Thiacloprid (THI), a cyano-substitued neonicotinoid, is widely used in agriculture and has been identified as a critical factor responsible for the global decline of non-target insects due to its persistent environmental residues. Here, we investigated the developmental and reproductive toxicity of THI in Drosophila melanogaster using sublethal concentrations (0.5, 5, 10 mg/L) referenced to previously reported sublethal THI levels in laboratory. The results showed that THI exposure significantly reduced larval body length by 22.6 % and pupation rate by 30.4 %. LC-MS analysis revealed a pronoun

Traditionally, testosterone and estrogen have been considered to be male and female sex hormones, respectively. However, estradiol, the pred … trogen, starting with the hypothalamus-pituitary-gonadal axis, followed by the Leydig, Sertoli, and germ cells, and finishing with the ductal epithelium, epididymis, and mature sperm. The third molar teeth (wisdom teeth) represent the last eruption of the teeth in the human dentition. Throughout evolution, the mandible has had a tendency to decrease in size; the third molar teeth are often impacted, resulting in incomplete tooth eruption that often causes clinical pericoronitis, dental caries, and pericemental abscess. Therefore, the wisdom teeth are often extracted. Moreover, wisdom teeth are often removed for clinical orthodontic treatment. On the other hand, tooth loss due to periodontal disease, dental caries, trauma, or a variety of genetic disorders continues to affec

Teratomas are fully formed germ cell tumors derived from multiple germ layers. While most commonly gonadal, they can occur in extragonadal sites, particularly the mediastinum. Isolated intrapulmonary teratomas without mediastinal extension are exceptionally rare, with fewer than 100 cases reported in the literature, and can be mistaken for other calcified lung lesions such as hamartomas. We report the case of a 57-year-old former smoker who presented with a three-month history of chest pain and dry cough. Chest X-ray and computed tomography (CT) scan revealed a large, well-circumscribed left l

Besides providing useful model systems for basic science, studies based on modification of the mammalian germ line are changing our understanding of pathogenetic principles. In this article, we review the most popular techniques for generating specific germ line mutations in vivo and discuss the impact of various transgenic models on the study of neurodegenerative diseases. The "gain of function" approach, i.e., ectopic expression of exogenous genes in neural structures, has deepened our understanding of neurodegeneration resulting from infection with papova viruses, picorna viruses, and human

INTRODUCTION: Mature teratomas are germ cell tumors composed of tissues derived from all three germ layers. These tumors are rare in the central nervous system, primarily occurring in the suprasellar and pineal regions. Rupture of intracranial teratomas is an exceptionally rare phenomenon, typically presenting on imaging as disseminated fatty droplets and occasionally associated with aseptic meningitis. We describe a case of a ruptured cystic teratoma in the pineal region, manifesting postoperatively with severe neurological symptoms consistent with aseptic meningitis.

Myelodysplastic syndrome (MDS) and myeloproliferative disorders are rare in children; they are divided into low-grade MDS (refractory cytopenia of childhood [RCC]), advanced MDS (refractory anemia with excess blasts in transformation), and juvenile myelomonocytic leukemia (JMML), each with different characteristics and management strategies. Underlying genetic predisposition is recognized in an increasing number of patients. Germ line GATA2 mutation is found in 70% of adolescents with MDS and monosomy 7. It is challenging to distinguish RCC from aplastic anemia, inherited bone marrow failure,

By definition, myeloproliferative disorders (MPDs) are caused by an acquired somatic mutation of a hematopoietic progenitor/stem cell and have sporadic occurrence. However, well-documented families exist with first-degree relatives acquiring one or several MPDs. It is reasonable to assume that the germ-line mutation(s) or genetic background must facilitate or predispose for one or several somatic mutation(s) that lead to the MPD that is indistinguishable from the sporadic form. This is best documented in familial polycythemia vera (PV), which appears to be inherited as an autosomal dominant di

Germline mutations in genes involved in perforin-granzyme-mediated cytotoxicity such as PRF1, UNC13D, STX11, and STXBP2 were known to cause familial hemophagocytic lymphohistiocytosis (FHL). In this study, we reported a unique group of 3 patients with germline mutations of UNC13D and STX11 genes and presented as adult-onset peripheral T-cell lymphoma (PTCL) with cytotoxic T-cell phenotype and atypical lymphoma presentations. CD107a degranulation assay and NK-cell activity analysis demonstrated impaired cytotoxic function of the NK/T-cells of the patients with FHL-related mutations. Gene expres

Numerical chromosomal aberrations are highly frequent in cancer cells. However, tumor-associated mutations in regulators of the mitotic machinery that controls chromosome segregation are rather rare. By sequencing families with hereditary cancer, Chen and colleagues report two novel heterozygous mutations in CDC20, a coactivator of the anaphase-promoting complex (APC/C) and a target of the spindle assembly checkpoint (SAC) that prevents chromosome missegregation during mitosis. CDC20 mutations result in partial SAC functionality and segregate with tumor susceptibility in families with aneuploi

BACKGROUND: The Hippo signaling pathway plays fundamental roles in stem cell maintenance in a variety of tissues and has thus implications for stem cell biology. Key components of this recently discovered pathway have been shown to be associated with primordial follicle activation. However, whether the Hippo signaling pathway plays a role in the development of Ovarian Germline Stem Cells (OGSCs) during physiological and pathological ovarian aging in mice is unknown.
METHODS: Mice at the age of 7 days (7D), or of 2, 10, or 20 months (2M, 10M, 20M) and mice at 2M treated with TPT and CY/BUS dru

This report describes the establishment of a cell line of a human uterine mixed mesodermal tumor. The tumor of origin derived from a hysterectomy specimen, has been maintained for 14 months in vitro and continues to grow as an established cell line. The original tumor as well as the cell line exhibited no estrogen receptors. alpha-Fetoprotein was not detected in the cultured cells or in the spent culture medium. Karyotyping revealed 46 XX chromosome complement with a balanced 11-16 translocation. This is the first documentation of such a chromosome abnormality in a genital tract carcinoma. Ste

Hyaluronan-binding protein 1 (HABP1), alternatively designated as p32, C1QBP, gC1qR, or HIV TAP (Tat-associated protein), is a highly conserved eukaryotic protein expressed across taxa from yeast to mammals, and is localized on the cell surface of diverse cell types. This protein exists in two distinct functional forms: firstly, a pro-protein exclusively found in male germ cells, suggesting its crucial role in spermatogenic differentiation; and secondly, a mature, post-translationally cleaved form localized on the sperm surface. Due to its specific affinity for hyaluronan (HA) a key constituen…

Mature cystic teratoma(MCT)2 is the most prevalent neoplasm derived from germ cells in females; its progression to malignancy is infrequent. The coexistence of papillary thyroid carcinoma(PTC)3 and strumal carcinoid derived from MCT is exceptionally rare. To the best of our knowledge, only six cases of this condition have been reported to date, however, the molecular information and immune cell infiltration for the mixture has never been studied. We present a case of a patient exhibiting a combination of these two components in the MCT. The clinical and pathological characteristics of the pati…

Mature cystic teratoma is the most common type of ovarian germ cell neoplasm, but occasionally, it can undergo malignant transformations, especially in postmenopausal women. These secondary malignant neoplasms are most commonly squamous cell carcinomas. The absence of clinical and radiological specificity of this transformation means that the diagnosis remains purely histological. Data is insufficient regarding the appropriate management given their rarity. However, the treatment is multidisciplinary and is based on surgery and a platinum-based chemotherapy regimen. We report the case of a 53-

Almost 50% of infertility cases are due to male factors, and spermatogenesis failure is one of the most severe forms of male infertility. Sertoli cell-only syndrome (SCOS) also known as germ cell aplasia is characterized by azoospermia in which the seminiferous tubules of testicular biopsy are lined only with Sertoli cells. The definitive diagnosis of SCOS is by diagnostic testicular biopsy. Although SCOS may be a result of Klinefelter syndrome, most of the SCOS men have a normal karyotype. Along with genetic aberrations, signaling pathways and endocrine processes might be major factors in the

Noonan syndrome (NS) is an autosomal dominant disorder caused by mutations in PTPN11, KRAS, SOS1, and RAF1. We performed SOS1, RAF1, BRAF, MEK1, and MEK2 mutation analysis in a cohort of 102 PTPN11- and KRAS-negative NS patients and found pathogenic SOS1 mutations in 10, RAF1 mutations in 4, and BRAF mutations in 2 patients. Three novel SOS1 mutations were found. One was classified as a rare benign variant and the other remains unclassified. We confirm a high prevalence of pulmonic stenosis and ectodermal abnormalities in SOS1-positive patients. Three patients with SOS1 mutations presented wit
