Sequential binary gene ratio tests define a novel molecular diagnostic strategy for malignant pleural mesothelioma.

PURPOSE: To develop a standardized approach for molecular diagnostics, we used the gene expression ratio bioinformatic technique to design a molecular signature to diagnose malignant pleural mesothelioma (MPM) from among other potentially confounding diagnoses and differentiate the epithelioid from the sarcomatoid histologic subtype of MPM. In addition, we searched for pathways relevant in MPM in comparison with other related cancers to identify unique molecular features in MPM. EXPERIMENTAL DESIGN: We conducted microarray analysis on 113 specimens including MPMs and a spectrum of tumors and

Isolation and molecular characterization of Newcastle disease virus genotypes II and VIId in Egypt between 2011 and 2012.

The current study was conducted to isolate and characterize Newcastle disease virus (NDV) from recent outbreaks affecting poultry farms in Egypt between 2011 and 2012. Trachea, spleen, liver, proventriculus and caecal tonsils were collected from clinically infected NDV ten different vaccinated broiler farms in Fayoum, Behira and Giza Provinces. Inoculation of all the collected samples in 10-day-old embryonated chicken specific-pathogen-free eggs resulted in isolation of haemagglutinating agents in three samples. These haemagglutinating agents were confirmed as NDV by real-time reverse transcri

Molecular characterization of a rabies virus isolate from a rabid dog in Hanzhong District, Shaanxi Province, China.

A canine rabies virus, Shaanxi-HZ-6, was isolated in Shaanxi Province, China, in 2009. Its genome has been completely sequenced and found to be closely related to the China I rabies virus strains widely circulating in China. The genomic length was 11,923 base pairs, and the overall organization of the genome was similar to that of other rabies virus isolates. Compared with isolates CQ92 and J, 84 amino acid substitutions (7 in the N gene, 15 in P, 6 in M, 25 in G, 31 in L) were observed in strain Shaanxi-HZ-6. Amino acid substitutions of R264H and V332I were noted in the G protein antigenic si

Differential protein expression profiles between Plasmodium falciparum parasites isolated from subjects presenting with pregnancy-associated malaria and uncomplicated malaria in Benin.

BACKGROUND: Plasmodium falciparum is responsible for severe malaria, including pregnancy-associated malaria (PAM). During intra-erythrocytic maturation, the infected erythrocyte (iE) membrane is modified by insertion of parasite-derived proteins, primarily consisting of variant surface antigens such as P. falciparum erythrocyte membrane protein-1. METHODS: To identify new PAM-specific parasite membrane proteins, we conducted a mass spectrometry-based proteomic study and compared the protein expression profiles of 10 PAM and 10 uncomplicated malaria (UM) samples. RESULTS: We focused on the 45

Full-length genome characterization and genetic relatedness analysis of hepatitis A virus outbreak strains associated with acute liver failure among children.

Clinical infection by hepatitis A virus (HAV) is generally self-limited but in some cases can progress to liver failure. Here, an HAV outbreak investigation among children with acute liver failure in a highly endemic country is presented. In addition, a sensitive method for HAV whole genome amplification and sequencing suitable for analysis of clinical samples is described. In this setting, two fatal cases attributed to acute liver failure and two asymptomatic cases living in the same household were identified. In a second household, one HAV case was observed with jaundice which resolved spont

Distribution and hybridization of Culex pipiens forms in Greece during the West Nile virus outbreak of 2010.

In 2010, an outbreak of West Nile virus (WNV) infections occurred in the region of Thessaloniki, Central Macedonia, in northern Greece. During this period, Culex pipiens sensu stricto mosquitoes were found infected by WNV lineage 2. Cx. pipiens s.s. presents two distinct biological forms, denoted molestus and pipiens. Hybrids between the two forms may potentiate the accidental transmission of WNV to humans. We have genetically characterized the form composition of Cx. pipiens s.s. samples collected during the outbreak from the region of Thessaloniki, where WNV cases occurred, and from the regi

Familial occurrence of cervical artery dissection--coincidence or sign of familial predisposition?

UNLABELLED: BACKGROUNDAND PURPOSE: The etiology of spontaneous cervical artery dissection (CeAD) is poorly understood in most patients. Mild cervical trauma preceding the dissection event is a common finding, but many CeAD occur spontaneously. It is likely that genetic factors may increase the risk for CeAD. However, familial cases are excedingly rare. Familial clustering of CeAD may be accidental or associated with genetic or environmental risk factors shared between affected relatives. In this explorative study, we aim to show that specific risk factors for familial CeAD exist. METHODS: Age

Triage of frail elderly with reduced exercise tolerance in primary care (TREE). A clustered randomized diagnostic study.

BACKGROUND: Exercise reduced tolerance and breathlessness are common in the elderly and can result in substantial loss in functionality and health related quality of life. Heart failure (HF) and chronic obstructive pulmonary disease (COPD) are common underlying causes, but can be difficult to disentangle due to overlap in symptomatology. In addition, other potential causes such as obesity, anaemia, renal dysfunction and thyroid disorders may be involved. We aim to assess whether screening of frail elderly with reduced exercise tolerance leads to high detection rates of HF, COPD, or alternative

First time molecular detection and phylogenetic relationships of torque teno sus virus 1 and 2 in domestic pigs in Uganda: further evidence for a global distribution.

BACKGROUND: Torque teno sus virus 1 (TTSuV1) and 2 (TTSuV2) are small, single-stranded circular DNA viruses belonging to the Anelloviridae family. Available studies clearly show that both viruses are widely distributed in the pig populations in America, Europe and Asia, although the impact of the infection is still unclear. Currently, the situation in domestic pig populations on the African continent is not known. Therefore, the aim of this study was to investigate the possible presence of the two viruses in domestic pigs in Uganda, and describe the phylogenetic relationships to those in the r

A panel of serum microRNAs as specific biomarkers for diagnosis of compound- and herb-induced liver injury in rats.

BACKGROUND: Drug-induced liver injury (DILI) has been a public, economic and pharmaceutical issue for many years. Enormous effort has been made for discovering and developing novel biomarkers for diagnosing and monitoring both clinical and preclinical DILI at an early stage, though progress has been relatively slow. Additionally, herb-induced liver injury is an emerging cause of liver disease because herbal medicines are increasingly being used worldwide. Recently, circulating microRNAs (miRNAs) have shown potential to serve as novel, minimally invasive biomarkers to diagnose and monitor human

Genetic analysis of the P1 region of human enterovirus 71 strains and expression of the 55 F strainVP1 protein.

Enterovirus 71 (EV71) is a member of the Entero-virus genus of the Picornaviridae family and is the major cause of Hand, foot, and mouth disease (HFMD) in children. Different strains from Gansu were cloned and the P1 protein was sequenced and analysed. Results indicate that there are three kinds of EV71 infections prevalent in Gansu. The VP1 protein from one of these strains, 55F, was expressed. The recombinant protein was expressed with high level and reacted specifically with the EV71 patient antibody, the recombinant protein was also applied to raise antiserum in rabbits and after the fourt

Evidence that Cryptosporidium parvum populations are panmictic and unstructured in the Upper Midwest of the United States.

Cryptosporidium parvum is a zoonotic protozoan parasite that causes cryptosporidiosis, an infectious diarrheal disease primarily affecting humans and neonatal ruminants. Understanding the transmission dynamics of C. parvum, particularly the specific contributions of zoonotic and anthroponotic transmission, is critical to the control of this pathogen. This study used a population genetics approach to better understand the transmission of C. parvum in the Upper Midwest United States. A total of 254 C. parvum isolates from cases of human cryptosporidiosis in Minnesota and Wisconsin and diarrheic

Evaluation of type-specific HPV persistence and high-risk HPV viral load quantitation in HPV positive women under 30 with normal cervical cytology.

The persistence of high-risk HPV (HR-HPV) infection is necessary for the development of cervical intraepithelial neoplasia. The aim of this study was to evaluate if HR-HPV typing and HPV16, 18, 31, and 33 quantitation are predictive for type-specific infection persistence and/or the development of CIN in women under 30 with normal cervical cytology. Young women (under 30) attending a family planning clinic who were HPV positive with normal cervical cytology were included. HPV genotyping was assessed by MY09/MY11 PCR, sequencing, phylogenetic analysis, and cloning when necessary. HR-HPV viral l

Identification and characterization of acute infection with parvovirus B19 genotype 2 in immunocompromised patients in Poland.

Parvovirus B19 (B19V) is divided into three genotypes. Genotypes 2 and 3 may cause diagnostic difficulties and their epidemiology is not well understood. In the present study the prevalence of B19V genotypes in patients with symptomatic infection in Poland was evaluated and the course of infection in patients infected with non-genotype 1 strains is described. Real-time PCR, able to detect all three genotypes of B19V was used to screen patient plasma samples. Sixty-nine, mainly acute-phase B19V DNA positive cases were identified in patients from hematological and obstetric/gynecological wards b

Unsupervised segmentation of overlapped nuclei using Bayesian classification.

In a fully automatic cell extraction process, one of the main issues to overcome is the problem related to extracting overlapped nuclei since such nuclei will often affect the quantitative analysis of cell images. In this paper, we present an unsupervised Bayesian classification scheme for separating overlapped nuclei. The proposed approach first involves applying the distance transform to overlapped nuclei. The topographic surface generated by distance transform is viewed as a mixture of Gaussians in the proposed algorithm. In order to learn the distribution of the topographic surface, the pa

Molecular identification of the parasites causing cutaneous leishmaniasis on the Caribbean coast of Colombia.

All clinical manifestations of leishmaniasis exist in Colombia, the cutaneous form being the most frequent in the department of Sucre, where the Leishmania species associated with cutaneous leishmaniasis (CL) is unknown. This study was carried out to determine which Leishmania species was responsible for CL in Sucre, based on amplification and sequencing of the Cyt b gene. Isolates of Leishmania were obtained after CL diagnosis of eight patients who received attention in several health care centers of the study area. The nucleotide sequences obtained from patients were compared to Leishmania r

Microsatellite typing of clinical and environmental Cryptococcus neoformans var. grubii isolates from Cuba shows multiple genetic lineages.

BACKGROUND: Human cryptococcal infections have been associated with bird droppings as a likely source of infection. Studies toward the local and global epidemiology of Cryptococcus spp. have been hampered by the lack of rapid, discriminatory, and exchangeable molecular typing methods. METHODOLOGY/PRINCIPAL FINDINGS: We selected nine microsatellite markers for high-resolution fingerprinting from the genome of C. neoformans var. grubii. This panel of markers was applied to a collection of clinical (n = 122) and environmental (n = 68; from pigeon guano) C. neoformans var. grubii isolates from Cu

A high-risk signature for patients with multiple myeloma established from the molecular classification of human myeloma cell lines.

BACKGROUND: Multiple myeloma is a plasma-cell tumor with heterogeneity in molecular abnormalities and treatment response. DESIGN AND METHODS: We have assessed whether human myeloma cell lines have kept patients' heterogeneity using Affymetrix gene expression profiling of 40 human myeloma cell lines obtained with or without IL6 addition and could provide a signature for stratification of patient risk. RESULTS: Human myeloma cell lines, especially those derived in the presence of IL6, displayed a heterogeneity that overlaps that of the patients with multiple myeloma. Human myeloma cell lines s

A novel hepatitis B virus (HBV) subgenotype D (D8) strain, resulting from recombination between genotypes D and E, is circulating in Niger along with HBV/E strains.

Niger is a west African country that is highly endemic for hepatitis B virus (HBV) infection. The seroprevalence for HBV surface antigen (HBsAg) is about 20%; however, there are no reports on the molecular epidemiology of HBV strains spreading in Niger. In the present study, HBV isolates from the sera of 58 consecutive, asymptomatic, HBsAg-positive blood donors were characterized. Genotype affiliation was determined by amplification, sequencing and phylogenetic analysis of the preS1, polymerase/reverse transcriptase (RT/Pol) and precore (preC)/C regions. The first series of results revealed th
