Ultrastructure of dopaminergic varicosities revealed by cryo-correlative light and electron microscopy.

Dopaminergic neurons are fundamental in governing motivation, movement, and many aspects of cognition. The targeted modulation of dopaminergic signaling serves as a cornerstone in developing therapeutic interventions for conditions such as Parkinson's disease, schizophrenia, and addiction. Despite the pivotal role of dopaminergic neurons, the ultrastructure of dopaminergic synapses remains poorly understood. Here, we establish and utilize a cryo-correlative light and electron microscopy process to investigate the micro/nanoscale architecture and organelle content of dopaminergic varicosities.

Loss of SREBF1 disrupts lipid signaling and causes slow muscle fiber degeneration.

SREBF1, encoding the lipogenic transcription factor SREBP1, has been linked to lean mass by human genomics. We used a CRISPR-Cas9 srebf1 zebrafish knockout combined with LC-MS/MS lipidomics, RNA-seq, Lugol-enhanced micro-CT imaging, and functional assays. Srebp1 loss disrupted slow-fiber organization, increased locomotion in larvae, and reduced muscle mass in adults. This knockout muscle showed altered fatty acid composition, reduced arachidonoylethanolamide (AEA), elevated palmitoylethanolamide (PEA), broad remodeling of polyunsaturated fatty (PUFA) acid-derived mediators, and differential ex

Short-Chain Fatty Acids as Endogenous Inhibitors of DPP-IV Enzyme: Implications in GLP-1 Stability and Metabolic Health.

Dipeptidyl peptidase-IV (DPP-IV/CD26) is a circulating serine protease that rapidly degrades incretin hormones, including glucagon-like pept … n (Ki = 2.6 nM). SCFAs protected GLP-1 from DPP-IV mediated degradation and preserved GLP-1-induced calcium signaling in INS-1 cells. Molecular docking indicated preferential SCFA interactions within the DPP-IV β-propeller domain. These findings identify SCFAs as direct, reversible DPP-IV modulators, providing a molecular link between gut microbial metabolites and …

Polydatin alleviates vascular calcification via the TP53/PI3K/AKT axis.

Vascular calcification (VC) is a major complication of chronic kidney disease (CKD), diabetes, atherosclerosis, and aging that significantly increases cardiovascular mortality, yet effective therapeutic interventions are currently limited. In this study, we demonstrated that Polydatin (PD), a natural polyphenolic compound, effectively alleviated VC across in vitro, ex vivo and in vivo models. PD treatment markedly reduced calcium deposition, decreased alkaline phosphatase (ALP) activity, and downregulated the expression of osteogenic markers (RUNX2 and BMP2). Network pharmacology analysis iden

The tubulin code in acute ischemic stroke: post-translational reprogramming of the neuronal cytoskeleton and implications for phase-specific neuroprotection.

Acute ischemic stroke initiates a rapid convergence of excitotoxicity, calcium overload, oxidative stress, neuroinflammation, and energetic failure. Although reperfusion therapies remain central to acute care, they do not directly address the structural failure of injured neuronal networks. This narrative review examines the neuronal microtubule cytoskeleton as an active regulatory platform whose function is governed by the tubulin code: tubulin isotype composition and post-translational modifications. The review integrates evidence that ischemia alters alpha-tubulin K40 acetylation, detyrosin

Electrical Microenvironment Reconstruction and the Application of Biomaterials in Spinal Cord Injury.

The key challenge in restoring neural function after spinal cord injury stems from a vicious cycle triggered by the collapse of the bioelectrical microenvironment at the injury site: an 'electrical silence-neuronal degeneration-glial proliferation' cascade that conventional therapies fail to reverse. This review systematically summarizes the pathological mechanisms of electrical microenvironment imbalance and its critical role in neural regeneration. Furthermore, current intervention strategies based on biomaterials are outlined: evolving from passive reconstruction of electrical pathways usin

Self-Assembled Nanomaterials for ER-Targeted Cancer Therapy: From Molecular Design to Therapeutic Applications.

The endoplasmic reticulum (ER) is essential for protein folding, lipid metabolism, calcium homeostasis, and cellular stress signaling. Cancer cells endure chronic ER stress from elevated metabolic demands and oxidative conditions, adapting ER pathways to evade apoptosis, while promoting growth, survival, and drug resistance. This dysregulated ER state presents a strategic therapeutic target. Self-assembled nanomaterials provide precise ER localization, significantly enhancing treatment efficacy while reducing systemic toxicity. This review details recent advances in their design for ER-targete

Hepatocyte-to-intestinal stem cell remote communication regulates blood glucose homeostasis.

The liver is known to play a pivotal role in modulating blood glucose homeostasis through intrahepatic glucose metabolism. Here, we reveal a unique mechanism by which fatty liver exacerbates hyperglycemia through remote communication from hepatocytes to intestinal stem cells (ISCs), independent of enhanced intrahepatic gluconeogenesis. Mechanistically, hepatocyte-derived alkaline phosphatase (ALP) targets α2δ-1 in ISCs to promote the membrane translocation of Cav1.2. This process triggers increased intracellular calcium levels, which subsequently activates the calcineurin/NFATC2 signaling axis

CaMKIIα holoenzymes self-organize into chain-like mesoscale clusters.

Calcium- and calmodulin-dependent protein kinase II (CaMKII) is highly enriched in dendritic spines at concentrations comparable to those of cytoskeletal proteins and plays a central role in synaptic plasticity. During long-term potentiation, CaMKIIα further accumulates in spines. However, the mechanisms governing its higher-order organization remain poorly understood. Here, we use high-speed atomic force microscopy to visualize interholoenzyme interaction of CaMKIIα at mesoscopic scales (5 to 500 nanometers). Under freely diffusible conditions, CaMKIIα holoenzymes do not form stable clusters.

Human iPSC-derived engineered heart tissue model of diastolic dysfunction in heart failure with preserved ejection fraction.

The prognosis for heart failure (HF) with preserved ejection fraction (HFpEF) remains poor, with treatment evidence and studies at the human cellular level limited. Here, we aimed to model HFpEF-associated diastolic dysfunction in vitro by generating human engineered heart tissues (hEHTs) using human induced pluripotent stem cells and culturing the tissues under high fatty acid and L-NG-nitroarginine methyl ester supplementation. Medium-loaded hEHTs showed a marked reduction in relaxation function while preserving contraction function; secreted high levels of the HF marker, BNP; exhibited abno

Cerebellar hypoplasia caused by calcium/calmodulin-dependent serine protein kinase deficiency.

Calcium/calmodulin-dependent serine protein kinase (CASK) is an X-linked multidomain scaffolding protein originally identified as an intracellular binding partner for Neurexins, a family of presynaptic cell-adhesion molecules. Loss-of-function mutations in CASK cause microcephaly with pontine and cerebellar hypoplasia (MICPCH), a severe neurodevelopmental disorder predominantly affecting females. Although CASK has been implicated in synaptic organization and transcriptional regulation, the mechanisms underlying the cerebellar hypoplasia have remained unsolved. Recent studies using genetically

Magnetically Actuated Iron Oxide Nanoparticles Sensitize Acute T-Lymphocyte Leukemia Cells to Vincristine via Reprogramming Cellular Homeostasis.

Vincristine (VCR) is a first-line chemotherapeutic agent for pediatric T-cell acute lymphoblastic leukemia (T-ALL), while its clinical efficacy is limited by dose-dependent peripheral neurotoxicity. As T cells in normal physiological environments experience various mechanical forces and VCR targets a mechanosensitive cellular microtubule, in this study, we developed a magnetically actuated mechanical stimulation (MAMS) strategy utilizing superparamagnetic Fe3O4 nanoparticles (IONPs) and an external static magnetic field (MF), which combined with VCR to significantly enhance the chemosensitivit

A Rare De Novo Missense Mutation in IFT122 Confers a Genetic Susceptibility Factor of Idiopathic Pediatric Uveitis Via Trio-based Whole-Exome Sequencing.

Idiopathic pediatric uveitis (IPU) is a leading cause of irreversible vision loss in children; however, the genetic and molecular mechanisms underlying this condition remain unclear. Herein, trio-based whole-exome sequencing was performed in 28 affected families and targeted sequencing was performed in 1953 sporadic cases from a Han Chinese cohort. A rare missense mutation, A773E in intraflagellar transport 122 (IFT122), was identified in one trio and absent from sporadic cases. Functional assays showed that deleterious IFT122-A773E increased inflammatory factor secretion and exacerbated barri

Recent advances in TMEM16F: structural plasticity, functional versatility, and implications for human diseases.

TMEM16F is a calcium-activated membrane protein that functions as both a non-selective ion channel and a phospholipid scramblase, linking intracellular Ca²⁺ signaling to dynamic regulation of plasma membrane lipid asymmetry. Recent studies have substantially advanced our understanding of TMEM16F structure and regulation, revealing pronounced conformational heterogeneity and allosteric mechanisms through which Ca²⁺, pH and membrane context coordinate its dual functions. Cell- and tissue-specific analyses indicate that TMEM16F-mediated membrane remodeling contributes to diverse physiological pro

Connexin 26 in Hearing Health and Disease: StructuralFoundations, Mutation Mechanisms, and Therapeutic Perspectives.

Mutations in gap junction protein β-2 (GJB2), encoding Connexin 26 (Cx26), are the most common genetic cause of hearing loss, responsible for up to 50% of inherited non-syndromic cases worldwide. This review covers Cx26 from three perspectives: protein structure, mutant disease mechanisms, and treatment approaches. Structurally, 12 Cx26 subunits assemble into a gap junction channel connecting neighboring cells, enabling exchange of ions and signaling molecules; activity is regulated by calcium, pH, and CO2. In the cochlea, Cx26 channels are required for the development of sound-sensing hair ce

Pathophysiology of Ischemic Cell Death in the Central Nervous System.

The high metabolic demand and complex blood supply of the central nervous system (CNS) render it susceptible to both global and focal ischemic insults, which represent a leading cause of morbidity and mortality in humans. The effects of ischemia in the CNS are determined by duration, severity, and neuroanatomical location of the insult. The pathophysiology of ischemic cell death represents a dynamic process in which necrosis, apoptosis, and other forms of regulated cell death (i.e., necroptosis, pyroptosis, ferroptosis, and autophagic cell death) can be involved. Multiple molecular signaling p
