Hydrogels as advanced therapeutic delivery systems for improved cancer immunotherapy: current progress, challenges, and future prospects.

Immunotherapy has emerged as a new clinical paradigm for cancer management, leveraging immunomodulation to enhance systemic antitumor responses. Despite its significant potency and distinct advantages, challenges such as limited patient response rates and immune-related toxicities persist. Although nanomaterial-based delivery systems have been explored to improve local drug retention within tumor tissues, the efficacy of systemic nanomaterial strategies is constrained by low bioavailability and poor tumor penetration. As innovative localized delivery systems, injectable hydrogels hold signific

Current Insights into the Potential Misuse of Platelet-based Applications for Doping in Sports.

Platelet-based applications are currently used for the delivery of growth factors and other biomolecules as autologous biomaterials in regenerative medicine and cosmetic therapies. Many studies have revealed that platelet-based applications such as platelet-rich plasma and platelet releasate exhibit beneficial biological effects after a sports injury or trauma when administered locally by intramuscular injections. At present, treatment of the public, patients and athletes with platelet-based applications is permitted and regulated by the Food and Drug Administration and the World Anti-Doping A

Proton beam therapy induces protective immunity via HMGB1-dependent signaling.

Proton beam therapy is widely regarded for its cost-effectiveness, precision, and protection of normal tissues. Emerging evidence shows that conventional radiotherapy can inhibit primary tumors and promote immunogenicity of distant tumors, possibly via damage-associated molecular patterns (DAMPs) like calreticulin (CRT) and high mobility group box 1 (HMGB1) released during immunogenic cell death (ICD). However, the role of proton beams in inducing DAMPs and enhancing immunogenicity remains unclear. This study aimed to investigate the effects of proton beam-induced DAMPs on the colonization of

Circulating plasma-derived extracellular vesicles expressing bone and kidney markers are associated with neurocognitive impairment in people living with HIV.

BACKGROUND: Although effective antiretroviral therapy (ART) has improved the life expectancy of people with HIV (PWH), the prevalence of milder forms of HIV-associated neurocognitive disorders (HAND) persist, and it is associated with systemic and neuro-inflammatory processes that could impact other organ systems. However, the complex signaling mechanisms between the bone-kidney systems and the brain in HAND remain unknown. Extracellular vesicles (EVs) play a potential role in inter-organ communication and are involved in regulating cell activity in distant tissues. In this study, we examined

Analysis and validation of abnormal signaling pathways and immune cell infiltration characteristics in digestive system cancers based on peroxisome-related genes.

BACKGROUND: Although emerging evidence suggests a role for peroxisomes in tumorigenesis, their functions in digestive cancers remain unclear. This study aims to investigate the association between peroxisomes and digestive tract tumors. METHODS: To systematically investigate peroxisomal functions in digestive cancers, we first constructed and validated tumor-specific prognostic signatures based on peroxisome-related genes (PRGs) through univariate Cox, least absolute shrinkage and selection operator (LASSO), and multivariate Cox regression analyses. We then characterized the tumor immune micr

Uterine wound healing after caesarean section: A systematic review.

The rate of caesarean section (CS) is increasing worldwide. Defects in uterine healing have a major gynaecological and obstetric impact (uterine rupture, caesarean scar defect, caesarean scar pregnancy, placenta accreta spectrum). The complex process of cellular uterine healing after surgery, and specifically after CS, remains poorly understood in contrast to skin wound healing. This literature review on uterine wound healing was mainly based on histological observations, particularly after CS. The primary objective of the review was to examine the effects of CS on uterine tissue at the cellul

A mitochondrion-targeted natural polyphenolic copper carrier overcomes tumor resistance to cisplatin by potentiating cuproptosis.

BACKGROUND: Platinum–based drug resistance remains a major obstacle in cancer therapy. Cuproptosis, a novel form of copper–dependent cell death regulated through mitochondrial pathways, represents a promising strategy to counteract drug resistance in tumors. However, its efficacy is constrained by several physiological barriers, including elevated intracellular glutathione (GSH) levels, inadequate copper accumulation both cytoplasmically and within mitochondria, and the overexpression of copper efflux transporters such as ATP7A/B. METHODS: A series of material characterization techniques (FT-I

Enhancing skin-implant integration in lower-limb transcutaneous prostheses: From interface biology to bioactive, antimicrobial and cell-based strategies.

UNLABELLED: Transcutaneous prosthetic systems, which directly connect an external limb prosthesis to the skeleton, offer substantial biomechanical and clinical advantages over conventional socket-based devices. By eliminating the soft tissue loading inherent to the socket interfaces, these systems enable improved load transfer, comfort, and more natural sensory feedback through direct skeletal attachment. However, their widespread clinical adoption remains limited by complications at the skin-implant interface, including infection, marsupialization, and epithelial down-growth.Advances in surfa

3D connective micro-fragment enriched with stromal vascular fraction in osteoarthritis: chondroprotective evidence in a preclinical in vivo model.

INTRODUCTION: Adipose-derived cell therapies are one of the most common regenerative therapeutic options to alleviate the multi-component damage of osteoarthritis (OA). Adipose stromal vascular fraction (SVF) has gained scientific consensus for its ability to interact protectively with the joint microenvironment. Recently, the wide range of enzyme-free tissue processing systems has outperformed classical treatments, because of their ability to produce connective micrografts enriched with the SVF (mctSVF). This preclinical in vivo study evaluates the chondroprotective potential of a newly gener

Iron-mediated oxidative stress induces PD-L1 expression via activation of c-Myc in lung adenocarcinoma.

Introduction: The PD-1/PD-L1 axis is hijacked by lung adenocarcinoma (LUAD) cells to escape immune surveillance. PD-L1 expression in LUAD is affected, among others, by the metabolic trafficking between tumor cells and the tumor microenvironment (TME). Methods: Correlation between PD-L1 expression and iron content within the TME was established on FFPE LUAD tissue samples. The effects of an iron rich microenvironment on PD-L1 mRNA and protein levels were assessed in vitro in H460 and A549 LUAD by using qPCR, western blot and flow citometry. c-Myc knockdown was performed to validate the role of

Potential of CAR-Macrophages Derived from Induced Pluripotent Stem Cells (iMacs) for Solid Tumor Therapy.

Macrophages, a type of immune cell, perform essential functions in the immune system, including controlling inflammation, facilitating tissue healing, and defending the body against disease. However, in solid tumors, they are often reprogrammed into tumor-associated macrophages (TAMs) that support tumors through promotion of blood vessel growth, suppressing immune responses, and promoting tumor spread. Macrophages derived from blood-based sources, including monocytes and peripheral blood mononuclear cells (PBMCs), suffer from three major limitations: inconsistency across different donors, limi

Infectious bursal disease virus (IBDV) as a novel oncolytic virotherapy in glioblastoma.

BACKGROUND: Glioblastoma (GBM) is the most aggressive form of cancer of the central nervous system. Despite advances in immunotherapies and standard-of-care treatments for GBMs, clinical outcomes remain limited-owing to the immunosuppressive tumor microenvironment and the intrinsic resistance of GBM to conventional approaches. As a result, there is growing interest in rational combination strategies, particularly those pairing oncolytic viruses with immune-based therapies or established treatment modalities. Oncolytic viruses, by displaying conditionally enabled tumor cell-restricted replicati

Functional restoration of ex vivo model of pylorus: Co-injection of neural progenitor cells and interstitial cells of Cajal.

Transplantation of neural stem cells is a promising approach in treatment of intestinal dysfunctionality. The interstitial cells of Cajal (ICCs) are also critical in conditions such as pyloric dysfunctionality and gastroparesis. The objective of this study was to replenish neurons and ICCs in a dysfunctional pylorus as cell-based therapy to restore functionality. ICCs and enteric neural progenitor cells (NPCs) were isolated from rat duodenum and transduced with fluorescent proteins. Rat pylorus was harvested, and an ex-vivo neuromuscular dysfunctional model was developed by selective ablation

Reprogramming glioblastoma multiforme cells into neurons by protein kinase inhibitors.

Источник пометил публикацию как отозванную

BACKGROUND: Reprogramming of cancers into normal-like tissues is an innovative strategy for cancer treatment. Recent reports demonstrate that defined factors can reprogram cancer cells into pluripotent stem cells. Glioblastoma multiforme (GBM) is the most common and aggressive malignant brain tumor in humans. Despite multimodal therapy, the outcome for patients with GBM is still poor. Therefore, developing novel therapeutic strategy is a critical requirement. METHODS: We have developed a novel reprogramming method that uses a conceptually unique strategy for GBM treatment. We screened a kinas

[Clinical applications of the study of immunoglobulin and T-cell receptor gene rearrangements in acute leukemia and non-Hodgkin's lymphoma in children].

In addition to conventional morphological, histological and immunological marker studies, cells from 150 children with leukemia or non Hodgkin's lymphoma were analysed using the Southern blot hybridization technique to examine immunoglobulin- (Ig) and T-cell receptor (TCR) gene rearrangements. Patients with B-lineage leukemia or NHL demonstrated in 90% an Ig heavy chain gene rearrangement, 6% with an additional light chain kappa gene rearrangement. Combined Ig- with TCR-beta-gene rearrangements were mainly found in patients with common ALL: 19% at first presentation, and 33% in relapse. Moreov

[Molecular Mechanisms of Antitumor Effect in Targeted Alpha-radionuclide Therapy].

Targeted α-radionuclide therapy (TAT) is a systemic therapy for cancer with cancer-targeting compounds conjugated with α-emitters. Since the linear energy transfer (LET) of α-particles are high, relative biological effectiveness is higher than low LET radiation, such as X-rays and β-particles, and the damage to the normal tissues surrounding the tumor is minimal because path length of α-particle is short (a few cells). TAT has demonstrated remarkable therapeutic effects in patients and has attracted considerable attention from physicians and researchers worldwide. In particular, a study publis

Next-generation Mesoporous Silica Nanoparticles: Precision-engineered Platforms for Ovarian Cancer Therapy.

Silica nanoparticles (SiNPs) with antifouling properties offer a promising approach for enhancing ovarian cancer (OC) therapy. OC remains one of the deadliest gynecological malignancies due to late-stage diagnosis, high recurrence rates, and limited treatment efficacy. Conventional therapies, such as chemotherapy, often face challenges due to drug resistance and limited targeting ability. The development of SiNPs with antifouling capabilities aims to address these issues by improving drug delivery efficiency, reducing non-specific interactions, and enhancing biocompatibility. Antioxidant Therapies as Emerging Adjuncts in Rheumatoid Arthritis: Targeting Oxidative Stress to Enhance Treatment Outcomes.

Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by persistent inflammation and progressive joint destruction. Recent data underscore oxidative stress as a primary factor in the pathophysiology of rheumatoid arthritis, intensifying inflammatory processes and tissue damage via the overproduction of reactive oxygen species (ROS) and compromised antioxidant defenses. Current therapies, including disease-modifying antirheumatic drugs (DMARDs), primarily target immune dysregulation but fail to address oxidative stress, necessitating novel adjunctive treatment strategies. Ocular Delivery of Therapeutic Proteins: A Review.

Therapeutic proteins, including monoclonal antibodies, single chain variable fragment (ScFv), crystallizable fragment (Fc), and fragment antigen binding (Fab), have accounted for one-third of all drugs on the world market. In particular, these medicines have been widely used in ocular therapies in the treatment of various diseases, such as age-related macular degeneration, corneal neovascularization, diabetic retinopathy, and retinal vein occlusion. However, the formulation of these biomacromolecules is challenging due to their high molecular weight, complex structure, instability, short half-
