[A Seven-Year Evaluation of Viral Central Nervous System Infections].

Identification of viral agents causing central nervous system (CNS) infections increased by the application of nucleic acid tests. In this s … ns' values. In the seven-year period, 7.7% of the1185 patients' CSF samples were positive for viral nucleic acids. As expected, enteroviruses were the most common pathogens in children and detected mainly in summer-autumn period. Syndromic approach in CNS infections could increase the viral pathogen detection.

Amino acid-anticodon binding specificity: rationale for a new class of therapeutic agent.

In this article a new class of anticancer and antiviral drugs is discussed. These new drugs consist of small di- and tri-peptides, designed to bind to single-stranded (ss) regions that are crucial for the expression of genes such as the c-myc oncogene in cancers and start sites (and other ss regions) of viral pathogenic genes. The components (i.e. the amino acids and the sequences they form) of these peptides could be dictated by the specific binding of amino acids to their ss anticodons in tRNA. Cancer cell viability depends on the continued overexpression of the c-myc oncogene, and thus this

CPP2Vec: A representation learning approach for cell-penetrating peptides prediction.

BACKGROUND: Cell-penetrating peptides (CPPs) facilitate the delivery of a variety of therapeutic molecules across the plasma membrane, from small chemical substances to nucleic acid-based macromolecules, such as antisense oligonucleotides (ASOs). Among neutral ASOs, peptide nucleic acids (PNAs) and phosphorodiamidate morpholino oligomers (PMOs) have been extensively studied as potential medical treatments for Duchenne Muscular Dystrophy (DMD), a severe genetic disease that causes muscle degeneration progressively. Over the last few decades, many in silico methods have emerged to detect novel C…

A De Novo Noncoding RARB Variant Associated with Complex Microphthalmia Alters a Putative Regulatory Element.

Retinoic acid receptor beta (RARB) is a transcriptional regulator crucial for coordinating retinoic acid- (RA-) mediated morphogenic movements, cell growth, and differentiation during eye development. Loss- or gain-of-function RARB coding variants have been associated with microphthalmia, coloboma, and anterior segment defects. We identified a de novo variant c.157+1895G>A located within a conserved region (CR1) in the first intron of RARB in an individual with complex microphthalmia and significant global developmental delay. Based on the phenotypic overlap, we further investigated the possib

[Anti-deoxyribonucleic acid antibodies in podagra].

Antibodies to native (n) and denatured (d) DNA were studied in 70 patients with primary gout and 40 experimental animals with hyperuricemia. Titres of anti-nDNA in patients with gout considerably exceeded similar indices in donors and were in inverse relationship with the severity of arthritis and markedness of morphological manifestations of renal affection. Anti-nDNA directly correlated with the count of B-lymphocytes in the blood and indirectly with other immunological indices. Anti-dDNA directly correlated with the count of B-cells, reaction of lymphocytes to Kon-A and the titres of antire

De novo genes originate in regions of ancestrally closed chromatin.

The evolution of de novo genes from ancestrally nontranscribed sequences has been reported in a wide range of taxa. However, the DNA sequence changes responsible for the origin of de novo genes are poorly understood. In this study, we investigated whether de novo genes evolve by exploiting ancestral regulatory sequences or are associated with the evolution of new regulatory regions. We focused on the youngest stratum of de novo genes that have polymorphic expression in the male accessory gland of Drosophila melanogaster and show no evidence of transcription in other Drosophila species. DNA conformational flexibility descriptors improve transcription factor binding prediction across diverse transcription factor families.

Precise transcription factor (TF) binding to DNA governs gene regulation, yet nucleotide sequence alone often fails to fully capture binding specificity. Although static DNA shape is a recognized determinant of indirect readout, the role of intrinsic conformational flexibility remains underexplored across TF families. Here, we demonstrate that integrating sequence-derived DNA flexibility descriptors into predictive models improves both prediction and mechanistic interpretability of TF-DNA affinity. Across large-scale in vitro data sets encompassing HT-SELEX and protein-binding microarrays for

Low-polar solvent strikingly stiffens double-stranded RNA and reverses its twist-stretch coupling.

Cellular environments are crowded systems with reduced solvent polarity, yet how solvent polarity shapes RNA elasticity remains unclear. In this work, our high-precision magnetic tweezers and all-atom molecular dynamics simulations showed that decreasing solvent polarity with ethanol as a model cosolvent produces a biphasic response for double-stranded (ds) RNA: moderate ethanol concentration softens dsRNA, causing a slight decrease in bending persistence length P and stretch modulus S, but high ethanol concentration markedly stiffens dsRNA, reflected by the about twofold increase in P and abo

Cascaded SDA-Exo III amplification with machine learning-driven multi-modal data fusion for self-powered biosensing.

BACKGROUND: Pokkah boeng disease, caused by Fusarium spp., is a destructive disease that can leads to substantial sugarcane yield losses under epidemic conditions. Early detection is extremely challenging due to the absence of visible symptoms during early-stage infection. Conventional diagnostic methods rely on sophisticated instrumentation and external power sources, which fundamentally restrict their practical application in resource-limited agricultural settings. To address these limitations, we develop a smart biosensing platform that breaks through the constraints of traditional methods,

An intimate view of Leishmania infantum chromosome ends reveals less conserved subtelomeric regions and variations in the telomeric repeat.

Leishmania infantum is an intracellular protozoan parasite that causes visceral leishmaniasis, predominantly affecting marginalized populations in tropical and subtropical countries. The parasite genome comprises 36 chromosomes whose ends have not been characterized. In most eukaryotes, chromosome termini are capped by telomeres and associated proteins, forming nucleoprotein structures that maintain genome stability and prevent the ends from being mistaken as broken DNA. Leishmania telomeres are composed of G-rich repetitive DNA replenished by telomerase activity. Here, we showed a detailed vi

DNA Nanostructure Self-Assembly in an Aqueous Ionic Liquid Solution with Enhanced Stability and Target Binding Affinity.

DNA nanostructure-enabled functional constructs have shown potential to improve healthcare outcomes by offering advanced disease diagnostic and therapeutic strategies. Translating this potential of DNA nanostructure-based constructs to real life applications relies on maintaining and enhancing the structural integrity and functions of the surface-anchored moieties. In this study, we explored the possibility of utilizing choline dihydrogen phosphate (CDHP) solution, an aqueous solution of ionic liquid, to assemble DNA nanostructures of different sizes and complexities with enhanced biostability

Cell-Free DNA and Mitochondria in Parkinson's Disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder marked by the gradual and irreversible loss of neurons, especially within the substantia nigra region of the midbrain. Early and accurate diagnosis remains a significant challenge in both research and clinical practice. This difficulty is further compounded by the substantial clinical and molecular heterogeneity of PD, emphasizing the urgent need for reliable biomarkers to enhance diagnostic precision and guide therapeutic strategies. One promising candidate biomarker is cell-free DNA (cfDNA), comprising short DNA fragments c

Co2+-mediated adsorption facilitates atomic force microscopy of DNA molecules at double-helix resolution.

Atomic force microscopy (AFM) has demonstrated the ability to resolve single DNA molecules in liquid at a spatial resolution that is sufficient to visualize the double helix structure and variations therein. Such variations can be due to inherent configurational flexibility and may be related to, e.g., DNA sequence, ionic screening, supercoiling, or protein binding. These AFM experiments require DNA to be adhered to a solid and preferably flat support. For high-resolution, in-liquid AFM studies so far, such adhesion has commonly been achieved using Ni2+ ions to electrostatically bridge between

Rolling Circle Amplification Boosts Ultrasensitive Liquid Biopsy.

Liquid biopsy is a minimally invasive and convenient diagnostic and monitoring method for diseases, involving the collection and analysis of samples such as blood, urine, and cerebrospinal fluid. However, due to the intricate matrix composition of biological samples, there remains a need for further enhancement of sensitivity and specificity in current liquid biopsy methods to adequately meet clinical requirements. Rolling circle amplification (RCA) is an efficient isothermal enzymatic reaction capable of rapidly producing numerous ultra-long single-stranded DNA molecules with repetitive seque
