Plant-Derived Peptide-Polymer Therapeutics for Cutaneous Infections and Inflammation: Mechanistic Basis, Delivery Design and Translational Considerations.

Cutaneous infections and chronic inflammatory wounds remain difficult to treat because antimicrobial resistance, polymicrobial biofilms, excessive protease activity, oxidative stress, and impaired barrier repair collectively reduce the effectiveness of conventional topical therapies. Plant-derived antimicrobial peptides (AMPs) and peptide-associated bioactives offer antimicrobial, antibiofilm, immunomodulatory, and tissue reparative potential; however, their clinical translation is limited by proteolytic instability, poor stratum corneum penetration, short cutaneous residence time, formulation

Integrating delivery systems and microenvironmental cues to accelerate clinical translation of cardiac reprogramming.

Direct cardiac reprogramming, which converts cardiac fibroblasts into functional cardiomyocytes, has emerged as a promising regenerative strategy for heart failure. Preclinical studies have demonstrated its potential to mitigate adverse remodeling; however, critical challenges remain in delivery precision, reprogramming efficiency, and scalability for clinical translation. Here, we review innovations in delivery systems and microenvironmental engineering aimed at overcoming these barriers. Integration-free viral vectors (e.g. Sendai virus and adeno-associated virus 5) and non-viral platforms (

Stem cell-derived extracellular vesicles for rare diseases: A clinical‑bottleneck‑to‑mechanism framework for translation.

Rare diseases impose a disproportionate clinical burden, and yet therapeutic progress is hindered by small cohorts, biological heterogeneity, and limited disease-specific options. Stem cell-derived extracellular vesicles (EVs), and especially exosome-enriched products, are emerging as adaptable cell-free therapeutics that preserve key paracrine activities of parent cells while offering improved controllability, engineering flexibility, and potentially lower acute immunogenicity than living-cell products. This review proposes a clinically driven bottleneck-to-mechanism framework for rare-diseas

Advances in 3D culture systems for maintaining prostate tissue architecture and functional ex vivo prostate organ culture for biomedical applications.

Model systems are inevitable for biomedical research, and advanced prostate three dimensional (3D) models help bridge the gap between in vitro and in vivo systems. Prostate diseases are biologically complex and regulated by hormone signaling, stromal-epithelial interactions, immunological infiltration, and glandular microenvironmental factors. Traditional two-dimensional (2D) cell culture techniques and prostatic cell lines exhibit limitations for translational research, as they reflect varied phenotypic and genotypic characteristics compared to native prostate tissue. Recent advancements in p

From molecular networks to the clinic: a systems biology-nanomedicine roadmap for osteoarthritis diagnosis, regeneration, and safety-by-design.

Osteoarthritis (OA) is a severe, whole-joint illness that is characterized by the permanent destruction of articular cartilage. The failure of traditional therapies and early tissue engineering efforts is mainly due to their reductionist biological strategies and reliance on bulk, non-biomimetic materials that cannot replicate the complex nanoscale architecture of the native extracellular matrix. This review proposes a closed-loop, multidisciplinary roadmap that integrates systems biology and biomimetic nanomedicine to advance OA diagnosis, cartilage regeneration, and nanomaterial safety-by-de…

Microfluidic engineering of polymer-based drug delivery systems: From particle synthesis to organ-on-a-chip validation and application.

The convergence of microfluidic engineering and organ-on-a-chip (OoC) technology is redefining the development and preclinical validation of polymer-based drug delivery systems and biomedical preparations. This review presents a bidirectional framework: first, how microfluidics enables precision synthesis of polymer particles with controlled size, morphology and multifunctionality; second, how OoC platforms serve as physiologically relevant testbeds to evaluate these particles under dynamic, human-mimetic conditions. We examine droplet microfluidics principles for generating monodisperse polym

Engineering manganese ferrite shell on iron oxide nanoparticles for enhanced T1 magnetic resonance imaging.

Doping Mn (II) ions into iron oxide (IO) as manganese ferrite (MnIO) has been proved to be an effective strategy to improve T1 relaxivity of IO nanoparticle in recent years; however, the high T2 relaxivity of MnIO nanoparticle hampers its T1 contrast efficiency and remains a hurdle when developing contrast agent for early and accurate diagnosis. Herein, we engineered the interfacial structure of IO nanoparticle coated with manganese ferrite shell (IO@MnIO) with tunable thicknesses. The Mn-doped shell significantly improve the T1 contrast of IO nanoparticle, especially with the thickness of ∼0.

Phosphorous pentoxide-free bioactive glass exhibits dose-dependent angiogenic and osteogenic capacities which are retained in glass polymeric composite scaffolds.

Bioactive glasses (BGs) are attractive materials for bone tissue engineering because of their bioactivity and osteoinductivity. In this study, we report the synthesis of a novel phosphorous pentoxide-free, silicate-based bioactive glass (52S-BG) composed of 52.1% SiO2, 23.2% Na2O and 22.6% CaO (wt%). The glass was thoroughly characterized. The biocompatibility and osteogenic properties of 52S-BG particles were analyzed in vitro with human adipose-derived mesenchymal stem cells (AdMSCs) and human osteoblasts. 52S-BG particles were biocompatible and induced mineralized matrix deposition and the …

Tissue-mimetic hybrid bioadhesives for intervertebral disc repair.

Intervertebral disc (IVD) degeneration and herniation often necessitate surgical interventions including a discectomy with or without a nucleotomy, which results in a loss of the normal nucleus pulposus (NP) and a defect in the annulus fibrosus (AF). Due to the limited regenerative capacity of the IVD tissue, the annular tear may remain a persistent defect and result in recurrent herniation post-surgery. Bioadhesives are promising alternatives but show limited adhesion performance, low regenerative capacity, and inability to prevent re-herniation. Here, we report hybrid bioadhesives that combi

The systemic homeostatic integration-field theory (SHIFT): Lipid-based particle ensemble as a candidate ascending observational layer for whole-body homeostasis.

Classical physiology elegantly explains acute homeostatic regulation via descending neuroendocrine feedback loops. However, a fundamental conceptual gap remains: how do central regulators continuously integrate spatially distributed, slowly accumulating tissue stresses-such as oxidative damage and lipotoxic drift-within a strongly mixed circulatory system? Free molecular markers face major physical limits, including rapid clearance kinetics and high-dimensional complexity, precluding them from serving as stable, long-term integrators. To bridge this gap, I propose the Systemic Homeostatic Inte

T. gondii excretory proteins promote the osteogenic differentiation of human bone mesenchymal stem cells via the BMP/Smad signaling pathway.

BACKGROUND: Bone defects, resulting from substantial bone loss that exceeds the natural self-healing capacity, pose significant challenges to current therapeutic approaches due to various limitations. In the quest for alternative therapeutic strategies, bone tissue engineering has emerged as a promising avenue. Notably, excretory proteins from Toxoplasma gondii (TgEP), recognized for their immunogenicity and broad spectrum of biological activities secreted or excreted during the parasite's lifecycle, have been identified as potential facilitators of osteogenic differentiation in human bone mar…

Synthesis and characterization of a magnetic bacterial cellulose-chitosan nanocomposite and evaluation of its applicability for osteogenesis.

INTRODUCTION: Natural biopolymers are used for various purposes in healthcare, such as tissue engineering, drug delivery, and wound healing. Bacterial cellulose and chitosan were preferred in this study due to their non-cytotoxic, biodegradable, biocompatible, and non-inflammatory properties. The study reports the development of a magnetic bacterial cellulose-chitosan (BC-CS-Fe3O4) nanocomposite that can be used as a biocompatible scaffold for tissue engineering. Iron oxide nanoparticles were included in the composite to provide superparamagnetic properties that are useful in a variety of appl…

Injectable microspheres adhering to the cartilage matrix promote rapid reconstruction of partial-thickness cartilage defects.

An effective treatment for the irregular partial-thickness cartilage defect in the early stages of osteoarthritis (OA) is lacking. Cartilage tissue engineering is effective for treating full-thickness cartilage defects with limited area. In this study, we designed an injectable multifunctional poly(lactic-co-glycolic acid) (PLGA) microsphere to repair partial-thickness cartilage defects. The microsphere was grafted with an E7 peptide after loading the microsphere with kartogenin (KGN) and modifying the outer layer through dopamine self-polymerization. The microsphere could adhere to the cartil…

Chondrogenic potential of PMSCs cultured on chondroitin sulfate/gelatin-modified DBM scaffold.

INTRODUCTION: Osteoarthritis is one of the most common orthopedic diseases that gradually causes wear and damage to the articular Subchondral bone due to the destruction of articular cartilage. One of the basic challenges in cartilage tissue engineering is the choice of scaffold. In the design of the cartilage scaffold, it is useful to consider parameters such as porosity, water absorption, high mechanical resistance, biocompatibility, and biodegradability. Therefore, in this study, demineralized bone matrix (DBM), which inherently has these characteristics to some extent, was chosen as the ba…

Osteogenic differentiation and proliferation potentials of human bone marrow and umbilical cord-derived mesenchymal stem cells on the 3D-printed hydroxyapatite scaffolds.

Mesenchymal stem cells (MSCs) are a promising candidate for bone repair. However, the maintenance of MSCs injected into the bone injury site remains inefficient. A potential approach is to develop a bone-liked platform that incorporates MSCs into a biocompatible 3D scaffold to facilitate bone grafting into the desired location. Bone tissue engineering is a multistep process that requires optimizing several variables, including the source of cells, osteogenic stimulation factors, and scaffold properties. This study aims to evaluate the proliferation and osteogenic differentiation potentials of …

Cells Derived from Human Long Bone Appear More Differentiated and More Actively Stimulate Osteoclastogenesis Compared to Alveolar Bone-Derived Cells.

Osteoblasts derived from mouse skulls have increased osteoclastogenic potential compared to long bone osteoblasts when stimulated with 1,25(OH)2 vitamin D3 (vitD3). This indicates that bone cells from specific sites can react differently to biochemical signals, e.g., during inflammation or as emitted by bioactive bone tissue-engineering constructs. Given the high turn-over of alveolar bone, we hypothesized that human alveolar bone-derived osteoblasts have an increased osteogenic and osteoclastogenic potential compared to the osteoblasts derived from long bone. The osteogenic and osteoclastogen
