Utilizing human-induced pluripotent stem-cell-derived vascular smooth muscle cells (hiPSC-VSMCs) to address seed cell source in tissue-engineered vascular grafts necessitates promoting cellular maturation. Morphological changes and functional remodeling of mitochondria are crucial to cellular differentiation and maturation. However, our current understanding of these changes during the transition from hiPSCs to VSMCs is limited. Our findings revealed that after hiPSC-VSMC differentiation, mitochondria transitioned from perinuclear to cytoplasmic distribution, increased in number, and changed s

The immunogenicity of DNA vaccines can be improved by designing plasmids that encode secreted vaccine fusion proteins that (i) target antigen-presenting cells and (ii) display bivalent antigen. We extended this principle to a combinatorial and simultaneous delivery of 2 influenza virus antigens, hemagglutinin and neuraminidase. Using distinct dimerization motifs prevented mixing of the two antigens in the endoplasmic reticulum of transfected cells, maintaining antigen-presenting cell targeting and bivalency of each. Such a combinatorial DNA vaccine induced B and T cell responses against hemagg

Tumor intracellular bacteria are a key driving force of epithelial-mesenchymal transition (EMT) in tumor cells, and EMT is a critical process in the metastasis cascade. Eliminating intracellular bacteria is a prospective therapeutic target for metastasis. However, existing targeted antibacterial approaches against intracellular bacteria, such as antibiotics, face challenges including drug resistance, biological barriers, or side effects. Here, we develop a tumor-targeted nanoplatform (HSO-OMDs) loaded with autophagy-induced metformin, docosahexaenoic acid, and chemotherapeutic drug oxaliplatin

The crosslinking of IgE-bound FcεRI by multivalent allergens initiates mast cell and basophil signaling underlying Type 1 allergy. Yet, how allergen properties and IgE occupancy impact receptor aggregation and downstream signaling remains unclear. We used Phl p 1-specific IgE and recombinant fusion proteins presenting a Phl p 1-derived peptide in defined valencies and positions (MB1N, MB1N1C, MB2N, and MB4N) to probe antigen-dependent signaling. Tetravalent MB4N evoked stronger degranulation and Ca2+ response than bivalent antigens MB1N1C and MB2N. MB4N was also capable of signaling at low IgE

In 2025, the 50th anniversary of the invention of hybridoma technology, the United States Food and Drug Administration (US FDA) approved a total of 44 new molecular entities (NMEs), of which 10 were therapeutic antibody-based molecules. Additionally, the US FDA approved 31 small molecules (of which one was a peptide), and 3 new nonantibody therapeutic proteins, but no new chimeric antigen receptor (CAR)-T-cell therapeutics. Of the new antibody-based biologics, two were antibody-drug conjugates, one was a T-cell engager-bispecific antibody, two were Fc-modified, half-life extended antibodies, a

Three unprecedented nitrogenated azaphilone hybrids, talamangins A-C (1-3), were isolated from the endophytic fungus Talaromyces mangshanicus SB17. Their structures, along with their absolute configurations, were determined by extensive spectroscopic data analysis and single-crystal X-ray diffraction. Compounds 1-3 represent the first examples of nitrogenated azaphilone hybrids, distinguished by a novel fused scaffold formed through the angular fusion of nitrogenated azaphilone and a benzophenone unit, with the azaphilone fragment featuring a pyridine ring. Preeclampsia (PE) is a severe pregnancy-specific disorder and a leading cause of maternal and perinatal morbidity and mortality. Its complex pathogenesis and the lack of effective therapies beyond delivery represent a major unmet clinical need. The syncytiotrophoblast (STB), the multinucleated cell layer at the outermost layer of the placental villous structure formed by the fusion of villous cytotrophoblasts (VCTs), serve as a critical functional barrier and regulatory hub at the maternal-fetal interface. Recent research highlights the central role of STB in the development of PE. This review

Desmoplastic small round cell tumor (DSRCT), an aggressive malignancy classically seen in adolescents and young adults with a strong male predominance, is exceptionally rare in the ovary. We report a 33-year-old woman with ovarian DSRCT presenting as a pelvic mass and peritoneal metastases. Histopathology revealed small round cell nests within desmoplastic stroma, exhibiting atypical architectural patterns (single-file, follicular-like cystic, microcystic, and solid). Immunohistochemistry demonstrated coexpression of epithelial (pan-keratin) and mesenchymal markers (vimentin, desmin), while mo

Prolonged infections in immunocompromized individuals are associated with accumulation of unexpected combinations of mutations, as exemplified by the Omicron variant. Here, we show that intra-host evolution during a chronic SARS-CoV-2 infection in the pre-Omicron era was associated with mutations that modulate antibody evasion, cell entry efficiency, spike processing, and fusogenicity. Single spike N-terminal mutations S13I and W64G exerted deleterious impacts on S1/S2 cleavage, syncytia formation, and cell entry but combined through an inferred recombination event with P330S to restore functi

The placenta is vital for fetal development, and altered placental vascularization, the most common placental pathology, underlies prevalent disorders, including fetal growth restriction, prematurity, and pregnancy complications. Impaired placental vascularization is associated with Vitamin A deficiency, but the mechanisms are undefined. To investigate this, we used retinoic acid (RA)-deficient Raldh2-/- embryos, and found they exhibit allantoic and placental endothelial hyperproliferation and impaired arterial-venous remodeling, which were rescued by providing all-trans-RA (ATRA) via maternal

Porcine contagious pleuropneumonia (PCP) is caused by Actinobacillus pleuropneumoniae (APP) and inflicts heavy economic losses on the swine industry. However, existing inactivated vaccines provide limited cross-protection, highlighting the need for improved vaccine strategies. In this study, we combined pangenome analysis with subtractive proteomics to screen the APP core genome and identified 11 potential antigens. Seven of them showed immunoreactivity by ELISA and Western blotting. These antigens, together with the ApxI-III toxins, were used for T and B cell epitope prediction. On this basis

Parkinson's disease (PD) is driven by mitochondrial impairment and oxidative stress-induced dopaminergic cell death. Phosphoglycerate mutase 5 (PGAM5) augmented accelerating mitophagy and attenuating neuronal apoptosis. Here, we present a novel cell-penetrating PEP-1-PGAM5 fusion protein designed to suppress oxidative damage and apoptosis. We assessed its transduction efficiency and neuroprotective efficacy in vitro and in vivo. PEP-1-PGAM5 was efficiently transduced into SH-SY5Y cells without cytotoxicity. In MPP+-exposed cells, PEP-1-PGAM5 attenuated ROS generation, prevented DNA fragmentati

The SARS-CoV-2 spike protein's membrane-binding domain bridges the viral and host cell membrane, a critical step in triggering membrane fusion. Here, we investigate how the SARS-CoV-2 spike protein interacts with host cell membranes, focusing on a membrane-binding peptide (MBP) located near the TMPRSS2 cleavage site. Through in vitro and computational studies, we examine both primed (TMPRSS2-cleaved) and unprimed versions of the MBP, as well as the influence of its conserved disulfide bridge on membrane binding. Our results show that the MBP preferentially associates with cholesterol-rich memb

The pleiomorphic structure and dynamic behavior of cellular endomembrane systems have been extensively studied using classical electron microscopy. However, fixation and staining constraints limit the in situ visualization of transient interactions, such as membrane fusion, scission, and intraluminal vesicle formation, potentially overlooking intermediate structures like membrane hemifusion. Using in situ cryo-electron tomography in four mammalian cell lines, we identify heterotypic hemifused vesicles featuring an extended hemifusion diaphragm consistently associated with a 42-nanometer proteo

Colibactin, a metabolite produced by gut bacteria carrying the polyketide synthase (pks) island, is associated with host genotoxicity and tumorigenesis. However, no Food and Drug Administration-approved therapeutics directly target colibactin. Here we show that expression of the intracellular colibactin self-resistance protein (ClbS) on the surface of engineered bacteria shields the host from genotoxic effects across multiple pks+ isolates. The surface display, due to the fusion of ClbS with outer membrane protein A (ClbS-OmpA) in Escherichia coli, effectively reduced colibactin-induced DNA da

Chimeric antigen receptor-T cell (CAR-T) immunotherapy has shown remarkable results for the treatment of certain hematologic malignancies. A redirection strategy that utilizes clinically relevant CAR-T cells in combination with adapter proteins may be an effective strategy to target other hematologic and solid cancers. We established a fusion antibody-based strategy with flexibility to target multiple tumor types in combination with a novel anti-leukocyte immunoglobulin-like receptor-B 4 (LILRB4) CAR-T cell. Specifically, we engineered switch protein (SwP) adapters containing the LILRB4 extrac

In Duchenne muscular dystrophy (DMD), muscle stem cells' (MuSCs) regenerative capacities are overwhelmed leading to fibrosis. Whether MuSCs have intrinsic defects or are disrupted by their environment is unclear. We investigated cell behavior and gene expression of MuSCs from DMD or healthy human muscles. Proliferation, differentiation, and fusion were unaltered in DMD-MuSCs, but with time, they lost their myogenic identity twice as fast as healthy MuSCs. The rapid drift toward a fibroblast-like cell identity was observed at the clonal level, and resulted from altered expression of epigenetic

The introduction and development of biologics such as therapeutic proteins, gene-, and cell-based therapy have revolutionized the scope of treatment for many diseases. However, a significant portion of the patients develop unwanted immune reactions against these novel biological modalities, referred to as immunogenicity, and no longer benefit from the treatments. In the current review, using Hemophilia A (HA) therapy as an example, we will discuss the immunogenicity issue of multiple biological modalities. Currently, the number of therapeutic modalities that are approved or recently explored t
