Neonatal isolation delays the developmental decline of long-term depression in the CA1 region of rat hippocampus.

Activity-dependent alterations of synaptic efficacy or connectivity are essential for the development, signal processing, and learning and memory functions of the nervous system. It was observed that, in particular in the CA1 region of the hippocampus, low-frequency stimulation (LFS) became progressively less effective at inducing long-term depression (LTD) with advancing developmental age. The physiological factors regulating this developmental plasticity change, however, have not yet been elucidated. Here we examined the hypothesis that neonatal isolation (once per day for 1 h from postnatal

Pharmacological dissection of calcium channel subtype-related components of strontium inflow in large mossy fiber boutons of mouse hippocampus.

Several subtypes of voltage-dependent calcium channels (VDCCs) are present in the presynaptic terminals. In the mammalian hippocampus, P/Q-, N-, and R- but not L-type VDCCs are involved in the fast transmitter release from large mossy fiber (MF) boutons, which are associated with CA3 pyramidal cell dendrites. We investigated whether L-type VDCCs are indeed absent in these large MF boutons. With the use of Sr2+ as the Ca2+ substitute, the stimulus-evoked Sr2+ increment (delta[Sr2+]pre) was evaluated fluorometrically. Delta[Sr2+]pre appeared to be proportional to Sr2+ inflow through VDCCs and wa

Of rodents and humans: A comparative review of the neurobehavioral effects of early life SSRI exposure in preclinical and clinical research.

Selective serotonin reuptake inhibitors (SSRIs) have been a mainstay pharmacological treatment for women experiencing depression during pregnancy and postpartum for the past 25 years. SSRIs act via blockade of the presynaptic serotonin transporter and result in a transient increase in synaptic serotonin. Long-lasting changes in cellular function such as serotonergic transmission, neurogenesis, and epigenetics, are thought to underlie the therapeutic benefits of SSRIs. In recent years, though, growing evidence in clinical and preclinical settings indicate that offspring exposed to SSRIs in uter

The structural localization of galanin, and its function in modulating acetylcholine release in the olfactory bulb of adult rat.

The localization of galanin immunoreactivity was analyzed within the olfactory bulb of adult rats. Galanin-positive neurons were differentially distributed among the bulb layers. The density of stained neurons was highest in the glomerular and external plexiform layers. According to morphology, size, location and arrangement, a large proportion of galanin-immunoreactive neurons corresponds to external tufted cells and short-axon neurons in the superficial part of the external plexiform and glomerular layers. A smaller number were middle tufted cells and short-axons neurons while only a few sho

GABAergic synaptic inputs of locus coeruleus neurons in wild-type and Mecp2-null mice.

Rett syndrome is an autism spectrum disorder resulting from defects in the gene encoding the methyl-CpG-binding protein 2 (MeCP2). Deficiency of the Mecp2 gene causes abnormalities in several systems in the brain, especially the norepinephrinergic and GABAergic systems. The norepinephrinergic neurons in the locus coeruleus (LC) modulate a variety of neurons and play an important role in multiple functions in the central nervous system. In Mecp2(-/Y) mice, defects in the intrinsic membrane properties of LC neurons have been identified, while how their synaptic inputs are affected remains unclea

Wallerian degeneration of injured axons and synapses is delayed by a Ube4b/Nmnat chimeric gene.

Axons and their synapses distal to an injury undergo rapid Wallerian degeneration, but axons in the C57BL/WldS mouse are protected. The degenerative and protective mechanisms are unknown. We identified the protective gene, which encodes an N-terminal fragment of ubiquitination factor E4B (Ube4b) fused to nicotinamide mononucleotide adenylyltransferase (Nmnat), and showed that it confers a dose-dependent block of Wallerian degeneration. Transected distal axons survived for two weeks, and neuromuscular junctions were also protected. Surprisingly, the Wld protein was located predominantly in the

Derivation of an intrinsic brain activity biomarker for the earliest prediction of cognitive decline.

Early identification of cognitive decline in the elderly is essential for timely and effective intervention. Current neuroimaging and clinical biomarkers have largely focused on patients with mild cognitive impairment (MCI) and dementias, and though effective, have not been demonstrated to be sensitive to the earliest signs of functional abnormalities. The electroencephalogram (EEG), with a millisecond time base, allows a precise exploration of synaptic dysfunction even at the initial stages of the damage. This study introduces a novel non-invasive EEG-based biomarker to predict cognitive decl

Effects of Electroacupuncture on Syt3 and GluA2 in Rats With Limb Spasms After Intracerebral Hemorrhage.

BACKGROUND: Clinical studies have confirmed that electroacupuncture (EA) has the potential to improve spasticity after intracerebral hemorrhage (ICH), yet its precise mechanism remains unclear. Synaptotagmin-3 (SYT-3), by mediating the internalization of the glutamate AMPA receptor GLUA2, may be related to EA's mechanism. This study aims to explore the mechanism by which EA improves limb spasticity after ICH, providing scientific evidence for its clinical application. METHODS: ICH models were established using stereotaxic injection of autologous tail blood into the right striatum. SD rats wer

A hypothalamic digoxin-mediated model for autism.

The isoprenoid pathway and its metabolites--digoxin, dolichol, and ubiquinone--were assessed in autism. The isoprenoid pathway and digoxin status was also studied for comparison in individuals of differing hemispheric dominance to determine the role of cerebral dominance in the genesis of autism. There was an upregulation of the isoprenoid pathway as evidenced by elevated HMG CoA reductase activity in autism. Digoxin, an endogenous Na+-K+ ATPase inhibitor secreted by the hypothalamus, was found to be elevated and RBC membrane Na+-K+ ATPase activity was found to be reduced in autism. Membrane N

Additive Enhancement of Neurovascular Regeneration and Functional Recovery by Combined MSC-Exosomes and Low-Intensity Pulsed Ultrasound in Ischemic Stroke Rats.

Cerebral ischemic stroke (CIS) leads to high mortality and disability due to cerebrovascular obstruction. Current therapeutic approaches such as thrombolysis have limitations, and many patients suffer from long-term functional impairments, so new treatment strategies are urgently needed. Both low-intensity pulsed ultrasound (LIPUS) and mesenchymal stem cell-derived exosomes (MSC-EXOs) have demonstrated some therapeutic potential for CIS, but their efficacy is less pronounced when used alone. Therefore, this study aims to evaluate whether the combination of MSC-EXOs and LIPUS exerts an additive

Cryo-EM Structures of Apo, Agonist- and Antagonist-Bound Heteromeric Kainate Receptors.

UNLABELLED: Kainate receptors (KARs) mediate excitatory synaptic transmission and regulate neurotransmitter release. In the central nervous system, KARs predominantly exist as heterotetramers comprising low-affinity (GluK1-3) and high-affinity (GluK4-5) subunits, with GluK2/GluK5 being the most abundant. To elucidate their conformational transitions, we determined the cryo-electron microscopy (cryo-EM) structures of GluK2/GluK5 KARs in the apo and distinct ligand-bound states. The apo structure revealed compact packing with extensive inter-subunit interactions between the ligand-binding domain…

Spinal 7-dehydrocholesterol reductase dependent cholesterol overload regulates astrocyte activation and mGluR5/ERK cascades via monocyte chemotactic protein 3 in tibial fracture-induced chronic pain in mice.

Patients with bone fractures commonly experience an exasperating and long-lasting pain following orthopedic procedures. Spinal monocyte chemotactic protein 3 (MCP-3)-encoding astrogliosis and mGluR5-mediated excitatory synaptic plasticity are essential for central nociceptive transmission. Recent studies highlight the requirement of 7-dehydrocholesterol reductase (DHCR7) for cholesterol overload and neuroinflammation. This present study investigates the potential associations among DHCR7, MCP-3 and mGluR5 in chronic pain using a mouse model of tibial fracture with intramedullary pinning. Here,

Cocaine-induced interindividual variability in female rats: From behavior to gene expression.

Cocaine use disorder is marked by strong interindividual variability, yet the molecular determinants underlying vulnerability in females remain poorly characterized. Here, we adapted a statistical model previously validated in males to classify female rats exposed to a cocaine-induced conditioned place preference paradigm into two subgroups: those expressing cocaine reward (CPP expression (CPPE), score >74 s) and those showing no preference ((nCPPE), score between -19 and 53 s), independently of estrous-cycle influences. Despite identical cocaine exposure, two clear behavioral phenotypes emerg
