OBJECTIVE: To determine whether Coxiella burnetti is able to survive within free-living amoeba.
METHODS: When C. burnetii was co-cultivated with the free-living amoeba species Acanthamoeba castellanii.
RESULTS: Viable bacteria were detected for 18 days. Microscopic studies confirmed the presence of bacteria within vacuoles, some of which appeared to be spore-like forms.
CONCLUSION: These results indicate that free-living amoebae could provide an intracellular niche for the spore formation and survival of C. burnetii in the environment.

The authors report a case of intracranial meningioma with granulofilamentous inclusions. A 50-year-old man had right trigeminal neuralgia due to trigeminal nerve compression by a petroclival tumor and received tumor resection. Microscopically, tumor cells containing eccentric nuclei and intracytoplasmic hyaline inclusions were arranged in sheets and whorls. The inclusions were negative for periodic acid-Schiff reaction. Eight cases of central neurocytomas were studied by immunohistochemistry and electron microscopy. Seven tumors were located in the lateral ventricles and one in the subependymal region. Cellular retinol binding protein (CRBP) was localized in the rat retina by means of light and electron microscopic immunocytochemistry. The peroxidase-antiperoxidase (PAP) method employed at the light microscopic level showed that CRBP is sharply localized to the retinal pigment epithelium (RPE). A striking network thought to consist of glycosaminoglycan in neonatal cartilage has recently been reported following glycol methacrylate (GMA) dehydration and embedding. This study attempted to learn why the network was not seen in other preparations previously reported. Comparative light and transmission electron microscopy of the network structures was carried out to determine their biological significance. We have shown the networks to be reproducible artefact, attained only following GMA dehydration of glutaraldehyde fixed neonatal cartilage.
