A multi-metric profiling of OFC subregion connectivity identifies system-level alterations in major depressive disorder.

BACKGROUND: Orbitofrontal cortex (OFC) dysfunction contributes to major depressive disorder (MDD), particularly through disrupted network interactions. Current studies lack whole-brain coverage and fine-grained OFC mapping. We employed OFC parcellation and multi-metric connectivity to comprehensively profile OFC-related network disturbances in MDD. METHODS: OFC was parcellated into medial (MOFC), anterior middle (aMidOFC), posterior middle (pMidOFC), and lateral (LOFC) subregions via community analysis of resting-state fMRI from 586 healthy individuals. Using these subregions and whole OFC as

VAD-EmotionNet: a spatial-spectral attention network for EEG emotion recognition in the 3D affective space.

Electroencephalography (EEG) signals are of great significance in human-computer interaction and affective computing, as they can objectively reflect transient neural dynamics in the brain. However, existing methods still face several challenges, including limited training samples, susceptibility of EEG signals to noise interference, and insufficient granularity in conventional two-dimensional emotion models. To address these issues, this study proposes VAD-EmotionNet, an EEG-based emotion recognition model built upon the three-dimensional valence-arousal-dominance (VAD) emotional space. Speci

Spontaneous Activity Patterns in Human Attention Networks Code for Hand Movements.

Recent evidence suggests that, in the absence of any task, spontaneous brain activity patterns and connectivity in the visual and motor cortex code for natural stimuli and actions, respectively. These "resting-state" activity patterns may underlie the maintenance and consolidation (replay) of information states coding for ecological stimuli and behaviors. In this study, we examine whether replay patterns occur in resting-state activity in association cortex grouped into high-order cognitive networks not directly processing sensory inputs or motor outputs. Fifteen participants (7 females) perfo

DeepNEU: cellular reprogramming comes of age - a machine learning platform with application to rare diseases research.

BACKGROUND: Conversion of human somatic cells into induced pluripotent stem cells (iPSCs) is often an inefficient, time consuming and expensive process. Also, the tendency of iPSCs to revert to their original somatic cell type over time continues to be problematic. A computational model of iPSCs identifying genes/molecules necessary for iPSC generation and maintenance could represent a crucial step forward for improved stem cell research. The combination of substantial genetic relationship data, advanced computing hardware and powerful nonlinear modeling software could make the possibility of

Machine learning enhanced prediction of optical properties via reverse Monte Carlo method.

We present a machine learning (ML) approach for the accelerated extraction of optical properties from the experimental reflection electron energy loss spectroscopy (REELS) spectra by the reverse Monte Carlo (RMC) method. Taking platinum (Pt) as an example, experimental REELS spectra recorded at incident electron energies of 2.0 and 1.5 keV over an energy loss range of 0-200 eV are processed by one-dimensional convolutional neural networks trained specifically for each incident energy. The training processes incorporate the physical constraints based on the perfect-screening (ps-) and oscillato

Future horizons: Toward an integrated neuroscience of sleep.

Sleep is now recognized as a vital biological process with roles beyond rest, including neurodevelopment, memory, emotional regulation, metabolism, immune function, and neuroprotection. Advances in neuroscience and technology reveal that sleep is an active, multifaceted process involving multiple brain and body systems. This chapter explores how neural networks, waste clearance, circadian rhythms, immune responses, and cognition interact to influence brain health and disease. We examine sleep's connections to neurodegeneration, mental illness, aging, and cognitive resilience. We also highlight

ChIP-Seq-Based Approach in Mouse Enteric Precursor Cells Reveals New Potential Genes with a Role in Enteric Nervous System Development and Hirschsprung Disease.

Hirschsprung disease (HSCR) is a neurocristopathy characterized by intestinal aganglionosis which is attributed to a failure in neural crest cell (NCC) development during the embryonic stage. The colonization of the intestine by NCCs is a process finely controlled by a wide and complex gene regulatory system. Several genes have been associated with HSCR, but many aspects still remain poorly understood. The present study is focused on deciphering the PAX6 interaction network during enteric nervous system (ENS) formation. A combined experimental and computational approach was performed to identi

Understanding the molecular basis of herbal medicines for cough variant asthma under the guidance of traditional herbal theories.

INTRODUCTION: Despite the clear clinical efficacy of the herbal formula Suhuang in treating cough variant asthma (CVA), its underlying mechanisms of action (MOAs) remain poorly understood. Traditional Chinese Medicine (TCM) offers a unique framework for disease treatment based on traditional herbal theories. However, the molecular basis of these theories remains largely unexplored. METHODS: To address this gap, we proposed a novel computational paradigm to understand how herbal medicines exert therapeutic effects on CVA under the guidance of TCM theories. Our approach integrates transcription

Combined Yoga and Transcranial Direct Current Stimulation Increase Functional Connectivity and Synchronization in the Frontal Areas.

Transcranial direct current stimulation (tDCS) is a non-invasive neuro-stimulation technique that can modulate cortical excitability. Similarly, yoga is shown to affect the brain's neural activity and networks. Here, we aimed to investigate the effect of combined yoga and tDCS on brain oscillations and networks using resting-state electroencephalography recordings. In a randomized, cross-over, double-blind design, twenty-two healthy subjects participated in a yoga/active tDCS session (2 mA; 20 min; anode-F3, cathode F4) or yoga/sham tDCS on 2 separate days. Resting-state EEG data were collecte

Computational modeling of fear and stress responses: validation using consolidated fear and stress protocols.

Dysfunction in fear and stress responses is intrinsically linked to various neurological diseases, including anxiety disorders, depression, and Post-Traumatic Stress Disorder. Previous studies using in vivo models with Immediate-Extinction Deficit (IED) and Stress Enhanced Fear Learning (SEFL) protocols have provided valuable insights into these mechanisms and aided the development of new therapeutic approaches. However, assessing these dysfunctions in animal subjects using IED and SEFL protocols can cause significant pain and suffering. To advance the understanding of fear and stress, this st

GenSo-FDSS: a neural-fuzzy decision support system for pediatric ALL cancer subtype identification using gene expression data.

OBJECTIVE: Acute lymphoblastic leukemia (ALL) is the most common malignancy of childhood, representing nearly one third of all pediatric cancers. Currently, the treatment of pediatric ALL is centered on tailoring the intensity of the therapy applied to a patient's risk of relapse, which is linked to the type of leukemia the patient has. Hence, accurate and correct diagnosis of the various leukemia subtypes becomes an important first step in the treatment process. Recently, gene expression profiling using DNA microarrays has been shown to be a viable and accurate diagnostic tool to identify the

Programmable optoelectronic memristors for energy-efficient adaptive binarized spiking neural networks.

Hardware realization of brain-inspired vision critically relies on neuromorphic devices that couple sensing, computation, and adaptive learning, yet such integration remains limited by unstable ion dynamics, poor controllability of device plasticity, and the learning-rate sensitivity of binarized networks. Here, we introduce an energy-efficient optoelectronic memristive platform based on Ag/Sb2Se3/SnO2/FTO devices, in which a 10 nm SnO2 interfacial layer confines Ag-ion migration, enabling stable bipolar resistive switching with an on/off ratio of ∼10 and retention exceeding 104 s. The device

Human superior colliculus pathways represent the form and motion of looming objects.

The superior colliculus is a conserved midbrain structure critical for detecting and responding to threats, yet its role in human cognition and emotion remains poorly understood. Using task-based fMRI and computational modeling in two experiments, we investigated how the superior colliculus responds to looming threats. We found that the human superior colliculus encodes information about the motion and category of looming objects present in naturalistic videos and controlled experimental stimuli. Encoding models based on task-optimized neural networks for collision detection, object recognitio

The direct influence of retinal degeneration on electrical stimulation efficacy: Significant implications for retinal prostheses.

Photoreceptor loss and inner retinal network remodeling severely impacts the ability of retinal prosthetic devices to create artificial vision. We developed a computational model of a degenerating retina based on rodent data and tested its response to retinal electrical stimulation. This model includes detailed network connectivity and diverse neural intrinsic properties, capable of exploring how the degenerated retina influences the performance of electrical stimulation during the degeneration process. Our model suggests the possibility of quantitatively modulating retinal ON and OFF pathways

Multi-plasticity synergy with adaptive mechanism assignment for training spiking neural networks.

Spiking Neural Networks (SNNs) are promising brain-inspired models known for low power consumption and superior potential for temporal processing, but identifying suitable learning mechanisms remains a challenge. Despite the presence of multiple coexisting learning strategies in the brain, current SNN training methods typically rely on a single form of synaptic plasticity, which limits their adaptability and representational capability. In this paper, we propose a biology-motivated computational framework that incorporates multiple synergistic plasticity mechanisms for more effective SNN train

Patterns of Neural Functional Connectivity in Infants at Familial Risk of Developmental Dyslexia.

IMPORTANCE: Developmental dyslexia is a heritable learning disability affecting 7% to 10% of the general population and can have detrimental impacts on mental health and vocational potential. Individuals with dyslexia show altered functional organization of the language and reading neural networks; however, it remains unknown how early in life these neural network alterations might emerge. OBJECTIVE: To determine whether the early emergence of large-scale neural functional connectivity (FC) underlying long-term language and reading development is altered in infants with a familial history of

Spiking Neural Networks for Temporal Processing: Status Quo and Future Prospects.

Temporal processing is fundamental for biological and artificial intelligence systems, as it enables the comprehension of dynamic environments and facilitates timely responses. Spiking neural networks (SNNs) are well-suited for efficient temporal processing owing to their rich neuronal dynamics and event-driven computation. Given the recent surge in the development of SNNs, there is an urgent need for a comprehensive evaluation of their temporal processing capabilities. In this article, we first conduct an in-depth assessment of commonly used neuromorphic benchmarks, revealing critical limitat

Rapid computation of high-level visual surprise.

Predictive processing theories propose that the brain continuously generates expectations about incoming sensory information. Discrepancies between these predictions and actual inputs, sensory prediction errors, guide perceptual inference. A fundamental yet largely unresolved question is which stimulus features the brain predicts, and therefore, what kind of surprise drives neural responses. Here, we investigated this question using electroencephalography and computational modeling based on deep neural networks (DNNs). Participants viewed object images whose identity was probabilistically pred

Visual Imagery and Perception Share Neural Representations in the Alpha Frequency Band.

To behave adaptively with sufficient flexibility, biological organisms must cognize beyond immediate reaction to a physically present stimulus. For this, humans use visual mental imagery [1, 2], the ability to conjure up a vivid internal experience from memory that stands in for the percept of the stimulus. Visually imagined contents subjectively mimic perceived contents, suggesting that imagery and perception share common neural mechanisms. Using multivariate pattern analysis on human electroencephalography (EEG) data, we compared the oscillatory time courses of mental imagery and perception
