Continuous monitoring of pH in the tissue mode: evaluation of a miniature sensor during acidosis and tissue hypoperfusion.

The in vivo performance of a 20G copolymer pH sensor, needlelike in configuration, was studied in the normal dog, and dogs made acidotic by the constant infusion of lactic acid, or by the induction of tissue perfusion defects. Sensors were placed at two extravascular sites in the leg, deep subcutaneous (pHe/sc), and intramuscular in the adductor (pHe/im). This pH sensor is a silver wire capped by a H+-specific polymer; it has a built-in reference system. Its electrochemical characteristics and in vivo performance are similar to those of glass pH electrodes. The continuously monitored values we

Surface properties of lymphocyte subpopulations in autoimmune NZB/NZW F1 hybrid mice: alterations correlated with the immunodeficiency of aging.

Partitioning in a two-polymer aqueous phase system was used to probe the surface properties of lymphoid cell subpopulations in aged male NZB/NZW F1 hybrid (B/W) mice, an important model of autoimmunity, immunodeficiency, and lymphoid malignancy. Spleen cells were fractionated by countercurrent distribution (CCD, a multiple-step extraction procedure) in a charged dextran-polyethylene glycol system. CCD of spleen cells from young, clinically normal male B/W mice yielded several broad distribution patterns which frequently had two or more peaks. Analysis of differentiation antigens and functional

Thiamine transport in thiamine-deficient rats. Role of the unstirred water layer.

As part of a systematic study of alcoholism and thiamine absorption, the effect of diet-induced thiamine deficiency and the role of the unstirred water layer on the thiamine transport were investigated. Using 3H-labeled dextran as a marker of adherent mucosal volume, jejunal uptake of 14C-labeled thiamine hydrochloride was measured, in vitro, in thiamine-deficient rats and pair-fed controls. Uptake of low thiamine concentrations (0.2 and 0.5 muM) was greater in the thiamine-deficient rats than in the controls. In contrast, uptake rates for high thiamine concentrations (20 and 50 muM) were simi

Granulocytic myeloid derived suppressor cells derived extracellular vesicles ameliorate DSS-induced colitis via modulation of Th17/Treg balance.

Colitis is closely associated with immune system dysregulation in the gastrointestinal tract. The present study aimed to evaluate the impact of Granulocytic Myeloid-Derived Suppressor Cells (G-MDSCs)-derived extracellular vesicles (EVs) on disease activity and the immunologic profile in a murine model of colitis. G-MDSCs-EVs were administered to colitis-bearing mice and were evaluated for changes in body weight, disease activity index, colon histopathology, frequencies of Th17 and Tregs, and cytokine release from lymphocytes. The impact of G-MDSCs-EVs was also evaluated on the expression of im

Purified Diets Lacking Fermentable Fiber Reduce Microbial Diversity, Alter Epithelial Transcriptome, and Exacerbate Colitis.

Background/Objectives: Dietary fibers play key roles in shaping gut microbiome and intestinal homeostasis. While purified diets offer experimental precision and reproducibility in rodent models, they omit the complex mixture of fermentable and non-fermentable fibers found in grain-based chow diets. We hypothesized that excluding fermentable fiber impairs intestinal homeostasis by reducing microbial metabolites and altering the colonic epithelial transcriptome, thereby increasing susceptibility to inflammation. Methods: Wildtype male C57BL/6 mice were maintained on either a standard grain-based

Ginsenoside compound K up-regulates β-subunits of long-chain L-3-hydroxyacyl-CoA (HADHB) to promote fatty acid oxidation recovery and improve the balance of macrophage polarization in mice with ulcerative colitis.

BACKGROUND AND PURPOSE: Ulcerative colitis (UC) is characterized by mucosal inflammation and a range of clinical symptoms, including abdominal pain, diarrhoea and bloody stools. Evidence indicates that UC is associated with an imbalance M1/M2 polarization of macrophages. Ginsenoside compound K (GCK) has anti-inflammatory potential the mechanism of action remains unclear. Whether GCK regulates macrophage polarization via β-subunits of long-chain L-3-hydroxyacyl-CoA (HADHB)-mediated fatty acid oxidation (FAO)b was investigated. EXPERIMENTAL APPROACH: The expression of HADHB and macrophage polar

Fibroblast-specific Gpx4 deletion exacerbates IBD via lipid peroxidation.

Broad antioxidant strategies in inflammatory bowel disease (IBD) have had limited success, likely because they indiscriminately quench both harmful and physiological reactive oxygen species. In our recent work, we demonstrated that fibroblast-specific overexpression of acyl-CoA synthetase long-chain family member 4 reprogrammed lipid metabolism and sensitized adjacent epithelial cells to ferroptosis in IBD models, pointing to heterocellular lipid cross talk as a driver of epithelial injury. Building on that insight, here we test the hypothesis that fibroblast glutathione peroxidase 4 (GPX4), a

ETHNOPHARMACOLOGICAL RELEVANCE: Cortex Phellodendri (Huangbai), the dried bark of Phellodendron chinense C.K.Schneid. (Rutaceae), represents a classic botanical drug in Traditional Chinese Medicine (TCM) historically prescribed to "clear heat and dry dampness" for treating gastrointestinal inflammatory conditions such as dysentery. However, its multi-component cooperative mechanisms remain largely enigmatic. AIM OF THE STUDY: This study aimed to elucidate the material basis of Cortex Phellodendri total alkaloids (CPA) and investigate its systemic mechanisms and cooperative interactions in ame

High-Resolution Free-Breathing Chemical-Shift-Encoded MRI for Characterizing Lymph Nodes in the Upper Abdomen.

OBJECTIVES: Accurate lymph node (LN) staging is crucial for managing upper abdominal cancers. Ultrasmall superparamagnetic iron oxide (USPIO)-enhanced magnetic resonance imaging effectively distinguishes healthy and metastatic LNs through fat/water and -weighted imaging. However, respiratory motion artifacts complicate detection of abdominal LNs. This study evaluates if a free-breathing radial stack-of-stars acquisition can match or outperform Cartesian reference scans to visualize LNs and depict uptake of USPIO nanoparticles. MATERIALS AND METHODS: Five volunteers with USPIO and 20 patients
