Biallelic and monoallelic ESR2 variants associated with 46,XY disorders of sex development.

PURPOSE: Disorders or differences of sex development (DSDs) are rare congenital conditions characterized by atypical sex development. Despite advances in genomic technologies, the molecular cause remains unknown in 50% of cases. METHODS: Homozygosity mapping and whole-exome sequencing revealed an ESR2 variant in an individual with syndromic 46,XY DSD. Additional cases with 46,XY DSD underwent whole-exome sequencing and targeted next-generation sequencing of ESR2. Functional characterization of the identified variants included luciferase assays and protein structure analysis. Gonadal ESR2 expr

Mapping of the hepatitis B virus attachment site by use of infection-inhibiting preS1 lipopeptides and tupaia hepatocytes.

BACKGROUND & AIMS: Studies on the early steps in the life cycle of hepatitis B virus have been hampered by the lack of readily available target cells. In this study, we mapped a defined virus attachment site to primary hepatocytes that is essential for infection. METHODS: We used purified virus particles from human carrier plasma as an inoculum and primary cultures of tupaia hepatocytes as susceptible target cells and studied the inhibitory effect of amino-terminally acylated preS1-derived lipopeptides on infection interference. RESULTS: Infectivity of virus could be blocked efficiently in t

Imatinib targets other than bcr/abl and their clinical relevance in myeloid disorders.

Imatinib mesylate is a small molecule drug that in vitro inhibits the Abelson (Abl), Arg (abl-related gene), stem cell factor receptor (Kit), and platelet-derived growth factor receptor A and B (PDGFRA and PDGFRB) tyrosine kinases. The drug has acquired therapeutic relevance because of similar inhibitory activity against certain activating mutations of these molecular targets. The archetypical disease in this regard is chronic myeloid leukemia, where abl is constitutively activated by fusion with the bcr gene (bcr/abl). Similarly, the drug has now been shown to display equally impressive thera

Pathogenicity of the BRCA1 missense variant M1775K is determined by the disruption of the BRCT phosphopeptide-binding pocket: a multi-modal approach.

A number of germ-line mutations in the BRCA1 gene confer susceptibility to breast and ovarian cancer. However, it remains difficult to determine whether many single amino-acid (missense) changes in the BRCA1 protein that are frequently detected in the clinical setting are pathologic or not. Here, we used a combination of functional, crystallographic, biophysical, molecular and evolutionary techniques, and classical genetic segregation analysis to demonstrate that the BRCA1 missense variant M1775K is pathogenic. Functional assays in yeast and mammalian cells showed that the BRCA1 BRCT domains c

Crystal structure of SEDL and its implications for a genetic disease spondyloepiphyseal dysplasia tarda.

SEDL is an evolutionarily highly conserved protein in eukaryotic organisms. Deletions or point mutations in the SEDL gene are responsible for the genetic disease spondyloepiphyseal dysplasia tarda (SEDT), an X-linked skeletal disorder. SEDL has been identified as a component of the transport protein particle (TRAPP), critically involved in endoplasmic reticulum-to-Golgi vesicle transport. Herein, we report the 2.4 A resolution structure of SEDL, which reveals an unexpected similarity to the structures of the N-terminal regulatory domain of two SNAREs, Ykt6p and Sec22b, despite no sequence homo

Malignant-hyperthermia susceptibility is associated with a mutation of the alpha 1-subunit of the human dihydropyridine-sensitive L-type voltage-dependent calcium-channel receptor in skeletal muscle.

Malignant hyperthermia susceptibility (MHS) is characterized by genetic heterogeneity. However, except for the MHS1 locus, which corresponds to the skeletal muscle ryanodine receptor (RYR1) and for which several mutations have been described, no direct molecular evidence for a mutation in another gene has been reported so far. In this study we show that the CACNL1A3 gene encoding the alpha 1-subunit of the human skeletal muscle dihydropyridine-sensitive L-type voltage-dependent calcium channel (VDCC) represents a new MHS locus and is responsible for the disease in a large French family. Comparative genome assemblies reveal distinct allelic and structural variation at blast resistance loci in rice CT21375-F4-4 and Moroberekan.

Rice blast, caused by Magnaporthe oryzae, remains a major constraint to rice (Oryza sativa L.) production, and durable resistance will require broader use of genetic diversity beyond commonly studied elite cultivars. We present chromosome-scale genome assemblies of two agronomically significant but genetically divergent rice cultivars: CT21375-F4-4 (hereafter, CT21) and Moroberekan. Despite their genetic divergence, both cultivars show consistently strong resistance to blast under multiyear field conditions. We combined field phenotyping with genome-wide protein-to-genome alignment, predicted

Elevational melanism in East African Honey Bees involves a Major-Effect locus.

Thermal melanism, whereby darker pigmentation occurs in colder environments, is a widespread pattern, yet its genetic and physiological basis in eusocial insects remains poorly understood. East African honey bees (Apis mellifera) occupy elevational gradients in which highland populations are darker, larger, and more cold tolerant than lowland conspecifics. Here, we integrated population genomics, structural variations, and functional genomics to investigate elevational melanism. We sampled, phenotyped and resequenced 139 workers from five East African mountain systems. A genome wide associatio

Evaluation of genetic parameters affecting the reliability and effectiveness of kinship reconstruction in Forest and Tundra Nenets populations using X-STR markers.

X-chromosomal STR markers (X-STR) are informative tools for kinship reconstruction, particularly in cases where standard autosomal panels have limited discriminatory power: when reconstructing kinship between full sisters, paternal half-sisters, paternal grandmother and granddaughter, maternal aunt and niece. However, the use of X-STR markers in forensic genetics requires consideration of linkage disequilibrium structure and the use of haplotype frequencies in calculating the likelihood ratio (LR). In this work, the genetic structure of Forest and Tundra Nenets populations was assessed using 1

Chromosome-level genome and population resequencing reveal genetic diversity, demographic history, and climate risk in the East Asian relict tree Perkinsiodendron macgregorii.

Relict trees are key to understanding macroevolution, biogeography, and extinction risk under environmental change, yet genome-wide data on diversity and demography remain scarce. Perkinsiodendron macgregorii, a monotypic East Asian relict within Styracaceae, is of notable horticultural and conservation significance. Here, we assembled a chromosome-level genome (∼1.15 Gb) and resequenced 167 individuals from 30 populations across its range. We performed a comparative analysis of single nucleotide polymorphisms (SNPs) and structural variants (SVs) to assess population structure, mutation load,

The genomic architecture of local adaptation in two connected populations of three-spined stickleback.

Populations often adapt to their local environments despite the homogenizing effects of gene flow, but the genomic mechanisms enabling this process remain unclear. Theory predicts that adaptive divergence under high connectivity is favored when beneficial alleles cluster in regions of reduced recombination, a pattern that can be reinforced by structural variants (SVs). We investigated this in three-spined sticklebacks (Gasterosteus aculeatus) from the St. Lawrence Estuary, where distinct freshwater and marine ecotypes meet and interbreed along a short ecological gradient. Using long- and short

Genomic Diversity and Footprints of Selection in Hamdani Sheep.

This study aimed to characterize the genomic diversity, population structure, and candidate regions consistent with selection in Hamdani (HAM) sheep using high-density SNP generated by double-digest restriction site-associated DNA sequencing (ddRADseq). A total of 20 animals were sampled, and after quality filtering, 310,663 SNPs from 18 individuals were retained for downstream analyses. Genetic diversity parameters indicated a relatively high level of genomic variation within the breed (HO = 0.323 ± 0.01 and HE = 0.326 ± 0.01) and low levels of inbreeding (FHOM = 0.029 ± 0.02 and FROH = 0.008

Genomic Structure Predicts Granulovirus Resistance Without a Detectable Life-History Trade-Off in Plodia interpunctella.

Understanding the genetic architecture of pathogen resistance and its relationship to host life-history traits is a fundamental problem in evolutionary biology, yet the conditions under which resistance costs manifest as detectable trade-offs remain poorly understood. Here, we examine resistance in Plodia interpunctella (Indian meal moth) to its granulovirus (PiGV) across 12 inbred lines maintained without deliberate pathogen exposure, alongside an independently assembled outbred stock population as a genomic reference. We quantify resistance using LD50 values from dose-response assays, summar

Genetic origin and forensic analysis of han populations from four districts in Shanghai, China, based on Y-chromosome STR and SNP markers.

Y-short tandem repeats (STRs) and Y-single nucleotide polymorphisms (SNPs) on the Y chromosome are genetic markers unique to men. They are widely used in identification, forensic medicine and paternal lineage tracing. Because of their strict paternal genetic characteristics, they have become a powerful means to trace the evolution of paternal lineages and populations. In this study, 38 Y-STR loci and 250 Y-SNP loci were genotyped, and a dataset was constructed for Qingpu Han (n = 396), Songjiang Han (n = 386), Jinshan Han (n = 384), and Fengxian Han (n = 372) populations in Shanghai. Inhibition of the atypical kinase WNK1 as a therapeutic strategy in TAL-related T-cell acute lymphoblastic leukemia.

Driver mutations in T-cell acute lymphoblastic leukemia (T-ALL) rarely affect druggable kinases. However, these kinases can be aberrantly activated or repressed as secondary oncogenic events. Thus, integrating unbiased phosphoproteomics with genomic approaches may offer novel opportunities for target discovery and therapeutic interventions. In our study, we identified WNK1 (with no lysine [K]) as a potential target in T-ALL by pairing a list of vulnerable kinases with data from a phosphoproteomic screen of T-ALL cell lines. We subsequently validated WNK1 by loss-of-function-based studies and t

Distinct ALK Expression Patterns Are Associated with Canonical and Noncanonical STRN::ALK Transcript Architectures in Oncocytic Thyroid Neoplasms.

Oncocytic thyroid carcinomas are characterized by a unique chromosomal landscape and frequent resistance to radioactive iodine therapy, underscoring the need for improved molecular characterization. Although ALK rearrangements, particularly STRN::ALK fusions, have been described in thyroid carcinomas, their biological and clinical significance in oncocytic thyroid neoplasms remains unclear. This study investigated STRN exon 3-ALK exon 20-derived transcripts in 56 oncocytic thyroid neoplasms using RT-PCR, followed by cloning and Sanger sequencing of positive cases. Transcript-positive cases wer

Identification of the HSP gene family in wild Daurian ground squirrel (Spermophilus dauricus) and analysis of their expression during different hibernation stages.

Hibernation is an extreme form of seasonal heterothermy that enables animals to survive under harsh environmental conditions and provides valuable natural models for investigating the physiological mechanisms underlying stress tolerance. The Daurian ground squirrel (Spermophilus dauricus) is a small burrowing mammal that exhibits pronounced seasonal changes in activity and naturally undergoes hibernation, making it a useful model for elucidating the molecular mechanisms underlying hibernation. Heat shock proteins (HSPs) are highly conserved molecular chaperones involved in protein homeostasis

Genome-wide identification of the cystatin gene family in Japanese flounder (Paralichthys olivaceus) and functional characterization of PoCystatin B and PoCystatin C.

Members of the cystatin superfamily serve as key regulators of cysteine protease activity that play critical roles in immune defense and inflammatory regulation, yet systematic studies on this family in Japanese flounder (Paralichthys olivaceus) remain limited. In this study, five cystatin family members were identified from the P. olivaceus genome, designated as Pocystatin B, Pocystatin C, Pocystatin, Pocystatin F, and Pokininogen 1. Phylogenetic analysis, gene structure characterization, conserved motif identification, and multiple sequence alignment collectively revealed the highly conserve
