG-precipitable activity (%PPA) of monomeric enzymes. The aim of this study was to verify a PEG precipitation protocol for the detection of macroenzymes in our laboratory by establishing upper reference limits (URLs)

Species specificity of phosphate triester anticholinesterases.

Two widely distributed enzyme systems degrade phosphate triester anticholinesterase agents to inactive phosphate diesters by nonhydrolytic mechanisms. The substrate specificity of these enzymes is discussed and the level of activity in various mammalian livers is described. The distribution of these enzymes is an important factor in accounting for the species specificity of at least some anticholinesterase agents.

The non-canonical CDK2 activator SPY1 uses separable mechanisms to activate phosphorylated and unphosphorylated CDK2.

The cyclin-dependent kinases (CDKs) regulate cell cycle progression subject to strict control by activating cyclin subunits and by site-specific phosphorylation on the activation loop. These mechanisms are highly conserved across eukaryotic organisms. The non-canonical CDK activator Speedy A (SPY1) is unrelated in sequence to the cyclins and activates CDK2 in G1/S phase in somatic cells and promotes meiosis in germ cells without requiring activation-loop phosphorylation of CDK2 on T160. This has been attributed to a single aspartic acid residue of SPY1 (D136) that mimics T160 phosphorylation b

A 16 × 8 Focused Compound Library Comprising All Configurational Isomers of the Archetypal Iminosugar, 1-Deoxynojirimycin.

Deoxynojirimycin is the archetypal iminosugar, from which many structural and configurational isosteres have been derived and studied in past decades. In total, 16 configurational isomers of deoxynojirimycin exist, but these have not yet been collected in a single library. This paper describes the assembly of such a library with each configurational isomer present in eight N-substituted forms. Screening of this library against the three glucosylceramide processing enzymes, glucosylceramide synthase (GCS), lysosomal glucosylceramidase (GBA1), and nonlysosomal glucosylceramidase (GBA2), confirms

N-aryl sulfonamide inhibitors of the bacterial enzyme diaminopimelate desuccinylase (DapE) as potential new antibiotics.

A series of N-aryl sulfonamides was designed, synthesized, and evaluated for their ability to inhibit the bacterial enzyme N-succinyl-L,L-diaminopimelic acid desuccinylase (DapE, EC 3.5.1.18). Our initial lead compound, N-(o-methoxyphenyl)-toluenesulfonamide (4a), identified through a high-throughput screen, showed modest inhibition, with an IC₅₀ of 72.9 μM. Through lead optimization, we improved the potency by modifying key functional groups. Replacing the o-OCH₃ on the aniline ring and p-CH₃ on the sulfonamide ring with p-CF₃ and p-OCH₃, respectively, while retaining the N-CHF₂ hydrophobic m

Protein Arginine Methyltransferase Inhibitors Target Multiple Stages of Plasmodium falciparum Parasites In Vitro.

The ongoing rise in antimalarial drug resistance underscores the urgent need for new drug candidates that specifically target novel mechanisms. Malaria parasites employ various epigenetic strategies to regulate gene expression throughout their complex life cycle, with histone lysine acetylation and methylation being well-studied and targeted by new antiplasmodials. By contrast, arginine methylation remains poorly explored. Plasmodium falciparum possesses three protein arginine methyltransferases (PRMTs) that maintain a unique and combinatorial histone arginine methylation landscape. Here, we p

Design and synthesis of novel quinazoline derivatives as KDM6B selective inhibitors.

The abnormal function of histone lysine demethylase 6B (KDM6B) is closely associated with the development and progression of various human diseases, including cancer, inflammatory disorders, and psychiatric conditions, supporting KDM6B as a significant therapeutic target. However, the development of potent and selective KDM6B inhibitors remains a critical unmet need. Based on the hit compound (A01) discovered by enzyme-level screening, a series of derivatives with quinazoline scaffold were designed, synthesized and identified as KDM6B inhibitors. Among these, compound 13k exhibited optimal pot
