Outbreeding alleviates senescence in hermaphroditic snails as expected from the mutation-accumulation theory.

Senescence, the decline in fitness components of an organism with age [1], is a nearly universal characteristic of living beings [2-6]. This ubiquity is challenging because natural selection does not favor the evolution of traits decreasing fitness [1, 7, 8]. Senescence may result from two nonexclusive mechanisms: the accumulation of deleterious mutations acting late in life, when the strength of natural selection against them declines [9-11] (mutation accumulation or MA hypothesis [12]) and the delayed cost of genes having beneficial effects early in life (antagonistic pleiotropy or AP hypoth

An isoenzyme survey of Trypanosoma brucei s.l. from the Central African subregion: population structure, taxonomic and epidemiological considerations.

In order to improve our knowledge about the taxonomic status and the population structure of the causative agent of Human African Trypanosomiasis in the Central African subregion, 169 newly isolated stocks, of which 16 came from pigs, and 5 reference stocks, were characterized by multilocus enzyme electrophoresis, for 17 genetic loci. We identified 22 different isoenzyme profiles or zymodemes, many of which showed limited differences between them. These zymodemes were equated to multilocus genotypes. UPGMA dendrograms revealed one main group: Trypanosoma brucei gambiense group I and 3 T. bruce

Estimating quantitative genetic parameters in haplodiploid organisms.

Haplodiploid organisms (diploid females and haploid males) are genetically distinct in that they have asymmetrical genetic segregation, gene dosage compensation in the haploid male, maternal effects, and a general propensity to inbreed. Taking these characteristics into account, we demonstrate how conventional experimental designs and statistics derived to measure quantitative genetic parameters for diploid organisms can be applied to haplodiploid ones. First, the C matrix of the joint-scaling test is modified to test the additive-dominance model and then the calculations for fractions in the

Intraspecific variation in key morphological characters of Culiseta melanura (Diptera:Culicidae).

Culiseta melanura (Coq.), the enzootic vector of eastern equine encephalitis in North America, is polymorphic for a trait used as a key diagnostic character. The absence of white abdominal bands distinguishes this species in several prominent keys to North American mosquitoes. However, this is an environmentally induced, nongenetic trait that cannot be used as a key character for diagnosing Cs. melanura. In light trap collections, banded specimens occur in early spring and summer, and nonbanded adults appear in late summer-autumn. Larvae reared in laboratory conditions produce nonbanded adults

Mutation and linkage disequilibrium analysis in genetic counselling of Spanish cystic fibrosis families.

We have analysed haplotypes for four DNA polymorphisms, closely linked to the cystic fibrosis (CF) gene, in 82 Spanish families, in which the CF probands are either homozygous for non-delta F508 mutations or heterozygous for the delta F508 deletion and other CF mutations. The analysis provides genetic data for a new polymorphism for the closely linked marker pKM.19, which is very strongly associated with CF. Haplotypes generated with the four marker loci are also in strong disequilibrium with the non-delta F508 CF chromosomes. The data reported here are useful in 1 in 4 risk pregnancies of par

Male frequency in Caenorhabditis elegans increases in response to chronic irradiation.

Outcrossing can be advantageous in a changing environment because it promotes the purge of deleterious mutations and increases the genetic diversity within a population, which may improve population persistence and evolutionary potential. Some species may, therefore, switch their reproductive mode from inbreeding to outcrossing when under environmental stress. This switch may have consequences on the demographic dynamics and evolutionary trajectory of populations. For example, it may directly influence the sex ratio of a population. However, much remains to be discovered about the mechanisms a

Deletion of the late cornified envelope LCE3B and LCE3C genes as a susceptibility factor for psoriasis.

Psoriasis is a common inflammatory skin disease with a prevalence of 2-3% in individuals of European ancestry. In a genome-wide search for copy number variants (CNV) using a sample pooling approach, we have identified a deletion comprising LCE3B and LCE3C, members of the late cornified envelope (LCE) gene cluster. The absence of LCE3B and LCE3C (LCE3C_LCE3B-del) is significantly associated (P = 1.38E-08) with risk of psoriasis in 2,831 samples from Spain, The Netherlands, Italy and the United States, and in a family-based study (P = 5.4E-04). LCE3C_LCE3B-del is tagged by rs4112788 (r(2) = 0.93

Antenatal gene tests in low-risk pregnancies: molecular screening for aspartylglucosaminuria (AGU) and infantile neuronal ceroid lipofuscinosis (INCL) in Finland.

Approximately one in five subjects in Finland carries some gene defect associated with 30 diseases belonging to the Finnish disease heritage, and about one in 500 children born is affected. Almost all carriers, women and men, are unaware of their condition. Recent advances in molecular medicine have offered the possibility of population-based carrier screening for recessive disorders. We studied acceptance and attitudes to antenatal screening for aspartylglucosaminuria (AGU) and infantile neuronal ceroid lipofuscinosis (INCL). From January 1995 until December 1996 carrier tests were offered at

Cystic fibrosis screening.

The integration of universal cystic fibrosis screening in women's health has considerably altered the way we care for obstetric patients and likely will be the foundation for incorporating other genetic tests into routine women's health care. Prior to this change in the standard of care, screening for genetic disease was primarily limited to those individuals who had a personal or family history of the genetic condition or who belonged to a particular ethnic or racial group characterized by a high frequency of carrier and affected individuals. However, technological advances have resulted in f

Genetic Variation and Demographic History of Green Weevil Hypomeces pulviger (Herbst, 1795) (Coleoptera: Curculionidae) in Thailand Examined by Mitochondrial DNA Sequences.

The population genetic diversity and demographic history of Hypomeces pulviger in Thailand were examined based on mitochondrial cytochrome c oxidase subunit 1 (CO1) and 16S ribosomal DNA (16S rDNA) sequence data. A total of 171 and 104 individuals from multiple populations were analyzed using CO1 and 16S rDNA sequences, respectively. The CO1 sequence dataset revealed high haplotype diversity (Hd = 0.999) and moderate nucleotide diversity (Nd = 0.0323), whereas the 16S rDNA showed lower diversity (Hd = 0.750, Nd = 0.0026). Population structure analyses showed low to moderate differentiation in

Population-scale genomic medicine with the Hong Kong Genome Project.

The Hong Kong Genome Project (HKGP) aims to build a foundational resource for precision medicine in the Chinese population through large-scale genome sequencing and integrated analyses. Here we report findings from over 20,000 HKGP participants across two cohorts: a rare disease cohort including 2,227 patients with suspected genetic diseases and a population cohort including 18,261 participants undergoing genomic screening for medically actionable findings. The rare disease cohort achieved a diagnostic rate of 25%. When benchmarked against panels designed for European ancestries, the analysis

Profiles of global mutations in the human intercellular adhesion molecule-1 (ICAM-1) shed light on population-specific malaria susceptibility.

Plasmodium falciparum is responsible for malaria-related morbidity and mortality. PfEMP1 (P. falciparum erythrocyte membrane protein 1) mediates infected erythrocytes adhesion to various surface vascular receptors, including intercellular adhesion molecule-1 (ICAM-1), associating this interaction with severe malaria in several studies. Genetic variation in host ICAM-1 plays a significant role in determining susceptibility to malaria infection via clinical phenotypes such as the ICAM-1Kilifi variant which has been reported to be associated with susceptibility in populations. Our genomic and str

Usefulness of genetic characterization of narcolepsy and hypersomnia on phenotype definition: a study in Portuguese patients.

INTRODUCTION: The determination of human leukocyte antigen (HLA) class II genotype is widely used to confirm the diagnosis of narcolepsy with or without cataplexy. The HLA genotyping is reliable, easy to perform and reassures the clinician. It is also less invasive than other methodologies and is in accordance with the autoimmune hypothesis for the origin of narcolepsy. AIM. To assess the usefulness of genetic markers (HLA) in the differential diagnosis between different sleep disorders and their relevance in the context of our population. [Refinement and role of the diagnosis of Gilbert disease with molecular biology].

Gilbert syndrome (GS), characterized by mild, chronic and isolated unconjugated hyperbilirubinemia is due to a partial deficiency of bilirubin-UDP-glucuronosyltransferase (UGT1A1). Recently, the genetic basis of GS has been identified in caucasian populations : it is related to the insertion of a dinucleotide (TA) in the promoter region of the UGT1A1 gene. In Asian populations, GS is due to missense mutations (either homozygous or heterozygous) in the coding sequence. The aim of this study was to develop a simple and rapid method to detect both genetic polymorphisms and mutations. This techniq

Prenatal Diagnosis and Screening of the Haemoglobinopathies.

The most important aspects of carrier detection procedures, genetic counselling, population screening and fetal diagnosis of the thalassaemias and sickle cell anaemia are reviewed. Carrier detection can be made retrospectively, i.e. following the birth of an affected child, or prospectively. Most carrier detection and genetic counselling in population at risk for alpha-thalassaemia an sickle cell anaemia is retrospective. However, some prospective carrier screening programmes for sickle cel anaemia are ongoing in Cuba and Guadeloupe and very limited screening for alpha-thalassaemia is in progr

A genetic register for Huntington's chorea in South Wales.

A regional genetic register for Huntington's chorea in South Wales is described, based on previous family studies in this area, which is one of high prevalence for the disorder. The primary role of the register is to help in the efficient delivery of services, including genetic counselling, to affected subjects and relatives, and to monitor changes in the population at risk. The mode of operation of the register is described and the essential importance of strict confidentiality is stressed.

Genotypic and phenotypic variability of 22q11.2 microduplications: An institutional experience.

Duplications in the 22q11.2 region can cause 22q11.2 duplication syndrome and encompass a variety of phenotypes including developmental delays, facial abnormalities, cardiovascular defects, central nervous system delays, and other congenital abnormalities. However, the contribution of these contiguous duplicated regions to the clinical phenotypes has not been fully elucidated. In this study, we identified nine patients carrying different 22q11.2 microduplications detected by chromosomal microarray. Of these patients, seven pediatric patients presented with various clinical features including t

Identification of novel loci associated with maturity and yield traits in early maturity soybean plant introduction lines.

BACKGROUND: To continue to meet the increasing demands of soybean worldwide, it is crucial to identify key genes regulating flowering and maturity to expand the cultivated regions into short season areas. Although four soybean genes have been successfully utilized in early maturity breeding programs, new genes governing maturity are continuously being identified suggesting that there remains as yet undiscovered loci governing agronomic traits of interest. The objective of this study was to identify novel loci and genes involved in a diverse set of early soybean maturity using genome-wide assoc

Variants in autophagy-related genes and clinical characteristics in melanoma: a population-based study.

Autophagy has been linked with melanoma risk and survival, but no polymorphisms in autophagy-related (ATG) genes have been investigated in relation to melanoma progression. We examined five single-nucleotide polymorphisms (SNPs) in three ATG genes (ATG5; ATG10; and ATG16L) with known or suspected impact on autophagic flux in an international population-based case-control study of melanoma. DNA from 911 melanoma patients was genotyped. An association was identified between (GG) (rs2241880) and earlier stage at diagnosis (OR 0.47; 95% Confidence Intervals (CI) = 0.27-0.81, P = 0.02) and a decrea

History of settlement of villages from Central Tunisia by studying families sharing a common founder Glycogenosis type III mutation.

Glycogen storage disease type III (GSD III; Cori disease; Forbes disease) is an autosomal recessive inherited metabolic disorder resulting from deficient glycogen debrancher enzyme activity in liver and muscle. In this study, we focused on a single AGL gene mutation p.W1327X in 16 Tunisian patients from rural area surrounding the region of Mahdia in Central Tunisia. This constitutes the largest pool of patients with this mutation ever described. This study was performed to trace the history of the patients' ancestries in a single region. After extraction of genomic DNA, exon 31 of AGL gene was
