Evaluation of polymorphic splicing in the mechanism of the association of the insulin gene with diabetes.

The association of type 1 diabetes with the insulin gene (IDDM2 locus) has been mapped to a short haplotype encompassing two single nucleotide polymorphisms (SNPs) in perfect linkage disequilibrium (r(2) = 1) with each other and with the two allele classes at the variable number of tandem repeats (VNTR) polymorphism upstream of the transcription site. Although it is believed that the genetic effect is mediated through transcriptional effects of the VNTR, an alternative mechanism has been recently proposed: In transfected cells, the common A allele at one of the SNPs (-23A-->T, in relation to t

Association study of tryptophan hydroxylase 2 gene polymorphisms in panic disorder.

Experimental studies on serotonin (5-HT) availability suggest a role for 5-HT synthesis rate in panicogenesis. Recently, it has been discovered that the tryptophan hydroxylase gene isoform 2 (TPH2), rather than TPH1, is preferentially expressed in the neuronal tissue and, therefore, is primarily responsible for the regulation of brain 5-HT synthesis. In the present case-control genetic association study we investigated whether panic disorder (PD) phenotypes are related to two single nucleotide polymorphisms (SNP) of TPH2, rs1386494 A/G and rs1386483 C/T. The study sample consisted of 213 (163

Normal ICOS, ICOSL and AID alleles in Danish patients with common variable immunodeficiency.

Common variable immunodeficiency, CVID, is a primary antibody deficiency characterized by decreased levels of serum immunoglobulin G (IgG), decreased IgA and/or IgM and recurrent infections. It is assumed to be heterogeneous group of disorders caused by different genetic defects. Some patients have decreased levels of class switched memory B cells and/or decreased levels of somatic hypermutation which points to defects in the germinal centre (GC) reactions as cause of the disease in these patients. The inducible costimulator, ICOS, and its' ligand, ICOSL, are both involved in and necessary for

Linkage disequilibrium analysis identifies an FGFR1 haplotype-tag SNP associated with normal variation in craniofacial shape.

Mutations in FGFR1 and TWIST1 have been reported to affect the timing of calvarial suture fusion resulting in craniosynostosis and facial abnormalities. We screened nonpathologic populations for genetic polymorphisms that may associate with normal craniofacial variation. We identified 17 single-nucleotide polymorphisms (SNPs) in FGFR1, 6 of which were novel (g.8591855G-->A, g.8593685G-->A, g.8602303C-->T, g.8602475A-->G (p.Ile293Val), g.8605849C-->T, g.8607868G-->A). No SNPs were found in TWIST1. FGFR1 SNP haplotypes were reconstructed for Caucasian, Asian, Australian Aboriginal, and African A

The organic anion transporter family: from physiology to ontogeny and the clinic.

The organic anion transporter (OAT) family handles a wide variety of clinically important compounds (antibiotics, nonsteriodal anti-inflammatory drugs, etc.) and toxins. However, little is known about their appearance during development despite documented differences in the handling of anionic drugs among neonates, children, and adults. A similar spatiotemporal pattern of mRNA expression of the OATs (OAT1-4) during kidney development suggests that OAT genes may be useful in understanding the mechanisms of proximal tubule maturation. Moreover, OAT expression in unexpected extrarenal sites (e.g.

Tracing the recent evolution of Group A Streptococcus M1 serotype.

The twenty-first century has seen global surges in scarlet fever and invasive Group A Streptococcus (GAS) infections, partly driven by the emergence of the toxigenic M1UK lineage. Characterized by increased SpeA superantigen expression and the stepwise accumulation of 27 single nucleotide polymorphisms (SNPs), M1UK has become the dominant GAS emm1 lineage in Europe, Australia and Canada, representing a notable shift in GAS molecular epidemiology. Interestingly, other distinct emm1 variants have emerged in China and Denmark but are yet to expand globally in the same manner. This review examines

5-HT2AR and BDNF gene variants in eating disorders susceptibility.

Evidence from family and twin studies points to a genetic contribution to the etiology of eating disorders (EDs), confirmed by the association of several single nucleotide polymorphisms (SNPs) with this group of disorders. Previous reports have suggested that the serotonin receptor (5-HT2AR) and brain-derived neurotrophic factor (BDNF) genes could be both involved in EDs susceptibility. In order to provide further evidence about such association, we focused our attention on two SNPs located in these genes carrying out a genetic association study on a large Italian cohort composed of 556 ED pat

Minor allele frequency of myeloproliferative neoplasm mutations in the Irish blood donor population.

Myeloproliferative neoplasms (MPNs) are rare diseases that include classic entities; polycythaemia vera, essential thrombocythaemia and primary myelofibrosis. In this short report, minor allele frequencies of common MPN mutations are compared between the Irish blood donor population and other populations of European descent using data from the Haplotype Map project. The Affymetrix array 6.0 platform was utilised identifying nine single nucleotide polymorphisms (SNPs) and six proxy SNPs. The variability of allele frequencies for MPN mutations could account for the different incidence rates seen

An overview of genetics of paediatric rheumatic diseases.

The evidence so far suggests that the paediatric inflammatory diseases encountered in rheumatology practice may be largely genetic in origin, where common single nucleotide polymorphisms (SNPs) in multiple genes contribute to risk, with real but variable environmental components. As far as genetic susceptibility to common paediatric rheumatic diseases is concerned, only juvenile idiopathic arthritis (JIA) has been investigated in any substantial way so far. This article discusses susceptibility for different types of JIA, the different methods used and their advantages and disadvantages. The g

Absence of TGFBR1 and TGFBR2 mutations in patients with bicuspid aortic valve and aortic dilation.

Mutations in the genes encoding transforming growth factor-beta receptor types I and II (TGFBR1 and TGFBR2, respectively) are commonly identified in patients with Loeys-Dietz syndrome, as well as some patients with Marfan's syndrome or familial thoracic aortic aneurysms and dissections. This suggests that there is considerable phenotypic heterogeneity associated with mutations in these genes. Because bicuspid aortic valve (BAV) is a congenital heart defect in patients with Loeys-Dietz syndrome, this study was conducted to investigate whether variants in TGFBR1 or TGFBR2 are responsible for spo

Integrated gene expression and mutation profiling of chemoresistance in canine mammary carcinoma cells.

Mammary gland tumors (MGTs) are common in female dogs, and are usually not treated with chemotherapy due to the lack of evidence of benefit for the patients. In clinical practice, when a female dog receives neoadjuvant or adjuvant chemotherapy, minimal improvement is achieved. Therefore, chemotherapy is not usually applied for these patients. We hypothesized that some canine MGTs do not respond to chemotherapy due to intrinsic chemoresistance. Chemoresistance can be a challenge and is often associated with overexpression of ABC transporters, such as MDR1, MRP1, MRP2, MRP3 and BCRP, which reduc

Interleukin23 Receptor Genetic Variants Associate With Crohn's Disease Risk and Microbiome Changes in Healthy First-Degree Relatives.

BACKGROUND & AIMS: Single nucleotide polymorphisms in the interleukin23 receptor gene are associated with Crohn's disease, suggesting a role in pathogenesis, and several biologic agents targeting this pathway are now established therapies. Interleukin23 has been suggested to be involved in regulation of intestinal barrier function and may impact gut microbial composition. We investigated whether interleukin23 receptor genetic variants predict Crohn's disease risk and influence gut barrier function and microbiome composition in healthy first-degree relatives. METHODS: A total of 3055 healthy f

Genetic insights into aging: Obesity's impact on frailty risk uncovered.

Frailty is an age-associated condition characterized by a state of vulnerability that compromises the quality of life and independence of older adults. To date, no studies have employed Mendelian randomization (MR) to investigate the causal relationship between obesity and frailty risk. Therefore, this study utilized a two-sample MR approach to elucidate the potential causal link between obesity and frailty. A two-sample MR analysis was conducted to assess the causal relationship between body mass index (BMI) or waist circumference and frailty. Independent genetic variants associated with obes

Swine wastewater drives dissemination of high-risk bla NDM-positive Escherichia coli clones.

INTRODUCTION: Swine wastewater can accelerate the spread of antibiotic-resistant bacteria among animals, the environment, and humans. Therefore, the molecular epidemiological characteristics of carbapenemase-producing Escherichia coli isolates from swine wastewater was investigated. METHODS: Antimicrobial susceptibility testing was used to determine metallo-enzyme activity. Conjugation experiments were used to determine transmissibility of the novel carbapenemase-encoding gene. Whole-genome sequencing (WGS) with short reads and ERIC-PCR genotyping were performed to characterize the genomic fe

The causal relationship between Omega-3 fatty acids and Achilles Tendinitis risk: A two-sample Mendelian randomization study in European populations.

Omega-3 fatty acids, found in fish oil, flaxseed oil, and walnuts, are widely known for their anti-inflammatory properties and are used in the treatment of various inflammatory diseases. While their benefits in cardiovascular diseases and arthritis are well-documented, their role in musculoskeletal conditions, particularly Achilles tendinitis, remains unclear. This study investigates the causal relationship between Omega-3 fatty acids and the risk of Achilles tendinitis using a two-sample Mendelian randomization approach. Genetic variants associated with Omega-3 levels were selected from publi

Autophagy functions in lung macrophages and dendritic cells to regulate allergen-dependent inflammatory responses.

Asthma affects 260 million people worldwide, with severe asthma cases that are associated with TH17 and TH1 responses and neutrophil-dominated inflammation being the most difficult to treat due to corticosteroid insensitivity. Single nucleotide polymorphisms in the ATG5 gene, which codes for a protein required for the cellular recycling process of macroautophagy/autophagy, are associated with higher risk for developing severe asthma. We explored the mechanistic basis for the genetic association of ATG5 variants with severe asthma. We identified an autophagy-dependent role for ATG5 in lung macr

Antimicrobial use on Campylobacter revealed by next-generation sequencing in patients with common variable immunodeficiency.

Because few studies have focused on recurrent Campylobacter bacteremia, we investigated two clinical cases of patients with common variable immunodeficiency and repeated Campylobacter bacteremia over a period of 6-10 years. We analyzed and compared genomes from isolates obtained from both patients during follow-up. For patient #1, 18 isolates of Campylobacter coli and 17 isolates of Campylobacter jejuni were obtained from 2014 to 2024. For patient #2, 10 isolates of C. coli were obtained from 2019 to 2024. Next-generation sequencing was used to identify species, characterize antimicrobial resi

Study on the causal relationship between educational attainment and delirium: A two-sample Mendelian randomization study.

This study aimed to investigate whether there is a causal relationship between educational attainment and delirium at the genetic level using the Mendelian randomization method, and provide new evidence for studies in this field. We found a causal relationship between educational attainment and delirium at the genetic level after excluding confounders using Mendelian randomization. The inverse variance weighting method of random effects was the main analysis method. The weighted median and Mendelian Randomization-Egger methods, as well as simple, and weighted modes were used as supplementary a
