Nanomaterial-Mediated Targeting of Mitochondrial Metabolism: Strategies and Applications in Cancer Therapy.

Mitochondria serve as cellular powerhouses and function as central hubs for oxidative metabolism and signaling regulation. These organelles produce ATP primarily through oxidative phosphorylation (OXPHOS), thereby fueling cellular growth and function. In cancer, metabolic reprogramming drives malignant progression, with mitochondria playing a pivotal role. To meet heightened energy and biosynthetic demands, cancer cells modulate mitochondrial OXPHOS activity while enhancing fatty acid oxidation and amino acid metabolism, thereby maintaining redox balance and supporting survival and proliferati

Butyrate epigenetically licenses sustained epithelial-T cell crosstalk for intestinal immune tolerance.

Intestinal epithelial cells (IEC) are the primary cell type in direct contact with stimuli from the luminal microbiota, playing a critical role in host-microbe interactions. However, how IECs communicate with underlying immune cells to maintain homeostasis remains poorly understood. In addition, the mechanisms by which IECs sense microbiota-derived stimuli to initiate this crosstalk are not yet fully understood. Here, we demonstrate that oral administration of the gut microbiota metabolite butyrate induces sustained IL-10 production in CD4⁺ T cells, conferring long-lasting protection against i

Research Progress on Arbuscular Mycorrhizal Fungi-mediated Selenium Uptake, Transport, and Metabolism in Plants.

Selenium (Se) is an essential micronutrient for human health, playing a crucial role in antioxidant defense and immune regulation. However, the uneven global distribution of bioavailable Se in soils has led to widespread dietary Se deficiency, posing a potential public health risk. Although plants are the primary dietary source of Se, their intrinsic capacity for Se enrichment is limited. This necessitates the exploration of microbial-assisted strategies for enhancing Se biofortification. Arbuscular mycorrhizal fungi (AMF) are keystone symbionts in the plant-soil continuum and are known to sig

Patterns and drivers of sedimentary carbon in global aquatic ecosystems during the Anthropocene.

Aquatic ecosystems serve as critical carbon sinks, with sediments playing a key role in long-term organic carbon sequestration. However, the global patterns and trajectories of sedimentary total organic carbon concentration (TOC) and organic carbon burial rates (OC-BR) during the Anthropocene remain poorly constrained. Here, we present a comprehensive global synthesis of sediment core records from lakes, reservoirs, inland wetlands, and coastal systems, integrating over 18,000 rows data from 585 sites across diverse climatic and geographic settings. Generally, our results show that both TOC an

Interplay Between Poly(ADP-ribosyl)ation and Specific Inner Cellular Events That Suggest Combination Strategies for Overcoming PARP Inhibitor Resistance.

Therapeutic resistance remains a major obstacle to durable cancer control, with functional reprogramming of the DNA damage response (DDR) network playing a central role. The poly(ADP-ribose) polymerase (PARP) family, particularly PARP1 and PARP2, is crucial for maintaining genomic integrity. By exploiting synthetic lethality, PARP inhibitors (PARPi) selectively target tumors with homologous recombination deficiency (HRD) and are integral to precision therapy in ovarian, breast, and prostate cancers. However, over 40% of patients with BRCA1/2 alterations develop resistance, and patient eligibil

Increasing rail transit ridership by improving passenger ride comfort.

In urban rail transit systems, passenger ride comfort (PRC) significantly influences ridership levels. Various factors affect PRC, with track geometry playing a crucial role. Among geometry parameters, the minimum required tangent length between consecutive railway curves is particularly significant. While this parameter affects passenger comfort considerably, it has received less attention in current design practices. This study addresses this gap by optimizing tangent length requirements to improve passenger comfort. To this end, a vehicle-track interaction model was established and subseque

Microfluidic Gd3+-chelated hydrogel microspheres enable MRI-visible, image-guided photothermal chemotherapy for pancreatic cancer.

Innovative biomaterials are playing an increasingly important role in biomedicine as drug-delivery carriers, tissue-repair scaffolds, and beyond. However, effective approaches for in situ, longitudinal monitoring of lesion initiation and progression in vivo during biomaterial-based interventions remain limited. Magnetic resonance imaging (MRI) offers high spatial resolution and excellent soft-tissue contrast. Here, gadolinium ions (Gd3⁺) were incorporated into a double-network gelatin methacryloyl (GelMA) hydrogel microsphere platform for photothermal-chemotherapy. Monodisperse microspheres we

Biosynthesis of Glycine from One-Carbon Resources Using an Engineered Escherichia coli Whole-Cell Catalyst.

Carbon dioxide (CO2) is a cost-effective, abundant, and renewable carbon source, but its utilization technologies face several issues. The reductive glycine pathway (RGP) is recognized as one of the most efficient one-carbon (C1) assimilation routes in nature, with its core component-the glycine cleavage system (GCS: GcvP, GcvH, GcvT, and GcvL)-playing an essential role in C1 metabolism. To develop efficient CO2 conversion and utilization pathways, we identified NhFtfL and AmFchA-MtdA with high catalytic efficiency through gene mining and constructed a four-plasmid co-expression system in E. c

Conserved mRNP remodeling mechanism of the TREX-2.1 complex.

Processing, packaging, and nuclear export of messenger ribonucleoprotein particles (mRNPs) are critical for eukaryotic gene expression, with the DEAD-box ATPase DDX39B (yeast Sub2) playing a central role in mRNP processing and remodeling. Our recent studies identified human TREX-2 (GANP•PCID2•DSS1), yeast TREX-2 (Sac3•Thp1•Sem1), and a related human TREX-2.1 complex (LENG8•PCID2•DSS1) as key regulators of DDX39B/Sub2. Here, we characterize the yeast TREX-2.1 (scTREX-2.1) complex, composed of Thp3, Csn12, and Sem1. We show that the scTREX-2.1 complex directly interacts with Sub2 and co-occupies

Large and low-PEG lipid nanoparticles enable efficient dendritic cell targeting for potent mRNA Cancer vaccines.

Efficient delivery of messenger RNA (mRNA) to dendritic cells (DCs) remains a major challenge limiting the efficacy of mRNA cancer vaccines. Here, we report a large-sized lipid nanoparticle (LLNP) platform specifically engineered for enhanced DC targeting. By reducing PEG-lipid content to 0.3% and proportionally increasing the concentrations of structural lipids and mRNA sixfold relative to the Moderna classic formulation, we generated LLNPs with enlarged size and optimized surface properties that favor DC uptake. Following intravenous administration, LLNPs achieved markedly enhanced mRNA expr

Plant Metabolites Screening Identifies 7-epi-10-Deacetyltaxol Inhibiting Cell Division by Interfering With the Cell Cycle and Microtubule Organization.

Plants produce a considerable number of metabolites, and many of them appear to interfere with certain cellular processes such as cell division. For example, taxanes produced by Taxus are known as anti-cancer drugs that stabilize microtubules and prevent the cell from dividing normally. Disorder of cell division also leads to abnormal growth and development in plants. It is worthy of consideration whether these metabolites have negative effects on plants per se. In this study, we developed an efficient screening system for identifying plant metabolites affecting rapeseed seedling root growth.

Macrophage metabolic reprogramming via HIF-1α-glycolysis drives osteoblast ferroptosis and bone loss through an IL-6-STAT3-dependent redox axis.

BACKGROUND: Postmenopausal osteoporosis (PMOP) is characterized by exacerbated bone resorption and inadequate bone formation, with macrophage-driven inflammation playing a key role. However, how immunometabolic reprogramming of macrophages modulates osteoblast fate remains unknown. METHODS: Using integrated single-cell and bulk transcriptomics, we identified a hypermetabolic macrophage subpopulation in PMOP marrow reliant on HIF-1α-glycolysis. We pharmacologically disrupted this axis with the HDAC inhibitor valproic acid (VPA) and validated its function using the HIF-1α stabilizer DMOG. The p
